ABSTRACT

Objective:To explore the key targets and mechanism of Bielong Ruangan decoction in the treatment of liver cancer based on network pharmacology and molecular docking.Methods:Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) database, PubChem database and PharmMapper database were used to search and screen the chemical components and related targets of Bielong Ruangan decoction and the targets of liver cancer diseases. The network diagram of " Bielong Ruangan decoction-traditional Chinese medicine-active ingredient-predicted target-disease" was constructed; Protein-protein interaction (PPI) network were analyzed through String database; gene ontology (GO) enrichment analysis was performed through WebGestalt database; Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis was carried out through KEGG Orthology Based Annotation System (KOBAS) database; Molecular docking of the active components and core target proteins of Bielong Ruangan decoction was carried out by using PyMOL, Auto DockVina and other software.Results:Bielong Ruangan decoction had 67 active components, 154 liver cancer targets and 244 pathways. According to the analysis of network pharmacology, Bielong Ruangan decoction may play an anti-cancer role through key targets such as epidermal growth factor receptor (EGFR), mitogen activated protein kinase 1 (MAPK1), estrogen receptor 1 (ESR1), MAPK8, serine threonine protein kinase 1 (AKT1), MAPK14, cysteine protease 3 (CASP3), cyclin-dependent kinase 2 (CDK2), bone morphogenetic protein 2 (BMP2), aldose reductase (AKR1B1) and other key targets. KEGG enrichment analysis showed that the treatment of liver cancer by Bielong Ruangan decoction involved the regulation of vascular endothelial growth factor (VEGF) signaling pathway, tumor necrosis factor (TNF) signaling pathway, thyroid hormone signaling pathway, T cell receptor signaling pathway and other pathways. The results of molecular docking showed that the binding energy of all compounds to protein was less than -5.6 kcal/mol, indicating that each compound and each protein could bind well.Conclusions:Bielong Ruangan decoction participates in the treatment of liver cancer through " multi-component, multi-target and multi-channel" ways, and plays an anti-cancer role mainly by regulating the proliferation and invasion of tumor cells and tumor inflammatory microenvironment.

ABSTRACT

Objective To investigate the clinical effect of Bielong Ruangan decoction combined with transcatheter arterial chemoembolization (TACE) versus TACE alone in the treatment of hepatitis B virus (HBV)-related primary liver cancer.Methods The patients with HBV-related primary liver cancer who were hospitalized in Liver Research Center in The First Affiliated Hospital of Hunan University of Chinese Medicine from March 2014 to March 2016 were enrolled and randomly divided into study group and control group.The patients in the study group were treated with Bielong Ruangan decoction combined with TACE,and those in the control group were treated with TACE alone.The two groups were observed in terms of short-term therapeutic effect,traditional Chinese medicine (TCM) syndrome score,reduction in serum alpha-fetoprotein (AFP),incidence rate of post-TACE syndrome,and 1-year survival rate.The t-test was used for comparison of continuous data between groups;the chi-square test was used for comparison of categorical data between groups;the ridit analysis was used for comparison of ranked data between groups;the Kaplan-Meier method was used to calculate survival rate and plot survival curves;the log-rank test was used to compare survival rates between groups.Results There were significant differences between the study group and the control group in objective tumor response rate (77.4% vs 56.4%,x2 =9.898,P =0.020) and reductions in TCM syndrome score and serum AFP level (t =2.194,P =0.030).As for the incidence rate of embolism syndrome after TACE,the study group had significantly lower incidence rates of pyrexia,nausea,vomiting,and poor appetite than the control group (all P < 0.05).The study group had significantly higher quality of life and physical scores than the control group (t =-3.893 and-6.187,both P < 0.001).There was also a significant difference in 1-year survival rate between the study group and the control group [77.4% (48/62) vs 63.6% (35/55),x2 =4.228,P =0.040].Conclusion In patients with HBV-related primary liver cancer,Bielong Ruangan decoction combined with TACE can improve their short-term clinical outcome and quality of life.

ABSTRACT

This study was aimed to construct a clinical trial technological platform of traditional Chinese medicine (TCM) live disease in accordance with the international good clinical practice (GCP) standard. Through the con-struction of technological platforms such as clinical new medicine ethics committee of liver disease of TCM, ex-perimental methods, curative effect evaluation, data management, and etc., the clinical evaluation method and in-dex system of new drugs with the curative effect characteristics of liver disease of TCM were established. It was concluded that under the construction of relevant technological platform, an effect evaluation method and index system with TCM diagnosis and treatment characteristics were built . And a key clinical trial technological platform of new drugs of TCM liver disease was established to meet requirements of the international GCP standard.

phological changes in rat liver tissues. TLR4 and NF-κB expression in the liver tissues were measured by im-munohistochemistry . The results showed that compared with the normal group , the serum levels of transaminase ( ALT , AST ) and endotoxin of the model group were higher ( P < 0 . 01 ); and the degree of liver pathology injury was significantly increased; the TLR4 and NF-кB expression were increased (P < 0.01). Compared with the model group, the serum levels of transaminase (ALT, AST) and endotoxin of the experimental group were lower (P < 0.01), the degree of liver pathology injury was significantly lighter; the TLR4 and NF-кB expression were significantly lower (P < 0.01). It was concluded that the WYJDHY granules has a good role in the prevention and treatment of liver injury of rat model of hepatic failure IETM through the downregulation of liver expression of TLR4 and NF-кB in rat liver tissues , reducing serum levels of endotoxin , which may be one of the mecha-nisms on hepatic failure treatment .

OBJECTIVE@#To confirm the expression and distribution of aquaporin 1 in laryngeal carcinoma, and analyze the correlation to micro angiogenesis in the development of laryngeal carcinoma.@*METHOD@#The expression and distribution of aquaporin 1 were examined by immunohistochemical technique in laryngeal carcinoma. New vessels were labelled using CD105 monoclonal antibody. Systematic analysis had been done on IMVD and data of clinical pathology.@*RESULT@#Positive AQP1 cells and IMVD in primary carcinoma were higher than that in normal laryngeal mucosa. The expression of AQP1 was gradually increased with the decrease of differentiation grade of laryngeal carcinoma. The expression of AQP1 in lymphatic metastasis groups was higher than that of controls.@*CONCLUSION@#The expression of AQP1 in tumor cells and IMVD of primary laryngeal carcinoma are higher than normal. Not only is increased expression of AQP1 positive relevant to IMVD, but to lymphatic metastasis and malignant cell grading from laryngeal carcinoma. The increased AQP1 expression in epithelial cells of carcinoma and vascular endothelial cells may play a considerable role in growing, infiltrating, metastasis of malignant tumor by promoting vascular permeability.

OBJECTIVE@#To explore the expression of SleX in nasal epithelium in patients with chronic rhinosinusitis.@*METHOD@#Nine nasal epitheliums from patients with chronic rhinosinusitis and 7 normal controls were stained with SleX antibody by immunohistochemistry, and its expression were analyzed.@*RESULT@#Both nasal epithelial cells and neutrophils expressed SleX. Nearly 88.9% nasal epithelium from patients with chronic rhinosinusitis expressed SleX while 14.3% in normal control. Expression of SleX in nasal epitheliums from patients with chronic rhinosinusitis were higher than that in normal control. There were significant difference between two groups.@*CONCLUSION@#Nasal epithelium from patients with CRS is highly expressed SleX. It may involve the occurrence of chronic rhinosinusitis.

ABSTRACT

OBJECTIVE@#To investigate the mRNA level of IL-12, ICAM-1 and MCP-3 in human nasal epithelial cells.@*METHOD@#Firstly, human primary nasal epithelial cells were cultured, and then 4 pairs of primers were designed for detecting mRNA level of IL-12, ICAM-1 and MCP-3. The 938 bp PCR products of GAPDH were used as internal standards. The mRNA expression levels of IL-12, ICAM-1 and MCP-3 in primary nasal epithelial cells was measured with semi-quantitative reverse transcription-polymerase chain reaction.@*RESULT@#The round or irregular primary nasal epithelial cells were observed sticking to the bottom of cell culture plates under 400 times optical microscope. The expressions of IL-12 p35, ICAM-1 and MCP-3 mRNA were found in primary nasal epithelial cells while IL-12 p40 subunit was not detected.@*CONCLUSION@#IL-12 p35, ICAM-1 and MCP-3 mRNAs are expressed in primary nasal epithelial cells, whereas effective IL-12 with integrity is not present in nasal epithelial cells.

OBJECTIVE@#To analyze expression of TLR9 in human nasal epithelial cells by semi-quantitative reverse transcription-polymerase chain reaction, immunohistochemistry and flow cytometry.@*METHOD@#Human primary nasal epithelial cells (HNECs) were cultured, and then analyze the expression of TLR9 in primary cultured HNECs measured by semi-quantitative RT-PCR and immunohistochemistry and flow cytometry.@*RESULT@#Primary cultured HNECs were observed by 400 x optical microscopes. Round or irregular cells stick to the bottom of cell culture plates. RT-PCR showed that mRNA expressions of TLR9 were found in primary nasal epithelial cells by 1% agarose gel electrophoresis. Interestingly, by analyzing with gray level of GAPDH, expression of TLR9 mRNA in HNECs was higher than positive control PBMCs.@*CONCLUSION@#TLR9 is expressed by HNECs. Expression of TLR9 mRNA in nasal epithelial cells is higher than in PBMCs.

ABSTRACT

OBJECTIVE To investigate the expression of SleX and CD24 in nasal inverted papilloma and its pathologic features.METHODS HE staining were conducted to study the pathologic features in specimens of 11 cases with nasal inverted papilloma. Further,immunohistochemistry stain for SleX and CD24 were performed in total specimens.RESULTS One case(9.1%) was diagnosed as severe atypical hyperplasia but tumor cells did not invaded basal membranes.SleX staining located at cell membranes. Positive SleX staining was found in 9 specimens (81.8%) and 1 normal nasal epithelium (16.7%).CD24 staining located in cytoplasm.Positive CD24 staining was found in 8 specimens of nasal inverted papilloma (72.7%). CD24 was negative in nasal epithelial cells and only a few lymphocytes were positive.CONCLUSION Some cases of nasal inverted papilloma are diagnosed with severe atypical hyperplasia.Most of cases express CD24,so nasal inverted papilloma may be a borderline tumor.Expression of SleX and infiltration of inflammatory cells suggest that nasal inverted papilloma may be related to inflammatory reactions.

ABSTRACT

OBJECTIVE: To study the efficacy and safety of Fuzheng Huayu Capsule (FZHY Capsule) against liver fibrosis with chronic hepatitis B. METHODS: Multicentric, randomized, double blinded and paralleled control led trial was conducted on patients (aged between 18 and 65) with liver fibrosis in chronic hepatitis B Indexes observed: (1) hepatic histological changes and HBV markers were observed at 0 and 24th week during the treatment; serological indexes (HA, LN, P-III-P, IV-C) were determined and B ultrasound examination of spleen and liver was taken at 0, 12th, 24th week; liver function (during the period of follow-up, liver function and serological indexes for liver fibrosis were evaluated) were observed at 0, 6th, 12th, 18th, 24th week; (2) indexes for safety: blood and urine routine tests, renal function and ECG were examined. RESULTS: (1) Enrollment and demographic data: There was no significant difference between the trial (110 cases) and control group (106 cases) in demographic feature, vital signs, course of illness, history for drug anaphylaxis, history of previous therapy, liver function, serological indexes for liver fibrosis, liver histological examination (99 cases for test group, 96 cases for control group), HBV markers, and renal function, etc. (2) Histological pathological examination: 93 cases of liver histological examination were taken, of these 50 cases for the trial group and 43 cases for control group which turned out to be at S mean value of 2.33 and 2.11 respectively pretreatment according to criteria for liver fibrosis staging. Post-treatment, the trial showed a significant decrease with S value of 1.80 compared to that of pretreatment; however, there was no significant improvement in control group before and after the treatment with S mean value of 2.14. There was significant difference in reversing rate (decrease at least 1 stage according to criteria for liver fibrosis staging) between the trial (52%) and control (23.3%) after liver biopsy. The trial had a rather good effect on improving inflammatory activity and was superior to control group with a marked decrease of mean value of inflammatory activity and score of inflammation (P<0.05). (3)Serological indexes for liver fibrosis: There was a significant decrease in HA, LN, P-III-P, IV-C content in test group after 12 and 24 weeks' treatment compared to that of pretreatment; the differences of HA, LN, P-III-P, IV-C between 12, 24 weeks' treatment and pretreatment were significantly greater than control group (P<0.01 or 0.05); the effectual was defined as 2 of 4 indexes lowered more than 30% of the baseline, according to this criteria, the trial was 72.7%, while control group 27.4% (P<0.01). (4)Liver function: Obvious improvement of serum Alb, ALT, AST, GGT was seen in 2 groups; compared with control group, marked improvement of GGT and Alb in the trial (P<0.05); the effective rate of serum ALT in the trial group was 72.7%, while control 59.4%. (5)No changes of significant difference between pre- and post-treatment in routine tests for blood and urine, renal function and ECG, etc. There was also no difference in the stable rate of ALT and serological indexes for liver fibrosis between the trial and control group 12 weeks after withdrawal (P<0.05). CONCLUSION: Fuzheng Huayu Capsule has good effect on alleviating liver fibrosis in chronic hepatitis B without any adverse effect and is superior to Heluo Shugan Capsule. Fuzheng Huayu Capsule is a safe and effective medicine for the treatment of liver fibrosis in chronic hepatitis B.

ABSTRACT

Objective To analyze the characteristics of the Chinese Medical Equipment Journal's articles and authors as well as their spatial & temporal distribution,and then put forward some advices for the development of the journal.Methods 4 434 articles of the journal from 2002 to 2008 were sampled for bibliometrics analysis from the aspects of column,author,region,organization,publishing delay and foundation support.Results With an abundant manuscript source mostly from the developed areas such as Beijing,Guangdong Province,Shandong Province and Tianjin,the journal set its columns reasonably and flexibly,and thus gifted with a short publishing delay.The coauthor degree and cooperation rate were less than the journals in other fields,and the number of foundation-supported articles,also less than others,but was increasing yearly.Conclusion Chinese Medical Equipment Journal,with a high-quality manuscript source and great influence in the field,can be considered as a platform for communication and academic achievements publishing.
