Determination of Main Components and Related Impurities in Solanesd Extraction by HPLC-MS / 中国药房

ABSTRACT

OBJECTIVE:

To develop a method for the determination of solanesol and the related impurities in salanesol extraction by HPLC-MS.

METHODS:

A C18 column was used as analytical column with column temperature set at 25 ℃; the mobile phase was methanol-ethanol (50∶50) at a flow rate of 0.5 mL?min-1. The mass spectrometer equipped with APCI, and the ion source for chemical ionization was operated in negative mode. The detector voltage was set at 1.6 kV, heat block temperature at 200 ℃, CDL temperature at 250 ℃, and nitrogen was used as drying gas with a flow rate of 2.5 L?min-1. The scanning mass/charge ratio range was 150~800 m/z.

RESULTS:

The calibration curve for solanesol was linear over the concentration range of 0.006~114.0 ?g?mL-1(r=0.999 8), and the average recovery was 99.3%(RSD=1.6%,n=3).

CONCLUSIONS:

The HPLC-MS method is simple, rapid, sensitive, accurate, and suitable for the determination of the main components and related impurities in salanesol extraction.
