ABSTRACT

Objective To optimize the manufacture process for Zhibai Anshen oral liquid. Methods The orthogonal designed experiments were conducted to monitor the effects of three factors on the content of mangiferin. The three factors included the amount of water, extraction time and alcohol precipitation concentration. Six month accelerated stability study and twelve month long term stability study were performed. Results Optimum percolation process was boiling the mixture with 10 times of water for 1 hour, followed by deposition with 60% alcohol. Conclusion This optimized process can be used for mass production.

ABSTRACT

Oligodendrocytes (OLs) are myelinating glial cells that form myelin sheaths around axons to ensure rapid and focal conduction of action potentials. Here, we found that an axonal outgrowth regulatory molecule, AATYK (apoptosis-associated tyrosine kinase), was up-regulated with OL differentiation and remyelination. We therefore studied its role in OL differentiation. The results showed that AATYK knockdown inhibited OL differentiation and the expression of myelin genes in vitro. Moreover, AATYK-deficiency maintained the proliferation status of OLs but did not affect their survival. Thus, AATYK is essential for the differentiation of OLs.

ABSTRACT

Objective To study the fingerprint of Polygonum cuspidatum from various regions by HPLC and provide technical support for the identification. Methods HPLC conditions :the chromatographic column was Agilent ZORBAX SB-C18 (4.6 mm × 250 mm,5 μm);the mobile phase was 0.1% formic acid in water and acetonitrile (A-B);gradient elution ;the de-tection wavelength was 306 nm ;the flow rate was 1 ml/min;the column temperature was 30 ℃ ;injection volume was 20 μl. Results The HPLC gradient conditions could effectively separate polydatin, resveratrol, emodin and emodin ether,etc.Con-clusion This method is stable, effective and durable, which is instructive to the herbs data identification of Polygonum cuspi-datum.

ABSTRACT

OBJECTIVE:To establish a HPLC method for determining the contents of rhein and emodin in Jianfei Jiangzhi tablets.METHODS:Stationary phase was Waters Symmetry C 18 (250mm?4.6mm;5?m),mobile phase was menthol-0.01%H 3 PO 4 (90∶10),detection wavelength440nm,flow rate1.0ml/min and column temperature30℃.RESULTS:There was good linearity in range of0.108?g~0.468?g for rhein and in range of0.180?g~0.480?g for emodin. The average recoveries of rhein and emodin were99.7%and98.6%respectively;RSD were1.93%and2.19%respectively.CONCLUSION:This method is rapid and accurate. It can be applied to effective control of the quality of Jianfei Jiangzhi tablet.
