ABSTRACT

AIM To explore the mercury accumulation in KM mice after being given Zuotai at different doses and time.METHODS KM mice were randomly divided into blank group,Zuotai low-,middle-and high-dose (6.07,60.70 and 606.97 mg/kg,42 d;606.97 mg/kg,14 d) groups. The mercury contents in brain (olfactory bulb, cortex, hippocampus, hypothalamus, brain stem, cerebellum),heart, lung, kidney, liver, spleen, serum, muscle of mice were measured after administration.RESULTS Compared with the blank group,Zuotai at low-dose significantly increased the mercury contents in hippocampus, cerebellum, lung, kidney, liver and serum of mice after 42-day treatment;Zuotai at middle-dose markedly increased the mercury contents in olfactory bulb, cortex, hippocampus, brain stem, cerebellum, heart, lung, kidney, liver, spleen and serum of mice after 42-day treatment;the mice treated with high-dose of Zuotai for 42,14 days significantly increased the mercury contents in olfactory bulb, cortex, hippocampus, hypothalamus, brain stem, cerebellum, heart, lung, kidney, liver, spleen, muscle and serum.CONCLUSION Mercury can be accumulated in different tissues of mice after intragastric administration of Zuotai in a dose-and time-dependent manner, which suggests that Zuotai and its compound preparations should not be used in high-dose and long-term.

ABSTRACT

Ductular reaction (DR) is a phenomenon that refers to impaired hepatocellular proliferation and activated hepatic stem cells at times of demand such as hepatic regeneration. The components of ductular reactions are thought to be derived from activated hepatic progenitor cells, intermediate hepatobiliary cells, and reactive ductular cells.DRs are thought to play two roles in hepatocarcinogenesis:their hepatic progenitor cells may undergo malignant transformation and migration, and they may contribute to the development of the tumor's microenvironment.Exploring DR phenotype, cellular diversity, and relationship to hepatocarcinogenesis will further enhance our understanding of the pathogenesis of human hepatocellular carcinoma with an ultimate goal of developing new therapies.

ABSTRACT

Hepatocellular carcinoma (HCC) development is closely related to overexpression of tumor promoters or down-regulation of tumor suppressors. The mammalian sirtuin family was found to be a nicotinamide adenosine dinucleotide (NAD/NADH)-dependent histone deacetylase (HDAC),which implicated in the regulation of critical biological processes through deacetylation modifying on histone and nonhistone.SIRT1 can regulate metabolism, aging, inflammation and cancer progression. In particular, more and more evidence proves that SIRT1 can act as a tumor promot er in hepatocellular carcinoma through deacetylation on tumor suppressors. On the other hand,SIRT1 can strongly suppress metabolic syndrome-associated liver cancer in the mouse model. This review will discuss the expression of sirtuin family member in liver cancer and its clinical significance.

ABSTRACT

Objective To investigate the effects of macrophage inflammatory protein-1α (MIP-1α) combined with molecule 4-1BB L on the tumorigenicity of hepatocellular carcinoma cells in vivo. Methods Mouse MIP-1α (mMIP-1α) expressed Hepa 1-6 cells were transfected with m4-1BBL recombinant retrovirus, the anti-histidinol cells clones were selected and amplified. The expression of m4-1BB L was confirmed by flow cytometry. The growth curve of Hepa 1-6 cells transfected with mMIP-1α and m4-1BBL alone or together was drawn and compared. C57B/L Mice were randomly divided into 7 groups, 9 mice in each group, injected with mMIP-1α+m4-1BB L Hepa 1-6 cells, m4-1BB L Hepa 1-6 cells, mMIP-1α Hepa 1-6 cells, Hepa 1-6 cells, pLXSHD Hepa 1-6 cells or PBS respectively. The tumorigenicity of hepatocellular carcinoma cells and the mice survival rate were compared between each groups. Results Hepa 1-6 mMIP-1α+m4-1BB L cells which expressed both mMIP-1α and m4-1BB L were successfully established. The expression of mMIP-1α and m4-1BB L alone or together did not affect the growth curve of Hepa 1-6 cells. Observed for 5 weeks, no tumor developed in Hepa 1-6 mMIP-1α+m4-1BB L injected mice. The tumorigenicity of Hepa 1-6 mMIP-1α+m4-1BB L was lower than that of Hepa 1-6 mMIP-1α or Hepa 1-6 m4-1BB L in vivo. The survival rate of Hepa 1-6 mMIP-1α+m4-1BBL injected mice(9/9) was higher than that of Hepa 1-6 m4-1BB L injected mice (6/9)or Hepa 1-6 mMIP-1α injected mice (1/9). Conclusion Chemokine MIP-1α combined with costimulatory 4-1BB L lowered the tumorigenicity of hepatocellular carcinoma cells in vivo, and prolonged the mice survival period.

ABSTRACT

OBJECTIVETo observe the in vivo antitumor activity of murine liver tumor vaccine expressing MIP-1alpha mediated by recombinant adenoviral vector.

METHODSThe infection efficacy was measured by GFP expression 48 hours after infection of Hepa1-6, and the number of cells was counted daily for 14 days. 5 x 10(6) modified Hepa1-6 cells were inoculated subcutaneously to C57BL/6 mice and the tumor-free animals were rechallenged by 2 x 10(6) wild-type Hepa1-6 cells or syngenic EL4 cells four weeks later. The tumor volume was measured twice a week.

RESULTSAdenoviral vectors could efficiently infect Hepa1-6 cells in vitro, and the in vitro growth rate of AdmMIP-1alpha modified Hepa1-6 cells was not affected; however the in vivo tumorigenicity was significantly decreased, compared with that of control vector modified Hepa1-6. Rechallenge of the tumor-free mice four weeks after administration of AdmMIP-1alpha with the parental Hepa1-6 cells resulted in significant inhibition of tumor growth, but there was no significant difference when rechallenged with EL4.

CONCLUSIONSThe liver cancer cells expressing mMIP-1alpha mediated by recombinant adenoviral vector decrease tumorigenicity and elicit specific immunological protection, and could be used as an effective liver tumor vaccine.

ABSTRACT

OBJECTIVETo observe the characteristics of coronary microvascular lesions (CML) in the autopsied elderly cases with hypertensive left ventricular hypertrophy (LVH) and the difference of CML among the groups of essential hypertension (EHT), coronary heart disease (CHD) and diabetes (NIDDM) also with LVH.

METHODSA retrospective study was performed in 206 cases > or = 60 years old of EHT, CHD and NIDDM with LVH and 30 normal cases as control, out of 3195 consecutive autopsied cases from 1954 to 1996 in our hospital. Arterioles with diameters of 10 - 60 microm and capillaries in the muscular layer were shown by the methods of HE, Elastic fiber + VG staining and immunohistochemistry of CD31. Quantitative measurements on the arteriole density (AD), the ratio of arteriolar wall and cavity (RWC), capillary density (CD) and the area of endothelial cell (AEC) were performed with light microscope observation and image analysis by computer. According to the thickness of the left ventricle free wall, the severity of LVH was divided into four degrees from 0 to III. LVH of degree 0-III was observed in EHT group, while only LVH of degree I was found in CHD, EHT + CHD, and NIDDM groups. SAS system was used for statistical analysis.

RESULTSAD and RWC increased while CD and AEC decreased significantly with the progression of LVH in EHT groups (P < 0.05 - 0.01). There was a similar but more severe change in the (HT + CHD) group (P < 0.01); the AD increased (P < 0.05) while all other measurements did not show obvious changes in the CHD group. The AD increased, CD and AEC decreased (all P < 0.05), but RWC did not change very much in the NIDDM group.

CONCLUSIONCML in the EHT group was characterized by an increased AD and RWC, decreased CD and AEC, among which the increased RWC was the typical change in EHT groups compared with the groups of CHD and NIDDM. Damaged CML may be one of the main factors for decreased coronary flow reserve and myocardial ischemia in cases of EHT with LVH.

ABSTRACT

92 specimens from patients with invasive ductal carcinoma were investigated. The expression of vascular endothelial growth factor (VEGF) was demonstrated by immunocytochemical staining with an antibody against VEGF and was quantitatively estimated by using computerized image analysis system. Vessels were immunohistochemically highlighted by using an antibody to CD34, and microvessel density (MVD) was quantified. The postoperative survey in univariate analysis showed that the relapse-free-survival (RFS) time of patients with more than 87 microvessels in single microscopic field was significantly worse compared to that of patients with less than 87 microvessels in node-negative patients (P<0.05). Multivariate analysis confirmed that MVD was an independent prognostic indicator for RFS in node-negative patients and all patients (P<0.05~0.01). In addition, there was a closely positive correlation between VEGF and MVD (P<0.01). The results suggested that VEGF plays crucial roles in the promotion of angiogenesis in primary breast carcinoma. VEGF and MVD are closely correlated with biological behavior of primary breast carcinomas. MVD can provide a useful message in predicting the recurrence or metastasis of tumors.

ABSTRACT

Objective: To establish a mouse hepatocellular carcinoma cell line that can produce mMIP-1α and to evaluate the possibility of cancer gene therapy by mMIP-1α. Methods: mMIP-1α cDNA was cloned into retrovirus vector pBabe puro and pBabe puro-mMIP-1α was constructed, then pBabe puro-mMIP-1α was used to transfect packaging cells, anti-puromycin cells was proliferated, the supernatant was used to infect hepa1-6, the anti-puromycin clone (hepa1-6 mMIP-1α) and hepa1-6 were analysed for the expression of mMIP-1α mRNA and protein by RT-PCR and immunohistochemistry respectively. The growth curve of hepa1-6 and hepa1-6 mMIP-1α was drawn. The chemotaxis of mMIP-1α produced by hepa1-6 mMIP-1α to mouse spleen cells was observed on agarose gel. C57B/L mouse was inoculated with the tumor cell and the tumorigenicity was studied. Results: Recombinant retrovirus vector pBabe puro-mMIP-1α with mMIP-1α cDNA was constructed. Hepa1-6 did not produce mMIP-1α mRNA and protein, while hepa1-6 mMIP-1α could produce mMIP-1α mRNA and protein. The growth curve of hepa1-6 and hepa1-6 mMIP-1α showed no difference. The chemotaxis of mMIP-1α produced by hepa1-6 mMIP-1α to mouse spleen cells was observed. The tumorigenicity was reduced. Conclusion: A mouse hepatocellular carcinoma Hepa1-6 mMIP-1α is established and mMIP-1α can affect the tumorigenecity of hepa1-6.

ABSTRACT

Objective: To investigate the expression difference of TRAIL and its receptors between resting and activated peripheral blood lymphocytes, and to study their role in cellular growth and proliferation. Methods: Expression of TRAIL and its receptors were detected by RT PCR and DNA sequencing. Results: Neither the expression of TRAIL nor its receptors were detected in resting peripheral blood lymphocyte. However, both expression were found in activated peripheral blood lymphocytes of various degree. Conclusion: TRAIL and its receptors may play a role in regulation and control of the activated lymphocytes in immune reaction.
