Deoxynivalenol (DON) in wheat: occurrence in samples commercialized in the São Paulo city / Desoxinivalenol (DON) em trigo e farinha de trigo comercializados na cidade de São Paulo

ABSTRACT
Twenty-eight samples of wheat flour and 14 samples of wheat were purchased in the city of São Paulo and these products were analyzed for deoxynivalenol (DON). Samples were extracted with acetonitrile-water (84+16) followed by a cleanup with MycoSep column. The extract purification was carried out on TLC and the toxin was determined with AlCl 3  solution. The efficiency of MycoSep columns 225 and 227 was evaluated with samples of wheat and wheat flour spiked with DON at two levels, 80.0 and 100.0 »g/kg. The recovery averages on columns 225 and 227 were, respectively, 72% and 107% for wheat flour and 90% and 125% for wheat grain, respectively. The relative standard deviations were 11% and 18% for wheat flour and 8% and 20% for wheat grain, respectively. DON was detected in 19 (45%) of 42 analysed samples at levels ranged from 82 to 1500 »g/kg. Eight samples were confirmed by means of HPLC-UV.

Vinte e oito amostras de farinha de trigo e 14 amostras de trigo em grão foram adquiridos na cidade de São Paulo e analisados para determinação de desoxinivalenol (DON). As amostras foram extraidas com acetonitrila-água (84+16) seguidas de limpeza dos extratos com colunas MycoSep. A separação e a quantificação foram pela técnica da Cromatografia em Camada Delgada (CCD) e a toxina visualizada com solução de AlCl 3 . A eficiência das colunas MycoSep 225 e 227 foi avaliada com amostras de farinha de trigo e trigo em grão, contaminadas com DON em dois níveis, 80,0 e 100,0 »g/kg. As recuperações médias das colunas 225 e 227 foram, respectivamente, 72% e 107% para farinha de trigo e 90% e 125% para trigo em grão. Os desvios padrão relativos foram 11% e 18% para farinha de trigo e 8% e 20% para trigo em grão, respectivamente. DON foi detectado em 19 (45%) das 42 amostras analisadas em níveis que variaram de 82-1500g/kg.Algumas amostras (8) foram confirmadas por CLAE-UV.
