ABSTRACT

Plant stem cells are the cells that are located in meristems and are kept in a state of undifferentiation. Plant stem cell possesses lower vacuolization, higher mitochondrial activity, more genetic stability and stronger self-renewal capacity compared with calli. Plant stem cell culture has a wide application in pharmaceutical, functional food as well as cosmetic industries. Here we describe the procedure of induction, isolation and identification of plant stem cells, to provide a reference for further research in this field.

ABSTRACT

Objective To study relationship of expression of Survivin and cyclooxygenase(COX)-2 in the tissue of non-small cell lung cancer (NSCLC) with differentiation, lymph node metastasis and clinical stage. Method The expression of Survivin and COX-2 were detected by immunohistochemical SP staining in the specimens of 43 cases of NSCLC and 20 cases of para-cancerous lung tissue. Results The positive rate of Survivin and COX-2 was 67.4% (29/43) and 55.8% (24/43) respectively in NSCLC,was 0 respectively in the para-cancerous lung tissue. Compared with para-cancerous lung tissue,there was statistically significant difference(P< 0.01). Both Survivin and COX-2 had significantly higher expression in NSCLC with poor differentiation,lymph node metastasis and clinical stage of Ⅱ-Ⅲ than those with well differentiation,without metastasis and clinical stage of Ⅰ (P <0.05). Conclusion The expression of Survivin and COX-2 are high in NSCLC,they both have relationship with differentiation,lymph node metastasis and clinical stage of NSCLC.

ABSTRACT

Objective To study the application of stomach tube insertion in the lateral recumbent position in rescuing the unconscious patients,to reduce the discomfort of patients and increase the one-time success rate in the insertion of stomach tube. Methods 100 cases of unconscious patients were divided into the control group and the treatment group with 50 cases in each, they adopted routine tube insertion method and tube insertion in the lateral recumbent position.The success rate of tube insertion and incidence of complications were collected and analyzed in the two groups.Results The experimental group had a higher success rate and less complications compared with the control group.Conclusions The lateral recumbent position for stomach tube insertion is the ideal position for the unconscious patients.

ABSTRACT

Acid fibroblast growth factor (aFGF) has great potential in clinical application, but it is very expensive. In order to reduce the cost of production and to make full use of the merits integrated with plant bioreator, we have explored the aFGF in transgenic Tobacco expression. AFGF gene was inserted into plant expression vector pBI121; the acquired plants contained aFGF gene expression vector pBI121-TOAB-aF. Using Agrobacterium-mediated gene transformation of Tobacco and using transgenic Tobacco containing kanamycin and cephalosporin culture medium, we obtained kanamycin resistant transgenic Tobacco plants. PCR detection, RT-PCR detection and Western blot detection confirmed that foreign genes were successfully expressed in Tobacco. These data could serve as a theoretical foundation on which to use the plant bioreactor for production of aFGF.

ABSTRACT

<p><b>OBJECTIVE</b>To establish a high-frequency regeneration system of Astragalus and an aFGF transformation system.</p><p><b>METHOD</b>Cotyledon node of the Astragalus explants was used for organogenesis to establish a high-frequency regeneration system. GV3101 was used to transform cotyledon node, and aFGF gene was introduced into Astragalus, renewable strain was detected by PCR.</p><p><b>RESULT</b>All cotyledon node was explants, adventitious buds were induced in the medium of MS +2.0 mg x L(-1) BA +0.5 mg x L(-1) IBA, the root was taken in the medium of 1/2MS +5.0 mg x L(-1) NAA to give a high frequency regeneration system. All cotyledon node was precultured in medium for 3 days and infected with Agrobacterium (A600 0.3) for 10 min and then cocultured for 2 days. The aFGF gene was confirmed to transform into genome of Astragalus.</p><p><b>CONCLUSION</b>A high-frequency regeneration system of Astragalus and an aFGF transformation system were established.</p>
