ABSTRACT

The present study aimed to explore the effects of Eerdun-Wurile on brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) expressions in the prefrontal cortex of MCAO / R injury rats.Sixty SD male rats of SPF grade were selected to establish the model of MCAO / R with Zea-Longa thread occlusion,and divided into five groups at random:the sham operation group,the model group,the nimodipine group and the Eerdun-Wurile group.After modeling,rats were anesthetized for preparing the brains.The pathomorphological changes of the brains were evaluated by immunohistochemical techniques,such as HE staining and SP.The protein and mRNA expressions of BDNF and NGF in the prefrontal cortex of rats were detected by ELISA and RT-PCR,respectively.As a result,compared with the model group,it was found that the number of necrotic cells in the prefrontal cortex were significantly reduced in the Eerdun-Wurile group (P < 0.05),while the mRNA and protein expressions of BDNF and NGF were significantly increased (P < 0.05).In conclusion,the BDNF and NGF expressions in the prefrontal cortex were up-regulated for stimulating the activation of astrocytes and protecting the neurons with the treatment of Eerdun-Wurile in MCAO / R injured rats,which may be the mechanism of the treatment of Eerdun-Wurile for white vein disease.

ABSTRACT

Objective:To investigate the inhibitory effect and mechanism of different doses of Xiao Chai Hu Tang on C6 glioma cells cultured in vitro. Methods:C6 glioma cells were inoculated in 96 holes,24 holes and 6 holes,each culture plate was divided into 4 groups:control group, low dose group, middle dose group and high dose group, when the cells covered the bottom of culture plate 80%-90%,began adding,cultured for 24 hours after the ter mination of training. Cell proliferation activity,cell viability,protein content and protein positive expression intensity of VEGF and ESM-1,cell apoptosis in early and late stage were detected by CCK-8,in vivo staining,ELISA, immunocytochemistry and flow cytometry. Results: CCK-8 assay showed that the growth of C6 glioma cells was inhibited by low,medium and high dose group;ELISA and immunocytochemistry showed that the expression of VEGF and ESM-1 was lower in the lower, middle and high dose groups, and the levels of protein expression and protein levels were decreased. The flow cytometry showed that the low dose of small radix,middle and high dose group could promote the cell apoptosis. Inverted microscope ob-servation showed that with the increase of dose,the number of cells increased gradually,and the number of dead cells increased,and all kinds of detection methods showed that the inhibition effect increased with dose and dose dependence. The difference between the two groups was statistically significant(P<0. 01). Conclusion:The growth of C6 glioma cells was significantly inhibited by Xiao Chai Hu Tang. It may play a role in inhibiting tumor growth by down regulating ESM-1 and VEGF protein level and promoting cell apoptosis.

ABSTRACT

Objective:To investigate the influence of piperlonguminine on blood-fat of animal and its acute toxicity. Methods:The hyperlipidemic model rats were established by feeding with the high-lipid diet for 14 days.Stomach-perfusion with piperlonguminine was accompanied at the same time.Effects of piperlonguminine on the hyperlipidemic rats were investigated by measurement of the contents of total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-c),high- density lipoprotein cholesterol(HDL-c).The hyperlipidemic model rabbits were established by feeding with hypercholesterol. The influences of piperlonguminine on serum lipid,liver lipid and atheroma were observed.The response of the acute toxicity of mice was also observed.Results:Piperlonguminine remarkably lowered the serum TC、TG、LDL-c,and increased the serum HDL-c of rats,lowered the serum TC、TG、LDL-c and liver TC、TG,and remarkably improved atheroma of rabbits.The acute toxicity was tested on mice.During the test there was no death.Even more it caused little pathological change.Conclusion: Piperlonguminine not only had significant antihyperlipidemia effects,but also improved atheroma effects.And the toxicity of piperlonguminine was very low.

ABSTRACT

<p><b>OBJECTIVE</b>To observe sperm motion parameters in pre-freeze and post-thaw semen samples using computer-assisted sperm analysis (CASA) system.</p><p><b>METHODS</b>Semen analyses of 238 samples before freezing and after thawing were separately performed by Hamilton-Thorne Sperm Analyzer.</p><p><b>RESULTS</b>Sperm motility in post-thaw samples was significantly decreased. There was significant correlation and difference between pre-freeze and post-thaw samples in sperm motion parameters, including average path velocity (VAP), straight line velocity (VSL), curvilinear velocity (VCL), amplitude of lateral head displacement (ALH), straightness (STR) and linearity (LIN), except heat cross frequency (BCF). The percentage of sperm movement velocity parameters (VAP, VSL and VCL) and moving pattern parameters (ALH) significantly decreased, while that of LIN and STR significantly increased in post-thaw samples.</p><p><b>CONCLUSION</b>CASA system is of clinically applied value and is a useful tool for evaluating sperm motion parameters in pre-freeze and post-thaw semen samples.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the semen quality of the donors.</p><p><b>METHODS</b>The semen parameters of the primary samples of 512 donors were examined following the World Health Organization (WHO) guide and computer-aided sperm analysis (CASA, product by Harmilton Thorne) system.</p><p><b>RESULTS</b>Of the 246 (246/512, 48%) donors with potential fertility, only 146 (146/512, 28.5%) came up to the semen standard set by the Chinese Ministry of Health, while 266 (266/512, 52%) were below the WHO reference values of semen parameters.</p><p><b>CONCLUSION</b>Repeat semen analyses may increase the success rate in screening semen donors. The semen quality of the donors suggests that it is necessary to pay more attention to male reproductive health.</p>
