ABSTRACT

Phytochemical investigation of Kandelia candel resulted in the isolation of six triterpenes (1 - 5) and two glyceryl glycosides (6 and 7) and their structures were determined by comparing the spectroscopic data with those of reported values. In present study, we described the inhibitory effects of fractions and isolated compounds from K. candel on pro-inflammatory cytokines (IL-12 p40, IL-6, and TNF-alpha) production in lipopolysaccharide (LPS) stimulated bone marrow-derived dendritic cells (BMDCs). Results indicated that compounds 3, 6, and 7 showed potent inhibition on IL-6 production (IC50 values at less than 0.5 microM, respectively). Meanwhile, compounds 6 and 7 exhibited strong inhibitory effects on the production of TNF-alpha (IC50 values of 1.7 +/- 0.1 and 5.5 +/- 0.2 microM). Compounds 1 and 3 were also showed the inhibitory effects on IL-12 p40 production (IC50 values of 8.9 +/- 0.4 and 3.3 +/- 0.1 microM, respectively).

ABSTRACT

OBJECTIVE@#To identify inhibitors of nitric oxide production and NF-κB activity from Chromolaena odorata (C. odorata).@*METHODS@#The compounds isolated from the aerial parts of C. odorata by bioassay-guided fractionation were investigated for their inhibitory effects on the NO production and NF-κB activity in LPS-stimulated RAW264.7 cells.@*RESULTS@#Six fatty acids (S)-coriolic acid (1), (S)-coriolic acid methyl ester (2), (S)-15,16-didehydrocoriolic acid (3), (S)-15,16-didehydrocoriolic acid methyl ester (4), linoleamide (5) and linolenamide (6) were isolated. All compounds inhibited the NO production at concentrations consistent with those required for NF-κB inhibition. Compound 2 was the most active with the IC(50) values of 5.22 and 5.73 μM. The addition of a double bond in the fatty chain decreased the inhibitory effects while the methyl esterification increased the activities.@*CONCLUSIONS@#The fatty acid components in C. odorata with NF-κB inhibitory activity could explain the anti-inflammation property of this plant in traditional medicine. This study could also contribute to the better use of C. odorata for human health care.

ABSTRACT

Dys-1 and Dys-2 compounds were isolated and determined structure from extracts of n-hexane and dichloromethane of sponge Dysidea cinerea by normal and reverse phases of silica gel column chromatography. Dys-1 is a white crystallized agent. Through data and reference materials, the agent is determined as Isofucosterol (24-Stigmasta-5, 24(24')-dien-3β-ol). Dys-2 compound is a crystallized agent with crystal of white needle form. The compound is determined as 5, 8- epidioxycholest-6--en-3-ol. It is cyto-toxic against three tumor cell lines in human such as epithelium cancer, liver cancer, and uterus membrane cancer
