ABSTRACT

Acute kidney injury (AKI) refers to the clinical syndrome of rapid loss of renal function caused by various causes.AKI increases the risk of developing chronic kidney disease (CKD) and the mortality of patients.The increase in the rate has caused a great economic burden on the family and society.The pathogenesis of mediating AKI is numerous,and it is currently believed that innate and adaptive immune-mediated inflammatory reactions are involved in the initiation,progression and repair stage of AKI.T lymphocytes play a key role in it.This article will review the progress of the role of T cells in AKI.

ABSTRACT

Systemic lupus erythematosus (SLE)is an autoimmune disease whose pathogenesis is extremely complicated.With the further research,the role of inflammasome in the pathogenesis of Lupus nephritis has also been gradually emphasized.Among them,the nucleotide-binding oligomerization domain-like receptors family pyrin domain containing 3 (NLRP3) inflammasome is the most exhaustive inflammasome.We summarize the related studies on the role of NLRP3 inflammasome in Lupus nephritis in recent years.We found that NLRP3 inflammasome not only plays an important role in the pathogenesis of Lupus nephritis,but also participates in the process of kidney injury by circulating immune cells and renal innate cells.Finally,we introduced two specific inhibitors of NLRP3 inflammasome,β-hydroxybutyrate and MCC950,which provided a new strategy for the treatment of Lupus nephritis.

ABSTRACT

Transcription factor EB(TFEB)is a member of the MiTF/TFE family and plays an important role in cell stress,metabolism,cancer and so on.There are relatively few studies on the role of TFEB in renal diseases.TFEB was initially found to be highly expressed in TFEB-fusion renal cell carcinoma and plays a key role in the development of re-nal cell carcinoma.Blocking the downstream signaling pathway activated by TFEB would be a promising treatment for TFEB-fusion renal cell carcinoma.On the contrary,the expression of TFEB in renal intrinsic cells is decreased in diabetic kidney disease,leading to a blockage in the autophagy-lysosome pathway.TFEB enhances the ability of cell stress and self-repair,and then delays the progress of diabetic kidney disease.In cystine nephropathy,TFEB expression is reduced in re-nal tubular epithelial cells and compensatory activation is insufficient as well.TFEB over-expression effectively eliminates intracellular cystine and repairs damaged lysosome,which is expected to alleviate or cure the Fanconi syndrome.In summa-ry,TFEB plays a key role in different kidney diseases,and targeted regulation of TFEB provides new hope for the treatment of kidney diseases.

ABSTRACT

The effectiveness,safety and economy of metformin on type 2 diabetes mellitus therapy are well recognized, which has been used as the first-line oral hypoglycemic agent in recent dec-ades. Apart from hypoglycemic effect, recent studies show that metformin can exert renal protection via the mechanisms of auto-phagy induction, anti-senescence, antioxidative stress, against endoplasmic reticulum stress,anti-inflammation, and anti-fibro-sis through AMPK dependent or independent pathway, which prompt its therapeutic potential in acute kidney injury and chron-ic kidney disease. The non-hypoglycemic nephroprotective effects as well as their underlying mechanisms of metformin are summarized in this review.

ABSTRACT

Objective:To verify the dose of static and dynamic intensity modulated radiotherapy (IMRT) through 2-dimension ionization chamber matrix after external electrically-driven multi-leaf collimator (MLC) was installed in KB1800 medical linear accelerator. Methods: 40 patients who need receive IMRT were divided into static IMRT group (20cases) and dynamic IMRT group (20cases) as the random number table. The ionization chamber was applied to implement scale dose, and solid water and 2-D ionization chamber matrix was applied to verify dose. Besides, the feasibility of static and dynamic IMRT were compared and researched.Results: The verification results of plane dosage of static IMRT plan and dynamic IMRT plan showed that the passing rates both of them were above 90.0%, and the verification of intensity modulated dosimetry of medical electric linear accelerator KB1800 with external electric MLC was consistent with standard.Conclusion:After the external electrically-driven MLC is installed on the accelerator, the dosage of IMRT can achieve the verification standard, and it can be applied to clinical treatment and can satisfy the requirement of clinical radiotherapy.

ABSTRACT

OBJECTIVE: To explore the characteristics of seasonal distribution of active systemic lupus erythematosus (SLE) and the influences of meteorological factors including temperature and humidity on active systemic lupus erythematosus. METHODS: The characteristics of seasonal distribution of active SLE and its correlation with meteorological factors were retrospectively analyzed in 640 patients living in the city of Zhanjiang, China and had active SLE between January 1997 and December 2006. RESULTS: In winter, when there are weaker ultraviolet (UV) rays, the ratio of patients with active SLE to total inpatients was 3.89 percento, which is significantly higher than in other seasons with stronger UV rays, including 2.17 percento in spring, 1.87 0 in summer and 2.12 0 in autumn. The number of patients with active SLE had significant negative correlation with mean temperature and was not significantly related to mean humidity. CONCLUSION: Active SLE has the characteristics of seasonal distribution and is associated with temperature. The mechanism remains to be further studied.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of bone morphogenetic protein-7 (BMP-7) on aristolchic acid induced renal tubular epithelial cell trans-differentiation to look for new therapeutic approach for aristolchic acid nephropathy (AAN).</p><p><b>METHODS</b>In vitro cultured human proximal renal tubular epithelial cell line HK-2 cells were treated with different concentrations of BMP-7 (75 ng/mL, 150 ng/mL and 300 ng/mL) after trans-differentiation of the cells was induced by AA (10 microg/mL). Levels of alpha-SMA mRNA and protein expressions were detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) and Western blotting respectively.</p><p><b>RESULTS</b>BMP-7 reversed the AA inducing alpha-SMA expressions in HK-2 cells in a dose-dependent manner.</p><p><b>CONCLUSION</b>BMP-7 can inhibit the trans-differentiation of human renal tubular epithelial cell induced by AA, thereby might be a new potential drug for AAN prevention and treatment.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effect of p38MAPK signaling pathway in interleukin-18-induced transdifferentiation in renal proximal tubular cells.</p><p><b>METHODS</b>Human proximal tubular epithelial cell line (HK-2 cells) was cultured in vitro. After preincubated with SB203580 (0, 5, 10, 20 micromol/L) for 30 minutes, cells were exposed to IL-18 (100 ng/ml) for 24, 48 and 72 hours respectively. The expressions of a-smooth actin (alpha-SMA) in cultured HK-2 cells were assessed by RT-PCR and ELISA.</p><p><b>RESULTS</b>IL-18-induced expressions of a-SMA mRNA and protein were inhibited obviously by a dose-dependent manner when HK-2 cells were incubated with SB203580 (0, 5, 10, 20 micromol/L) and IL-18 (100 ng/ml) for different time (P < 0.05).</p><p><b>CONCLUSION</b>IL-18-induced transdifferentiation of renal tubular epithelial cells (RTECs) is suppressed obviously by blocking p38MAPK signaling pathways. IL-18-induced transdifferentiation of RTECs is probably mediated, at least in part, through the activation of p38MAPK signaling pathways.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the pathological and clinical features of IgA nephropathy (IgAN) in west Guangdong province.</p><p><b>METHODS</b>The pathological type and clinical features of 120 patients with IgAN were retrospectively analyzed.</p><p><b>RESULTS</b>Mesangial proliferative glomerulonephritis and focal segmental glomerulosclerosis were the most frequent features of IgAN. IgM deposit could be found in half of the IgAN patients, especially in the IgAN patients with focal segmental glomerulosclerosis.</p><p><b>CONCLUSION</b>The incidence of IgAN may vary between different regions. Clinically, misdiagnosis of other renal diseases as IgAN may often occur. The nature and severity of glomerular immunoglobulin deposition can be related to the pathogenesis and progression of IgAN.</p>
