ABSTRACT

Objective:To explore the application effects of case-based learning (CBL) based on Rain Classroom in surgical nursing teaching of large class.Methods:A total of 357 nursing undergraduates of Batch 2017 in Fenyang College of Shanxi Medical University were selected as the research objects and randomly divided into experimental group (176 cases) and control group (181 cases). The experimental group adopted the CBL based on Rain Classroom, while the control group adopted the traditional multimedia classroom CBL mode. After the teaching, the teaching effect was evaluated by interviewing, comparing the scores of theoretical examination and questionnaire survey. SPSS 19.0 was performed for t test. Results:The scores of memory type, comprehension type and comprehensive application type questions in the experimental group were significantly higher than those in the control group ( P<0.01). The scores of the experimental group were significantly higher than those of the control group in stimulating thirst for knowledge, learning initiative, classroom learning efficiency, memory of knowledge points, problem analysis, clinical thinking and team cooperation ability ( P<0.01). The interview information suggested that the interest of pre-study materials, the control of discussion links and the network effect were the problems affecting the classroom effect of the experimental group, while the control group had the problems of poor learning autonomy and enthusiasm. Conclusion:The CBL based on Rain Classroom can mobilize the initiative and participation of students in large class teaching, improve the learning effect and cultivate comprehensive ability, which is worthy of promotion.

ABSTRACT

Background and Objectives@#This study was to investigate the role of microRNA-29a-3p (miR-29a-3p) in human bone marrow mesenchymal stem cells (hBMSCs), and its relationship with steroid-associated osteonecrosis. @*Methods@#and Results: The online tool GEO2R was used to screen out the differentially expressed genes (DEGs) in GSE123568 dataset. Quantitative real time-polymerase chain reaction (qRT-PCR) was performed to detect the expression of miR-29a-3p, forkhead box O3 (FOXO3), alkaline phosphatase (ALP), bone gamma-carboxyglutamate protein (OCN) and RUNX family transcription factor 2 (Runx2) in the hBMSCs isolated from the patients with steroid-associated osteonecrosis. CCK-8 assay was executed to measure cell viability; western blot assay was utilized to detect FOXO3, ALP, Runx2, OCN and β-catenin expression. Cell apoptosis and cell cycle were detected by flow cytometry. Immunofluorescence assay was used to detect the sub-cellular localization of β-catenin. Bioinformatics analysis and luciferase reporter gene assay were performed to confirm whether miR-29a-3p can combine with FOXO3 3’UTR. MiR-29a-3p was markedly up-regulated in the hBMSCs of patients with steroid-associated osteonecrosis, while FOXO3 mRNA was significantly down-regulated. Transfection of miR-29a-3p mimics significantly inhibited the hBMSCs’ proliferation, osteogenic differentiation markers’ expressions, including ALP, Runx2, OCN, and repressed the ALP activity, as well as promoted cell apoptosis and cell-cycle arrest. FOXO3 was identified as a target gene of miR-29a-3p, and miR-29a-3p can inhibit the expression of FOXO3 and β-catenin, and inhibition of miR-29a-3p promoted translocation of β-catenin to the nucleus. @*Conclusions@#MiR-29a-3p can modulate FOXO3 expression and Wnt/β-catenin signaling to inhibit viability and osteogenic differentiation of hBMSCs, thereby promoting the development of steroid-associated osteonecrosis.

ABSTRACT

Objective To evaluate the value of GeneXpert MTB/RIF assay in the clinical diagnosis of tuberculosis and tuberculosis associated rifampin resistance. Methods A total of 851 patients admitted to the Guangxi Longtan Hospital from September 22, 2015 to May 25, 2016 were enrolled. The smear microscopic examination, solid culture test, and GeneXpert MTB/RIF assay was performed on each patient. Drug sensitivity test of rifampicin using proportion method was done on patients infected with mycobacterium tuberculosis isolated by solid culture, and rifampicin resistance detection was performed on GeneXpert MTB/RIF positive patients. Bacteriological positive consistency test was performed on GeneXpert MTB/RIF and solid culture, and rifampicin resistance detection was compared bwtween GeneXpert MTB/RIF and drug sensitivity using proportion method. Culture and drug susceptibility testing were used as the gold standard, and the MTB sensitivity and specificity of smear microscopic examination, solid culture, and GeneXpert MTB/RIF assay was compared. Results Culture was used as the gold standard. Compared with culture, the sensitivity, specificity, positive predictive value, and negative predictive value of GeneXpert MTB/RIF was 95.48%(211/241), 94.91%(559/569), 87.55%(211/241), and 98.24%(559/569) respectively. The sensitivity of Xpert MTB/RIF for culture positive smear positive specimens and culture positive smear negative specimens was 98.78% (162/164) and 85.96%(49/57) respectively. Drug susceptibility testing was used as the gold standard. Compared with drug susceptibility testing, the sensitivity, specificity, positive predictive value, and negative predictive value of rifampin resistance measured by GeneXpert MTB/RIF was 98.70%(152/154), 92.98%(53/57), 97.44%(152/156) and 96.36%(53/55) respectively. The consistency rate of GeneXpert MTB/RIF and solid culture was 93.83% (Kappa = 0.847), and that of GeneXpert MTB/RIF rifampin resistance detection and proportion method for drug sensitivity was 96.38% (Kappa = 0.904). Conclusions The GeneXpert MTB/RIF detection is easily operated with high sensitivity and specificity compared with smear microscopic examination and solid culture, and it also possesses good consistency with solid culture and traditional drug sensitivity. Moreover, it could also be applied to diagnose the resistance to rifampin, which is very promising in application.

ABSTRACT

[Objective] To investigate the construction and inhibitory effects of artificial microRNA targeted on hepatitis B virus.[Methods] Four artificial microRNA sequences for HBV gene were designed with Invitrogen online design software (BLOCK-iTTM RNAi Designer),amiRNA eukaryotic expression plasmid were constructed and transfected into human hepatocellular carcinoma cell line HepG2.2.15.Hepatitis B virus DNA in supernatant was detected by quantitative fluorescence PCR.HBsAg and HBeAg detected by ELISA.[Results] All of the four artificial microRNA sequences could down regulate the level of HBV-DNA,HBsAg and HBeAg.miR3 had the most significant effect on inhibiting HBV-DNA replication.[Conclusion] Artificial microRNA sequences can effectively inhibit the replication of HBV in vitro.

ABSTRACT

AIM:To investigate the effect of Sonic Hedgehog ( Shh) signaling blockade on the growth of hema-tocarcinoma cells and underlying mechanisms.METHODS: The expression of Shh signaling molecules in hematocarci-noma cell lines BEL-7402, Huh7 and HepG2 was detected by RT-PCR.The cell viability was detected by MTT assay.The cell cycle and apoptosis were analyzed by flow cytometry.The expression of apoptosis-related proteins was determined by Western blot.RESULTS:Shh signaling molecules were all expressed in BEL-7402, Huh7 and HepG2 cells.The mRNA expression of Patched ( Ptch) , Gli1 and Gli2 was down-regulated by anti-Shh antibody.Blockade of Shh signaling pathway inhibited the proliferation of hepatocarcinoma cells with increasing cells in G0/G1 phase and induced the apoptosis of hepa-tocarcinoma cells.Treatment with anti-Shh antibody down-regulated the protein expression of pro-caspase-3, pro-caspase-8 and pro-caspase-9, while up-regulated the protein levels of cleaved caspase-3, cleaved caspase-8 and cleaved caspase-9 in BEL-7402 cells.CONCLUSION:Blockade of Shh signaling pathway inhibits the growth of hepatocarcinoma at different levels by cell cycle arrest and inducing apoptosis of hematocarcinoma cells.

ABSTRACT

Objective To construct the evaluation index system of clinical nursing competence for higher vocational nursing skills contest. Methods A total of 31 nursing experts participated in the three-round consultation by Delphi technique, and empirically tested the final result. Results The response rates of questionnaire investigation for the three-round were 93.6%(29/31),100.0%(29/29) and 100.0%(29/29) respectively. The specialist authority coefficient was 0.861,and the coordination coefficients for each round were 0.207,0.291 and 0.371, respectively, P<0.01.The overall reliability coefficient was 0.921, evaluators reliability correlation coefficient was 0.942. There were significant correlation between each item and total score, P<0.05. The overall degree of differentiation was 0.523. Finally, the clinical nursing competence assessment system for higher vocational nursing skills contest was constructed,including 3 first-level indexes,10 second-level indexes and 39 third-level items. Conclusions The results of three-round expert consultation are reliable, the reliability, validity and differentiation are high. To evaluate clinical nursing ability higher vocational nursing students provide scientific basis.

ABSTRACT

Objective To explore the level of knowledge,attitude and practice in preventing sudden deafness and the influencing factors among nursing students in Taiyuan City.Methods By random cluster sampling,1 187 nursing students from 6 universities in Taiyuan City were surveyed about the level of knowledge,attitude and practice and the influencing factors by questionnaires in November 2013.The results underwent analysis.Results Total scoring rate of knowledge,attitude and practice of preventing sudden deafness in the nursing students was 48.36% (574/1 187),among which knowledge,attitude and practice was 28.30% (336/1 187),62.68% (744/1 187) and 54.34% (645/1 187),respectively.Logistic regression analysis showed that whether taking health education or not,different parents' education degree and household capita monthly income had significantly different influence on level of knowledge,attitude and practice of preventing sudden deafness,P<0.01.Significant correlation existed between knowledge and attitude,attitude and practice,r=0.467,0.149,P<0.05.Conclusions The level of knowledge,attitude and practice of preventing sudden deafness of the nursing students is low and has muhifaceted influencing factors.Colleges and universities should strengthen the health propaganda,and community should step up intervention to the families of low income and low education degree,at the same time to improve the compliance of health behavior in order to play a role in prevention and treatment of sudden deafness among nursing students.

ABSTRACT

Objective To evaluate clinical characteristics of acquired immunodeficiency syndrome (AIDS) patients infected with mycobacterium tuberculosis.Methods The clinical data of 440 AIDS patients infected with mycobacterium tuberculosis were collected and subjected to statistical analysis,including infectious type,sites,and situation.Results There were mainly multitopical or disseminated tuberculosis in 163 cases;the rate of extrapulmonary tuberculosis was 32.50%(143 cases);there were 48 cases of confidently extrapulmonay tuberculosis;there were 8 cases of adult primary pulmonary tuberculosis.Conclusion Acquired immunodeficiency syndrome patients infected with mycobacterium tuberculosis is the most opportunistic infection.There were multitopical or disseminated infection of mycobacterium tuberculosis in acquired immunodeficiency syndrome patients infected with mycobacterium tuberculosis.

ABSTRACT

This article evaluates the curative effect of intestinal sustenance of larygo-Carlinomactomee, the intestinal sustenance therapy has two stages. In the first stage (five days), patients are given nasal feeding liauid diet, takeing protein 44.7g, appropriate vitamin and ferralia every day, the total calorie is 1 631 large calories. In the second stage (eleven days), patients are given high caloric liguid diet, take in protein 65g, every day. The total caloric is 3 345 large calories. The result indicates: giving intestinal sustenance earlier has a good effect on laryngocarcinomactomee by improving the patients's immunity and accelerating the patients' early recover.
