ABSTRACT

The domestic and international research progress on the regulation of gut microbiota by Traditional Chinese Medicine (TCM) ingredients and their impact on intestinal absorption and transportation were summarized, which provided assistance for subsequent clinical rational drug use targeting gut microbiota. Literature on the relationship between gut microbiota and intestinal absorption and transportation in recent years were reviewed and analyzed, and the mechanism of TCM ingredients regulating gut microbiota on drug absorption and transportation was elucidated. Research has found that TCM ingredients alter gut microbiota, thereby affecting intestinal barrier function and absorption of transport proteins, which is of great significance for rational clinical medication.

ABSTRACT

OBJECTIVE@#To explore the effect and possible mechanism of dimethyl fumarate (DMF) on T-cell acute lymphoblastic leukemia (T-ALL), and provide experimental and theoretical basis for the clinical treatment of T-ALL.@*METHODS@#Jurkat cells were treated with different concentrations of DMF for 24 hours, and then the proportion and absolute count of Ki67-positive Jurkat cells were analyzed by flow cytometry. Meanwhile, the protein levels of nuclear factor-erythroid 2-related factor 2 (Nrf2) and E3 ubiquitin ligase HACE1 in Jurkat cells treated with DMF for 24 hours were evaluated by Western blot. Nrf2 proteins were co-immunoprecipitated in Jurkat cells, and then HACE1 protein was assessed by Western blot. Plasmids of Flag-Nrf2 and different gradients of Flag-HACE1 were transfected into HEK293T cells, and the levels of Flag-Nrf2 were detected by Western blot after 48 hours.@*RESULTS@#DMF could significantly inhibit the proportion and absolute count of Ki67-positive Jurkat cells, and DMF inhibited the proliferation of Jurkat cells in a dose-dependent manner (r=0.9595, r=0.9054). DMF could significantly up-regulate the protein levels of Nrf2 and E3 ubiquitin ligase HACE1 in Jurkat cells (P<0.01, P<0.01). HACE1 physically interacted with Nrf2 in Jurkat cells. Overexpression of Flag-HACE1 significantly increased the protein level of Flag-Nrf2 in a dose-dependent manner (r=0.9771).@*CONCLUSION@#DMF inhibits the proliferation of T-cell acute lymphoblastic leukemia cell. The mechanism may be that, DMF significantly up-regulates the protein levels of Nrf2 and E3 ubiquitin ligase HACE1, and HACE1 interacts with Nrf2 and positively regulates Nrf2 protein level.

ABSTRACT

Corticosteroid switching can reverse abiraterone resistance in some patients with metastatic castration-resistant prostate cancer (mCRPC). Here, we investigated the potential biomarkers for predicting the efficacy of corticosteroid switching during treatment with abiraterone acetate (AA). We retrospectively analyzed 101 mCRPC patients receiving corticosteroid switching from West China Hospital and Sun Yat-Sen University Cancer Center between January 2016 and December 2018. All cases received AA plus prednisone as first-line therapy during mCRPC. Primary end points were biochemical progression-free survival (bPFS) and overall survival (OS). The risk groups were defined based on multivariate analysis. A total of 42 (41.6%) and 25 (24.8%) patients achieved 30% and 50% decline in prostate-specific antigen (PSA), respectively, after corticosteroid switching. The median bPFS and median OS on AA plus dexamethasone were 4.9 (95% confidence interval [CI]: 3.7-6.0) months and 18.8 (95% CI: 16.2-30.2) months, respectively. Aldo-keto reductase family 1 member C3 (AKR1C3) expression (hazard ratio [HR]: 2.15, 95% Cl: 1.22-3.80, P = 0.008) and baseline serum alkaline phosphatase (ALP; HR: 4.95, 95% Cl: 2.40-10.19, P < 0.001) were independent predictors of efficacy before corticosteroid switching in the multivariate analysis of bPFS. Only baseline serum ALP >160 IU l-1 (HR: 3.41, 95% Cl: 1.57-7.38, P = 0.002) together with PSA level at switch ≥50 ng ml-1 (HR: 2.59, 95% Cl: 1.22-5.47, P = 0.013) independently predicted poorer OS. Based on the predictive factors in multivariate analysis, we developed two risk stratification tools to select candidates for corticosteroid switching. Detection of serum ALP level, PSA level, and tissue AKR1C3 expression in mCRPC patients could help make clinical decisions for corticosteroid switching.

ABSTRACT

Objective Tostudy the influence of pipe structures on the mixing uniformity of airborne effluents from nuclear power plant chimneys. Methods We used the computational fluid dynamics (CFD) method to simulate the velocity distribution and gas mixing in long straight pipes (I type) with square section and circular section, 90° single-bend pipes (L type) with square section and circular section, and 90° double-bend pipes (S type and U type) with square section and circular section. Results For the long straight pipe, due to the lack of flow disturbance caused by structural changes, the mixing effect was not good; when the pipe section was circular, it might take mixing distance 20 times the hydraulic diameter to achieve the uniformity index required by the relevant standard; for the square pipe, the distance might be longer. In the single bend pipe with square section, the velocity uniformity was improved more greatly after the bend, and the tracer gas met the mixing uniformity at a shorter distance (11 times the hydraulic diameter), as compared with the single bend pipe with circular section. For the S-type double-bend pipe, the tracer gas appeared uniformly mixed after a distance 6 times the hydraulic diameter in the square pipe, and 7 times the hydraulic diameter in the circular pipe. For the U-type double-bend pipe, the gas in the square pipe also achieved uniform mixing ata shorter distance downstream, and the airflow showed greater disturbance when passing through the bend. Conclusion The CFD method can make an accurate prediction for the change patterns of gas mixing uniformity in pipes with different structures, and can partially replace physical experiments to study the factors affecting the mixing uniformity of airborne effluents from the chimney of nuclear power plants.

ABSTRACT

Outbreak of COVID-19 is ongoing all over the world. Spine trauma is one of the most common types of trauma and will probably be encountered during the fight against COVID-19 and resumption of work and production. Patients with unstable spine fractures or continuous deterioration of neurological function require emergency surgery. The COVID-19 epidemic has brought tremendous challenges to the diagnosis and treatment of such patients. To coordinate the diagnosis and treatment of infectious disease prevention and spine trauma so as to formulate a rigorous diagnosis and treatment plan and to reduce the disability and mortality of the disease, multidisciplinary collaboration is needed. This expert consensus is formulated in order to (1) prevent and control the epidemic, (2) diagnose and treat patients with spine trauma reasonably, and (3) reduce the risk of cross-infection between patients and medical personnel during the treatment.

ABSTRACT

Even in the era of novel targeted agents, switching to a second-line nonsteroidal antiandrogen (NSAA) is still widely used in treating metastatic castration-resistant prostate cancer (mCRPC), especially in undeveloped countries. However, whether prior treatment with a second-line NSAA would impact the efficacy of abiraterone acetate (Abi) remains uncertain. In the current study, 87 mCRPC patients treated with Abi were analyzed. Among them, 21 were treated with a second-line NSAA (from bicalutamide to flutamide) before receiving abiraterone, while the remaining 66 received Abi directly. Therapeutic efficacy of Abi was compared between those with and without prior second-line NSAA using Kaplan-Meier curves, log-rank test, and Cox regression models. The therapeutic efficacy of Abi was similar between those with or without the prior switching treatment of flutamide, in terms of either prostate-specific antigen progression-free survival (PSA-PFS, 5.5 vs 5.6 months, P = 0.967), radiographic progression-free survival (rPFS, 12.8 vs 13.4 months, P = 0.508), overall survival (OS, not reached vs 30.6 months, P = 0.606), or PSA-response rate (71.4% [15/21] vs 60.6% [40/66], P = 0.370). This is the first time that the impact of prior switching of treatment to a second-line NSAA on the efficacy of Abi in mCRPC patients has been addressed. Our data support that, use of prior sequential bicalutamide and flutamide does not seem to preclude response to abiraterone, although larger cohort studies and, ideally, a randomized controlled trial are needed. These findings will facilitate doctors' decision-making in the treatment of mCRPC patients, especially for those with previous experience of switching NSAA second-line treatments in the clinic.

ABSTRACT

Objective To investigate the feasibility and efficacy of combinational acetabular quadrilateral surface plates in treating elderly patients with acetabular fractures.Methods A retrospective case series study was conducted on the clinical data of eight elderly patients with acetabular fractures admitted between August 2016 and June 2017.There were five males and three females,with an average age of 64.2 years (range,60-68 years).According to the Letournel-Judet's classification,there were four patients with both column fractures,two T-shaped fractures,two anterior column and posterior hemitransverse fractures,among which three were combined with posterior wall fractures.Before operation,the artificial pelvis model was prepared by 3D printing for simulating the placement of combinational plates in the quadrilateral surface,and the plates were shaped for operation use.During operation,through the high iliac inguinal approach,the infrapectineal plate,pubo-ischiatic plate,and ilio-ischiatic plate were placed successively to fix anterior column," middle column",and posterior column,respectively.The pubo-ischiatic and ilio-ischiatic plates were combined with the infrapectineal plate.After fracture reduction,internal fixation was completed at one time.The incision length,operation time,and intraoperative blood loss were recorded.Postoperative fracture reduction was evaluated by Matta standard,and function of hip joint by modified Merle d'Aubigné and Postel scoring system.The complications were recorded.Results All the patients were followed up for 3 12 months (mean,6.8 months).The average incision length was 10 cm (range,8-12 cm).The average operation time was 130 min (range,100-180 min).The average blood loss was 600 ml (range,350-1 000 ml).According to the Matta standard,six patients got excellent result,one good,and one fair,with an excellent and good rate of 7/8.With respect to the hip joint function at the last follow up,the result was excellent in four patients,good in two,fair in one and bad in one,with an excellent and good rate of 6/8.No iatrogenic vascular injury,inguinal hernia or pulmonary embolism occurred during operation.No traumatic arthritis or avascular necrosis of femoral head occurred after operation.Conclusions The fixation from inside to outside by using combinational quadrilateral surface plates is in accordance with the biomechanical characteristics of hip joint,which enables better function recovery,especially for elderly patients with acetabular fractures involving quadrilateral surface.Combined with 3D printing technique for preoperative planning,it can reduce operation time,blood loss and postoperative complications.

Acid-sensing ion channels ( ASICs) are proton-gated channels expressed widely in the central nervous systems and pe-ripheral tissues, among which ASIC1a is a core part and plays an important role in many physiological and pathological proces-ses.As a key receptor for extracellular protons , ASIC1a is in-volved in a variety of pathophysiological processes involving tis-sue acidosis, such as pain, inflammation, seizures and multiple sclerosis.Autoimmune disease is a chronic inflammatory dis-ease , and the excessive activation of T , B cells leads to multiple tissue and organ damage when the body responds to autoantigen immune response . In recent years , studies have found that ASIC1a plays an important role in the development of various au-toimmune diseases.In this paper, the biological characteristics of ASIC1a are briefly reviewed , and the research progress of ASIC1a in the development and progression of autoimmune dis-eases is discussed .

<p><b>OBJECTIVE</b>To explore the role of eukaryotic translation elongation factor 1A1 (eEF1A1) in regulating the invasion and metastasis of hepatocellular carcinoma (HCC) cells and the possible mechanism.</p><p><b>METHODS</b>qRT-PCR and Western blotting were used to detect the mRNA and protein expression of eEF1A1 and NOB1 in different HCC cell lines and normal liver cells. The invasion and migration abilities of HCC cells with eEF1A1 knockdown or overexpression were examined using Transwell chamber assay and RTCA assay, and the changes in NOB1 mRNA and protein expressions in the cells were detected. The effects of increasing NOB1 expression in HCCLM3-sheEF1A1 cells and decreasing NOB1 expression in eEF1A1-overexpressing MHCC97h cells on eEF1A1 expression and cell invasion and migration abilities were analyzed using Western blotting, Transwell chamber assay and RTCA assay.</p><p><b>RESULTS</b>The expressions of eEF1A1 and NOB1 were significantly increased in positive correlation in HCC cells as compared with normal hepatocytes. Knockdown of eEF1A1 significantly decreased the invasion and migration of HCC cells and reduced the mRNA and protein expression of NOB1 ( < 0.01). Overexpression of eEF1A1 significantly enhanced invasion and migration of HCC cells and increased NOB1 mRNA and protein expressions ( < 0.01). Increasing NOB1 expression in HCCLM3-sheEF1A1 cells led to the restoration of NOB1 expression and cell invasion and migration abilities ( < 0.01), whereas decreasing NOB1 in MHCC97h-eEF1A1 cells resulted in inhibition of NOB1 expression and cell invasion and migration ( < 0.01).</p><p><b>CONCLUSIONS</b>eEF1A1 positively regulates the expression of NOB1 to promote the invasion and migration of HCC cells .</p>

ABSTRACT

Objective To explore the effect of enriched environment on neuroregeneration in subventricle zone(SVZ)of rats with vascu-lar dementia.Methods All rats were divided into sham group(n=5),vascular dementia group(VD group,n=12)and enriched environment group(EE group,n=12).Then,2-VO method was applied to make the vascular dementia model.The sham group and VD group received conventional breeding environment for 30 days,while EE group was subjected to enriched environment for 30 days.Immunohistochemistry was applied to detect the BrdU and Doublecortin(DCX)expression in SVZ,and DCX/Ki67,DCX/p-cAMP-response element binding pro-tein(CREB)expression in SVZ was assessed by immunofluorescence double-labeling technique.Results Compared with the sham group, the number of DCX+cells(t=2.989,P=0.026)and BrdU+cells(t=3.069,P=0.005)increased in VD group,and increased more in EE group (t=3.067, P=0.027; t=2.907, P=0.011). Besides, the number of the DCX/Ki67 (t=2.994, P=0.040) and DCX/p-CREB (t=4.707, P=0.009) cells was significantly higher in EE group than in VD group.Conclusion Enriched environment could up-regulate the neuroregeneration ca-pacity in SVZ of rats with vascular dementia through CREB signal pathway.

Objective To analyze the mRNA expression level of lymphoid enhance factor 1 (LEF-1), and to investigate its clinical significance in bone marrow mononuclear cells of patients with chronic myeloid leukemia chronic-phase (CML-CP) after initial diagnosis and chemotherapy, and to analyze its clinical significance. Methods The real-time fluorescence quantitative polymerase chain reaction was used to measure the expression level of LEF-1 gene in 38 CML-CP patients after initial diagnosis and chemotherapy and 20 persons without blood system diseases and neoplastic diseases as normal control. The difference of LEF-1 expression level between the patients and healthy control was compared, and the effect of imatinib on the main molecular response (MMR) was analyzed. Results The expression of LEF-1 mRNA in 38 newly diagnosed patients [0.00214 (0.00020 - 0.02120)] was significantly higher than that in normal controls [0.00101 (0.00009 - 0.00233)] (U= 163.0, P 0.05). The level of LEF-1 mRNA expression of non-MMR group was also higher than that of the normal control group (U= 14.0, P<0.01). The rate of acquiring MMR was significantly higher in high LEF-1 mRNA expression group [84.2 %(16/19)] than that in low expression group [47.4%(9/19)] (χ2=4.209, P<0.01). The time of acquiring MMR was significantly shorter in the high LEF-1 mRNA expression group [(10.0 ± 4.5) months] than that in the low expression group [(14.6 ± 3.8) months] (t= 2.705, P< 0.01). Conclusions LEF-1 may be involved in the occurrence and development of CML, and reflects the tumor burden. It may be one of the indicators to predict the efficacy of imatinib.

ABSTRACT

Objective: To put an end to events of sulfur content over-standard in Zhaotong Gastrodia elata Bl. f. glauca S. Chow (ZGE) and to improve the drying efficiency. Methods: Effects of steaming time and drying parameters on phenomenal and quality characteristics of ZGE were investigated. Drying process of ZGE adapt to modern process equipment was explored. Results: Optimal steaming time for second grade fresh ZGE was 4 min under 95℃. Suitable drying process parameters were as follows: ZGE was natural cooling at room temperature for 12 h after steamed. ZGE was drying at 30℃ for 1 d, and then the temperature was raised with the speed of 2℃/12 h to 40℃. Then with the speed of 5℃/12 h until to 60℃, all the work can be stopped until the ZGE was dried completely. It totally took about 6 d. ZGE appeared light brown and less deformation, and the fold was shallow, without hollow. The content of gastrodin was 0.93%, soluble extract content was 20.1%, and the content of total ash was 2.65%. All quality indexes were corresponded to the standard of Chinese Pharmacopoeia 2010. Conclusion: Drying process of ZGE was applied to modern processing equipment. It can not only guarantee the phenomenal but also the quality of ZGE. Meanwhile, it can improve the production efficiency and was suited to demonstrate and extend in Zhaotong region.

ABSTRACT

The physiological response and bioaccumulation of 2-year-old Panax notoginseng to cadmium stress was investigated under a hydroponic experiment with different cadmium concentrations (0, 2.5, 5, 10 μmol · L(-1)). Result showed that low concentration (2.5 μmol · L(-1)) of cadmium could stimulate the activities of SOD, POD, APX in P. notoginseng, while high concentration (10 μmol · L(-1)) treatment made activities of antioxidant enzyme descended obviously. But, no matter how high the concentration of cadmium was, the activities of CAT were inhibited. The Pn, Tr, Gs in P. notoginseng decreased gradually with the increase of cadmium concentration, however Ci showed a trend from rise to decline. The enrichment coefficients of different parts in P. notoginseng ranked in the order of hair root > root > rhizome > leaf > stem, and all enrichment coefficients decreased with the increase of concentration of cadmium treatments; while the cadmium content in different parts of P. notoginseng and the transport coefficients rose. To sum up, cadmium could affect antioxidant enzyme system and photosynthetic system of P. notoginseng; P. notoginseng had the ability of cadmium enrichment, so we should plant it in suitable place reduce for reducing the absorption of cadmium; and choose medicinal parts properly to lessen cadmium intake.

ABSTRACT

The physiological effects of Panax notoginseng seedlings under simulated drought stress by PEG 6000 on antioxidant enzymes, osmotic substances and root activities were studied. The results showed that the activity of POD and APX in roots and leaves kept rising with increasing processing concentration and time. However, on the one hand, at the same processing time, SOD in roots and leaves firstly increased and then decreased with the increase of processing concentration. On the other hand, at the same processing concentration, SOD kept rising with the extension of processing time. In addition, the activity of CAT in roots and leaves tended to increase with the increasing concentration at the same processing time, while it increased at first and then decreased with the extension of time at the same concentration. The activity of SOD and APX in stem did not change obviously, whereas CAT activity in stem increased with the increasing processing time and concentration. With the increase of processing concentration and the extension of processing time, the MDA, soluble protein, proline content and root activity in leaves and roots apparently rose. Moreover, fluorescence signal of H2O2 and NO in root tip enhanced as the processing concentration increased after treated for 1 d. In summary, P. notoginseng seedlings could deal with drought stress by means of adjusting the system of antioxidant enzyme, permeating stress substances and impeded stress signal substances. Thus, when the concentration of PEG 6000 was more than 5%, it would have harm on P. notoginseng seedlings.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the influence of Kudou Shencha decotion on INF-y, ICAM-1, MCP-1 levels of prostate tissue homogenate in immunity prostatitis model rats.</p><p><b>METHOD</b>Forty Wistar male rats were divided into 5 groups randomly: Kudou Shencha decotion group with high dosage and low dosage, Qianleitai group, the model control group and normal group. The rat model of chronic nonbacterial prostatitis was established by multiple hypodermical injection of the suspension of prostatic protein purification with Freund's completed adjuvant. The level of intercellular adhesion molecule (ICAM-1), interferon gamma (INF-gamma) and monocyte chemotactic protein-1 (MCP-1) were measured by enzyme linked immunosorbent assay (ELISA).</p><p><b>RESULT</b>The content of ICAM-1 and MCP-1 in the model group was higher than that of the normal group (P < 0.05), the content of ICAM-1 was obviously decreased in Kudou Shencha decotion group with high dosage (P <0.05), the contents of MCP-1 were all obviously decreased in Kudou Shencha decotion groups and Qianlietai group. Compared with the model group, the contents of INF-gamma in all treatment groups were decreased insignificantly.</p><p><b>CONCLUSION</b>Kudou Shencha decotion has the action of lowering the level of ICAM-1 and MCP-1, which may be one of the mechanisms of Kudou Shencha decotion in the therapy of chronic prostatitis.</p>

ABSTRACT

Anthocyanin from purple sweet potato (PSP) extracted by microwave baking (MB) and acidified electrolyzed water (AEW) exhibited antioxidant activity. After further purification by macroporous AB-8 resin, the color value of PSP anthocyanin (PSPA) reached 30.15 with a total flavonoid concentration of 932.5 mg/g. The purified extracts had more potent antioxidant activities than the crude extracts. After continuously administering the PSP extracts to 12-mo-old mice for 1 mo, the anti-aging index of the experimental group was not significantly different from that of 5-mo-old mice. To a certain degree, PSPA was also effective for controlling plasma glucose levels in male Streptozocin (STZ)-treated diabetic mice. In addition, the extracts inhibited Sarcoma S180 cell growth in ICR mice. Mice consuming the PSP extracts formed significantly fewer and smaller sarcomas than mice consuming the control diets. The highest inhibition rate was 69.03%. These results suggest that anthocyanin extracts from PSP not only exert strong antioxidant effects in vitro, but also had anti-aging, anti-hyperglycemic, and anti-tumor activities.

ABSTRACT

It is now established that CD4(+)CD25(+)regulatory T (Treg) cells expressing transcription factor FOXP3, a regulatory subpopulation of T cells, is indispensable for the maintenance of immunological self-tolerance and immune homeostasis. FOXP3 expression in Treg cells is specific and it is the key control factor for the development, activation and function of Treg cells. At present, CD4(+)FOXP3(+)T lymphocytes are often used to define Treg cells for scientific research. But recent studies show that human CD4(+)FOXP3(+)T cells are phenotypically and functionally heterogeneous, including suppressive and non suppressive T cells. The different functions of these cell subsets can be distinguished by phenotypic differences. This review discusses the recent research progress about phenotypic characteristics and functional heterogeneity of CD4(+)FOXP3(+)T cell subsets.

ABSTRACT

Objective To investigate selcnium(Se) levels of environment and human body in Gaomi City and Zichuan District of Shandong. Methods Lijiaying Township in Gaomi City of Weifang City, Zhaili Township and Longquan Township in Zichuan District of Zibo City were selected. Two farming soil samples at different spot, local wheat and corn, residents nail samples from 3 to 4 families were collected in each natural village in the investigated towns. The contents of Se were detected by 2,3-diamino naphthalene fluorescence method. Results Se level of the soil, wheat, corn, and nails in Lijiaying [(0.054 ± 0.019), (0.022 ± 0.009), (0.018 ± 0.007), (0.365 ± 0.108)mg/kg] was significantly lower than that in Zhaili [(0.425 ± 0.080), (0.130 ± 0.043), (0.098 ± 0.026), (0.751 ± 0.134)mg/kg] and Longquan[(0.487 ± 0.153), (0.112 ± 0.030), (0.097 ± 0.029), (0.735 ± 0.145)mg/kg;P < 0.01]. In Lijiaying, Se was deficient in soil, wheat, corn(< 0.200, < 0.025 mg/kg), above Se deficiency diagnosis and below Se-adequate level in the nail, while in Zhaili and Longquan, the Se level in the soil (0.425, 0.487 mg/kg), wheat(0.130, 0.112 mg/kg), corn (0.098, 0.097 mg/kg), nails (0.751, 0.735 mg/kg) was adequate (≥0.400 mg/kg). Conclusions The external environment is Se-deficient in Lijiaying, Se-adequate in Longquan and Zhaili. The selenium level in human body is consistent with the external environment.

ABSTRACT

<p><b>OBJECTIVE</b>To study the effect of Hewei Xiaopi Capsule (HXC) in treating patients with dyskinesis functional dyspepsia (FD).</p><p><b>METHODS</b>Sixty-three patients with FD were randomly assigned to two groups, 33 in the treated group and 30 in the control group, who were treated respectively with HXC and domperidone for 4 weeks to observe the clinical effect and changes in electrogastrogram (EGG) before and after treatment.</p><p><b>RESULTS</b>Symptoms were alleviated in both groups after treatment, 3 patients in the treated group were cured, the efficacy was judged as markedly effective in 11, effective in 14 and ineffective in 5, while in the control group, 1 cured, 8 markedly effective, 14 effective and 7 ineffective, difference between the two groups showed no statistical significance. EGG showed that in the treated group, the normal slow-wave percentage was 41.93 +/- 18.22 before treatment and 50.86 +/- 16.03 after treatment, showing significant difference (P < 0.05).</p><p><b>CONCLUSION</b>HXC is markedly effective in treating patients with FD.</p>
