ABSTRACT

As an important method of systems biology, metabolomics not only plays an important role in life science but also has been increasingly widely used in radiation protection research. Based on the clinical studies of metabolomics and metabolomics methods in rodent and primate models, this article summarizes the methods and techniques of metabolomics in the diagnosis of radiation damage, the study of radiation damage mechanisms, and the development of radiation protection drugs.

ABSTRACT

Objective:To compare the dosimetric difference between volumetric modulated arc therapy (VMAT) plans with flattening filter (FF) and flattening filter-free (FFF) for whole-breast radiation therapy after breast conserving surgery for left breast cancer.Methods:Twenty patients with left breast cancer who underwent breast conserving surgery in Xuzhou Central Hospital from August 2017 to August 2018 were selected by random number table method, and the CT data were obtained. Both FF-VMAT plan and FFF-VMAT plan were designed with ECLIPSE 10.0.4 treatment planning system (TPS) of American Varian company. The prescription dose of the planned target volume (PTV) was 50 Gy (2 Gy/time), and the local tumor bed was irradiated with 10 Gy/5 times. The dosimetric parameters of the two plans were compared.Results:The two plans of all patients met the prescription dose requirements. Compared with FF-VMAT plan, FFF-VMAT plan had better conformity (conformal index: 0.87±0.04 vs. 0.77±0.05) and homogeneity (homogeneity index: 7.36±0.88 vs. 10.89±3.00) (both P < 0.01); FFF-VMAT plan had lower average dose of heart [(7.73±1.44) Gy vs. (9.16±4.24) Gy] and contralateral lung [(3.61±0.74) Gy vs. (8.42±0.45) Gy] (both P < 0.01), FFF-VMAT plan had higher average dose of contralateral breast [(3.92±1.08) Gy vs. (2.02±1.42) Gy] ( P < 0.01), and V 2 Gy in the heart [(79.27±1.18)% vs. (58.94±21.25)%] and V 5 Gy in the ipsilateral lung [(44.89±2.17)% vs.(37.86±8.33)%] (all P < 0.01). The monitor unit of the FF-VMAT plan was (973±89) MU, which was lower than that of the FFF-VMAT plan [(1 356±201) MU] ( P < 0.01). Conclusion:FFF-VMAT plan can effectively reduce the cardiac dose after breast conserving surgery for left breast cancer, but it increases the risk of organ damage at low dose area.

ABSTRACT

Objective:To investigate the effect of Ranae Oviductus (RO) on ovarian follicular development, phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt) signaling pathway, and pregnancy function in rats, and the estrogen-like mechanism of OR. Method:Seventy female Wistar rats were randomly divided into a normal group, a progynova+ luteohormone group (1 mg·kg<sup>-1</sup>+40 mg·kg<sup>-1</sup>), a clomiphene group (10 mg·kg<sup>-1</sup>), and high-dose(400 mg·kg<sup>-1</sup>) and low-dose(200 mg·kg<sup>-1</sup>) RO groups. Rats were administered correspondingly by gavage for eight weeks. After seven weeks of intragastric administration, the estrus cycle of all rats was measured. After eight weeks of intragastric administration, four rats from each group were selected to give birth. For other rats, blood was collected on the day of estrus, and the serum levels of estradiol (E<sub>2</sub>),progesterone (P), testosterone (T), follicle-stimulating hormone (FSH), and luteotropic hormone (LH) were detected. Uterus and ovaries were extracted and weighed to calculate organ index. One ovary was made into pathological sections, and the follicles at different developmental stages and corpus luteum were counted. Real-time fluorescence-based quantitative polymerase chain reaction(Real-time PCR)and Western blot were performed on the other ovary to detect mRNA and protein changes in the PI3K/Akt signaling pathway. Forty female Kunming mice were randomly divided into a normal group and RO groups (400 mg·kg<sup>-1</sup>) with 14 days, 28 days, and 56 days of intervention. Mice in the RO groups were raised with male mice in cages after intragastric administration of OR for 14, 28, and 56 days, respectively. After 18 days, the number of intrauterine fetuses on both sides and the number of stunted fetuses were counted. Result:After eight weeks of intragastric administration of OR, the rats showed decreased uterine index (<italic>P</italic><0.05), declining serum LH (<italic>P</italic><0.05), reduced luteum (<italic>P</italic><0.01), dwindled primary follicles (<italic>P</italic><0.05), and increased rate of follicle atresia (<italic>P</italic><0.01). Additionally, more luteal or interstitial glands degenerated into interstitial structures in the ovarian cortex in a short time. The mRNA expression levels of PI3K and Akt in the ovary were elevated (<italic>P</italic><0.01), while the mRNA expression levels of mTOR and PTEN were reduced (<italic>P</italic><0.01). The phosphorylation level of Akt protein showed a downward trend without significant difference. For the rats, the number of fetuses was decreased (<italic>P</italic><0.05). The pregnancy rate of mice was decreased to varying degrees after administration of RO for different durations, with the lowest in the 14 day RO group, as low as 30%. After 28 days of intragastric administration of RO, the difference in left and right uterine pregnancy increased (<italic>P</italic><0.05). Conclusion:Long-term administration of RO can lead to premature ovarian failure by over-stimulating the ovary, which is similar to clomiphene. Short-term administration can result in decreased pregnancy rate, excessive ovulation on one side, and inhibition of ovulation on the other side. The influence on follicles needs further exploration.

ABSTRACT

Objective To analyze the current status of mental symptoms and related influencing factors in patients with schizophrenia, and to provide reference for helping patients achieve better home rehabilitation. Methods Cluster extraction was done of 371 home schizophrenia patients registered in the community, and follow-up surveys were carried out for general demographic data, family status, current status of the disease, and treatment status.Univariate and multivariate logistic regression analysis was used for each factor in affecting the patient′s mental symptoms. Results All of the 371 patients completed follow-up surveys, and 121 patients with positive psychotic symptoms (positive rate 32.61%).Univariate analysis showed that differences in the economic situation, course of illness(years), risk behavior level, self-knowledge, hospitalization and working status were statistically significant (P < 0.05), while differences in gender, age, education level, marital status, disease course, family history were not.And there was no significant difference in the disability identification, monitoring status, medication compliance, and free medication (P>0.05).Multivariate logistic analysis showed that complete self-knowledge(OR=0.488, 95%CI:0.284-0.838), and participation in work(OR=0.469, 95%CI:0.257-0.857) were protection factors (P < 0.05). Conclusion The service management of patients with schizophrenia at home should be strengthened, regular follow-up and nursing care mechanisms should be improved, and patients with positive mental symptoms should be actively mobilized for hospitalized treatment.Improving the patient′s self-knowledge and helping patients improve their ability to work and work is the key to preventing mental symptoms in patients with schizophrenia.

ABSTRACT

Objective:To investigate the ecological environment and associated plants of mountain cultivated Panax ginseng were investigated in Xifeng county,Kuandian Manchu autonomous county and Huanren Manchu autonomous county of Liaoning province,in order to provided suggestions on the development of the mountain cultivated P. ginseng in these areas and necessary reference materials for the analysis of ecological suitability of mountain cultivated P. ginseng. Method:The field survey was conducted through sample plot survey in the survey pattern ofbased on representative area-sample plot-quadrat,literature review and data collation, were conducted to record indicators, calculate relevant parameters and make summary. Result:The associated plants were investigated based on 42 samples of mountain cultivated ginseng collected. A total of 95 plant species were identified. The correlation analysis was made on the six main species with an occurrence frequency of higher than 33%, and they were all positively correlated. Asarum heterotropoides var. mandshuricum was the strongest,which was followed by Schisandra chinensis, and Tilia tuan was the strongest in the arbor layer. The survey also found many problems in these areas,such as fewer large-scale planting bases, more scattered households,mixed germplasms,irregular planting techniques,simple planting mode and less sources of economic income generation. Conclusion:Xifeng county,Kuandian county and Huanren county of Liaoning province boast a superior ecological environment and abundant resources of mountain cultivated P. ginseng. The survey gets the whole picture of the ecological environment and associated plants of mountain cultivated P. ginseng in representative areas of Liaoning province,so as to provide the basis for the further development of mountain cultivated P. ginseng cultivation industry and the promotion of mountain cultivated Panax ginseng economy.

ABSTRACT

Brain metastases are common in oncologic patients. Stereotactic radiosurgery (SRS) is an effective treatment for oligo brain metastases. However, SRS is not suitable for large metastatic lesions. Fractionated stereotactic radiation therapy (FSRT) is a new option for the treatment of brain metastases. It can deliver high doses of radiation to tumor while protecting the normal tissues and organs outside the irradiation field as much as possible and maintaining good local control rate of tumors. This article reviews the dose-response relationship and adverse reactions of FSRT with different fractionated dose schemes, and the progress of FSRT combined with targeted therapy and immunotherapy.

ABSTRACT

Objective: To screen the best compatibility proportion of total alkaloids of Coptidis Rhizoma and Notoginseng Radix et Rhizoma total saponins (HS composition)by uniform design and pharmacological model and to observe the effect on diabetic complications. Method: The total alkaloids of Coptidis Rhizoma and Notoginseng Radix et Rhizoma total saponins were used as the research objects, U6(62) table was choosed for grouping design.The content of triglyceride fasting blood-glucose (FBG), prothrombin time (PT) and active partial thromboplastin time (APTT) was chosen as index. The best dose ratio was obtained by multipleregression analysis. Rats with type 2 diabetes mellitus induced by high-fat diet combined streptozotocin were divided into blank group, model group, metformin group (150 mg·kg-1), HS composition group (total alkaloids of Coptidis Rhizoma 360 mg·kg-1+ Notoginseng Radix et Rhizoma total saponins 40 mg·kg-1). Rats were administered orally for 10 weeks.By observing the blood glucose, glucose tolerance,area under the curve (AUC), triglyceride (TG), high-density lipoprotein (HDL-C)and low-density lipoprotein (LDL-C), hemorheological indexes and pathological changes of pancreas, heart, kidney and retina in rats of each group, the effect of this composition on diabetic complications was verified. Result: Combination of 625 mg·kg-1 total alkaloids of Coptidis Rhizoma and 60 mg·kg-1 Notoginseng Radix et Rhizoma total saponins was the optimal dosage ratio of HS composition.The validation test showed that compared with blank group, the fasting blood glucose and lipid levels in the model group were significantly increased (PPPPPPPPPPPConclusion: The optimum compatibility dose of HS composition have a good therapeutic effect on diabetic complication rats induced by high-fat diet and streptozotocin.

ABSTRACT

Multi-walled carbon nanotubes-tungsten oxide (MWCNTs-WOx) nanocomposites were fabricated on glassy carbon electrode (GCE) through a simple electrodeposition method,in which WOx were fabricated on MWCNTs. The morphology and constitution were characterized by field emission scanning electron microscopy(SEM) and X-ray photoelectron spectroscopy(XPS). Electrochemical characterization of modified electrode was done by electrochemical impedance spectroscopy (EIS). The cyclic voltammogram (CV)method was adopted to investigate the electrochemical behavior of dopamine (DA) on MWCNTs-WOx-modified glassy carbon electrode, and a new detection method for DA was developed by differential pulse voltammetry (DPV). The results showed that MWCNTs-WOx nanocomposites had obvious electrocatalytic effect on DA in phosphate buffer solution (pH=6.5). Under the optimized experimental conditions, the DA peak current demonstrated a good linear relationship with concentration in the range of 0.05-1.00 mmol/L, and the detection limit was 17 μmol/L(S/N=3). Effects of different experimental parameters on the response current of the modified electrode were investigated,and it was found that the prepared electrochemical sensor displayed good reproducibility,high selectivity and strong anti-interference ability. UA did not interfere with the detection of DA. A new electrochemical method for the quantitative determination of DA was established and successfully applied to the determination of dopamine hydrochloride injection samples.

ABSTRACT

BACKGROUND: With a clear distinction from traditional computed tomography (CT) imaging with information absorption, phase-contrast CT with synchronous radiation has implemented the microstructure imaging of soft tissues in organisms with an unprecedented imaging mechanism. OBJECTIVE: To explore the synchrotron radiation phase-contrast CT imaging technology in the bone regeneration imaging after bone grafting. METHODS: Four New Zealand white rabbits were used to make a metaphyseal defect model. Then, model rabbits were randomized into a group with calcium phosphate bone grafting and a group with Bio-Oss bone grafting in the defects. The specimens were imaged by the synchrotron radiation phase-contrast CT and stained with hematoxylin-eosin and sirius red 2 weeks after bone grafting. RESULTS AND CONCLUSION: (1) Bio-Oss bone graft material group: Osteoid was observed not only around the graft material but also in the area far from the graft bone material as reticulate structure by the synchrotron radiation phase-contrast CT. Hematoxylin-eosin staining showed a large amount of red osteoid tissues arranged as trabecular bone, and a large amount of osteoblasts with obvious osteogensis. Sirius red-stained pathological sections were largely stained yellow, and there were round or oval osteoblasts with strongly expressed type I collagen. (2) Calcium phosphate bone graft material group: There was no reticulate structure shown by the synchrotron radiation phase-contrast CT, and the creep of osteoid tissues was only around the bone graft. Hematoxylin-eosin staining showed a large amount of red osteoid tissues, and sirius red-stained pathological sections were stained yellow and red. To conclude, the synchronous radiation phase-contrast CT can clearly display the regenerated structure of bone grafts.

ABSTRACT

Objective To investigate and compare the curative effect between delayed percutaneous coronary intervention (PCI) for patients with acute myocardial infarction presenting 12-24 hours from symptom onset and medical therapy on acute myocardial infarction patients presenting with ST-segment elevation (STEMI). Methods Using a prospective,open,parallel,controlled research approach,186 patients with STEMI were divided into delayed PCI group(n=89),which received PCI within 12-24 hours after STEMI and medical therapy group(n=97),which received medical therapy after STEMI. All patients were followed up 1-6 months with average follow-up (5.6 ± 1.4) months. Data of hospitalization period, the cardiac structures detected by echocardiography such as left atrial diameter (LAD), left ventricular diastolic diameter(LVDd),left ventricular ejection fraction LVEF,left ventricular fractional shortening(LVFS),composite end point events and major adverse cardiac events(MACE)were compared between the two groups.Results Compared with medical therapy group, the hospitalization cycle was significantly shorter in delayed PCI group. Data of the LAD and LVDd were significantly decreased,but LVEF and LVFS were increased in delayed PCI group compared with those of medical therapy group at 30 d and 6-month follow-up. The incidence of MACE and composite end point events were significantly less in delayed PCI group than those of medical therapy group (P<0.05). Conclusion Delayed PCI treatment can decrease the time of hospital stay and decrease the incidence rates of MACE and composite end point events,and improve left ventricular function and prognosis of patients.

Objective To review and investigate the malaria control history of Wuzhou City,Guangxi Zhuang Autonomous Region from 1950 to 2015,so as to provide the evidence for future malaria control and surveillance. Methods The data of ma?laria control in Wuzhou City from 1950 to 2015 were collected and analyzed. Results In 1950 decade,the malaria incidence in Wuzhou City was 1 435.55/100 000,higher than the average level in Guangxi,and the mortality of malaria was 0.95/100 000. The malaria incidence of local residents was reduced to 3.61/100 000 in 1979 and no local malaria case was found since. The im?ported malaria cases were found in Wuzhou City since 1980,and were more than local cases since 1981. In recent five years, 87.50%(7/8)of imported malaria cases were from south?east Asia. Conclusions Wuzhou City has reached the national criteri?on of malaria elimination,but the imported malaria is the recent threat. The surveillance and control work of malaria should be strengthened.

ABSTRACT

<p><b>OBJECTIVE</b>To detect the base sequences of all exons and part of introns in the GYPA gene of the glycophorin GPA and to investigate the polymorphism of M, N alleles in Chinese population.</p><p><b>METHODS</b>A total of 225 blood sample were randomly colleeted from unrelated Chinese volunteers and were detected by serology techniques. The primers were designed by self, the seguencing of GYPA gene related with sample exon 1-7 full length sequences of bases and intron-1-7 partial sequence was performed, the polymorphism of M, N gene mutation in mucleotide sequence was analysed.</p><p><b>RESULTS</b>The results of M and N genotyping were in agreement with the results of serological detection. The 23rd base of intron-2, the 55th base of intron-3, the 63rd base of intron-4, the 55th, 189th and 190th base of intron-6, the 712th base variation of exon-7 in the gene M and N were used to subdivide the gene M and N into the mutant M103, M201, M202, N101, N102, N103, N104, and N201. At the same time, it was found that 42th and 54th base were mutated, the base T was inserted between 59th and 60th base in the intron-2, the new mutations occurred in the alleles 28, 29, 65 and 102 in intron-3 in this study.</p><p><b>CONCLUSION</b>The polymorphism of the the Chinese population's GYPA gene occurs in all the exons and partly in the introns. The gene polymorphism of M and N blood group in Chinese population might provide the theoretical basis for the studies of clinical blood transfusion, human population genetics and molecular biology.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To identify more effective and less toxic drugs to treat animal toxoplasmosis.</p><p><b>METHODS</b>Efficacy of seven kinds of sulfonamides against Toxoplasma gondii (T. gondii) in an acute murine model was evaluated. The mice used throughout the study were randomly assigned to many groups (10 mice each), which either remained uninfected or were infected intraperitoneally with tachyzoites of T. gondii (strains RH and CN). All groups were then treated with different sulfonamides and the optimal treatment protocol was determined candidates. Sulfadiazine-sodium (SD) was used for comparison.</p><p><b>RESULTS</b>The optimal therapy involved gavaging mice twice per day with 250 mg/kg bw of sulfachloropyrazine-sodium (SPZ) for five days. Using this protocol, the average survival time and the time-point of 50% fatalities were prolonged significantly compared with SD treatment. Treatment with SPZ protected 40% of mice from death, and the heart and kidney tissue of these animals was parasite-free, as determined by nested-PCR. SPZ showed excellent therapeutic effects in the treatment of T. gondii in an acute murine model and is therefore a promising drug candidate for the treatment and prevention of T. gondii in animals.</p><p><b>CONCLUSIONS</b>It can be concluded that the effective drug sulfachloropyrazine may be the new therapeutic options against animal toxoplasmosis.</p>

ABSTRACT

OBJECTIVE@#To discuss the influencing factors of using late postmortem phenomena to estimate PMI and to provide experience for an accurate estimation.@*METHODS@#Forty-nine corpses of late postmortem were collected in Shaoxing City, Zhuji area from 2004 to 2011. The related factors were analyzed including season, scene, estimated PMI, exact PMI, cause of death and main factors effected PMI, etc.@*RESULTS@#Of all 49 cases, 20 corpses were outdoor, 11 were indoor and 18 were in water. Thirty-seven cases were successful to estimate PMI and 12 cases were unsuccessful. The main factors affected PMI were infection, poisoning, human destruction and high-pressure electric shock, etc.@*CONCLUSION@#In general, PMI can be correctly estimated by late postmortem phenomenon. When the cases included infection, poisoning and human destruction, we should estimate PMI with the comprehensive analysis.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effects of Coptis Chinensis on vasoconstrictive activity of isolated thoracic aorta rings of normoxic and chronic intermittent hypobaric hypoxic (CIHH) rats, and to investigate the underlying mechanisms.</p><p><b>METHODS</b>Young male Sprague-Dawley rats were randomly divided into normoxic group and CIHH group: the fonnrmer were not given any special treatment; the latter were exposed to hypoxia in a hypobaric chamber simulating 5000 m altitude (PB = 404 mmHg, PO2 = 84 mmHg, 11.1% O2), 6 hours daily for 28 days. The isolated thoracic aorta rings of rats were prepared and perfused in thermostat, and the effects of Coptis on vasoconstrictive activity of aorta rings were recorded, the mechanisms were investigated simultaneouly.</p><p><b>RESULTS</b>Coptis Chinensis significantly decreased NE and KC-induced vasoconstriction of normoxic and CIHH rats' isolated aortic rings, but the inhibitive effects had no obvious discrepancy between the two groups. The contractive amplitude had no marked change after the removal of endothelium. When calculated by Logit Loglinear analysis, IC50 of NE and KCl-induced contractive amplitude in normoxic group were respectively 2.99 g/L and 6.14 g/L, while they were 3.45 g/L and 5.81 g/L in CIHH group. The inhibitive effect of Coptis on vasoconstrictive activity of both groups could be partly decreased by Glibenclamide and nitro-L-arginine methyl ester; Indomethacin suppressed the effect on normoxic group as well. Also Coptis significantly inhibited NE-induced both intracellular and extracellular calciumion-depended vasoconstriction.</p><p><b>CONCLUSION</b>Coptis Chinensis obviously relaxes isolated thoracic aorta rings of normoxic and CIHH rats, but the effects are endothelium-independent and have no marked discrepancy between the two groups. The mechanisms of the effects may be related to the opening of ATP-sensitive K+ channel, raise of nitric oxide concentration in both groups, and the increasing of PGI2 in normoxic group. Besides, Coptis may inhibit sarcoplasmic reticulum releasing Ca2+ and decrease the inflow of extracellular Ca2+ via cell membrane.</p>

ABSTRACT

AIM:To research the chemical modification from berberine hydrochloride to berberrubine in Cortex Phellodendri before and after processing.METHODS:Berberrubine was isolated by column chromatography and the contents of berberine hydrochloride and berberrubine in different processed Cortex Phellodendri were determined by HPLC.RESULTS:With increacing processing temperature and time,part of berberine hydrochloride became berberrubine in processed Cortex Phellodendrin,and berberrubine content decreased under the condition of high temperature operation or overtime processing.CONCLUSION:The experimental data shows that the processed Cortex phellodendri has the maximum average content of berberrubine as processing temperature is maintained as 180℃ for 40 min.

ABSTRACT

In order to clone and identify differentially expressed genes in the sporogony stage of Eimeria tenella, the cDNAs from unsporulated oocysts and sporulated oocysts of E. tenella were used as driver, respectively, the cDNAs from sporozoites of E. tenella was used tester, Two subtractive cDNA libraries of sporozoites were constructed by using the technique of suppression subtractive hybridization (SSH). the cDNAs from unsporulated oocysts was used driver, the cDNAs from sporulated ooceysts was used tester, one subtractive cDNA library of sporulated oocysts was constructed. PCR amplification revealed that the two subtractive cDNA libraries of sporozoites and one subtractive cDNA library of sporulated oocysts contained approximated 96%, 96% and 98% recombinant clones, respectively. Fifty positive clones were sequenced and analyzed in GenBank with Blast search from three subtractive cDNA libraries, respectively, thirteen unique sequences were found from the subtractive cDNA library of sporulated oocysts, eight ESTs shared significant identity with previously described. A total of forty unique sequences were obtained from the two subtractive cDNA libraries, nine ESTs shared significant identity with previously described, the other sequences represent novel genes of E. tenella with no significant homology to the proteins in Genbank. These results have provided the foundation for cloning new genes of E. tenella and further studying new approaches to control coccidiosis.

ABSTRACT

Two-dimensional electrophoresis (2-DE) was employed to compare the proteome of Diclazuril-resistance Eimeria tenella with that of sensitive strains for identifying unique proteins of these stains. 5 protein spots were found to express differentially. Four spots which remarkably were measured by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS). The data obtained from peptide mass fingerprinting were used in NCBInr database search, two protein spots in gel were identified as Eimeria tenella sporulated oocyst TA4 antigen protein, Heat shock 70kD protein, two protein spots were functional proteins of Eukaryote. These proteins are potentially basic work for finding molecular mechanism about drug-resistance of Eimeria tenella and new marker in the detection of resistance of Eimeria tenella.
