RESUMEN
Actualmente no son del todo conocidos los mecanismos que intervienen en la "decisión" del sistema inmune para promover una respuesta fundamentalmente efectora y cuando hacerlo con una respuesta fundamentalmente tolerante. A lo largo de la historia el pensamiento ha ido variando a medida que las tecnologías disponibles han permitido entender el funcionamiento del sistema inmune. Por tales razones, se realizó la presente revisión bibliográfica con el propósito de resumir los cambios en el pensamiento inmunológico en torno al tema fundamental de la inmunología. Se abordaron los conceptos de horror autotóxico enunciado por Erlich, el significado de la teoría de la selección clonal en el entendimiento de cómo operaba la tolerancia central. También se abordó la teoría de las dos señales y las contribuciones de Jerner y su teoría de la red inmune. Finalmente se analizó el papel de la teoría del peligro y se concluyó con la teoría de la tolerancia dominante; se enuncian los aportes de cada uno de estas en el entendimiento de cómo puede operar el sistema inmune.
ABSTRACT
At present, the mechanisms involved in the "decision" of the immune system to promote an essentially effector response or a tolerant response are not fully known. Throughout history, immunological thinking has changed as the available technologies have led to a better understanding of the immune system. For these reasons, the present literature review was conducted to summarize the changes in immunological thinking regarding the fundamental problem of immunology. The concept of horror autotoxicus proposed by Erlich and the meaning of the clonal selection theory for understanding central tolerance were discussed. The two-signal model, Jerne’s contributions and his immune network theory were also addressed. Finally, the danger model and the theory of dominant tolerance were analyzed. The contributions of each theory to understanding how the immune system works were included.
