ABSTRACT

OBJECTIVE:

To study the protective effect of ethyl acetate extract from Japanese Artactylodes Rhizome on myocardial ischemical reperfusion injury in rats and its possible mechanism.

METHODS:

50 Wistar rats were divided randomly into 5 groupssham group, model group, medicine (ethyl acetate extract from Japanese Artactylodes Rhizome at dosage of 200mg?kg-1 and 400mg?kg-1,respectively) group and verapamil(positive medicine) group. Rats in medicine group, model group and positive group were subjected to myocardial ischemia for 40min followed by reperfusion for 30 min, then muscular tissues were sampled for the determination of the activity of GSH-Px, the content of mitochondrial calcium (Mit-Ca2+),and the activities of Na+-K+-ATPase and Mg2+-ATPase in myocardium.

RESULTS:

As compared with model group, in the group treated with ethyl acetate extract from Japanese Artactylodes Rhizome 30min after reperfusion, the activity of GSH-Px was significantly higher, the content of myocardial mitochondrial calcium was significantly lower, and the activities of Na+-K+-ATPase, Mg2+-ATPase were significantly higher.

CONCLUSION:

The protective effect of ethyl acetate extract from Japanese Artactylodes Rhizome on myocardial ischemical reperfusion injury might be achieved through enhancing the activity of myocardial GSH-Px, lowering the content of myocardial mitochondrial calcium and enhancing the activities of Na+-K+-ATPase and Mg2+-ATPase of cellular membranes.
