ABSTRACT

Objective:To establish an effective classification and identification method for sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) maps of Rana dybowskii,its analogues and counterfeits based on cluster analysis and multivariate statistical analysis. Method:SDS-PAGE maps of 18 batches of R. dybowskii,its analogues and 2 counterfeits were obtained by SDS-PAGE method. SDS-PAGE maps were transformed into data matrix. NTSYSpc 2.10e statistical analysis software was used for cluster analysis,and SMICA-P 14.1 software was used for multivariate statistical analysis. Unsupervised Principal Component Analysis (PCA),Supervised Partial Least Squares Discriminant Analysis (PLS-DA) and Orthogonal Partial Least Squares Discriminant Analysis (OPLS-DA) were performed for multivariate analysis and evaluation. Result:SDS-PAGE maps technology combined with cluster analysis and multivariate statistical analysis could accurately classify and identify R. dybowskii,its analogues and counterfeits. Cluster analysis could cluster four kinds of medicinal materials into four branches except No.1 medicinal materials. PCA results were superior to cluster analysis. Supervised PLS-DA and OPLS-DA results in multivariate statistical analysis were superior to unsupervised PCA. The classification and identification efficiencies of OPLS-DA were better than those of unsupervised PCA. OPLS-DA aggregated R. dybowskii,its analogues and 2 counterfeits into four groups. Six different protein components were obtained by comprehensive analysis of variable importance in projection (VIP) value, and OPLS-DA Bi load diagram,with relative molecular weights were 51.363,35.838,14.565,17.563,15.358 and 21.696 kDa,respectively. Conclusion:SDS-PAGE maps combined with cluster analysis and multivariate statistical analysis can be used as an effective method to classify and identify R. dybowskii,its analogues and counterfeits. This study provides a reference for the quality evaluation and screening of R. dybowskii.

ABSTRACT

Objective To understand the current overall status of implementation on the National Demonstration Areas of Comprehensive Prevention and Control of Non-communicable Diseases.Methods According to the scheme design of the questionnaires,all the National Demonstration Areas were involved in this study.For each National Demonstration Areas,eight departments were selected to complete a total of 12 questionnaires.Results Scores related to the implementation of the National Demonstration Areas accounted for 71.8% of the total 170 points.Based on the scores gathered from this study,the 23-items-index-system that represented the status of project implementation was classified into seven categories.Categories with higher percentile scores would include:monitoring (88.0%),safeguard measures (75.0%),health education and health promotion (75.0%).Categories with lower percentile scores would include:the national health lifestyle actions (67.7%),community diagnosis (66.7%),discovery and intervention of high-risk groups (64.7%),and patient management (60.9%).There were significant differences noticed among the eastern,central and western areas on items as safeguard measures,health education/promotion,discovery and intervention of high-risk groups.In all,the implementation programs in the eastern Demonstration Areas seemed better than in the central or western regions.As for the 23 items,five of the highest scores appeared on policy support,mortality surveillance,tumor registration,reporting system on cardiovascular/cerebrovascular events,and on tobacco control,respectively.However,the lowest five scores fell on healthy diet,patient self-management program,oral hygiene,setting up the demonstration units and promotion on basic public health services,respectively.The overall scores in the eastern region was higher than that in the central or the western regions.The scores in the central and western regions showed basically the same.Conclusions The overall status of implementation on the National Demonstration Areas was satisfactory.Future attention should be focusing on patient management as well as discovery and intervention of high-risk groups,which also presented the lowest scores,in this survey.

ABSTRACT

Objective To investigate the consumption of vegetables and fruits and related influencing factors among residents from the National Demonstration Areas of Comprehensive Prevention and Control of Non-communicable Diseases.Methods From November to December,2016,a total of 4 000 residents,aged 18 or above,from ten Demonstration Areas,were selected as participants for this study by multi-stage cluster random sampling method.Information on vegetables,fruits consumption and related influencing factors was collected via questionnaire.Results A total of 3 891 residents were involved in the final analysis.Daily consumption of vegetables and fruits accounted for 72.1% and 53.6% of the residents under study.The residents who were aware of the National Demonstration Areas activities were more willing to have adequate intake of vegetables (OR=3.017,95%CI:2.426-3.753) and fruits (OR=1.261,95%CI:1.007-1.580).Residents with higher degree of participation activities of the demonstration areas were more likely to have adequate fruits intake (high degree:OR=1.431,95%CI:1.210-1.694;medium degree:OR=1.573,95%CI:1.315-1.882).Conclusions The implementation of the National Demonstration Areas for Comprehensive Prevention and Control of Non-communicable Diseases has improved the adequate vegetables and fruits intake among residents.Relevant activities carried out in the Demonstration Areas appeared conducive to the healthy lifestyle of the residents.

ABSTRACT

Objective To understand the current situation on management of diabetes mellitus patients aged 35 and above in the National Demonstration Areas for Comprehensive Prevention and Control of Non-communicable Diseases,in China.Methods Local residents,aged 18 years and above were randomly selected by a complex,multistage,probability sampling method.Face-to-face questionnaire survey was carried out between November and December 2016.Rates regarding prevalence,treatment and management of diabetes were calculated,and influencing factors of diabetes were analyzed by using the non-conditional logistic regression model.Results A total of 3 213 residents aged ≥35 years were included in this study,of which 11.48% (369/3 213) reported that they had ever been informed by a doctor or other health worker that their blood sugar level was high or being diabetic.The rate of self-reported treatment among the diabetic patients was 83.20% (307/369).Rates on overall management and standardized management were 69.92% (258/369) and 53.66% (198/369),respectively.Higher rates were seen in residents aged 55 to 64 years,76.32% for overall management and 59.65% for standardized management.Through multiple logistic regression analysis,we found that standardized management for diabetes was much higher in the Demonstration Areas located in the eastern areas (OR=2.942,95%CI:1.547-5.594),or patients with characteristics including high implementation score (OR=3.499,95% CI:1.865-6.563),already signed family doctors (OR=5.661,95% CI:3.237-9.899),or without hypertension (OR=1.717,95% CI:1.010-2.920).Residents who were living in the first and second batch areas of implementation or responding to the NCDs with positive attitude were more likely to accept standardized management.Conclusion Prevention and management programs on diabetes had met the requirements set for the Demonstration Areas which had promoted the specific implementation and further development of standardized management on diabetes.

ABSTRACT

Objective To investigate the current status of prevalence,treatment,and management on hypertension among Chinese adults from the National Demonstration Areas for Comprehensive Prevention and Control of Non-communicable Diseases.Methods We selected a total of 4 000 residents aged ≥ 18 years for this questionnaire-based survey by multi-stage clustering sampling in 10 National Demonstration Areas between November and December,2016.Results There were 3 891 effective questionnaires.The self-reported prevalence of hypertension among aged ≥35 years was 31.47% (1 011/3 213).For the past two weeks,the self-reported treatment of hypertension was 86.75%(877/1 011),with the rates of guidance as 56.87% (575/1 011) on physical activity,40.95% (414/1 011) on diet,38.33% (385/1 011) on weight management,and 22.75% (228/1 011) on smoking cessation.For the past 12 months,74.68% (755/1 011) of the residents aged ≥35 years were under the proper management and 62.12% (628/1 011) of them were under the standardized management programs.The follow-up program lasted for 4 (P2s-P75:4-12) times per year,with 15 (P25-P75:10-20)minutes per each visit.Hypertensive patients would mainly visit the outpatient clinics (53.51%),followed by home visits (22.91%) and telephone calls (13.64%).Rate of satisfaction on management services was 94.83% (716/755) from the hypertensive patients.Multivariate analysis showed that the rate of self-reported treatment (OR=1.986,95% CI:1.222-3.228) and self-reported standardized management (OR=2.204,95%CI:1.519-3.199) on hypertension were higher in the Demonstration Areas with higher implementation scores of self-reported non-communicable diseases management.Conclusions Prevention and management on hypertension in the Demonstration Areas had met the requirement set for the Demonstration Areas during the "12th Five-Year Plan".Projects on setting up the National Non-communicable Diseases Demonstration Areas had played an active role in promoting the standardized management program on hypertension.

ABSTRACT

Objective To assess the implementation and impact of programs carried out by the National Demonstration Areas for Comprehensive Prevention and Control of Non-communicable Diseases.Methods Both sociological and epidemiological methods were used to collect qualitative and quantitative data in November and December,2016 in order to conduct on process and outcome evaluation of the above mentioned objective.In the meantime,case study was also conducted.Results All the National Demonstration Areas for Comprehensive Prevention and Control of Non-communicable Diseases were found well implemented across the country,with health education and health promotion,surveillance and safeguard measures in particular.A government-led and inter-sector coordination and communication mechanism had been well established,with more than 16 non-health departments actively involved.28.7% of the residents living in the National Demonstration Areas for Comprehensive Prevention and Control of Non-communicable Diseases were aware of the key messages related to chronic diseases.Among the residents,72.1% of them consumed vegetables and 53.6% consumed fruits daily,with another 86.9% walked at least 10 minutes per day.Over 70% of the patients with hypertension or diabetes reported that they were taken care of by the Community Health Centers,and above 50% of them were under standardized management.Residents,living in the National Demonstration Areas under higher ranking of implementation scores,were more likely to be aware of relevant knowledge on chronic disease control and prevention (OR=6.591,95%CI:5.188-8.373),salt reduction (OR=1.352,95%CI:1.151-1.589),oil reduction (OR=1.477,95%CI:1.249-1.746) and recommendation on physical activities (OR =1.975,95% CI:1.623-2.403).Conclusion The implementation of programs carried out by the National Demonstration Areas for Comprehensive Prevention and Control of Non-communicable Diseases had served a local platform for the control and prevention of non-communicable diseases,and thus become an important'carrier'for chronic disease prevention and control programs in China.

ABSTRACT

Objective To explore the mechanical properties of erythrocytes in the patients with hypertension.Methods Venous blood of 32 patients with hypertension were collected and divided into three groups of H1,H2,and H3 (degree Ⅰ,Ⅱ,Ⅲ),with 8 cases of healthy adults into a control group.Erythrocytes were isolated,the diameter,height and elastic modulus were detected by the atomic force microscope (AFM).Statistical analysis was carried out.Results The RDW-CV of H2,H3 were much larger than the control group (P<0.05,P<0.01);The height of erythrocyte in H2,H3 groups were lower than that of the control group (P<0.05).The elastic modulus of H 1,H2 groups were larger compared with normal erythrocytes (P<0.05).Furthermore,erythrocytes elastic modulus correlated with RDW-CV (R2=0.629).Conclusion Hypertension could affect the elasticity modulus of erythrocytes.AFM could be an effective tool in measuring the mechanical characteristics of erythrocytes at single cell level.This study investigates the relationship between hypertension and the structure and function of erythrocytes from a biomechanical aspect.

ABSTRACT

Objective To investigate the curative effects of damage control surgery in treatment of borderline severe multiple fractures.Methods The study involved 37 patients with severe multiple fractures(ISS of 16-42,Pape classification of borderline type)surgically treated from January 2009 to June 2010.All patients underwent staged surgeries according to damage control principles:hemostasis,debridement and provisional fixation were given in the first stage;antishock,anti-coagulation dysfunction,anti-hypothermy,anti-infection were performed in the second stage;definitive operations for fractures were performed as soon as possible in the third stage,provided that their physiological condition was permitted.Results All the patients received effective treatments such as debridement,provisional fixation and antishock in the first and second stages,with no iatrogenic secondary injury.The patients had significant recovery in the 6-14 months of follow-up,with total disability rate of 16%.Conclusion The staged operations according to damage control principles are effective and safe in treatment of borderline severe multiple fractures.

ABSTRACT

Objective To find out the status of prevention and control of iodine deficiency disorders and evaluate the iodine nutritional status of Jinan residents,to explore appropriate iodine level in drinking water,and to provide a scientific basis for adjustment of intervention strategies.MethodsAccording to the Monitoring Program of the National Iodine Deficiency Disorders (Trial),qualified iodized salt consumption rate,drinking water iodine content and urinary iodine levels of women of childbearing age were determined in iodine deficiency areas from 2003 to 2010.Salt iodine was detected by direct titrimetry,urinary iodine by As-Ce catalytic spectrophotometric assay and iodine in drinking water by cerous sulfate catalytic spectrophotometric method.Results Intake rate of qualified iodized salt was up to 90% and above from 2003 to 2010,median water iodine was 13.65 μg/L in the 10 counties(cities,districts),of which less than 100 μg/L accounted for 79.82%(4560/5713 ) and > 150 μg/L accounted for 12.73%(727/5713).With the increase of water iodine(0 ~ < 10,10 ~ < 50,50 ~ < 100,100 ~ < 150,150 ~ < 300 and ≥300 μg/L),urinary iodine levels of women of childbearing age increased successively(median 156.56,175.81,267.04,349.00,524.22,583.20 μg/L,respectively,x2 =121.20,P < 0.05),while the ratio of urinary iodine < 100 μg/L was significantly lower.The ratio of urinary iodine between 100 and 300 μg/L was decreased gradually,but the ratio of great than 300 μg/L was gradually increased.ConclusionsIodine deficiency areas in Jinan have reached the standard of elimination of iodine deficiency disorders.We should insist to carry out our measures to suit local conditions,classified guidances and scientific principals of iodine supplementation.

ABSTRACT

<p><b>OBJECTIVE</b>To apply the Wright-Giemsa stain in micronucleus test and to explore the stain outcomes of Wright-Giemsa dye of various proportions and staining times.</p><p><b>METHODS</b>Use Wright-Giemsa dye, Wright dye (staining time 3 min) and Giemsa dye (staining time 5 min) to stain HepG2 and then observe the staining effect. The Wright-Giemsa dye was applied under 5 different proportions (3:1-1:3) and different staining times (1, 3, 5, 10, 15 min).</p><p><b>RESULTS</b>After stained for 3-5 min with the proportion ratio of 3:1 of Wright-Giemsa dye, the HepG2 cells showed much better staining outcomes compared with the single stain of either Wright or Giemsa.</p><p><b>CONCLUSIONS</b>Wright-Giemsa stain can be used in cell micronucleus test to obtain good staining outcomes.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the epidemic characteristics of bacterial dysentery in Jinan municipality, and to provide scientific basis for effective strategy for bacterial dysentery control.</p><p><b>METHODS</b>The epidemiological characteristics of bacillary dysentery in Jinan from 1951 to 2005 were analyzed. A total of 485,333 cases in the span of 50 years were recorded, while the population-based case distribution was less than the total cases due to the data incompleteness during the Cultural Revolution.</p><p><b>RESULTS</b>The incidence of bacillary dysentery in Jinan has been decreasing by years with average incidence rate of 283.10/100,000. The significant differences were observed among the incidence rates of various ages(chi2 = 14.99, P < 0.05). There were four epidemic peaks, and all the incidence rates were about 1000/100,000. Age of onset mainly concentrated in the 0-4 years old, 20-years old and 30-years old. In terms of occupational distribution, workers accounted for 30.31%, the living-scattered children accounted for 22.71%, and the farmers accounted for 17.90%. The incidence focus was from July to September, which accounted for 71.57%. The peak of incidence emerged in August. The highest incidence in urban was 550.94/100,000.</p><p><b>CONCLUSION</b>Through the efforts of several generations of health workers, the incidence of bacillary dysentery in Jinan has been basically brought under control. Further step should be taken for the control of bacterial dysentery in urban areas and the management of bacterial dysentery in rural areas. Moreover, the biological characteristics of F2a should be a focus for the future study.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the efficiency of a peptide nucleic acid (PNA) delivery system by using liposome via PNA-DNA hybrids and to test the inhibitive action of antisense PNA on expression of multidrug resistance (MDR) related P-glycoprotein (P-gp) in human neuroblastoma cell line SK-N-SH.</p><p><b>METHODS</b>Two antisense PNAs were designed targeting at MDR-1 mRNA and then combined with partially complement DNAs respectively. The hybrids were delivered into cells using cationic liposome. The transfection efficiency, expression of P-gp and MDR-1 mRNA, intracellular adarimycin (ADM) were measured by flow cytometry, reverse transcription-polymerase chain reaction (RT-PCR), and high performance liquid chromatography (HPLC).</p><p><b>RESULTS</b>Transfection of PNA increased the cell average fluorescence intensity significantly and the extent of increase was dependent on the concentration of PNA. After being transfected by both PNAs, P-gp expression of SK-N-SH cells decreased significantly and the intracellular ADM level was increased by about 3 times. The level of MDR-1 mRNA expression slightly decreased after transfection, but no statistical significance was observed.</p><p><b>CONCLUSIONS</b>PNA can be delivered into tumor cells in form of PNA-DNA hybrids by cationic liposome. Properly designed antisense PNA can inhibit MDR related P-gp expression of SK-N-SH cells efficiently and specifically.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To construct human-SCID chimeric mice through implantation of mononuclear cells from human cord blood and study the immunoreaction of SCID-Hu IC mice immunized with rAd5HPV16L1-E7 vaccine.</p><p><b>METHODS</b>(1) Experiment groups were injected with the suspension of mononuclear cells from human cord blood through a tail vein; the control ones were injected with non serum RPMI 1640 medium. Eight weeks after implantation, blood was collected and human serum IgG level in the mice were tested, and human CD45, CD3 and CD19 were determined. (2) SCID-Hu IC mice were divided into two groups: in group A the mice were immunized intraperitoneally with rAd5HPV16L1-E7 virus and in group B the mice were immunized through nasal drip with rAd5HPV16L1-E7 virus. At the end of fourth week, the serum specific IgG antibody to rAd5HPV16L1-E7 virus, IFN-gamma in culture medium of spleen lymphocyte and T-lymphocyte propagation were tested.</p><p><b>RESULTS</b>(1) In the experiment groups, the number of mice positive for human IgG was 10/15, the average values of CD45, CD3 and CD19 were (9.39+/-4.21), (3.25+/-3.99) and (1.69+/-0.75), respectively. In the control ones, the human IgG, CD45, CD3 and CD19 were negative. (2) The results in the experiment groups showed that the IFN-gamma and T-lymphocyte stimulated by HPV16 protein were higher than those in the non-stimulated group (P less than 0.05).</p><p><b>CONCLUSION</b>(1) The results indicated that the construction of human-SCID chimaera through the implantation of mononuclear cells from human cord blood into SCID mice was successful. They also indicated that the reconstructed SCID-Hu IC mice has the ability to produce immune response against rAd5HPV16L1-E7 recombinant virus.</p>
