ABSTRACT

Objective:To study the alkaloids in Macleaya cordata. Methods:The compounds were isolated and purified by silica gel,recrystallization,and semi-preparative HPLC, and their strcuctures were confirmed by MS and NMR. Results: Nine alkaloids were obtained and identified as dihydrochelerythrine(1), 6-cyanodihydrosanguinarine(2), R-6-((R)-1-hydroxyethyl)dihydrochel-erythrine(3),8-demethyl-dihydrochelerythrine(4),cheilanthifoline(5),canadine(6),8-demethyl-chelerythrine(7),protopine(8) and allocryptopine(9). Conclusion:Compounds 4-6 are isolated from the genus of Macleaya for the first time.

ABSTRACT

Objective To establish HPLC characteristic chromatogram ofKanglao Qingfei Granules.Methods HPLC analysis of samples was performed on Kromasil C18 column (4.6 mm × 250 mm, 5μm), with acetonitrile-1% glacial acetic acid as the mobile phase of gradient elution (0-50 min, 5%→15% acetonitrile;50-70 min, 15%→25% acetonitrile;70-80 min, 25%→40% acetonitrile;80-90 min, 40%→65% acetonitrile, 90-120 min, 65%→95% acetonitrile);the volume flow rate was 1.0 mL/min;detection wavelength was set at 290 nm;column temperature was 30℃. Chromatographic peaks were identified by HPLC-MS/MS method.Results The similarity degrees of 10 batches of samples were all greater than 0.995, and 13 chromatographic peaks were determined as common characteristic peaks, of which 10 peaks were confirmed in the source attribution and 8 peaks were identified in chemical component.Conclusion The established HPLC characteristic chromatogram can be used for the quality control ofKanglao Qingfei Granules.

ABSTRACT

Objective To optimize the extraction process of Kanglao Qingfei granules. Methods Orthogonal test design was used to study the influence of soaking time, water content, extraction time on extraction technology with bergenin's extraction rate, total polysaccharides's extraction rate and extract yield as evaluation indexes. Results The optimum extraction process conditions were as follows:the raw materials were immersed in water for 1.0 h and extracted two times under reflux, and the ratio of water volume to raw materials weight was 10 and 8 (mL/g), and extraction time was 2.5 h and 2.0 h, respectively. Conclusion The optimized extraction process was simple and stable, which can provide foundation for industrial production of Kanglao Qingfei granules.

ABSTRACT

Objective To evaluate toxicity and safety of trans-resveratrol (t-RSV). Methods For assays of acute toxicity,genetic toxicity, and sub-chronic toxicity, Ames test, mice bone marrow erythrocyte micronucleus, and mice sperm abnormality were performed. Results In the acute oral toxicity tests, maximum tolerable dose (15 g/kg) in male and female Kunming mice showed no toxicological signs. For 90-d feeding of t-RSV at dosage range of 167-500 mg/(kg·d) in both male and female Sprague-Dawley rats, no noticeable toxicological effects were observed.Conclusion T-RSV has no acute toxicity and no genotoxicity, no harmful effects on the human body at the tested dosage range and thus resveratrol is safe for human consumption.

ABSTRACT

OBJECTIVE:To prepare compound alendronate sodium sustained-release tablet and to investigate its drug release profile in vitro. METHODS: Hydroxy propyl methylcellulose (HPMC), ethylcellulose (EC), and lactis anhydrous were used to prepare sustained-release tablets by wet granule compression technique with release rate in vitro as index. Orthogonal experiment was conducted to optimize the formula. The release rate of the tablets in vitro was investigated. RESULTS: The optimized formula was as follows: HPMC 80 mg, EC 20 mg, and lactis anhydrous 20 mg. 12 h drug release of the preparation was achieved, and the drug release behavior in vitro fitted Higuchi equation. CONCLUSION: The sustained-release tablet is reasonable in formula and satisfactory in slow release efficacy.

At present,medicinal plant extract industry in China develops rapidly.As a modern industry,it does not only use the traditional Chinese medicinal materials as raw materials in conception and content,but also use medicinal plant extract as a core,including worldwide medicinal plant extract.China is an important country of extract production in the world.The present status accompanied with the existed questions and induced factors,countermeasure for development,and suggestion for medicinal plant extract industry in China are discussed in this article.It lays the emphasis on present status of industry for medicinal plant extract which involves the production,application,technology,standard and policy of present status influenced on development stage of plant extract enterprise itself in domestic and international.The development countermeasure and suggestion are put forward from seven aspects.
