ABSTRACT

Objective To investigate vein pump injection of epinephrine on the effect of serum potassium during gastric cancer radical surgery. Method Forty patients with ASA grade Ⅱ~Ⅲ underwent surgery within a time limit of gastrointestinal under general anesthesia. All patients were randomly divided into the experimental group and the control group.Patients in the experimental group received continuous intravenous injections of adren-aline[0.03 to 0.1 μg/(kg·min)].Patients in the control group received equal dose of saline.Potassium concentra-tions at different time points were determined and compared between patients in two groups. Results No signifi-cant differences were observed in gender and age distribution of patients in the experimental group and the control group. Compared with the control group, potassium concentration was significantly decreased in patients at T3mo-ment(30 min pump injection of norepinephrine)(P<0.05),but was lower than that of patients in the experimen-tal group at T1moment(before anesthesia)(P<0.05).Compared with the control group, potassium concentration was significantly increased in the experimental group at T4moment(pump adrenaline injection for 10 min)(P <0.05),which was also significantly higher than that in the experimental group at T1moment(P < 0.05). Mean-while,compared with the control group, potassium concentration was also significantly increased in the experimen-tal group at T5moment(stop the pump adrenaline injection for 30 min),which was also significantly higher than that in the experimental group at T1moment(P < 0.05). Conclusions Intravenous injection of adrenaline can reduce potassium concentration in patients received gastrointestinal surgery, and potassium concentration can be increased after adrenalin injection was stopped.

ABSTRACT

Aim To investigate the roles of chloride channels in the apoptosis and apoptotic volume de-crease (AVD)induced by adriamycin in nasopharyn-geal carcinoma CNE-2Z cells. Methods Apoptotic rates were detected by flow cytometry, and the volume changes were measured by the time-lapse live cell ima-ging technique. The patch clamp technique was used to record whole-cell chloride currents. Results Adria-mycin induced apoptosis of CNE-2Z cells. An early ap-optotic volume decrease was observed in the cell trea-ted with adriamycin. The cell volume was decreased by about 10% in 2 h.Adriamycin activated a chloride current which showed outward rectification. The chlo-ride channel blocker 5-nitro-2-(3-phenylpropylamino)-benzoate (NPPB ) could inhibit the adriamycin-in-duced chloride currents, apoptosis and prevent cell shrinkage. Conclusions Our findings suggest that ad-riamycin causes cell apoptosis by activation of chloride channels.Chloride channels may be involved in the apoptosis and apoptotic volume decrease induced by adriamycin in CNE-2Z cells.

ABSTRACT

Objective To observe the therapeutic effect of surface functional electrical stimulation combined with electroacupuncture on neurogenic bladder after the spinal cord injury. Methods Forty cases of neurogenic bladder after the spinal cord injury were randomly divided into a control group (n =20) and a combination group (n =20).The combination group was treated with surface functional electrical stimulation combined with electroacupuncture on the basis of internal urethral catheterization, while the control group was treated with surface functional electrical stimulation on the basis of internal urethral catheterization. The bladder urination was tested using urodynamic study before and after two months of treatment. Results There was no significant difference between the 2 groups in the average maximum bladder capacity, detrusor pressure at filling time, maximal urethral closure pressure, maximum urine flow rate and residual urine volume before the treatment. After the treatment, however, the average maximum bladder capacity, maximal urethral closure pressure and maximum urine flow rate in the combination group were significantly higher than those of the control group, while the average detrusor pressure at filling time and the residual urine volume of the combination group were significantly lower than the control group. Conclusion Surface functional electrical stimulation combined with electroacupuncture could remarkably improve bladder function of patients with the neurogenic bladder after the spinal cord injury.

ABSTRACT

To develop a timesaving and easy operating Reverse Phase (RP) chromatography method, we adopted Thermo Pierce RP C18 Tip to separate small amount hippocampus peptide mixtures and to compare with high performance liquid chromatography (HPLC). According to the separation performance of 4 ACN gradient optimization methods, we determined the best ACN concentration gradient. The results showed that, the experiment took only 10 min by separating with eight ACN concentration gradient, which accounted 1/4 for HPLC. But as for the identified proteins, RP C18 Tip accounted 85.5% for HPLC. ACN gradient of 5%, 15%, 20% and 90% had best repeatability (P = 0.429) and result for separating 30 μg peptides. This method is easy to operate, timesaving and has low cost. It could be used into pretreatment of small amount complex proteomic samples.

ABSTRACT

Objective To study the mechanism by which a high-intensity pulsed electromagnetic field (HIPEMF) (0.1 Hz,4 T,8 pulses) facilitates the proliferation of neural stem cells by detecting the expression of β-catenin genes and protein. Methods Neural stem cells (NSCs) were isolated from the sub-ventricular zone (SVZ) of neonatal rats and cultured in supplemented,serum-free medium for two weeks. The NSCs were then divided into an experimental group exposed to a HIPEMF for 8 pulses and a control group given sham stimulation. The gene and prorein expression of β-catenin in the NSCs were assayed by RT-PCR and Western blotting on the 1st,3rd,5th and 7th day after the stimulation. Results The NSCs' cloned spheres were round and translucent, and showed red fluorescence after staining with anti-nestin (cy3).The RT-PCR results showed β-catenin genes were highly expressed in the exposed group (significantly more than in the controls). The Western blotting showed that expression of β-catenin protein was also higher in the experimental group, especially at the 7th day after stimulation,a difference which was also statistically significant Conclusion HIPEMF at 0.1 Hz,4 T,in 8 pulse trains can promote NSC proliferation, perhaps through the Wnt/β-catenin signaling pathways.
