ABSTRACT

Methotrexate(MTX) is a commonly used antimetabolite, which can be used in the treatment of a variety of diseases. However, hepatotoxicity in the use of MTX severely limits its clinical use. Therefore, how to prevent and treat hepatotoxicity of MTX has become an urgent clinical problem. This paper summarizes and analyzes relevant literatures on the prevention and treatment of hepa-totoxicity caused by MTX with traditional Chinese medicines and natural medicines in recent years. MTX-induced hepatotoxicity mechanisms include folate pathway, oxidative stress damage and adenosine pathway, of which oxidative stress theory is the main research direction. A total of 14 kinds of traditional Chinese medicine and natural medicine extracts including white peony root, and 21 kinds of natural monomer compounds, including berberine, play an anti-MTX-induced hepatotoxic effect by resisting oxidative stress, inhibiting inflammation and regulating signal pathways. According to current studies on the prevention and treatment of hepatotoxicity induced by MTX with traditional Chinese medicines and natural medicines, there are insufficiencies, such as partial and superficial mechanism studies, inadequate combination of experimental research and clinical practice, non-standard experimental design and lack of application of advanced technologies and methods. This paper systematically reviewed the effects and mechanisms of traditional Chinese medicines and natural medicines against hepatotoxicity induced by MTX and defined current studies and deficiencies, in the expectation of proposing new study strategies and directions and providing scientific basis for rational clinical use of MTX and development of new drugs against MTX hepatotoxicity.

ABSTRACT

Objective:To explore the molecular mechanism of spleen-strengthening traditional Chinese medicine (TCM) Weichang'an granule in inhibiting the invasion and metastasis of human gastric cancer MKN45 cells. Method:MKN45 cells were cultured in vitro and incubated with different concentrations(600, 900, 1 200, 1 500 mg·L-1)of Weichang'an granule for 24, 48, 72 h. Cell counting kit-8 (CCK-8) assay was used to detect its effect on the cell proliferation. Western blot was used to detect the expression of RUN and FYVE domain containing 3(RUFY3) in normal gastric mucosa cells and different gastric cancer cell lines. The expression of RUFY3 in the gastric cancer cells after Weichang'an granule intervention (600, 900, 1 200 mg·L-1) was detected by Western blot. Lentivirus transfection technique was used to achieve the stable and silenced expression of RUFY3 in gastric cancer MKN45 cells. Transwell assay was used to evaluate the influence of Weichang'an granule and silenced RUFY3 on the metastasis and invasion ability of MKN45. E-cadherin,N-cadherin,Vimentin,Zinc-finger transcription factor (SNAIL1 and SNAIL2) protein expression levels were detected by Western blot. Result:RUFY3 expression in human gastric cancer cells was significantly higher than that in normal gastric mucosa. The protein expression of RUFY3 in MKN45 cells of silenced RUFY3 group was significantly lower than that in Lentivirus negative group (P<0.01). Weichang'an granule inhibited the expression of RUFY3 in human MKN45 gastric cancer cells (P<0.05, P<0.01) in a concentration-dependent manner. As compared with the blank group, both Weichang'an granule and silenced RUFY3 inhibited the metastasis and invasion ability of MKN45 (P<0.01). After Weichang'an granule and silenced RUFY3 treatment, the protein expression of epithelial marker gene E-cadherin was up-regulated (P<0.01), the protein expression of Vimentin and N-cadherin decreased, but with no statistical difference, while SNAIL1 and SNAIL2 were both significantly down-regulated (P<0.01). Conclusion:By targeting RUFY3 to regulate epithelial mesenchymal transformation, the spleen-strengthening TCM compound Weichang'an granule can inhibit the invasion and metastasis of gastric cancer.

ABSTRACT

This study aimed to investigate the effect of methyl eugenol(ME) on hypoxia/reoxygenation(H/R)-induced injury of human renal tubular epithelial HK-2 cells and its mechanism. The viability of HK-2 cells cultured with different concentrations of ME and exposed to H/R was detected by cell counting kit-8(CCK-8) assay. The effect of ME on the morphology of HK-2 cells was observed under an inverted microscope. The content of intracellular reactive oxygen species in different groups was detected after 2',7'-dichlorodihydrofluorescein diacetate(DCFH-DA) fluorescence staining. Cell apoptosis was determined by flow cytometry. Changes in mitochondrial membrane potential were monitored by JC-1 dye. The concentrations of nuclear factor erythroid 2 related factor 2(Nrf2), heme oxygenase-1(HO-1), and nicotinamide adenine dinucleotide phosphatase oxidase 4(Nox4) were measured by Western blot, followed by the assay of Nrf2 concentration changes in cytoplasm and nucleus by confocal fluorescence staining. The results showed that when the concentration of ME was 0-40 μmol·L~(-1), the activity of HK-2 cells was not affected. Compared with the model group, ME enhanced the activity of HK-2 cells and the cell morphology was normal. As revealed by further experiments, ME inhibited the release of reactive oxygen species and the decline in mitochondrial membrane potential of HK-2 cells after H/R injury, promoted Nrf2/HO-1 expression and Nrf2 translocation to the nucleus, and down-regulated the expression of Nox4, thereby significantly reducing apoptosis. This protective effect of ME could be reversed by the specific Nrf2 inhibitor ML385. These findings have preliminarily proved that ME effectively protected HK-2 cells against H/R injury, which might be related to its promotion of Nrf2/HO-1 signaling pathway and inhibition of Nox4. Such exploration on the possible mechanism of ME in the treatment of renal ischemia-reperfusion injury(IRI) and protection of organ function from the perspective of antioxidant stress has provided reference for related research on the treatment of acute kidney injury with traditional Chinese medicine.

ABSTRACT

<b>Objective::To explore the effect of Weichang' an (WCA) on the autophagy of human gastric cancer MKN45 cells and its possible anti-cancer mechanism. <b>Method::MKN45 cells were cultured in vitro and incubated with different concentrations (250, 500, 1 000, 2 000 mg·L-1) of WCA for 24, 48, 72 h. Cell counting kit-8 (CCK-8) assay was used to detect the cell proliferation. AO/EB Dyeing (AO) staining and monodansylcadaverin (MDC) staining were used to observe the changes of the effect of WCA on autophagosome and autophagic vesicles in gastric cancer MKN45 cells at 48 h. Real-time polymerase chain reaction (Real-time PCR) and Western blot were used to detect microtubule-associated protein 1 light chain 3 (LC3), Beclin1, sequestosome 1 (p62), human autophagy-related gene 5 (ATG5), human autophagy-related gene 7 (ATG7) mRNA and protein expression levels. <b>Result::WCA showed a dose-and-time-dependent growth inhibition at the concentration above 1 000 mg·L-1. Compared with the blank group, WCA (500, 1 000, 2 000 mg·L-1) significantly inhibited the proliferation of MKN45 cells (P<0.05, P<0.01). AO staining and MDC staining showed that autophagosomes and autophagic sacs increased with the rise of WCA concentration compared with the blank group. Real-time PCR and Western blot showed that the expressions of autophagy-related proteins LC3-Ⅱ, Beclin1, ATG5, ATG7, mRNA and protein increased gradually after WCA (1 000 mg·L-1) intervention, while p62 expression decreased (P<0.05, P<0.01). <b>Conclusion::WCA induces the autophagy of human gastric cancer MKN45 cells in a time-and dose-dependent manner in vitro, which may be related to the up-regulation of LC3-Ⅱ, Beclin1, ATG5 and ATG7 as well as the down-regulation of p62 and LC3-Ⅰ.

ABSTRACT

The application of prostate-specific antigen (PSA) in the screening and diagnosis of prostate cancer (PCa) has improved the clinical management of PCa patients. However, the PSA assay has been faced with criticism due to its potential association with over-diagnosis and subsequent overtreatment of indolent patients.Matrix metalloproteinase-26 (MMP26) is a member of matrix metalloproteinases (MMPs) and has been reported to be highly expressed in many cancers. This investigation evaluated the potential of serum MMP26 as a biomarker for PCa.The level of serum MMP26 was measured by enzyme-linked immunosorbent assay (ELISA) in 160 subjects including PCa group (n=80),benign prostatic hyperplasia (BPH) group (n=40) and control group (n=40).Furthermore,we evaluated the expression of MMP26 in tissues by immunohistochemistry. The results showed the serum MMP26 levels were significantly higher in PCa group than in BPH group and control group.Similarly, the MMP26 protein was positive in PCa tissues and negative in BPH tissues and control tissues. In conclusion, these results suggested MMP26 could be used as a potential serum biomarker in the diagnosis of PCa.

ABSTRACT

Glycosides are the active ingredients (AIs) of many Chinese herbs and have become hot spots along with the findings of their new functions, such as anti-inflammatory,antivirus,enhanced immunity and anti-cancer.It has been found that glycosides exert their effects by converting to aglycons or other AIs in vivo. Therefore, the transformation of glycosides to the corresponding AIs in vitro becomes very important to enhance their bioavailabilities. The microbial transformation has an unparalleled advantage in the transformation of Chinese herbs in vitro for its reaction specificity, less by-products,mild reaction conditions and environmental protection. This paper summarized and prospected researches of glycosides' microbial transformation.

ABSTRACT

Objective To analyze the differences of coagulation parameters between patients with advanced schistosomiasis cirrhosis and hepatitis B cirrhosis under different degrees of liver function,so as to provide the evidence for further guidance of judging clinical condition and prognosis. Methods Sixty?three patients with advanced schistosomiasis cirrhosis and eighty pa?tients with hepatitis B induced cirrhosis hospitalized in Jingzhou Central Hospital from January 2014 to June 2016 were served as an advanced schistosomiasis cirrhosis group and a hepatitis B cirrhosis group, respectively, and ninety?six gastropathy pa?tients excluded from other diseases that might affect coagulation in the same period were served as a control group. The levels of PT,INR,Fib,TT,APTT,and PLT of the patients among the above 3 groups and the cirrhosis patients with different Child?Pugh classes were detected and compared. Results The differences of the levels of PT,INR,Fib,TT,APTT,and PLT among the 3 groups were statistically significant(F=84.512,81.672,37.612,104.475,52.497,102.233;all P 0.05). Under all the grades of liver function,TT and APTT of the hepatitis B induced cirrhosis group was longer than those of the advanced schistosomiasis cirrhosis group ,and the Fib level of the former was lower than that of the latter. Among those with the liver function of Grade A and B ,PT and INR of the hepatitis B induced cirrhosis group were longer than those of the advanced schistosomiasis cirrhosis group;while for those with the liver function of Grade C,the difference between the two groups was not statistically significant(all P > 0.05). Conclu?sions There exist differences of the damage degrees of coagulation function between the patients with advanced schistosomiasis cirrhosis and hepatitis B cirrhosis. When the damage degree of the liver function is lighter, the coagulation function in the pa?tients with hepatitis B cirrhosis decreases more significantly;when the damage degree becomes more severe,APPT in the pa?tients with hepatitis B cirrhosis extends more significantly, while the differences of PLT and PT between them are little.

ABSTRACT

OBJECTIVE: To enhance the anticancer activity of doxorubicin(DOX) by conjugating DOX and vitamin E succinate (VES) and loading the conjugate into hyaluronic acid-octadecylamine (HA-C18) copolymer micelles. METHODS: DOX and VES were conjugated by amide reaction. DOX-VES/HA-C18micelles were prepared via a probe-type ultrasonication technique. The morphology of the micelles was determined using a transmission electron microscopy (TEM).Dynamic light scattering (DLS) technique was used to determine the particle size distribution, hydrodynamic diameters, and stability of the micelles. Ultracentrifugation was exploited for measuring the drug loading (DL) and encapsulation efficiency (EE), and the in vitro release was investigated using a dialysis tubing. The cellular uptake and cellular distribution of drug-loaded micelles in MCF-7 cells were observed by fluorescence microscope, and the fluorescence intensity of DOX was evaluated by flow cytometer. The cytotoxicity of free DOX and drug-loaded micelles was tested by MTT assay against MCF-7 cells. RESULTS: DOX-VES/HA-C18 showed a nearly spherical morphology and good stability in PBS (pH 7.4) and 10% FBS. The particle size and zeta potential were (184.6±9.42) nm and (-20.7±1.23) mV, respectively. The DL and EE were (15.8±2.85)% and (94.2±1.32)%, respectively. DOX-VES/HA-C18had a good controlled drug release property. Furthermore, DOX-VES/HA-C18with accumulation in nucleipresented higher anti-tumor activity than free DOX and DOX/HA-C18. CONCLUSION: DOX-VES conjugate has synergistic anti-tumor effect and good application prospects.
