ABSTRACT

Studies have investigated the effects of androgen deprivation therapy (ADT) use on the incidence and clinical outcomes of coronavirus disease 2019 (COVID-19); however, the results have been inconsistent. We searched the PubMed, Medline, Cochrane, Scopus, and Web of Science databases from inception to March 2022; 13 studies covering 84 003 prostate cancer (PCa) patients with or without ADT met the eligibility criteria and were included in the meta-analysis. We calculated the pooled risk ratios (RRs) with 95% confidence intervals (CIs) to explore the association between ADT use and the infection risk of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and severity of COVID-19. After synthesizing the evidence, the pooled RR in the SARS-CoV-2 positive group was equal to 1.17, and the SARS-CoV-2 positive risk in PCa patients using ADT was not significantly different from that in those not using ADT (P = 0.544). Moreover, no significant results concerning the beneficial effect of ADT on the rate of intensive care unit admission (RR = 1.04, P = 0.872) or death risk (RR = 1.23, P = 0.53) were found. However, PCa patients with a history of ADT use had a markedly higher COVID-19 hospitalization rate (RR = 1.31, P = 0.015) than those with no history of ADT use. These findings indicate that ADT use by PCa patients is associated with a high risk of hospitalization during infection with SARS-CoV-2. A large number of high quality studies are needed to confirm these results.

ABSTRACT Introduction The discussion about the impact on the physical and mental health of obese college students is a current issue. The implementation of "weight loss exercise prescription" seeks to discuss weight loss techniques and their effects, problems most faced by obese girls. Objective Explore the effect of sports intervention on the physical and mental health of obese female students. Methods 20 female college students with simple obesity were selected for an 18-week intervention experiment of fitness exercise associated with long-distance running. The physical and mental health of the volunteers was measured before and after the experiment for statistical analysis. Results After 18 weeks of exercise, it was observed that interpersonal sensitivity, depression, anxiety, and SCL-90 scores decreased significantly, indicating that the implementation of the exercise prescription had a positive impact on the mental health of obese girls. Across the study, compared to a single exercise method, associated aerobic exercise had a significant effect on improving BMI indicators, body weight, and body fat rate in obese adolescent girls. Conclusion By prescribing exercise for weight loss, one can significantly improve the cardiopulmonary function of obese college students and promote physical health, manifesting in the indices of vital capacity and cardiopulmonary function. RESUMO Introdução A discussão sobre o impacto na saúde física e mental de estudantes universitárias obesas é um tema atual. Através da implementação da "prescrição de exercícios de emagrecimento", busca-se discutir tópicos sobre as técnicas de perda de peso e seus efeitos, problemas mais enfrentados pelas meninas obesas. Objetivo Explorar o efeito da intervenção esportiva sobre a saúde física e mental das estudantes obesas. Métodos 20 estudantes universitárias com obesidade simples foram selecionadas para uma experiência de intervenção de 18 semanas de exercício físico de aptidão física associada a corrida de longa distância. A saúde física e mental das voluntárias foi medida antes e depois do experimento para uma análise estatística. Resultados Após 18 semanas de exercícios observou-se que a sensibilidade interpessoal, a depressão, a ansiedade, e a pontuação de SCL-90 diminuíram significativamente, indicando que a implementação da prescrição de exercícios teve um impacto positivo sobre a saúde mental das meninas obesas. Através do estudo, comparado com um único método de exercício, o exercício aeróbico associado apresentou um efeito significativo na melhoria dos indicadores de IMC, no peso corporal e na taxa de gordura corporal das adolescentes obesas. Conclusão Através da prescrição de exercício para perda de peso, pode-se melhorar significativamente a função cardiopulmonar de estudantes universitárias obesas e promover a saúde física, resultado manifestado nos índices de capacidade vital e função cardiopulmonar. RESUMEN Introducción El debate sobre el impacto en la salud física y mental de las estudiantes universitarias obesas es un tema de la actualidad. A través de la aplicación de la "prescripción de ejercicios para adelgazar", se pretende debatir temas sobre las técnicas de adelgazamiento y sus efectos, los problemas a los que más se enfrentan las chicas obesas. Objetivo Explorar el efecto de la intervención deportiva sobre la salud física y mental de las estudiantes obesas. Métodos Se seleccionaron 20 estudiantes universitarias con obesidad simple para un ensayo de intervención de 18 semanas de ejercicio físico asociado a carreras de larga distancia. Para el análisis estadístico se midió la salud física y mental de los voluntarios antes y después del experimento. Resultados Tras 18 semanas de ejercicio se observó que las puntuaciones de sensibilidad interpersonal, depresión, ansiedad y SCL-90 disminuyeron significativamente, lo que indica que la aplicación de la prescripción de ejercicio tuvo un impacto positivo en la salud mental de las chicas obesas. En todo el estudio, en comparación con un único método de ejercicio, el ejercicio aeróbico asociado mostró un efecto significativo en la mejora de los indicadores del IMC, el peso corporal y el índice de grasa corporal en chicas adolescentes obesas. Conclusión Mediante la prescripción de ejercicio para la pérdida de peso, se puede mejorar significativamente la función cardiopulmonar de las estudiantes universitarias obesas y promover la salud física, resultado que se manifiesta en los índices de capacidad vital y función cardiopulmonar. ABSTRACT

Objective:To compare the chemical constituents of Puerariae Flos from three different varieties of <italic>Pueraria montana</italic> var. <italic>lobata</italic>, <italic>P. montana</italic> var. <italic>thomsonii</italic> and <italic>P</italic>. <italic>montana</italic> var<italic>. montana</italic>. Method:Ultra-performance liquid chromatography-quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF-MS) was used with the mobile phase of 0.1% formic acid aqueous solution (A)-acetonitrile (B) for gradient elution (0-20 min, 10%-30%B; 20-30 min, 30%-55%B; 30-35 min, 55%-95%B; 35-37 min, 95%B; 37-40 min, 95%-10%B), the flow rate was 0.25 mL·min<sup>-1</sup>. Electrospray ionization (ESI) was used to scan and collect MS data in positive and negative ion modes with scanning range of <italic>m</italic>/<italic>z</italic> 50-1 500. The chemical components from different sources of Puerariae Flos were identified in combination with the chemical composition database and literature information. After the obtained data were normalized by MarkerView<sup>TM</sup> 1.2.1, they were imported into SICMA-P 14.1 software for principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) to select the main differentiated components among the three different varieties. Result:A total of 35 compounds were identified from three different varieties of Puerariae Flos, including 22 isoflavones, 6 flavonoids and 7 saponins. The flowers of <italic>P</italic>. <italic>lobata</italic>, <italic>P. montana</italic> var. <italic>thomsonii</italic> and <italic>P</italic>. <italic>montana</italic> var<italic>. montana</italic> contained 32, 35, 33 compounds, respectively. And 18 differential compounds were screened under the positive and negative ion modes, including kakkalide, tectoridin, 6″-<italic>O</italic>-xylosyl-tectoridin, 4'-methyltectorigenin-7-glucoside, glycitin, 6″-<italic>O</italic>-xylosyl-glycitin, irisolidone, kaikasaponin Ⅲ, 6″-<italic>O</italic>-malonylglycitin, kakkalidone, tectorigenin, rutin, soyasaponin BB, vitexin, biochanin A, genistin, kakkatin, azukisaponin Ⅱ. Conclusion:This research is the first to systematically study the chemical constituents of the flower of <italic>P</italic>. <italic>montana</italic> var<italic>. montana</italic>, although the flower of <italic>P</italic>. <italic>montana</italic> var<italic>. montana</italic> is used as adulterants, it has high contents of tectoridin and 6″-<italic>O</italic>-xylosyl-tectoridin, which has great potential for development. The efficacy components such as kakkalide and tectoridin in Puerariae Flos from the three sources of varieties are obviously different, and it is necessary to carefully consider the application of these three varieties as Puerariae Flos.

ABSTRACT

Objective:To determine the current status of multiple antibiotic residues in meat and meat products in Shanghai based on a 5-year surveillance, and perform the health risk assessment. Methods:We performed the examination in accordance with the Manual for National Food Contamination and Harmful Factor Risk Monitoring, and conduct health risk assessment according to the national limit standards on the monitoring data of 2016‒2020. Results:The total detection rate of multiple antibiotics in meat and meat products in Shanghai was determined to be 16.03%, in which the total unqualified rate was 1.97%. Moreover, the detection rate of quinolones was 2.78% and its unqualified rate was 0.83%. The unqualified rate of loxacin in cooked meat products was 2.12%. The detection rate of tetracyclines was 17.06% and its unqualified rate was 0.34%, in which the highest detection rate was identified in doxycycline (11.64%). The detection rate of sulfonamides was 3.16%, in which the highest detection rate was in sulfamethazine (1.05%). The detection rate of florfenicol was 5.15% and its unqualified rate was 0.12%. The difference of ofloxacin residues between diverse food categories (χ2=17.44, P<0.05) and processing links (χ2 =14.10, P<0.05) was statistically significant. In addition, the sum amount of ofloxacin, enrofloxacin and ciprofloxacin in cooked meat products was higher than other food categories; the unqualified rate and residual amount of ofloxacin available in online stores and catering links were both higher. The residual amount of doxycycline and the unqualified rate in the online store link were significantly higher than those in other links. Based on preliminary assessment, the high exposure values in the 97.5 percentile of meat and meat products accounted for a very low proportion of the corresponding acceptable daily intake (ADI) and posed a low health risk to the population. Conclusion:The total detection rate of tetracyclines in meat and meat products is relatively high, which obviously accumulates in the offals of livestock and poultry. In addition, some antibiotics, such as ofloxacin and doxycycline, are relatively high in catering and online stores. It is recommended to strengthen the supervision of quinolones in cooked meat products, especially ofloxacin, enrofloxacin, and ciprofloxacin, and improve the supervision of doxycycline in meat and meat products in online stores.

ABSTRACT

Objective@#To observe the effect of functional high-intensity intermittent training combined with breathing and diet intervention on body shape, blood lipid and liver function of obese college students, so as to verify and improve the relevant intervention methods.@*Methods@#Fifty-eight obese college students recruited by Chongqing Telecom Vocational College were selected as participants. According to gender and body mass index (BMI), functional high-intensity intermittent training, breathing training and diet control intervention were carried out for 6 weeks. BMI, body fat percentage, total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C), non-alcoholic fatty liver disease (NAFLD), alanine aminotransferase (ALT), aspartate aminotransferase (AST), gamma glutamyltransferase (GGT) and other physical indicators were compared before and after intervention.@*Results@#Before and after the intervention, the changes of body indexes were as follows: BMI, body fat percentage of male and female were significantly decreased (t=7.57,4.40;4.17,5.61,P<0.01); TC, TG and LDL-C of male and female were significantly decreased, while HDL-C increased significantly (t=7.56,5.80;2.58,4.76;12.26,11.07;3.72,3.37,P<0.05); NAFLD, abnormal ALT , abnormal ast , abnormal GGT were significantly reduced(P<0.05).@*Conclusion@#The comprehensive intervention method can significantly reduce body weight and regulate blood lipid metabolism and liver function in obese college students, and the intervention experience is better. It can be applied and promoted as an effective way to lose weight for obese college students.

ABSTRACT

Molecular pharmacognosy is a science of classification and identification, cultivation and protection, and production of active ingredients of graduated drugs at the molecular level. The proposal of molecular pharmacognosy allows the research of crude drugs to advance from the microscopic level to the genetic level. Pueraria lobata root, as a medicinal and edible plant, has high application value and economic value. There are many varieties that are easy to cause confusion, and it is not easy to distinguish and identify according to traditional identification methods. Moreover, the research of P. lobate root at the genetic level is still relatively shallow. the study received extensive attention of scholars. This article reviews recent research on molecular identification of P. lobate, transcriptome sequencing, cloning and synthesis of functional genes of P. lobate root in recent years in order to provide references for further promoting the development and utilization of P. lobate root and its active ingredients.

OBJECTIVE@#To investigate the effect of continuous adductor block on pain control after bilateral knee joint Ⅰ stage replacement.@*METHODS@#A retrospective analysis was made of the data of 24 patients with bilateral knee joint I stage replacement who were treated in our hospital from January 2018 to January 2019, and who underwent continuous adductor block analgesia. There were 6 males and 18 females, aged 60 to 72 (65.05±5.82) years old. The patients underwent continuous block of adductor canal with patient-controlled analgesia system. At 4, 6, 12, 24, 36 and 48 hours after operation, visual analogue score(VAS) of resting state and passive motion state was performed;the knee joint activity was followed up for 1 week, 1, 3 and 6 months after operation;the knee joint function was scored at 6 months after operation, using the knee joint scoring standard of American Special Surgery Hospital(HSS);adverse reactions and complications were recorded.@*RESULTS@#The VAS scores under resting state and passive motion state at each time point were less than 3 points in patients with continuous adductor block. The patients had better postoperative exercise of knee joint activity. The score of HSS was excellent in 20 cases, good in 2 cases, fair in 1 case and poor in 1 case. There were only 4 cases of nausea and vomiting, none of them had serious adverse reactions and complications such as bradycardia and deep vein thrombosis.@*CONCLUSION@#Continuous adductor block has a significant effect on pain control and less adverse reactions after bilateral knee jointⅠ -stage replacement.

BACKGROUND: Caspofungin, a novel echinocandins systemic antifungal agent, has been shown to exert broad-spectrum antibacterial effect on deep fungal infections, which is superior to or equivalent with the role of amphotericin B, but there is no report on its application for preventing fungal infection after renal transplantation.OBJECTIVE: To analyze the difference in high risk factors of fungal infection after kidney transplantation using donation after cardiac death donors and living-related donor kidney transplantations, and to explore the feasibility and safety of caspofungin to prevent fungal infection after kidney transplantation using donation after cardiac death donors.METHODS: This was a prospective, single-center, controlled trial finished at the Department of Kidney Transplantation,the First Affiliated Hospital of Zhengzhou University, Henan Province, China. Totally 188 patients undergoing primary kidney transplantation without history of fungal infection and use of antifungal drugs between January 2012 and August 2013 were enrolled, including kidney transplantation with donation after cardiac death donors (n=102, trail group), and kidney transplantation with living-related donors (n=86, control group). The CYP3A5 genotype was determined preoperatively. All patients received tacrolimus+mycophenolate mofetil+prednisone triple immunosuppression after transplantation. The trial group was subjected to caspofungin therapy for 2 weeks. The risk factors for fungal infection in the two groups were compared, and the effects of caspofungin on the tacrolimus concentration, tacrolimus concentration/dose were detected in the recipients with same CYP3A5 genotype recipients at 1 and 2 weeks, and 1, 3 and 6 months postoperatively. The liver and kidney function, adverse events and fungal infections were recorded at different time points. This trial was registered with the Chinese Clinial Trial Registry (Regitration number:ChiCTR-OON-17013342).RESULTS AND CONCLUSION: The survival rate of patient/kidney was 98.4% and 97.3% respectively, 97 cases in the trial group and 86 controls competed 6-month follow-up. Preoperative hemodialysis time, hemoglobin value, cold ischemia time, warm ischemia time, intraoperative blood transfusion volume, time of central venous catheter kept in situ,methylprednisolone usage, ATG usage, serum creatinine reduced level at 1 week, thrombocytopenia and duration of postoperative body temperature > 38 ℃ were the risk factors for fungal infection in the trail group relative to the control group. The fungal infection rate in the trial and control groups was 0% and 2.3%, respectively, at 6 months of follow-up.The serum creatinine level in the trail group was significantly higher than that in the control group at 1 month postoperatively (P < 0.05), and the level showed no significant difference between two groups at other time points (P >0.05). After 2 weeks of caspofungin treatment, the concentrations of tacrolimus and tacrolimus concentration/dosage did not differ significantly in different CYP3A5 genotype recipients (P > 0.05). Caspofungin might induce some adverse reactions, especially electrolyte disturbance with an incidence of 21.6%, but there was no significant difference between two groups (P > 0.05). These findings imply that kidney transplantation using donation after cardiac death donors presents with various risk factors for fungal infection compared with living-related donor kidney transplantation.Furthermore, caspofungin is effective and safe for preventing fungal infection and has no effect on tacrolimus concentration; therefore, it can be used as a new anti-fungal agent after kidney transplantation.

OBJECTIVE: To analyze the microRNA levels in the serum of dust exposure population. METHODS: By cluster random sampling method,479 dust exposure workers were selected as dust exposure group. Four hundred and thirty-four normal healthy people without occupational dust exposure history were selected as control group. Forty-three pneumoconiosis patients were selected as pneumoconiosis group. The peripheral venous blood of the objects of 3 groups were collected,and the plasma serum were separated for detecting the relative expression of miR-16,miR-21,miR-29 a,miR-155,miR-200 c,miR-204 and miR-206 in serum by real time quantitative polymerase chain reaction. The difference of microRNA expression in the above 3 groups was observed. RESULTS: From high to low,the relative expression levels of miR-16,miR-29 a and miR-200 c were pneumoconiosis group > dust exposure group > control group( P < 0. 05),while the relative expression levels of miR-204 were control group > dust exposure group > pneumoconiosis group( P < 0. 05).The relative expression levels of miR-21 in dust exposure and pneumoconiosis groups were higher than that of control group,respectively( P < 0. 05). The relative expression level of miR-155 in dust exposure group was lower than that of control group( P < 0. 05). The relative expression level of miR-206 in pneumoconiosis group was higher than those of dust exposure group and control group respectively( P < 0. 05). CONCLUSION: The miR-16,miR-29 a,miR-200 c and miR-204 could be used as biomarkers in early diagnosis of pneumoconiosis disease.

ABSTRACT

OBJECTIVES: Remote ischemic perconditioning is the newest technique used to lessen ischemia/reperfusion injury. However, its effect in hypertensive animals has not been investigated. This study aimed to examine the effect of remote ischemic perconditioning in spontaneously hypertensive rats and determine whether chronic treatment with Olmesartan could influence the effect of remote ischemic perconditioning. METHODS: Sixty rats were randomly divided into six groups: vehicle-sham, vehicle-ischemia/reperfusion injury, vehicle-remote ischemic perconditioning, olmesartan-sham, olmesartan-ischemia/reperfusion and olmesartan-remote ischemic perconditioning. The left ventricular mass index, creatine kinase concentration, infarct size, arrhythmia scores, HIF-1α mRNA expression, miR-21 expression and miR-210 expression were measured. RESULTS: Olmesartan significantly reduced the left ventricular mass index, decreased the creatine kinase concentration, limited the infarct size and reduced the arrhythmia score. The infarct size, creatine kinase concentration and arrhythmia score during reperfusion were similar for the vehicle-ischemia/reperfusion group and vehicle-remote ischemic perconditioning group. However, these values were significantly decreased in the olmesartan-remote ischemic perconditioning group compared to the olmesartan-ischemia/reperfusion injury group. HIF-1α, miR-21 and miR-210 expression were markedly down-regulated in the Olmesartan-sham group compared to the vehicle-sham group and significantly up-regulated in the olmesartan-remote ischemic perconditioning group compared to the olmesartan-ischemia/reperfusion injury group. CONCLUSION: The results indicate that (1) the protective effect of remote ischemic perconditioning is lost in vehicle-treated rats and that chronic treatment with Olmesartan restores the protective effect of remote ischemic perconditioning; (2) chronic treatment with Olmesartan down-regulates HIF-1α, ...

This study was aimed to identify the mutation of the whole coding region of shock transcription factor 4 (HSF4) gene in a Chinese family with autosomal dominant congenital cataract (ADCC). All exons of HSF4 were amplified by PCR. Sequence analysis of PCR products was performed. Restriction fragment length polymorphism (RFLP) analysis was conducted to confirm the pathogenic mutation. The results showed that a C to T substitution occurred at nucleotide 331 in patients of this family, leading to the replacement of the amino acid arginine-111 with cysteine in exon 3. RFLP analysis showed that the amino acid change was co-segregated with all affected individuals. It was concluded that the new mutation of c.331C>T in HSF4 DNA may be responsible for the autosomal dominant congenital cataract in this family.

In this paper, biotin photoaffinity labeled molecule with propargyl group 1 has been designed and synthesized, followed it's labeling of N2-acetyl-2'-O-propargyl guanosine 9 by "click chemistry". This technology presents delight development potential in labeling of second messenger cyclic nucleotide, antisense oligonucleotide or siRNA.

ABSTRACT

Objective:To discuss the effect of AngⅡ on insulin gene expression IN RIN-m cells and its molecular mechanism. Methods:RIN-m cells were cultured and divided into three groups, including the control group, 100 nmol·L-1 AngⅡ group and losartan pretreatment group. After 24-hour incubation, insulin gene expression in RIN-m cells was detected by RT-PCR, the mean flu-orescent intensity of 2', 7'-dichlorofluorescein ( DCF) was detected by flow cytometry, PDX-1 and MafA mRNA expression were detec-ted by RT-PCR and the protein expression was detected by Western-blot. Results: Insulin expression in RIN-m cells, cellular ROS level, PDX-1 expression and MafA expression in 100 nmol · L-1 AngⅡ group were significantly different from those in the control group and losartan pretreatment group (P0. 05). Conclusion:AngⅡ can down-regulate PDX-1 and MafA expression inβ-cells through oxidative stress pathway, and then inhibit insu-lin gene expression. Pretreatment with losartan can antagonize the effect of AngⅡ, and has protective effect onβ-cells in the aspect of insulin gene expression.

ABSTRACT

Cyclic diguanylate (c-di-GMP) is a ubiquitous second messenger present in a wide variety of bacteria, which is responsible for cell differentiation, biofilm formation, pathogenic factor generation, and so on. The level of c-di-GMP in bacteria is regulated by two opposing active domains, diguanylate cyclase (DGC) and phosphodiesterase (PDE), which are present in the same bifunctional protein, and in charge of the synthesis and the degradation of c-di-GMP, respectively. The target of c-di-GMP in the bacterial cell consists of PilZ domain and GEMM riboswitch, the only riboswitch that involved in signal transduction. This article gives an overview of c-di-GMP, focusing on its metabolic pathway, regulatory mechanism, biological function of c-di-GMP, and the synthesis of c-di-GMP analogues and their biological activity.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the sensitivity and specificity of Golgi protein 73 (GP73) monoclonal antibody in the diagnosis of hepatocellular carcinoma (HCC).</p><p><b>METHODS</b>Self-prepared GP73 monoclonal antibody was used as the primary antibody for detecting the serum GP73 levels in healthy controls(n=31)and HCC patients (n=59). The baseline level of the healthy controls was determined by semiquantitative analysis. The results were compared with those from GP73 polyclonal antibody and alpha-fetoprotein (AFP).</p><p><b>RESULTS</b>The GP73 level of healthy controls was 1.2 (0.9-1.7) relative unit (RU), which was significantly lower than that of HCC patients [5.7 (2.5-7.8) RU] (P<0.001) with monoclonal antibody. Using polyclonal antibody, the GP73 level of HCC patients was also significantly higher than healthy controls [7.8 (3.0-12.4) RU vs. 1.1 (1.0-2.0) RU, P<0.001]. The sensitivity and specificity of GP73 monoclonal antibody in diagnosis of HCC were 84.7% and 93.5%; on the contrary, those of GP73 polyclonal antibody were 78.0% and 93.5%, respectively. The sensitivity and specificity of AFP (67.8% and 74.2%, respectively) in the HCC patients were markedly lower than those of GP73. Logistic regression analysis showed that the odds ratio (OR) of GP73 monoclonal antibody was 7.18 and that of GP73 polyclonal antibody was 1.51.</p><p><b>CONCLUSIONS</b>Our self-prepared monoclonal antibody can effectively detect GP73 serum level in HCC patients, and has higher sensitivity and specificity than AFP. It may be superior to the currently used GP73 polyclonal antibody. The results lay the foundation for the further development of ELISA methods by using this monoclonal antibody.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To establish a gas chromatography method for detecting the concentration of 1,1-dichloro-1-nitroethane in air of workplaces.</p><p><b>METHOD</b>1,1-dichloro-1-nitroethane in air of workplaces was collected by activated charcoal tube, absorbed using carbon disulfide and analyzed by Gas Chromatography (FID) with FFAP capillary column.</p><p><b>RESULTS</b>The linear rang of 1,1-dichloro-1-nitroethane in this method was 4.0-858.2 microg/ml, the linear regression formula was Y = 283X-1076, the correlation coefficient was 0.9999, the lowest detection concentration was 0.4 mg/m3 (3L sampling air), the relative standard deviation (RSD) was 1.8%-4.1%, the desorption efficiency was 88.5%-90.6%, the breakthrough volume was > 0.7 mg, the sampling efficiency was 100%, the samples could be kept at ambient temperature for at least 7 days.</p><p><b>CONCLUSION</b>The indicators of this method were conformed to the requirements of "Guide for establishing occupational health standards--Determination methods of air chemicals in workplace". This method could be used to detect 1,1-dichloro-1-nitroethane in air of workplaces.</p>

ABSTRACT

MircoRNAs (miRNAs) are widely existing inorganisms, non-coding small RNA. MircoR-NAs play important role in cell proliferation, apoptosis, stress resistance and fat metabolism. In vitro studies indicate that miRNAs are important factors in platinum resistance of ovarian cancer cell lines. However, the pathway of platinum resistance of ovarian cancers is complex. Studies find miRNA expression play important roles in the sensitivity of platinum resistance of ovarian cancer cell lines , and can be used as an early alert for platinum resistance. Changing the expression of miRNA even can reverse the platinum resistance . But the regular role of miRNAs in target genes network is still unconfirmed. Thus, more studies about miRNA in vivo and animal tumor models are needed to identify the function of miRNAs in platinum resistance of ovarian cancer.

ABSTRACT

<p><b>OBJECTIVE</b>To express and purify Schistosoma japonicum ribosomal protein S4(SjRPS4) in Escherichia coli, and assess its value in immunodiagnosis of Schistosomiasis japonica.</p><p><b>METHODS</b>Gene fragment of SjRPS4 was amplified by screening the cercaria cDNA library of Schistosoma japonicum. The target gene was cloned into the expressive vector pQE30 and transformed into E. coli M15. The recombinant protein expression was induced by isopropylthio-β-D-galactoside (IPTG). This fusion protein was purified by Ni(2+)-NTA chromatography and identified by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), Western blot and ELISA.</p><p><b>RESULTS</b>The plasmid pQE30/SjRPS4 was constructed successfully and expressed a SjRPS4 fusion protein in E. coli as showing a single special band on SDS-PAGE gel at Mr 30 × 10(3) position. It reached a purity of above 90% after purification. The Western blot result confirmed that the recombinant protein could specifically react with the serum samples from patients of schistosomiasis. Detecting the serum of Schistosomiasis japonica patients by ELISA, the sensitivity and specificity of the ELISA method were 90.91% (70/77) and 92.59% (25/27), the positive rate of recombinant protein expression was 67.30% (70/104). There was no cross-reaction with paragonimiasis patients' serum.</p><p><b>CONCLUSION</b>Protein SjRPS4 was successfully cloned and expressed, and it was confirmed that SjRPS4 antibodies were valuable in the diagnosis of Schistosomiasis japonica.</p>

ABSTRACT

OBJECTIVE To analyze the isolation rate and four consecutive years′(2004-2007) antibiotic-resistance trend of extended-spectrum ?-lactamases(ESBLs) producing Escherichia coli in urinary tract infection and to provide evidence for anti-infection therapy.METHODS The anti-microbial susceptibility test results of E.coli cultured from the midstream urine of outpatients and inpatients of our hospital from 2004 to 2007 were collected.Whonet 5.4 statistical analysis software was used for analysis.RESULTS During four years,no apparent change has been shown about antibiotic resistant rate of ESBLs producing E.coli.The isolation rate of this kind of E.coli increased year by year from 31% to 44% and showed high resistance to third generation cephalosporins and quinolones antibiotics,but was 100% sensitive to imipenem.CONCLUSIONS With the ESBLs-producing E.coli increasing year by year,the antibiotic-resistance rate of E.coli isolated from urinary tract infection is apparently increased and thus screening of antibiotics sensitive to enzyme-producing bacterial is the key point when the empirical therapy especially when the third and fourth generation cephalosporins are ineffective.
