ABSTRACT
A TLC-UV method for the determination of lignin in polysaccharides of cone of Pinus koraiensis was reported. Active lignin was obtained by acid hydrolysis,and developed on GF-254 TLC-Plate by mixed Solventof dich1oro methane-ethylether. UV absorbance of the eluate was determined at 23Onm. The concentration oflignin was linearly related to absorbance within the range of 2~70pg/ml. The recovery was 100. 2% ~ 101.4% and RSD was 6. 5 %.
ABSTRACT
Acid polysaccharides in pinecone were determined quantitatively by sulfuric acid-carba zole colorimetry and the influence of amount of sulfuric acid carbazole, reaction temper ature and time forcolor-reaction were investigated. The maximum abstrption was found to be at 520nm. The absorbance was a linear function of concentrations between 0 and 7 ?g/ml acid polysaccharides (r = 0.9999, n = 7). Analytical recovcry was 91.0%~99.8% . RDS was 1.3%~2.3% (n = 6 ). The minimum detectable concentration was 0.05?g/ml.
ABSTRACT
Quantitative determination of polysaccharides in Pine cones by phenol-sulfuric acid mehod was studied. and the effects of concentration of phenol-sulfuric acid and reaction temperature on color formation were investigated. It was found that the absorbance is linearly correlated to polysaceharide concentrations between 10~78?g/ml (r=0.9999). The analytical recovery was 99.8%, CV% was 1.2% and the minimal detectable concentration was 5?g/ml.
