Research on Dynamic Adsorption Technology of Total Alkaloids from Cortex Phellodendri with AB-8 Macroporous Resin / 广州中医药大学学报

ABSTRACT
Objective To optimize the adsorption and desorption conditions for total alkaloids from Cortex Phellodendri with AB-8 macroporous resin.Methods With the adsorption and desorption volume of total alkaloids as the observation indexes,the experiment of dynamic adsorption with AB-8 macroporous resin was carried out to observe the effect of the sample volume,flow rate,water volume for removing impurity,and the concentration and volume of eluant on the total alkaloids from Cortex Phellodendri.Results The optimal adsorption conditions were as followssample volume being 50mL for each milliliter of the resin,flow rate being 20mL/h(equal to 2 column volume),water for impurity eluent being 30 mL(equal to 3 column volume),and the eluent being 60 mL(equal to 6 column volume) of 40% ethanol.Under the above condition,the purity of the total alkaloids was 40.06%,which is 2.56 times as much as the total alkaloids before loading the column.Conclusion This methed is simple and shows good purifying effect.
