ABSTRACT

One of the main challenges of tissue engineering in dentistry is to replace bone and dental tissues with strategies or techniques that simulate physiological tissue repair conditions. This systematic review of in vitro studies aimed to evaluate the influence of the addition of nanohydroxyapatite (NHap) to scaffolds on cell proliferation and osteogenic and odontogenic differentiation of human mesenchymal stem cells. In vitro studies on human stem cells that proliferated and differentiated into odontogenic and osteogenic cells in scaffolds containing NHap were included in this study. Searches in PubMed/MEDLINE, Scopus, Web of Science, OpenGrey, ProQuest, and Cochrane Library electronic databases were performed. The total of 333 articles was found across all databases. After reading and analyzing titles and abstracts, 8 articles were selected for full reading and extraction of qualitative data. Results showed that despite the large variability in scaffold composition, NHap-containing scaffolds promoted high rates of cell proliferation, increased alkaline phosphatase (ALP) activity during short culture periods, and induced differentiation, as evidenced by the high expression of genes involved in osteogenesis and odontogenesis. However, further studies with greater standardization regarding NHap concentration, type of scaffolds, and evaluation period are needed to observe possible interference of these criteria in the action of NHap on the proliferation and differentiation of human stem cells.

ABSTRACT

The role of cyclooxygenase (COXs) isoforms in maintaining colonic mucosal integrity is not fully understood. This study aimed to evaluate the role of COX-1 and -2 on colonic mucosal integrity in an experimental colitis model. Colitis was induced in Wistar rats by intracolonic administration of 2,4,6-trinitrobenzenesulfonic acid (20 mg + 50% ethanol). The control group (sham group) received saline only. After 7, 14, or 28 days, colonic samples were removed, and macroscopic lesion scores, wet weight, myeloperoxidase activity, and transepithelial electrical resistance (TER) were determined. In other rat groups, colonic samples from the sham group and a 7th day post-colitis group were mounted in Üssing chambers with the luminal side exposed to a buffer solution (control), acetylsalicylic acid (ASA), SC-560 (COX-1 inhibitor), or celecoxib (COX-2 inhibitor). TER and epithelial permeability to fluorescein were measured. The 7th day colitis group had higher macroscopic damage scores, wet weight, and myeloperoxidase activity and lower basal TER than the sham, 14th day colitis, and 28th day colitis groups. Inhibition of COX-1 but not COX-2 significantly decreased TER and increased permeability to fluorescein in the 7th day post-colitis group compared to the sham group. Additionally, ASA decreased the colonic mucosal integrity on day seven post-colitis compared to the sham group. A decrease in the colonic mucosa integrity in the experimental colitis model can be aggravated only by the inhibition of COX-1, which demonstrated the importance of this enzyme in the maintenance of colonic mucosal integrity.

ABSTRACT

Adipose tissue-derived mesenchymal stromal/stem cells (ASCs) are considered important tools in regenerative medicine and are being tested in several clinical studies. Porcine models are frequently used to obtain adipose tissue, due to the abundance of material and because they have immunological and physiological similarities with humans. However, it is essential to understand the effects and safe application of ASCs from pigs (pASCs) as an alternative therapy for diseases. Although minipigs are easy-to-handle animals that require less food and space, acquiring and maintaining them in a bioterium can be costly. Thus, we present a protocol for the isolation and proliferation of ASCs isolated from adipose tissue of farm pigs. Adipose tissue samples were extracted from the abdominal region of the animals. Because the pigs were not raised in a controlled environment, such as a bioterium, it was necessary to carry out rigorous procedures for disinfection. After this procedure, cells were isolated by mechanical dissociation and enzymatic digestion. A proliferation curve was performed and used to calculate the doubling time of the population. The characterization of pASCs was performed by immunophenotyping and cell differentiation in osteogenic and adipogenic lineages. The described method was efficient for the isolation and cultivation of pASCs, maintaining cellular attributes, such as surface antigens and multipotential differentiation during in vitro proliferation. This protocol presents the isolation and cultivation of ASCs from farm pig as an alternative for the isolation and cultivation of ASCs from minipigs, which require strictly controlled maintenance conditions and a more expensive process.

RESUMO: Introdução: A cabine de segurança biológica (CBS) é um equipamento de proteção coletiva, utilizado para efe-tuar a contenção de aerossóis produzidos nos procedimentos laboratoriais. A CBS protege tanto os trabalhadores, quanto o material manipulado e o meio ambiente. Dispõe de lâmpadas de luz ultravioleta (UV) que possui ação germicida, alterando os ácidos nucleicos dos micro-organismos. Objetivo: O objetivo do presente estudo foi avaliar a ação germicida da luz UV da CBS classe II, tipo A2, frente à cultura de duas espécies de bactérias com diferentes condições de exposição a luz UV. Material e Métodos: Para o desenvolvimento da pesquisa foram utilizadas as bactérias Staphylococcus aureus ATCC 25923 e Klebsiella pneumoniae ATCC 10031, na concentração de 1,5x108 Unidades Formadoras de Colônias/ml (UFC/ml), que após a semeadura em biplacas de Petri foram expostas à luz UV em diferentes condições e tempos. Resultados: Os resultados obtidos revelaram que as espécies de bactérias apresentaram igual perfil de crescimento ou inibição quando submetidas às diferentes condições de exposição. As biplacas de Petri com as tampas abertas e protegidas ou não com embalagem de esterilização apresentaram, nos tempos 15 e 20 minutos, inibição bacteriana. Nas biplacas protegidas pelo papel alumínio e nas biplacas com a tampa fechada, independentemente do tempo e da condição, os micro-organismos apresentaram crescimento bacteriano. Conclusão: Com os resultados obtidos, sugere-se que a ação germicida da luz UV foi eficaz, garantindo a descontaminação adequada e assegurando a qualidade na biossegurança laboratoria. (AU)


ABSTRACT: Introduction: The biological safety cabinet (BSC) is a collective protective device used to hold aerosols produced in laboratory procedures. The BSC protects workers, material handling, and the environment. It relies on ultraviolet light (UV) lamps that have germicidal action, altering the nucleic acids of microorganisms. Objective: The objective of the present study was to evaluate the germicidal action of BSC class II type A2 UV light against the culture of two bacterial species with different conditions of exposure to UV light. Methods: For the research, the bacteria Sta-phylococcus aureus ATCC 25923 and Klebsiella pneumoniae ATCC 10031 were used, at a concentration of 1.5x108Colony Forming Units/ml (CFU/ml), which were seeded in Petri biplates. Use UV light under different conditions and times. Results: The bacterial species showed the same growth or inhibition profile when subjected to different exposure conditions. Petri dishes with the cover open and protected or not with sterilization packaging showed bacterial inhibition at 15 and 20 minutes. The microorganisms in the biplates protected by the aluminum foil and in the biplates with the cover on, regardless of the time and condition, showed bacterial growth. Conclusion: With the results obtained, it is suggested that the germicidal action of UV light was effective, ensuring adequate decon-tamination and ensuring quality in laboratory biosafety. (AU)

ABSTRACT

Objetivou-se avaliar as alterações fisiológicas de ovinos submetidos à restrição alimentar, expostos à radiação solar e à sombra. Dez borregas mestiças foram divididas igualmente em dois grupos: grupo radiação solar - GRS, animais expostos ao sol durante o dia; e grupo sombra - GS, animais alocados dentro de um galpão à sombra. Nos seis dias iniciais de experimento, os animais foram submetidos à restrição alimentar, com acesso apenas à água, já expostos ao sol ou à sombra. Nos seis dias subsequentes, foi ofertada dieta composta por ração e feno de alfafa (3%PV). Os parâmetros avaliados foram frequência respiratória (FR), temperatura retal (TR), ß-hidroxibutirato (BHB), pH urinário, ingestão de matéria seca (IMS) e dados bioclimáticos (temperatura, umidade relativa do ar e índice de temperatura e umidade - ITU). A FR foi o parâmetro em que houve maior diferença significativa entre os grupos (p<0,05). O balanço energético negativo foi constatado em todos os animais (BHB>0,8mmol/L; pH<7,0). Ovelhas que sofrem restrição alimentar durante períodos de alta temperatura e umidade têm menor frequência respiratória e temperatura interna, independentemente de estar na sombra ou pleno sol. Porém, quando normalizado a oferta de alimento as ovelhas em pleno sol têm menor ingestão de matéria seca.(AU)


We aimed to evaluate physiological changes in sheep submitted to food restriction, exposed to the sun or shade. Ten ewe lambs were equally divided into two groups: Sun Radiation Group - GRS, exposed to sun during day hours, and Shade Group - GS, with animals placed inside a barn. In the first six days of experiment, animals were submitted to food restriction, with access to fresh water, already exposed to sun and shade. In the following six days alfalfa hay and concentrate were provided to the animals (3%BW). Parameters assessed were respiratory rate (FR), rectal temperature (TR), ß-hydroxybutyrate, urinary pH, dry matter intake (IMS) and climate data (air temperature, humidity, and temperature-humidity index - ITU). The parameter that most differed between groups was FR. All animals presented negative energy balance (BHB>0.8mmol/L; pH<7,0). Ewe lambs submitted to food restriction during periods of high temperature and humidity have lower respiratory rate and intern temperature, independently of being exposed to sun or shade. However, when food was offered animals exposed to sun had lower dry matter intake.(AU)

ABSTRACT

Avaliações com o intuito de mensurar marcadores de eficiência na performance esportiva do cavalo Crioulo são escassas e de fundamental importância no que tange às possíveis especificidades da raça. O objetivo do presente trabalho foi avaliar e determinar os padrões de frequência cardíaca, velocidade, concentração de lactato e gasto energético de equinos da raça Crioula durante provas credenciadoras ao Freio de Ouro. Tais variáveis foram avaliadas durante a realização das etapas funcionais da competição. Observaram-se flutuações superiores da variável frequência cardíaca (FC) durante a realização das etapas de Andadura, Figura, Volta sobre Patas e Esbarradas (And/fig/VSP) (203bpm) e menores valores na etapa Paleteada II (185bpm) (P<0,05). Em relação à velocidade, o maior valor atingido foi registrado na etapa de Paleteada II (39,7km/h). A concentração de lactato sanguíneo aferida se mostrou elevada em todas as fases da competição, sendo o maior valor observado na etapa de Paleteada II (14,5mmol/L) (P<0,05) e o menor durante a etapa de Mangueira I (9,3mmol/L). Superior gasto energético foi atribuído à etapa de And/Fig/VSP (853,28kcal/kgPV/min). Portanto, todas as etapas funcionais podem ser classificadas como anaeróbias, por apresentarem concentrações de lactato sanguíneo acima de 4mmol/L, e demandam alto gasto energético pelos competidores.(AU)


Evaluations of athletic performance markers of Crioulo breed horses are scarce yet fundamentally important regarding possible unique characteristics of this breed. This study aimed to evaluate and determine heart rate, speed, blood lactate and energy expenditure patterns of Crioulo breed horses during qualifying tests in the functional phases of the "Freio de Ouro" championship. Higher values of heart rate during the phases "andadura, figura, voltas sobre patas, esbarradas" (And/Fig/VSP) (203bpm) and lower values at "Paleteada II" (185bpm) (P<0.05) were noticed. Regarding speed variable, the maximum value was registered at "Paleteada II" (39.7km/h). During all the phases, blood lactate concentration was high, with the highest value found at "Paleteada II" (14.5mm/L) and the lowest during "Mangueira I" (9.3mm/L) (P<0.05). Superior energy expenditure was noticed in the "And/Fig/VSP" phase (853.28Kcal/kgPV/min). Thus, all functional phases can be classified as anaerobic, as blood lactate concentrations remained above 4mmol/L, with high energy demand of the horses.(AU)

ABSTRACT

O presente estudo avaliou o perfil mineral de Cálcio (Ca), Cobre (Cu) e Zinco (Zn) no estojo córneo pré e pós-desmame e sua associação com pigmentação, gênero e idade de potros da raça Crioula criados em pastagens nativas no Rio Grande do Sul. Foram coletadas amostras do casco de potros da raça Crioula 41 dias pré e 28 dias pós-desmame. Os teores de Ca, Cu e Zn variaram no casco fetal (571,0±39,4; 14,5±7,6 e 130,0±30,5mg/kg, respectivamente; P<0,05) e no definitivo (653,0±169,2; 33,8±11,5 e 69,3±36,8mg/kg, respectivamente; P<0,05), no pré (620,0±184; 17,2±21,2 e 103,0±75,5mg/kg, respectivamente; P<0,05) e no pós-desmame (517,5±181; 0 e 79,0±41,7mg/kg, respectivamente; P<0,05). Houve associação positiva (P<0,05) com a faixa etária no pré-desmame para Ca (r=0,5) e Cu (r=0,57), e negativa para Zn (r=-0,69; P<0,05). No pós-desmame, houve associação positiva (P<0,05) para Ca (r=0,36) e Zn (r=0,64) e negativa para Cu (r=-0,39; P<0,05). Tanto a pigmentação quanto o gênero não afetaram o perfil mineral. Conclui-se que há diferenças nas concentrações de Ca, Cu e Zn na epiderme do casco de potros da raça Crioula no pré e pós-desmame, independentemente da pigmentação e do gênero, onde as concentrações de Ca e Cu aumentam com a idade, enquanto as de Zn diminuem.(AU)


The aim of this study was to evaluate the mineral profile of Ca, Cu and Zn in the hoof horny capsule at pre and post-weaning and its association with pigmentation, gender and age range of Crioulo foals raised in native pastures in RS. Samples were collected from the epidermis of the Crioulo foal's hoof at two times, 41 pre and 28 post-weaning. The contents of Ca, Cu, Zn Varied in the fetal hoof ( 571.0±139.4, 14.5±7.6 and 130.0±30.5mg/kg, respectively, P<0.05) and definitive (653.0±169.2, 33.8±11.5 and 69.3±36.8mg/kg, respectively, P<0.05), in the pre (620.0±184, 17.2±21.2 and103.0±75.5mg/kg, respectively, P<0.05) and post-weaning (517.5±181, 0.1 and 79.0±41.7mg/kg, respectively, P<0.05).There was a positive association (P<0.05) with a preweaning age for Ca (r=0.5) and Cu (r=0.57) and negative for Zn (r=-0.69, P<0.05). In the post weaning there was a positive association (P<0.05) for Ca (r=0.36) and Zn (r=0.64) and negative for Cu (r=-0.39; P<0.05). Neither pigmentation nor gender affected the mineral profile. It is concluded that there are differences in Ca, Cu, Zn concentrations in the epidermis of foals of the crioulo breed at the pre and post weaning, regardless of pigmentation and sex, where Ca and Cu concentrations increase with age, while Zn concentrations decrease.(AU)

ABSTRACT

O objetivo do presente estudo foi analisar a dissimilaridade fenotípica de amostras populacionais de três linhagens de éguas Crioulas, uruguaia (La Invernada), argentina (Cardal) e chilena, para caracteres morfológicos, apontando as características de maior variância intra e entre as diferentes linhagens que são passíveis de seleção. Foram avaliadas 22 características morfológicas de 113 éguas da linhagem uruguaia "La Invernada", 38 argentinas "Cardal" e 73 chilenas puras, totalizando 224 éguas de cria pertencentes à raça Crioula. Para determinação das variáveis lineares e de perímetro, foi utilizada fita métrica, prumo, paquímetro e hipômetro. Verificou-se variabilidade nas características morfológicas entre as três linhagens (P<0,05), incluindo as características lineares altura, perímetro de tórax e perímetro de canela (P<0,01), consideradas de caráter impositivo para fins de registro. Maior variação fenotípica foi observada nas éguas da linhagem chilena em relação aos outros dois grupos genéticos (P<0,01). As características morfológicas altura garupa, perímetro rostral pescoço, comprimento metatarso, largura garupa, profundidade do tórax, comprimento dorsal pescoço e comprimento do corpo foram passíveis de seleção entre as éguas Crioulas pertencentes às linhagens argentina (Cardal), uruguaia (La Invernada) e chilena. Em conclusão, não há homogeneidade entre as linhagens estudadas, o que identifica a diversidade entre as linhagens de éguas Crioulas estudadas.(AU)


The aim of the study was to analyze the phenotypic dissimilarity of morphological characteristics of three strains of Criollo mares, Uruguian (La Invernada), Argentine (Cardal), and Chilena, pointing to the characteristics of higher variance within and among the different strains that are selectable. We evaluated 22 morphological characteristics of 113 mares of the Uruguaian line, 38 Argentines and 73 pure Chilean, totaling 224 breeding mares belonging to the criollo breed. For the determination of the linear and perimeter variables, a metric tape, plumb, pachymeter and hypometer were used. There was variability in the morphological characteristics between the three strains (P<0.05), including the linear characteristics of height, chest circumference and perimeter of cinnamon (P<0.01), considered as taxation for registration purposes. Higher phenotypic variation was observed in the mares of the Chilean line in relation to the other two genetic groups (P<0.01). The morphological characteristics of height croup, rostral perimeter neck, metatarsal length, croup width, chest depth, dorsal neck length, and body length were the selectable characteristics among the breeding mares belonging to the Argentine (Cardal), Uruguaian (La Invernada) and Chilena strains. In conclusion, there is no homogeneity between the studied strains, which identifies the diversity among the lines of Criollo mares studied.(AU)

ABSTRACT

Ozone (O3) therapy has been used for medical procedures for centuries; however, there are no extensive studies on its utilization in horses. This study aimed to evaluate the application of transrectal O3 on horses by physical and laboratorial evaluation, and production of reactive oxygen species (ROS). Sixteen healthy horses were separated in two groups: a control group (CG) and a group treated with O3 (TG). The TG animals received 1L of an oxygen and O3 mixture transrectally. The initial dose was 10µg/ml for the first two applications, 15µg/ml for the following two applications, and 20µg/ml for the next six applications. The CG animals received 1L of oxygen transrectally. In TG animals no variations in the physical examination were detected; furthermore, TG animals did not exhibit changes in biochemical evaluation results, fibrinogen concentrations, or ROS production. TG animals had increased red blood cell counts, hemoglobin concentrations, and packet cell volume values in comparison to the baseline and CG values. We could infer that O3 affected the red blood cell counts and improved rhetological properties of the blood. The transrectal application of O3 in horses is safe and can indirectly improve the oxygenation and metabolism of tissues.(AU)


A utilização medicinal do ozônio (O3) é secular, contudo não existem estudos expressivos de sua utilização em equinos. O objetivo deste estudo foi avaliar o efeito da aplicação transretal de O3 em equinos por meio da avaliação física, laboratorial, e produção de espécies reativas de oxigênio (EROs). Dezesseis equinos hígidos foram separados em dois grupos: grupo controle (GC) e grupo tratado com O3 (GT). O GT recebeu por via retal 1L da mistura de oxigênio e ozônio, sendo a dose inicial de 10µg/ml por duas aplicações, 15µg/ml por mais duas aplicações e 20µg/ml por seis aplicações. O GC recebeu 1L de oxigênio via transretal. No GT não foram observadas alterações no exame físico, bem como não foram observadas alterações na avaliação bioquímica, concentração de fibrinogênio e produção de EROs. O GT apresentou aumento no número de hemácias, na concentração de hemoglobina, e nos valores de hematócrito em relação aos valores basais e GC. Podemos inferir que o O3 alterou os valores de eritrócitos e melhorou as propriedades reológicas do sangue. Conclui-se que a aplicação transretal de 03 em equinos é segura e pode melhorar indiretamente a oxigenação e metabolismo dos tecidos.(AU)

ABSTRACT

O objetivo deste trabalho foi determinar a digestibilidade do farelo de palmiste (Elaeis guineensis) para o tambaqui (Colossoma macropomum), em duas classes de peso: 1 (210 alevinos de 4,45±1,18g) e 2 (54 juvenis de 115,91±4,01g). Os coeficientes de digestibilidade aparente (CDA) da matéria seca, proteína bruta e energia bruta do farelo de palmiste foram avaliados pela metodologia de substituição da dieta referência, utilizando-se 0,1% de óxido crômico como indicador externo. Os dados foram analisados pelo teste t de Student, a 5% de probabilidade. Os CDAs da matéria seca, proteína bruta e energia bruta do ingrediente foram iguais (P>0,05) nas classes de peso avaliadas. Os CDAs observados nas classes 1 e 2, respectivamente, foram: matéria seca (17,52% e 20,75%), proteína bruta (62,83% e 63,75%) e energia bruta (14,16% e 22,43%). A capacidade do tambaqui para digerir os nutrientes do farelo de palmiste não foi influenciada pelo peso corporal, e o aproveitamento satisfatório da proteína (63,29%) faz desse ingrediente uma potencial fonte alternativa de proteína em dietas para a espécie.(AU)


The objective of this work was to determine the digestibility of palm kernel meal (Elaeis guineensis) in tambaqui (Colossoma macropomum), in two weight classes: 1 (210 fingerlings of 4.45±1.18g) and 2 (54 juveniles of 115.91±4.01g). The apparent digestibility coefficients (ADC) of dry matter, crude protein and crude energy of the palm kernel meal were evaluated by the substitution of the reference diet methodology, using 0.1% chromic oxide as an external indicator. Data were analyzed by Student's t-test at 5% probability. The dry matter, crude protein and crude energy ADCs of the ingredient were the same (P> 0.05) in the weight classes evaluated. The ADCs observed in classes 1 and 2, respectively, were: dry matter (17.52% and 20.75%), crude protein (62.83% and 63.75%) and crude energy (14.16% and 22.43%). The ability of tambaqui to digest the nutrients of palm kernel meal was not influenced by body weight, and satisfactory protein utilization (63.29%) makes this ingredient a potential alternative source of protein in diets for the species.(AU)

ABSTRACT

A fenda palatina é a comunicação entre a cavidade oral e a nasal através de um orifício no palato. Tem diversas etiologias, podendo ser congênita, traumática, por deficiência mineral ou por fatores hormonais. O diagnóstico é realizado por meio de exame físico da cavidade oral, e a correção cirúrgica é o tratamento de escolha. Em animais adultos, pode ser corrigida com o auxílio de retalho mucoperiosteal, apresentando bons resultados. Já em filhotes, a correção cirúrgica é mais complicada, com prognóstico menos favorável. Este trabalho relata o caso de um canino, fêmea, sem raça definida, adulta, com histórico de fenda palatina secundária, de origem traumática, no palato mole por ingestão de osso. Para a correção cirúrgica, primeiramente foi utilizada membrana biológica de pericárdio bovino, mas não se obteve êxito. O segundo procedimento foi realizado com retalho mucoperiosteal simples autólogo e, dois meses após o procedimento, já havia cicatrização completa. A técnica de retalho mucoperiosteal simples autólogo se mostrou eficaz no tratamento da fenda palatina, aliada aos cuidados adequados no pós-operatório.(AU)


The cleft palate is the communication between the oral and nasal cavity through an aperture in the palate, it's causes include an infinitude of factors: congenital, traumatic, mineral deficiency or hormonal. Examination of the oral cavity determines if the diagnosis and treatment is surgical. Correction in adult animals is performed with mucoperiosteal flap showing good results. However, surgical correction in puppies is more complicated with less favorable prognosis. This current work reports a case of an adult, female dog of undefined breed, with a history of secondary clef palate of traumatic origin in the soft palate due to bone ingestion. For correction, a biological membrane of bovine pericardium was used, but it was not successful, requiring a second surgical procedure performed with autologous simple mucoperiosteal flap. The last technique combined with adequate postoperative care was effective.(AU)

ABSTRACT

As lesões dos meniscos são uma das principais causas de dor e claudicação em equinos. O objetivo do presente estudo foi avaliar a morfometria dos meniscos da articulação femorotibial de equinos. Foram utilizados 48 meniscos de 12 animais de ambos os sexos, com idade entre cinco e 15 anos e com peso entre 400kg e 500kg. A medida da extensão periférica compreendeu desde a parte mais cranial à mais caudal e denominou-se circunferência externa (CE). A margem interna, com o mesmo tratamento, foi chamada de circunferência interna (CI). Os meniscos foram divididos em terços craniais, médios e caudais. A espessura foi obtida nos pontos médios de cada terço. Foi calculada a área dos meniscos em contato com os côndilos femorais. O menisco medial apresentou maior CE com média de 126,38mm, enquanto o menisco lateral apresentou média de 115,32mm. O menisco lateral mostrou maior espessura nos terços médio e caudal, com valores médios de 16,00mm e 19,85mm, respectivamente, contra 13,75mm e 14,99mm dos meniscos mediais. Os resultados deste estudo mostraram relação importante entre os dados morfométricos e os achados clínicos na tentativa de explicar a maior incidência de lesões envolvendo o menisco medial.(AU)


Meniscal lesions are one of the main causes of pain and lameness in horses. The objective of this study was to evaluate the morphometry of the meniscus of the femorotibial joint of horses. 48 meniscus were used from 12 animals, aged between five and 15 years and weighing between 400kg and 500kg. The measurement of the peripheral extension went from the most cranial part to the most caudal, denominated external circumference (CE). The internal margin, with the same treatment, was called internal circumference (IC). The meniscus was divided into cranial, middle and caudal thirds. The thickness was obtained at the midpoints of each third. The area of the meniscus in contact with the femoral condyles was also calculated. The medial meniscus presented a higher CE with a mean of 126.38mm, while the lateral meniscus presented a mean of 115.32mm. The lateral meniscus showed greater thickness in the middle and caudal thirds, with mean values of 16.00mm and 19.85mm respectively, against 13.75mm and 14.99mm of the medial meniscus. The results of this study showed an important relationship between the morphometric data and the clinical findings in an attempt to explain the higher incidence of lesions involving the medial meniscus.(AU)

There is currently a lack of information adjacent on the influence of sex and age in heart rate variability (HRV), adjusted according to accelerometer-based physical activity (PADL). We hypothesized that the effect of sex and age on the HRV should be reduced or absent in individuals with a suitable PADL level. We aim to evaluate the influence of sex and age on HRV, adjusted for the confounding effects of the PADL level. A total of 485 age-stratified subjects (18-39, 40-59, and ≥60 years) underwent HRV analyses at rest and 7-day assessments of accelerometer-based PADL. Multivariate analyses of covariance were done using log-transformed HRV indices as outcomes, age and sex as fixed factors, and PADL, cardiovascular risk, fat body mass, and heart rate (HR) at rest as covariates. Despite the adjustment for directly measured PADL, women had better indices of vagal tone, whereas men had higher sympathetic influence. Also, compared to middle-aged and older adults, younger individuals (ages 18-39 years) presented better HRV. Multiple regression analyses confirmed that age and sex were the main predictors of HRV indices, even after adjusting for PADL directly assessed by triaxial accelerometer and HR. We also observed that the correlation between some HRV indexes and the different indexes of physical activity directly evaluated was significant, but not very consistent. Thus, HRV indices are influenced by age and sex, regardless of accelerometer-based physical activity. Interventions with physical activity and exercise aimed at improving the autonomic modulation of asymptomatic adults should take such differences into account.

ABSTRACT

Métodos observacionais subjetivos, como escore de locomoção (EL) e escore de condição corporal (ECC), têm sido amplamente utilizados para avaliação populacional de diferentes graus de claudicação e estado nutricional em bovinos. Este estudo objetivou verificar a associação longitudinal entre o escore de condição corporal e o escore de locomoção em vacas da raça Holandesa submetidas a um regime alimentar semiextensivo. O período experimental compreendeu dois anos de observações, em uma propriedade localizada no sul do Rio Grande do Sul. O grupo experimental foi constituído por 900 vacas lactantes, que foram avaliadas trimestralmente por dinâmica ortopédica pelo EL e do estado metabólico pelo ECC. A coleta desses dados foi realizada por três veterinários treinados. A fim de se verificar a correlação entre variáveis, utilizou-se o programa SAS, por meio do método de correlação de Pearson, para determinar a associação entre as variáveis avaliadas (EL e ECC), com nível de significância de 5%. A distribuição dos diferentes escores de locomoção durante o período foi a seguinte: 57,2% dos animais tiveram o escore de locomoção 1, ou seja, estavam saudáveis em relação ao sistema locomotor. O restante dos animais que apresentou algum grau de dificuldade de locomoção distribuiu-se da seguinte forma, segundo a intensidade: 21,6% dos animais apresentaram EL2, 15,5% deles EL3, 3,5% deles EL4 e 2,2% deles EL5. Quanto ao resultado da correlação, observou-se associação negativa (r= -0,57) entre ECC e EL (P= 0,03), com ECC médio de 2,97±0,33 e EL de 1,35±0,24 durante o período experimental. Portanto, há uma correlação negativa entre o escore de condição corporal e o escore de locomoção em vacas da raça Holandesa submetidas ao regime alimentar semiextensivo.(AU)


Subjective observational methods such as locomotion score (LS) and body condition score (BSC) have been widely used for populations evaluation of different degrees of claudication and nutritional status in cattle. This study aimed to verify the longitudinal association between body condition score and locomotion score in Holstein cows submitted to a semi-extensive breeding regime. The experimental period comprised two years of observations at a property located in the South of Rio Grande do Sul. The experimental group consisted of 900 lactating cows, which were evaluated quarterly, being evaluated by orthopedic dynamics through LS and of the metabolic status through BCS. The collection of these data was performed by three trained veterinarians. To verify the correlation between variables, the SAS program was used using the Pearson Correlation method to determine the association between the variables evaluated (LS and BSC), at a significance level of 5%. The rest of the animals that showed some degree of locomotion difficulty, were distributed following intensity: 21.6% of LS2 animals, 15.5% of LS3 animals, 3.5% with LS4, and 2.2% with LS5, while 57,2% were without locomotion difficulty (LS1). Regarding the correlation result, a negative association (r= -0.57) was found between BSC and LS (P=0.03), with an overall of BSC of 2.97±0.33 and LS of 1, 35±0.24 during the experimental period. Therefore, there is a negative correlation between body condition score and locomotion score in Holstein cows submitted to the semi-extensive breeding regime.(AU)

ABSTRACT

A urolitíase obstrutiva em pequenos ruminantes é uma doença metabólica de etiologia multifatorial com distribuição mundial. A elevação da concentração urinária de solutos, minerais ionizados (cristaloides) que formam cristais insolúveis é citada por alguns autores como o fator mais importante. Assim, o conhecimento do perfil mineral dos animais submetidos a dietas calculogênicas e a composição química dos urólitos tornam-se ferramentas eficazes na prevenção da doença. Neste estudo, foram utilizados 14 ovinos hígidos, machos (não castrados), da raça Santa Inês, com idade aproximada de 90 dias, distribuídos em dois grupos (G1 - sem vitamina C e G2 - com vitamina C) e alimentados com dieta calculogênica. A análise dos perfis minerais, séricos e urinários revelou completo desbalanceamento na relação entre concentrações de cálcio, fósforo e magnésio, havendo elevação expressiva do fósforo e do magnésio e diminuição substancial do cálcio. Com isso, a análise bioquímica dos urólitos demonstrou que o cálcio esteve presente em 50% das amostras analisadas.(AU)


Urolithiasis in small ruminants is a metabolic disease of multifactorial etiology with worldwide distribution. Increased urinary concentration of solutes, ionized minerals (crystalloid) that form insoluble crystals is cited by some authors as the most important factor. Thus, knowledge of mineral profile of the animals fed calculogenic diets and chemical composition of uroliths becomes an effective tool in preventing the disease. In this study, we used 14 healthy, male, non-neutered sheep, of the Santa Ines breed, aged approximately 90 days, divided into two groups (G1-without vitamin C and G2-with vitamin C) fed calculogenic diet. Analysis of mineral profiles in serum and urine revealed complete imbalance in the relationship between concentrations of calcium, phosphorus and magnesium, with significant increase of phosphorus and magnesium and substantial reduction of calcium. Thus, biochemical analysis of uroliths showed that calcium was present in 50% of samples.(AU)

ABSTRACT

Abstract Antimicrobial resistance in Escherichia coli isolated from pet dogs can be considered a potential threat of infection for the human population. Our objective was to characterize the resistance pattern, extended spectrum beta-lactamase production and genetic relatedness of multiresistant E. coli strains isolated from dogs (n = 134), their owners (n = 134), and humans who claim to have no contact with dogs (n = 44, control), searching for sharing of strains. The strains were assessed for their genetic relatedness by phylogenetic grouping and pulsed-field gel electrophoresis. Multiresistant E. coli strains were isolated from 42 (31.3%) fecal samples from pairs of dogs and owners, totaling 84 isolates, and from 19 (43.1%) control group subjects. The strains showed high levels of resistance to ampicillin, streptomycin, tetracycline, trimethoprim and sulfamethoxazole regardless of host species or group of origin. The blaTEM, blaCTX-M, and blaSHV genes were detected in similar proportions in all groups. All isolates positive for bla genes were ESBL producers. The phylogenetic group A was the most prevalent, irrespective of the host species. None of the strains belonging to the B2 group contained bla genes. Similar resistance patterns were found for strains from dogs, owners and controls; furthermore, identical PFGE profiles were detected in four (9.5%) isolate pairs from dogs and owners, denoting the sharing of strains. Pet dogs were shown to be a potential household source of multiresistant E. coli strains.

ABSTRACT

Abstract Paca (Cuniculus paca Linnaeus, 1766) is the second largest rodent found in Brazil. The quality of the meat and a long tradition of hunting have contributed to the decline of the natural populations of this species. Hunting of paca is strictly prohibited in Brazil, but in spite of this restriction, no forensic tools are available for the identification of the meat. We describe an efficient method, based on single nucleotide polymorphisms of the cytochrome b gene, that can be used to differentiate biological material derived from paca from those of domestic species commonly used as sources of meat. The identification of the presence of C. paca in the samples was 100% reliable.


Resumo Paca (Cuniculus paca Linnaeus, 1766) é o segundo maior roedor brasileiro. A qualidade da carne e a forte tradição da caça de subsistência são fatores que contribuem significativamente para o declínio das populações. Apesar da proibição a caça no Brasil, no momento ainda não há ferramentas disponíveis para identificar a carne e seus produtos como prova forense. Neste trabalho propomos um método eficaz de identificação, baseado em polimorfismos de único nucleotídeo no gene Citocromo b, objetivando diferenciar material biológico de paca das espécies domésticas comumente utilizadas como alimento no Brasil. A identificação das amostras de paca foram possíveis em 100% das amostras analisadas.

ABSTRACT

The objective of this study was to evaluate the alterations in sleep and circadian parameters during the aging process. The study sample comprises volunteers older than 18 up to 90 years of age that answered the Pittsburgh Sleep Quality Index (PSQI) and the Horne and Östberg circadian preference questionnaire. We observed that the shift to morningness with increasing age is associated with a significant worsening in sleep quality. We discuss that this sleep profile characterized by morningness and worse sleep quality observed in elderly, when compared to younger people, reflects not necessarily a pathological state, but an expected profile for this age group.

ABSTRACT Seeds of Acacia farnesiana are commonly sold in the local markets of northeastern Brazil as a therapeutic agent. The present work aimed to evaluate the anti-inflammatory and analgesic activities of proteins obtained from A. farnesiana seeds. Five different protein fractions (albumin, globulin, prolamin, acidic and basic glutelins) were obtained and investigated for the protein pattern, the presence of hemagglutinating and proteolytic activities. The globulin fraction (GLB) was also evaluated for anti-inflammatory and analgesic activities. Globulins reduced the paw edema induced by carrageenan in a dose-dependent manner, which was accompanied by a reduction of myeloperoxidase activity (p < 0.05). Additionally, GLB reduced the neutrophil peritoneal migration induced by carrageenan. However, GLB was not able to inhibit the edema triggered by dextran. Pre-treatment with globulins reduced the abdominal constrictions induced by acetic acid as well as the paw licking time induced by formalin (69.1% at first phase). However, it did not produce a significant antinociceptive effect in the hot plate test (55-56 °C). Treating the GLB with heat (at 100 °C for 30 min) abolished its anti-edematogenic and hemagglutinating activities. Our results showed that seeds from A. farnesiana are a source of proteins with anti-inflammatory and analgesic properties.


RESUMO Sementes de Acacia farnesiana são comumente vendidas em feiras locais no nordeste do Brasil como agente terapêutico. O presente trabalho objetivou avaliar as atividades antiinflamatória e antinociceptiva de proteínas obtidas de sementes de A. farnesiana. Cinco frações protéicas distintas (albuminas, globulinas, prolaminas, glutelinas ácidas e básicas) foram obtidas e investigadas quanto o perfil de proteínas, presença de atividade hemaglutinante e proteolítica. A fração globulina (GLB) também foi avaliada quanto a presença de atividade antiinflamatória e analgésica. Globulinas reduziram o edema de pata induzido por carragenina de modo dependente da dose que foi acompanhada da redução da atividade da mieloperoxidase (p < 0,05). Em adição, GLB reduziu a migração de neutrófilos para cavidade peritoneal induzida por carragenina. Entretanto, GLB não foi capaz de inibir o edema induzido por dextrana. O pré-tratamento com globulinas reduziu as contorções abdominais induzidas por ácido acético, bem como o tempo de lambedura da pata induzida por formalina (69.1% na primeira fase). Por outro lado, GLB não produziu um efeito antinociceptivo significante no teste de placa quente (55-56 °C). O pré-tratamento de GLB com calor (100 °C por 30 min) aboliu sua atividade anti-edematogênica e hemaglutinante. Nossos resultados mostraram que sementes de A. farnesiana são fonte de proteínas com propriedades antiinflamatórias e analgésicas.

ABSTRACT

This report aimed to study the interference in molecular testing for Ehrlichia canis and Anaplasma platys in blood of 155 dogs from the coastal region of Rio de Janeiro. Five Anaplasmataceae positive samples but negative for E. canis and A. platys, from microfilaremic animals, were chosen for sequencing. These sequences, when compared to Gen et Bank database, showed 88% to 100% similarity with Wolbachia spp. denoting an interference in the detection of DNA from other members of Anaplasmataceae, possibly due to a high concentration of Wolbachia spp. DNA.(AU)
