ABSTRACT

Objective@#To investigate the brain function and their correlation with sleep beliefs and attitudes in adolescents with drug naive insomnia by using fractional amplitude of low frequency fluctuation, so as to provide a reference for the mechanism and treatment of insomnia.@*Methods@#An insomnia group ( n =21) recruited first episode, drug naive, adolescents with insomnia who met the diagnostic criteria of the American Diagnostic and Statistical Manual of Mental Disorders(DSM-V). Healthy subjects matched with age, gender, and educational background were selected as the control group ( n =20). Pittsburgh Sleep Quality Index Scale (PSQI), Brief Version of Dysfunctional Beliefs and Attitudes about Sleep (DBAS-16), 24 Items Hamilton Depression Scale (HAMD-24), 14 Items Hamilton Anxiety Scale (HAMA-14) were evaluated. Fractional amplitude of low frequency fluctuation was used for analysis, and Pearson correlation analysis was employed to quantify the correlation between peak values of brain regions with significant differences and the clinical scale scores of the two groups.@*Results@#Compared with the control group, ractional amplitude of low frequency fluctuation(fALFF) values in the insomnia group were significantly decreased ( P <0.01, Alphaism corrected) in the left dorsolateral prefrontal lobe (L-DLPFC, MNI coordinates: -12, 60, 21, t =-3.85, K =495) and the left precuneus (MNI coordinates: -3, -54, 51, t =-4.29, K =417). The fALFF value of L-DLPFC in the insomnia group was positively correlated with DBAS-16 score ( r= 0.47 , P = 0.04 ).@*Conclusion@#Abnormalities in the L-DLPFC region suggest that adolescents with insomnia may suffer from impaired regulation of emotional and cognitive activities related to sleep.

ABSTRACT

Objective:To explore the status quo of alexithymia, ruminant thinking and depression in infertility patients and their relationship. Methods:A cross-sectional survey was conducted among infertile patients who visited the reproductive center of the First Affiliated Hospital of Nanjing Medical University from September to December 2018. The data were analyzed by descriptive analysis, t-test, variance analysis and path analysis. Results:There was a moderate positive correlation between depression score (13.17 ±8.66), alexithymia score (51.62±10.91) and ruminant thinking score (18.28±10.65) in infertile patients ( r values were 0.442, 0.551, 0.569, all P < 0.05). Alexithymia played a mediating role between ruminant thinking and depression in infertile patients, and its indirect effect was 0.026 4, accounting for the total effect 24.81%. Conclusions:The depression status of infertility patients is not optimistic. Ruminant thinking has a great impact on infertility patients through alexithymia. The inner feelings of infertility patients can not be effectively expressed. It is easy to form negative emotional stress, which leads to depression. Medical staff and family members should pay attention to the ruminant thinking of patients' inner thoughts and timely dumping. Hearing and counseling can promote the physical and mental recovery of patients.

ABSTRACT

Objective@#To describe the molecular epidemiological characteristics of norovirus gastroenteritis outbreaks in Zhuhai from 2011 to 2016.@*Methods@#Anal swab specimens were collected from 576 cases with 56 outbreaks of acute norovirus gastroenteritis from 2011 to 2016. Specimens were tested by real-time RT-PCR. Three to four of norovirus positive specimens were selected from every outbreak to amplify the VP1 gene by RT-PCR and one strain was chosen randomly from every outbreaks to determine the genotype by phylogenetic tree analysis.@*Results@#Eight genotypes were identified from 56 outbreaks and all of them belonged to GⅡ genogroup. The genotype of norovirus strain changed with prevalence time. The GⅡ.4/2006b was dominant from 2011 to 2012, and replaced by GⅡ.4/Sydney _2012 during the 2012—2013 norovirus season, and both of them never appeared after Feb. 2013. GⅡ.17 was the only genotype during the 2014—2015 norovirus season. All the 7 outbreaks occurred from 2015 to 2016 were caused by GⅡ.3 norovirus. The GⅡ.17and GⅡ.3 were identified from Apr. to Sep. 2016; GⅡ.p16-GⅡ.2 were the only genotype in 12 outbreaks from Nov. to Dec. 2016. The GⅠ genogrope was never identified from 2011 to 2016 in Zhuhai.@*Conclusions@#The Norovirus GⅡ was the only pathogeny which caused the outbreaks of norovirus gastroenteritis. The recombinant norovirus strain GⅡ.p16-GⅡ.2 emerged and caused large outbreaks in the last two months of 2016 in Zhuhai; several recombinant strains of the GⅡ.p16 RdRp gene were found now, which suggests that attention should be focused on the prevalence and evolution of the recombinant norovirus.

ABSTRACT

Objective To explore the effect of BPDE on the expression of N-Ras in the human bronchial epithelial cell line. Methods The levels of mRNA and protein expression in BPDE transformed 16HBE cells(16HBE-T) and untransformed control 16HBE cells(16HBE-N) were examined by using RT-PCR and Western blot. Locations and expression levels of protein in both kinds of cells were analyzed by immunocytochemical method. Results Compared with 16HBE-N, the levels of mRNA and protein of N-Ras significantly increased to 3.616 and 1.600 times in 16HBE-T. Immunocytochemical method showed N-Ras protein in 16HBE-T and 16HBE-N expressed in the cytomembrane and cytoplasm, but the expression level of protein in 16HBE-T was significantly higher than that in 16HBE-N. Conclusion The up-regulated expression of N-Ras oncogene may play an important role in the malignant transformation of 16HBE induced by BPDE

ABSTRACT

OBJECTIVE: To study the antidepressant effects of total flavone of A( TFA) on post stroke depression ( PSD) in rats. METHODS: Middle cerebral artery occlusion ( MCAO) followed by isolation feed and irritable stimuli was used to induce PSD. Behaviours of rats were detected by open- field test . The effects of TFA on the indexes of hemorheology and brain homogenate lipid peroxidation were also tested. RESULTS: TFA could increase the crossing and rearing scores in open- field test, inhibit the elevation of whole blood viscosity as well as plasma viscosity at high- middle- low shear rates, increase the erythrocyte deformation, enhance the activities of Superoxide Dismutase ( SOD) and Glutathione Peroxidase ( GSH- Px) , and reduce Malondialdehyde ( MDA) content in brain homogenate in PSD in rats. CONCLUSIONS: TFA produces an antidepressant- like effect in PSD rats. This mechanism may be in association with attenuating hemorheological parameters and decreasing lipid peroxidative damage.

ABSTRACT

OBJECTIVE:To study the effect of the extracts of Paederia scandens (EPS) on acute gouty arthritis induced by monosodium urate in rats. METHODS: Monosodium urate crystals were used to establish gouty arthritis model in rats, with ankle joints' swelling degree at different time after modeling measured using paw volume measuring meter and gaits of rats scored at 24h after modeling. Pathological changes of articular tissues of rats were observed with HE staining and TNF-? and IL-1? levels were monitored by Radioimmunoassay. RESULTS:EPS could significantly inhibit joint swelling of rats and improve the gaits; histopathologically, EPS could abate tissue swelling of ankle joints, reduce inflammatory cell infiltration of articlar tissues and improve the pathological changes of synovial membranes. Radiommunoassay revealed that EPS could reduce levels of TNF-? and IL-1? in rats' synovial membranes. CONCLUSION:EPS have remarkable efficacy in improving symptoms of acute gouty arthritis induced by monosodium urate crystals in rats, which might be related to its inhibition on levels of IL-1? and TNF-?.

ABSTRACT

Objective To establish a system for rapidly detecting single nucleotide polymorphisms (SNPs) in mitochondrial DNA (mtDNA) using hybridization probes and melting temperature (Tm) curve analysis. This technique is suitable for population-based studies on the interaction between genetic factors and environmental exposures and the risk of Parkinson's disease (PD). Methods mtDNA was extracted from the blood. Rapid polymerase chain reaction (PCR) and melting curve analyses were performed with primers and fluorochrome-labeled probes on a LightCycler. Genotyping of 10 SNPs was based on the analysis of allele-specific Tm of detection probes. The results of melting curve analyses were verified by sequencing all 150 PCR products. Results Real-time monitoring showed optimal PCR amplification of each mtDNA fragment. The changes at nucleotide positions 1719, 4580, 7028, 8251, 9055, 10398, 12308, 13368, 13708, 16391 from wild-type to mutant genotype resulted in 6.51, 8.29, 3.26, 7.82, 4.79, 2.84, 2.73, 9.04, 8.53, 9.52℃ decline in Tm of the detection probes respectively. Genotyping of all detected genes was verified by 100% correspondence with the results of sequencing. Conclusion A rapid and reliable detection system for identifying mitochondrial polymorphisms and haplotypes has ben developed based on hybridization probe technology. This method may be suitable for mitochondrial genotyping of samples from large-scale epidemiology studies and may prove useful for exploring the molecular etiopathogenesis of PD, identifying markers of genetic susceptibility, and protecting susceptible individuals from PD.

ABSTRACT

Aim To study the antidepressant effects of bioactive compounds from entomogenous fungi(BCEF) in mouse models of depression. Methods The antidepressant effects of Bioactive compounds from entomogenous fungi was examined on the chronic unpredictable stress test, yohimbine induced lethality test, and 5-HTP induced head-twitches test. Results BCEF(50 mg?kg~-1 , ig, qd?21 d)could significantly increase the crossing and rearing score in open-field test. After administration yohimbine 1 h, BCEF 100 mg?kg~-1 group mice mortality rate rising rapidly; BCEF(50,100 mg?kg~-1 ) could distinctly increase the head-twitch number in the 5-HTP induced head-twitches test. Conclusion BCEF has antidepressant effects in depression mouse models. The mechanisms of its antidepressant effects may be related with the reinforcement of central monoamine neurotransmitter especially to 5-HT and NE nerves system.

ABSTRACT

AIM To study the antidepressant effects of bioactivecompounds from entomogenous fungi(BCEF) in mice models of depression. METHODS The antidepressant effects of Bioactive compounds from entomogenous fungi were examined in the learned helplessness model( forced swimming mice, tail suspension mice) and chronic unpredictable stress mice models. Spectrofluorometer and UV spectrophotometer were used to detect the activity of MAO, central monoamine neurotransmitter in mice brain mitochondria. RESULTS BCEF0083 25,50,100 mg?kg -1 could obviously shorten the immobility time in forced swimming mice, tail suspension mice and showed some extent of dose-effect relationship. BCEF0083 25,50,100 mg?kg -1 could obviously inhibite the activity of MAO-A,B on brain remitochondria in chronic unpredictable stress mice models and could rise the content of NE,5-HT,5-HIAA,DA in defferent degree. CONCLUSION The results suggested that BCEF0083 had antidepressant effects in mice depression models. The mechanisms of BCEF0083 antidepressant effects may be related with the inhibition of MAO-A,B activity and the increased content of central monoamine neurotransmitter.
