ABSTRACT

<p><b>OBJECTIVE</b>To retrospectively compare combined endovascular intervension with traditional bypass operation for the treatment of Budd-Chiari syndrome (BCS).</p><p><b>METHODS</b>From July 1989 to June 2001, 49 patients undergoing surgery for BCS were studied. 32 operations were performed by traditional bypass (from superior mesenteric vein or inferior vena cava to right auricle), and 17 by combined endovascular operation.</p><p><b>RESULTS</b>The data demonstrated a high incidence of perioperative complications, longer hospital stay, and expensive cost in the former group than in the latter group (P < 0.01). The mid-term effects were significantly better in the latter than in the former (P < 0.05). Severe complications occurred in the bypass group included hepatoencephalopathy, obtinacy ascites, cardiac dysfunction, and embolization of vascular grafts.</p><p><b>CONCLUSIONS</b>Combined endovascular intervention and shunting are the treatment of choice for BCS, with different combination according to its clinical type. This approach is simple, safe, effective, and economic.</p>

ABSTRACT

<p><b>OBJECTIVE</b>Analysis of differential gene expression profiles by cDNA microarray in pancreatic carcinoma with or without lymphatic metastasis.</p><p><b>METHODS</b>cDNA microarray was prepared by spotting polymerase chain reaction (PCR) products of 4 000 human genes onto specially treated glass slides. The cDNA probes were prepared by labeling normal tissue mRNA and cancer tissue mRNA with Cy3-dUTP and Cy5-dUTP, separately through reverse transcription. The mixed probes were, then, hybridized to the cDNA microarray. The chips were scanned by ScanArray 3000 laser scanner (General Scanning, Inc) on two wavelengths. The acquired image was analyzed by ImaGene 3.0 software (BioDiscovery, Inc). The intensity of each spot on the two wavelengths represented the quantity of Cy3-dUTP and Cy5-dUTP, with Cy5 to Cy3 ratio computed on each.</p><p><b>RESULTS</b>Fifty-six genes (including 24 previously reported) exhibited differential expressions in 2 specimens of pancreatic carcinoma with lymphatic metastasis and 2 without.</p><p><b>CONCLUSION</b>cDNA microarray provides an promising approach to specific gene expressions of the presence of lymphatic metastasis in human pancreatic carcinoma.</p>

ABSTRACT

To study the effect of intraarterial bolus of recombinant human angiogenin (rhANG) on stimulating revascularization in a rabbit hindlimb ischemic model,the limb ischemic model was induced by ligation and excision of every branches of the femoral artery in 28 rabbits. On the 11th day after operation, various doses of rhANG (0,5,20 and 40 ? g) as a single bolus were administered intraarterialy via the catheters inserted into the bifurcation of the iliac artery. Revascularization and perfusion of the ischemic limb were compared in each animal postoperatively by using 99 Tc MAA perfusion scan, serial angiography and thigh muscle biopsy. Rabbits in the 20?g group and the 40?g group had significantly better revascularization and blood perfusion than the control group and the 5?g group.It is concluded that rhANG can promote blood purfusion, revascularization and recovery.

ABSTRACT

ObjectiveTo construct the eukaryotic coexpression plasmid CEA/IL-2, and to lay the foundation for further studying CEA nucleiotide vaccine , adjuvant and their effects of special antitumor immunity.MethodsThe eukaryotic expression plasmid (pcDNA3-CEA)containing the gene coding for CEA was obtained by RT-PCR and gene recombination techniques.Using enzymolysis,ligation and other techniques,an eukaryotic coexpression plasmid (pIRES-CEA/IL-2)containing two expression unites of CEA and IL-2 gene connected with internal ribosome site was constructed.ResultsThe coexpression plasmids were transformed into COS7 cells and expression of two proteins were demonstrated by ELISA, and flow cytometer and elecsy.CEA and IL-2 were (23.73?0.26)ng/ml,and(20.17?0.13)ng/ml respectively.ConclusionsThe eukaryotic expression plasmids pIRES-CEA/IL-2 could be successfully constructed and transformed into COS7 cells.Expression of two proteins were demonstrated with no difference on expression.

Low shear stress and high shear stress of BRV in GC control group were (20.32?5.42)mPa?s and(7.96?3.16)mPa?s, respectively;those in GC LMWH group were (11.42?5.03)mPa?s and (3.96?3.07)mPa?s,respectively after operatin. Low shear stress and high shear stress of BRV were (21.82?6.17)mPa?s and ( 8.62 ?3.48) mPa?s,respectively in PC control group;those in PC LMWH group were (13.11?5.17)mPa?s and (4.96?3.61)mPa?s. After operation,there were no hemorrhagic complications in GC and PC LMWH groups. Conclusions Low shear stress and high shear stress of BRV rise generally seeing in GC and PC patients after operation,and in PC patients are higer than those in GC patients. The use of Low molecular weight heparin could reduce the occurrence of thrmbosis.

ABSTRACT

Objective To develop a new way to prevent vascular anastomotic intimal hyperplasia, we applied surgical suture soaked in c-myc antisense oligodeoxynucleotide solution to perform vascular anastomosis in rabbits. Methods8/0 sutures (coated Vicryl) soaked in antisense, sense and mismatched c-myc oligomer solution respectively were applied to accomplish vascular anastomoses in rabbits with interposition of external jugular veins between common carotid arteries. Twenty-four animals were randomly divided into four groups: control group, antisense group, sense group and mismatched group. Four weeks later, angiography was made to scrutinize the patency of anastomosis and specimens were harvested for microscopy, the image was digitalized by computer video analysis system, the intimal thickness and area, medial thickness and area and their ratios were calculated and analyzed statistically among groups. Resultsall the anastomoses were normal and patent.
