ABSTRACT

Objective: To investigate the molecular mechanism of circZNF609 targeting miR-153 to regulate the proliferation and apoptosis of diffuse large B-cell lymphoma. Methods: Fifty cases of lymphoma tissue from patients with diffuse large B-cell lymphoma who were diagnosed and treated in the First Affiliated Hospital of Zhengzhou University from July 2018 to December 2019 were collected. Thirty cases of normal lymph node tissues that were confirmed to be reactive hyperplasia by pathological diagnosis during the same period were selected as controls. Real time quantitative polymerase chain reaction (PCR) was used to detect the expression of circZNF609 in diffuse large B-cell lymphoma tissues and control hyperplasia lymph nodes. Diffuse large B-cell lymphoma OCI-LY19 cells were divided into control group (blank control), si-con group (transfected with siRNA control), si-ZNF609 group (transfected with circZNF609 siRNA), and si-ZNF609+ Anti-NC group (co-transfected with circZNF609 siRNA and inhibitor control) and si-ZNF609+ Anti-miR-153 group (co-transfected with circZNF609 siRNA and miR-153 inhibitor). Cell counting kit-8 (CCK-8) was used to detected proliferation, flow cytometry was used to detect cell cycle and apoptosis. Western blot was used to detect the protein expressions of C-caspase-3, cyclin D1, p21. The luciferase reporter system was used to identifie the relationship between circZNF609 and miR-153. Results: The expression level of circZNF609 in diffuse large B-cell lymphoma tissue was (1.44±0.22), higher than (0.37±0.14) in the control tissues (P<0.001). The cell survival rate of the si-ZNF609 group was (51.74±6.39)%, lower than (100.00±10.23)% of the control group and the (99.64±11.67)% of the si-con group (P<0.001). The proportion of cells in the G(0)/G(1) phase was (63.25±4.11)%, higher than (48.62±4.32)% of the control group and (47.12±3.20)% of the si-con group (P<0.001), the apoptosis rate was (13.36±1.42)%, higher than (3.65±0.47)% of the control group and (3.84±0.62)% of the si-con group (P<0.05). The expression levels of C-caspase-3 and p21 protein were (0.85±0.09) and (0.90±0.08), higher than (0.38±0.04) and (0.65±0.07) in the control group and (0.39±0.05) and (0.66±0.05) in the si-con group (P<0.001). The expression level of cyclin D1 protein was (0.40±0.03), lower than (0.52±0.06) of the control group and (0.53±0.04) of the si-con group (all P<0.001). CircZNF609 and miR-153 are mutually targeted. The cell survival rate of the si-ZNF609+ Anti-miR-153 group was (169.92±13.25)%, higher than (100.00±9.68)% of the si-ZNF609+ Anti-NC group (P<0.001), the ratio of cells in G(0)/G(1) phase and apoptosis rate were (52.01±3.62)% and (8.20±0.87)%, respectively, lower than (64.51±5.17)% and (14.03±1.17)% in the si-ZNF609+ Anti-NC group (P<0.001). The protein expression levels of C-caspase-3 and p21 were (0.42±0.06) and (0.52±0.06), lower than (0.80±0.07) and (0.92±0.10) of the si-ZNF609+ Anti-NC group (P<0.001). The protein expression level of cyclin D1 was (0.68±0.07), higher than (0.39±0.04) in the si-ZNF609+ Anti-NC group (P<0.001). Conclusion: Down-regulation of circZNF609 inhibits the proliferation of diffuse large B-cell lymphoma OCI-LY19 cells and induces apoptosis by targeting miR-153.

ABSTRACT

Objective To investigate effects of xylo-oligosaccharides with chitooligosaccharides on alcohol-induced liver injury and immune function in mice. Methods Different doses of chitosan oligosaccharide (0.045 g/kg-0.26 g/kgB.W) and xylo-oligosaccaride (0.055 g/kg-0.32 g/kgB.W) were feed to the mice for 30 days. The mice live-injury model was induced by alcohol. MDA、 GSH、 TG level in liver and T lymphocyte proliferation of spleen cells induced by conA, the antibody-producing cells, and natural killer (NK) activity were detected. Results Compared with the live-injury model group, MDA level of low/middle dose group, TG level of middle dose group and pathological evaluation score of liver steatosis of high dose group in mice liver were decreased because of chitosan oligosaccharide and xylo-oligosaccaride feeding. Compared with the control group, the ability of T lymphocyte proliferation of mouse spleen induced by ConA and the antibody-producing cells were increased in mice of middle and high dose group. The differences had statistical significances (P<0.05) . Conclusion Under this experimental condition, xylo-oligosaccharides in combination with chitooligosaccharides could protect the mice from alcohol-induced liver injury and enhance immune function in spleen of normal mice synergistically.

ABSTRACT

Objective To investigate the clinical feasibility and value of transumbilical single-port laparoscopic cholecystectomy with intra-operative ultrasonic knife for directly handling the cystic artery. Methods A retrospective analysis was performed on the clinical data of 23 1 patients who underwent laparoscopic cholecystectomy with intraoperative ultrasonic knife for directly handling the cystic artery in our depart-ment from March 201 1 to December 2012.According to the required surgical approaches, the patients were divided into transumbilical single-port laparoscopic group (n=125,group A)and three-port laparoscopic group (n=106,group B).The two groups were compared in terms of operative time, intraoperative blood loss, postoperative analgesia, postoperative food intake, length of postoperative hospital stay, and postoperative complications,as well as the performance of the ultrasonic knife in directly handling the cystic artery.Comparison of con-tinuous data between groups was made by t test, while comparison of categorical data was made by chi-square test. Results Group A had a significantly longer mean operative time than group B (20.21 ±1.86 min vs 18.43 ±1.37 min,P0.05).Group A had sig-nificantly fewer patients who needed postoperative analgesia than group B (5 vs 21,P0.05).The mean length of postoperative hospital stay was (2.98 ±0.23)d in group A,versus (3.02 ±0.18)d in group B (P>0.05).No indwelling drainage tube was used after opera-tion in either group. There were no postoperative bleeding, bile duct injury, bile leakage, incisional wound infection, and other complica-tions in the two groups. The ultrasonic knife had reliable hemostatic effect when directly sealing the cystic artery, without causing postopera-tive secondary bleeding. All patients were followed up for 2-12 months (mean,6.5 months);they recovered well without incisional herni-a,and the umbilical scar was not obvious, with relatively good cosmetic results. Conclusion Transumbilical single -port laparoscopic cholecystectomy has a comparable clinical effect to three-port laparoscopic cholecystectomy, causing little trauma, postoperative pain, and scar, and it is suitable for patients with a high demand for incision appearance.Directly sealing the cystic artery with the ultrasonic knife can reduce bleeding due to careful separation of the cystic artery and shorten the operative time. Therefore,it is a safe, feasible surgical ap-proach and worthy of clinical application.

ABSTRACT

OBJECTIVE: To explore an efficient heterocyclic bioisostere as the C3 carboxylic group of antibacterial fluoroquinolones for further development of antitumor fluoroquinolones. METHODS: Using the s-triazole ring as an isosteric replacement of the C3 carboxylic group with another different heterocyclic ring, oxadiazole, as a modified group, new bis-(different azole) methylsulfide derivatives, 6-fluoro-7-(4-methyl-piperazin-1-yl)-1, 8-(2, 1-oxpropyl)-5-[5-(aryl-[1, 3, 4]-oxadiazol-2-methylsulfanyl)-4H-[1, 2, 4]-tri-azol-3yl]-quinolin-4(1H)-ones (6a-6j), were designed from ofloxacin (1). The in vitro antitumor activity of 6a-6j against three cancer cell lines was evaluated by MTT assay. RESULTS: Ten title compounds (6a-6j) ere synthesized and their structures were characterized by spectral data. They exhibited significantly higher in vitro antitumor potency than the parent compound ofloxacin. CONCLUSION: The heterocyclic ring, s-triazole, could be used as an efficient isostere of the C-3 carboxylic group for further development of antitumor fluoroquinolone candidates.

ABSTRACT

Treatment with the combination of Chinese herbs and cytotoxic chemotherapies showed a higher survival rate in clinical trials. In this report, the results demonstrated that the tanshinone II A, a key component of Salvia miltiorrhiza bunge, when it is combined with the cytotoxic drug cisplatin showed synergistic antitumor effects on human prostate cancer PC3 cells and LNCaP cells in vitro. Antiproliferative effects were detected with MTT assay. Cell cycle distribution and apoptosis were detected by flow cytometer. Protein expression was detected by Western blotting. The intracellular concentration of cisplatin was detected by high performance liquid chromatography. The results demonstrated that tanshinone II A significantly enhanced the antiproliferative effects of cisplatin on human prostate cancer PC3 cells and LNCaP cells with the increase of the intracellular concentration of cisplatin. These effects were correlated with cell cycle arrested at S phase and cell apoptosis. The apoptosis might be achieved through death receptor pathway and mitochondrial pathway. Furthermore, the Bcl-2 family members were also involved in this apoptotic process. Collectively, these results indicated that the combination of tanshinone II A and cisplatin had a better treatment effect in vitro not only on androgen-dependent LNCaP cells but also on androgen-independent PC3 cells.

ABSTRACT

This study is to investigate the antitumor activity of ophiopogonin B (OP-B). MTT assay, flow cytometric analysis, acridine orange staining, Lyso-Tracker Red staining and HeLa-GFP-LC3 transfect cells assay were used to detect the proliferation activity, apoptosis and autophagy of HeLa cells. The results showed that OP-B exerted potent antiproliferative activity on HeLa cells, the cell growth inhibition effect of OP-B was not due to apoptosis and OP-B could induce autophagy of HeLa cells. OP-B also induced the protein expression up-regulation of Beclin-1 and promoted LC3 I transformation LC3 II, which were representative proteins of autophagy. Furthermore, 3-MA, an inhibitor of autophagy, not only inhibited OP-B-mediated autophagy but also almost completely reversed the antiproliferative effect of OP-B, suggesting that the growth inhibition effect of OP-B was autophagy dependent. Western blotting demonstrated that OP-B inhibited the phosphorylation of Akt and its' downstream vital protein, such as mTOR and p70S6K. In addition, OP-B also induced the protein expression up-regulation of PTEN, which is a negative regulation protein for Akt/mTOR signaling pathway. However, OP-B did not affect the protein expression of total Akt. Collectively, the antitumor effects of OP-B were autophagy-dependent via repression Akt/mTOR signaling pathway. Therefore, OP-B is a prospective inhibitor of Akt/mTOR and may be used as an alternative compound to treat cervical carcinoma.

Objective To explore curative effects of high-dose prednisone used for children with immune thrombocytopenia (ITP).Methods The children with ITP were divided into prednisone group,γ-globulin group and methylprednisolone group.Platelet levels in peripheral blood were recorded after treatments within the 5th day and the 30th day,and the blood pressure levels of children were also recorded during treatments.Thex2 test was used to compare the effective rate of different treatments as well as the incidence rate of hypertension in these 3 groups. Results In new diagnosis children, there was no significant difference in the effective rate among the 3 groups in the 5th day(P > 0.05).The effective rate of prednisone group in the 30th day was significantly higher than γ-globulin group (P <0.05),and there was no significant difference than methylprednisolone group (P > 0.05).In persistence and chronic ITP children, the effective rate of prednisone group were both significantly higher than those in γ-globulin group in the 5th days and the 30th day (P <0.05),and were also no significantly higher than those in methylprednisolone group (P > 0.05).The incidence rate of hypertension was significantly lower in prednisone group than that in methylprednisolone group (P < 0.05).Conclusion High-dose prednisone oral treatment is safe, effective and worth using in children with ITP.

ABSTRACT

Chemical constituents in ethyl acetate and butanol fractions of ethanol extracts from Acorus tatarinowii were separated by column chromatography. Bufo skeletal muscle fatigue model was established to study the anti-fatigue activity of separated compounds. Five compounds were separated and identified by spectroscopic analysis as acoramone(1),cycloartenone(2),2,4,5-trimethoxyl-2'-butoxy-1,2-phenyl propandiol(3),5-hydroxymethyl furfural(4), and 5-butoxymethyl furfural(5). Compound 3 was a new compound, and compounds 2 and 5 were separated from this plant for the first time. Compound 4 exhibited a notable anti-fatigue activity.
