ABSTRACT

Background In the context of improving urban environment for healthy aging, it is necessary to rationally plan and provide community living space and public service facilities suitable for the elderly, and constantly optimize the built environment towards an age-friendly city. Objective To understand the relationship between community built environment and obesity in the elderly in Longgang City, and to provide a reference basis for improving the health of the elderly. Methods Elderly adults aged 60-90 years (n=6527) who completed a physical examination during the period from October 2020 to January 2021 in Longgang City were surveyed, and data on height and weight, waist circumference (WC), and other sociological demographic characteristics were obtained. Overweight was determined by 24 kg·m−2 ≤ body mass index (BMI) < 28 kg·m−2 and obesity by BMI ≥ 28 kg·m−2. Men with WC ≥ 85 cm and women with WC ≥ 80 cm were considered central obesity. Based on the participants' residential addresses, geocoding was performed using a geographic information system, and built environment indicators such as restaurants, convenience stores, and basic medical facilities were obtained using Gaode Map. A binary logistic regression model with adjusted individual-level covariates was used to evaluate the relationship between obesity and built environment indicators among elderly adults by gender and age. Results Among the 6527 community elderly, 46.93% were male and 53.07% were female, with a mean age of (73.69±0.07) years, a mean BMI of (24.32±2.84) kg·m−2, and 51.92% of the elderly were overweight or obese. The regression results showed that for elderly men, the more convenience stores and the higher mixed land use in residential areas, the higher risk of central obesity; however, the increases in street connectivity and accessibility to parks and recreational areas were associated a decreased risk of central obesity. The prevalence of overweight/obesity was higher among elderly women with more convenience stores in residential areas, while increased street connectivity was associated with a lower prevalence of central obesity among elderly women. Accessibility to primary health care facilities was negatively associated with the risk of central obesity among the 60- to 70-year-olds. For elderly residents aged 71−80 years, higher mixed land use and better accessibility to transit stations were associated with a higher prevalence of overweight/obesity, while street connectivity was negatively associated with the central obesity. Proximity to parks and recreational areas was associated with a reduced risk of overweight/obesity among the 81- to 90-year-olds. Conclusion Among the variables of a 500-m neighborhood built environment, the number of convenience stores, mixed land use, street connectivity, accessibility to primary health care facilities, accessibility to public transit stations, and accessibility to parks and recreational areas are correlated with obesity among elderly residents, and the degree of influence varies by gender and age.

ABSTRACT

ObjectiveTo evaluate the lipid-lowering activity of Quansanqi tablets(QSQ), an innovative new drug of Panax notoginseng. MethodMice and golden hamsters were used to establish a hyperlipidemia model by injecting egg yolk milk and feeding high-fat diets. The levels of total cholesterol (TC),triglyceride (TG),low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C) were detected, and liver function indicators [alanine aminotransferase (ALT), aspartate amino-transferase (AST), and alkaline phosphatase (ALP)] of golden hamsters were detected. Hematoxylin-eosin (HE) staining was used to observe the degree of liver injury. In the experiments, a normal group, a model group, an atorvastatin calcium group, and low-, medium-, and high-dose QSQ groups (0.32, 0.64, 1.28 g·kg-1 for mice, and 0.16, 0.32, 0.64 g·kg-1 for golden hamsters) were set up. ResultCompared with the normal group, the acute hyperlipidemia model mice showed increased TC, TG, and LDL-C levels (P<0.01), and the hyperlipidemia model mice showed increased TC and LDL-C levels (P<0.01). Additionally, the hyperlipidemia model golden hamsters showed increased serum TC, TG, LDL-C, ALT, AST, and ALP levels (P<0.05, P<0.01). HE staining indicated the presence of fat accumulation in the liver, accompanied by inflammatory reactions. Compared with the model group, QSQ of various doses could reduce TC, TG, and LDL-C levels in acute hyperlipidemia model mice (P<0.05, P<0.01), and the high-dose QSQ could reduce TC and LDL-C levels (P<0.01) and increase HDL-C level (P<0.05) in hyperlipidemia model mice, as well as reduce TC, TG, and LDL-C levels in hyperlipidemia model golden hamsters (P<0.05, P<0.01), especially in the first two weeks. In addition, atorvastatin calcium could further increase ALT, AST, and ALP levels (P<0.05, P<0.01) and aggravate liver function damage, while low-dose QSQ could reduce ALT, AST, and ALP (P<0.05), and medium- and high-dose QSQ did not cause further liver function damage. ConclusionQSQ have a significant lipid-lowering effect on different hyperlipidemia model animals and can improve liver function and liver injury.

ABSTRACT

OBJECTIVE To establish the method for the content determination of 5-hydroxymethylfurfural (5-HMF) in glucosamine hydrochloride tablets, and to analyze its regularity and influential factors. METHODS Quantitative analysis of 5-HMF was performed using high-performance liquid chromatography. The analysis was conducted on Shim-pack GIST C18-AQ column with mobile phase consisted of 0.1% phosphoric acid solution-methanol (90∶10, V/V) at the flow rate of 1.0 mL/min. The column temperature was 30 ℃, and detection wavelength was 284 nm. The injection volume was 20 μL. Reaction kinetics test of different temperatures was adopted to analyze the relationship of 5-HMF content with reaction temperature and reaction time, and utilized to build its formation kinetic model. RESULTS The linger range of 5-HMF was 0.057-5.698 μg/mL (r=0.999 9). The limits of detection and quantitation were 5.70 and 17.09 ng/mL; RSDs of precision, repeatability and stability (24 h) tests were all lower than 1.0% (n=6). The average recoveries ranged from 99.38% to 99.73%(RSD=0.53%, n=9). The contents of the 5-HMF in 8 batches of samples ranged 4.10-35.13 μg/g. Results of data fitting in reaction kinetics test showed that the higher reaction temperature and the longer reaction time, the higher 5-HMF content in the sample. At 50, 60, 70 and 80 ℃ , the relationship between the content of 5-HMF and the reaction time was linear, in accordance with a zero-order kinetic model. The reaction rate constants were 6.789, 7.715, 8.815 and 11.430, respectively. CONCLUSIONS The established method has strong specificity, high sensitivity, and good accuracy; the reaction temperature and reaction time are important influential factors for the formation of 5-HMF in glucosamine hydrochloride tables. The change rule of its content conforms to the zero-order kinetic model.

ABSTRACT

RNA methylation is of great significance in the regulation of gene expression, among which the more important methylation modifiers are N6-methyladenosine (m6A) and 5-methylcytosine (m5C). The methylation process is mainly regulated by 3 kinds of proteins: methyltransferase, demethylase, and reader. m6A, m5C, and their related proteins have high abundance in the brain, and they have important roles in the development of the nervous system and the repair and remodeling of the vascular system. The neurovascular unit (NVU) is a unit of brain structure and function composed of neurons, capillaries, astrocytes, supporting cells, and extracellular matrix. The local microenvironment for NVU has an important role in nerve cell function repair, and the remodeling of NVU is of great significance in the prognosis of various neurological diseases.

ABSTRACT

Objective:To explore the etiology, pathogenesis and prescription of treatment methods of COVID-19 in traditional Chinese medicine (TCM), and to evaluate the value of integrated Chinese and western medicine for the treatment of the disease.Methods:According to the theory of five Yun and six Qi, regional climate characteristics and clinical evidence, the etiology and pathogenesis of COVID-19 were analyzed, and TCM treatment was established for clinical practices for 30 patients with COVID-19 in Huanggang Central hospital, during December 2019 and Janual 2020. Results:It is considered that COVID-19 belongs to the range of Plague in TCM theory, the etiology and pathogenesis are cold and dampness directly attacking the three yin channels, and internal constraint of deficient fire, so the disease name is called by cold-dampness pestilence. The prescription was Wumeiwan (Mume Pill) combined with Mahuang-Shengma Decoction (Ephedra and Cimicifuga Decoction) and Dayuanyin (Dayuan Beverage), and the treatment was modified according to pattern differentiation. A total of 30 patients were treated with pattern differentiation and treatment of TCM. Among which, one patient was treated only with herbal medicine, and the other 29 were treated based on the routine treatment of Western medicine. After treatment, 25 of the 30 patients had improved symptoms after taking 1 dose of TCM. After taking 7-10 doses, all patients had major symptoms ranging from improvement (8+4 cases) to complete disappearance (18 cases), 13 cases met the discharge criteria, including the one who was treated only with herbal medicine. Among all the patients, 5 of the 6 severe turned to moderate and 1 turned to mild. Conclusions:The pathogenesis of COVID-19 is cold and dampness directly attacking the three yin channels, and internal constraint of deficient fire, and then Jue-Yin Disease, which belongs to the category of cold-damp pestilence. The method of integrated Chinese and Western medicine can rapidly improve the symptoms and the clinical effect is satisfactory.

ABSTRACT

Objective Effect of hawthorn and melanoidins on the in-vitro growth of Bifidobacterium and E.coli. Methods According to methods of the Chinese pharmacopoeia (2015),the charred hawthorn was prepared. The melanoidins in charred hawthorn were separated and purified by the macroporous resin extraction process. Ultraviolet spectrophotometry was used to detect melanoidins. The gas chromatography was used to detect the effects of hawthorn, charred hawthorn and melanoidins on the content of the acetic acid in Bifidobacterium and E.coli during growth, stable and decay period. Results In the early stage, the effects of hawthorn and charred hawthorn on bacteria were greater than melanoidins. In the middle and late stage, melanoidins inhibited the growth and metabolism of E.coli by changing the generation of acetic acid, and contributed to that of Bifidobacterium and also promoted the generation of acetic acid and regulate the intestinal flora. Conclusion Hawthorn, charred hawthorn and melanoidins all promote digestion by promoting the growth and metabolism of intestinal flora. Among them, charred hawthorn has a better effect on intestinal flora.

ABSTRACT

Objective To study the friction between brackets and archwires and the influential factors. Methods To test the friction of six combinations of canine bracket and stainless steel archwire, and to calculate the critical contact angles for each combination. Results The friction maintains stability with slight changes when the contact angle is smaller than the critical contact angle. The friction increases linearly when the contact angle is larger than the critical contact angle. The friction of different combinations of bracket-archwire are different and the critical contact angles are also different. Conclusion The critical contact angle influences the friction between bracket and archwire significantly. The critical contact angle is relevant to both the structure of the bracket and the size of the archwire.

Lupus-like syndrome is characterized by positive anti-nuclear antibody and clinical manifestations similar with systemic lupus erythematosus (SLE).Primary immunodeficiencies (PID) are specific underlying causes of lupus-like syndrome in children.Various infections,drugs,and tumors can also have manifestations like SLE.Treatment for the underlying disease is recommended.Whether immunosuppressive therapies should be initiated depending on the condition of the patient.

ABSTRACT

Objective: To explore the effect of the Epstein-Barr virus (EBV) BZLF1 gene on the biological behavior of EBV-negative gas-tric cancer cells and the underlying molecular mechanism. Methods: Lentiviral overexpressing BZLF1 was used to infect AGS and HGC27 gastric cancer cell lines. And the cell proliferation, apoptosis and migration invasive ability, and expression changes of cell sig-naling pathway were detected by CCK8 assay, apoptosis detection, migration and invasion assay, as well as western blot. HGC27 cells overexpressing BZLF1 were injected into the dorsal of NOD/SCID mice to construct xenografts, and the effect of BZLF1 on tumor growth was observed. Results: The expression of BZLF1 protein was significantly up-regulated in AGS-BZLF1 and HGC27-BZLF1 infected by over-expressing BZLF1 lentivirus. The cell proliferation in vitro and the tumorigenic ability in mice were significantly increased (P<0.05). Apoptosis was inhibited by BZLF1 protein, and the apoptotic rate of AGS-BZLF1 and HGC27-BZLF1 was (2.40±0.14)% and (3.90± 0.14)%, which was significantly lower than (5.75±0.35)% and (9.70 ± 0.42)% of AGS and HGC cells (P<0.05); however, there was no sig-nificant change in cell migration and invasion ability. In-depth molecular mechanism studies found that PI3K/AKT signaling pathway was significantly activated with enhanced pAKT and pS6 expression. After blocking the PI3K/AKT signaling pathway with BEZ235 inhibi-tor, the growth of HGC27-BZLF1 and AGS-BZLF1 cells was inhibited. Conclusions: EBV BZLF1 may promote the growth of gastric cancer cells by activating the PI3K/AKT signaling pathway and targeting PI3K/AKT pathway inhibitors, and serve as a promising treatment op-tion for EBV-associated gastric carcinoma.

ABSTRACT

Objective To systematically evaluate the postoperative complications following open reduction and internal fixation combined with vascularized bone flap graft versus only reduction and internal fixation for femoral neck fractures.Methods CNKI,Wan Fang Chinese database,Pubmed,EMBASE and Google Scholar English database were searched for the randomized controlled trials from January 1,2000 through October 31,2017 which compared open reduction and internal fixation combined with vascularized bone flap graft (combined surgery group) with only reduction and internal fixation (simple surgery group) for femoral neck fractures.The data concerning postoperative nonunion and avascular necrosis of the femoral head were extracted.The 2 surgical treatments of the patients with femoral neck fracture were compared in terms of the 2 complications.Statistical analyses were conducted using software Stata 12 by relative risk (RR) and corresponding 95% confidence intervals (95% CI).Results According to our inclusion and exclusion criteria,a total of 23 studies were included,involving 2,162 cases (1,048 cases receiving combined surgery and 1,114 cases simple surgery).The Meta analyses showed that the fracture nonunion rate for the combined surgery group was significantly lower than that for the simple surgery group [RR =0.27,95% CI(0.19,0.38),P < 0.001] and the rate of avascular necrosis was also significantly lower for the former than for the latter [RR =0.31,95 % CI(0.24,0.42),P < 0.001].Conclusion In the treatment of femoral neck fractures,open reduction and internal fixation combined with vascularied bone graft may lead to lower rates of nonunion and avascular necrosis of the femoral head than simple open reduction and internal fixation.

ABSTRACT

Objective To investigate the efficacy of calf self weight traction reduction combined with locking plate fixation for the treatment of intertrochanteric fractures in the elderly.Methods A retrospective case series study was conducted on the clinical records of 174 elderly patients with the modified Evans Ⅰ-Ⅲ types of fresh intertrochanteric fractures treated with locking plate from January 2012 to December 2015.According to treatment method,the patients were assigned to traction bed reduction with locking plate fixation (Group A,62 patients) and calf self weight traction reduction with locking plate fixation (Group B,112 patients).Group A comprised 32 males and 30 females,with age range of 65-91 years [(72.47 ± 6.35) years].Group B comprised 68 males and 44 females,with age range of 65-95 years [(73.23 ± 6.05) years].The time of reduction,operation time,incision length,intraoperative blood loss,frequency of fluoroscopy,postoperative drainage volume,hospital stay,postoperative weightbearing standing time or walking time (ambulation time),surgical complications,and fracture healing were recorded.Harris and modified Barthel index score in Chinese (MBI-C) were used to evaluate the functional recovery of hip joint.Results All patients were followed up for 5-61 months (mean,15 months),and noted with fracture healing.The time of reduction in Group A was (13.27 ± 3.03) minutes,longer than that in Group B (0 minute) (P <0.05).The operation time in Group A was (63.63 ± 13.90)minutes,longer than that in Group B [(59.62 ± 8.38) minutes] (P < 0.05).Fluoroscopy in Group A was (5.35 ± 2.36) times,more than (4.28 ± 3.11) times in Group B (P < 0.05).There were no significant differences in the incision length,intraoperative blood loss,postoperative drainage volume,ambulation time,fracture healing time,Harris score,and MBI-C index between the two groups (P >0.05).There were no significant differences in the postoperative complications such as deep venous thrombosis,pulmonary infection,incisional infection,urinary tract infection,delirium,bed sores,cardiac insufficiency,electrolyte disturbance,and postoperative plate rupture between the two groups (P>0.05).The incidence of deep vein thrombosis was 9.7% (6/62) in Group A,and 4.5% (5/112) in Group B (P >0.05).No screw fracture,nail and plate combination failure,bone nonunion,or screw cut out of the femoral head were observed in both groups.Conclusions For the modified Evans Ⅰ-Ⅲ types of intertrochanteric fractures,both traction bed reduction and calf weight reduction with locking plate have equivalent efficacy.However,the latter method has advantages of shorter reduction and operation time and less intraoperative X-ray exposure,and hence is worthy of clinical application.

ABSTRACT

In 2014, The Cancer Genome Atlas (TCGA) classified gastric cancer into four types based on the genotype include EBV-infect-ed tumors, genomically stable tumors, chromosomally unstable tumors and MSI tumors, among which Epstein-Barr virus-associated gastric cancer (EBVaGC) is expected to be most suitable for immunotherapy because of its molecular characteristics, such as PD-L1/2 amplification or upregulation. Understanding the molecular characterizations of EBVaGC is very important in clinical practice. The re-ported molecular characteristics of EBVaGC include: PD-L1/2 upregulation, PIK3CA mutation, ARID1A mutation, DNA hypermethyl-ation, and rare mutation of TP53. Therefore, the possible therapeutic strategies for EBVaGC include the use of immune checkpoint in-hibitors, PI3K/AKT signaling pathway inhibitors, or antiviral therapy. This review summarizes the important molecular characterizations and possible treatment strategies for EBVaGC.

Neurogenesis is a process in which the neuronal stem cells differentiate into functional neurons including the cell proliferation,differentiation and migration.Previously,it was believed that neurogenesis is a prenatal process and the adult ependymal cells are incapable of regeneration.Now it is clear that mammalian brain retains the ability to generate new ceils in specific regions.One of the regions is subventricular zone of the lateral ventricles,new generated neurons and glial cells later migrate to olfactory and repair dysosmia through the RMS road.Here we will review the advances in adult neurogenesis in mammal subventficular zone.

ABSTRACT

Objective To explore the clinical and genetic characteristics of congenital generalized lipodystrophy (CGL). Method The clinical data of one child with CGL caused by BSCL2 gene mutation were analyzed retrospectively and relative literature were reviewed. Results A 2-year-9-month old girl had clinical manifestations of a lack of subcutaneous fat, acanthosis nigricans, hepatolienomegaly and mild hypophrenia. Laboratory examinations showed hypertriglyceridemia, hyperinsulinemia and cardiomyopathy. The peripheral blood from the child and her parents were collected and 4 genes, AGPAT2, BSCL2, CAV1 and PTRF, were sequenced by Sanger. The results showed a heterozygous mutation of BSCL2 gene from maternal frameshift (c.567-568delGA, p.E189EfsX12) and paternal nonsense mutation (c.565G>T,p.E189X) respectively in the child, and both mutations were pathogenic ones. By a literature review, it is known that BSCL2 gene mutation is the most common cause of in Asian. In CGL with BSCL2 gene mutation, the commom clinical manifestations include disappearance of systemic adipose tissue, acathosis nigricans and hepatomegaly, and the incidence of myocardial infarction and mental retardation were 40% and 30% respectively. Conclusion The main clinical manifestations of CGL caused by BSCL2 gene mutation were loss of systemic adipose tissue and metabolic disorder at an early age. It was often accompanied by myocardial lesions and mental retardation. Gene diagnosis analysis should be made as earliest possible time for the children suspected of this disease.

ABSTRACT

Objective To explore the clinical efficacy of 1251 stent implantation in treatment of malignant tracheal stenosis with the help of ventilation catheter under local anesthesia.Methods Totally 180 consecutive patients with malignant tracheal stenosis underwent 125I airway stent implantation were analyzed retrospectively.The anhelation grade,oxygen saturation and respiratory rate before and after operation were recorded and analyzed,125I stent situation,clinical symptoms and survival situation were followed up after operation.Results The success rate of 125I stent placement was 100% (180/180),and the stent type was tubular stent in 132 cases,Y-shaped stent in 34 cases,and L-shaped stent in 14 cases.Dyspnea was significantly relived in all patients.Oxygen saturation and respiratory rate improved from (80.60±3.87)% and (29.36± 3.20)times/min before operation to (94.31±3.40)% and (19.29±2.19)times/min after operation (t=-30.52,35.09,both P<0.01).Patients were followed up 3-13 mouth after operation,and stent restenosis were occurred in 6 patients.The lifetime was 49-401 days and the average lifetime was (182±94) days.The 60-day survival rate was 0.99 and the 180-day survival rate was 0.65.Conclusion 125I stent implantation in treatment of malignant tracheal stenosis with the help of ventilation catheter under local anesthesia is a safe and effective method.

Objective To evaluate the safety and feasibility of 125I seeds stent implantation with assistance of airway tube under local anesthesia in treating malignant tracheobronchial stenosis.Methods The clinical data of 24 patients with malignant tracheal and bronchial stenosis,who underwent the treatment of 125I seed stent implantation at authors' institute during the period from March 2014 to January 2015,were retrospectively analyzed.The primary lesions included tracheal and bronchial cancer (n=6),esophageal carcinoma (n=8),pulmonary cancer associated with mediastinal lymph node metastasis (n=9),and malignant mediastinal tumor (n=l).All patients had different degrees of shortness of breath,cough and other symptoms due to airway constriction caused by malignant tumors.Grade Ⅱ of breath shortness was seen in 2 patients,grade Ⅲ in 16 patients,and grade Ⅳ in 6 patients.First,a 4F single-curve catheter was placed into one of the main bronchi,which was used as the ventilation catheter,then implantation of 125I seed stent in the diseased trachea and bronchus was carried out,finally,after the implantation of 125I seed stent was successfully accomplished the ventilation catheter was removed.The technical success rate of 125l seed stent implantation,the procedure-related complications and the intraoperative patient tolerance were calculated and the results were analyzed.Results Successful implantation of 125I seed stent with single procedure was accomplished in all 24 patients and no severe operation-related complications occurred.The technical success rate was 100%.Under the assistance of ventilation catheter,the oxygen saturation maintained at the level around 92.9% when the stent release device was passing through the airway stenotic site.Neither intraoperative asphyxia nor severe decrease in blood oxygen saturation occurred in all patients.After the treatment,6 patients had bloody sputum,2 patients developed irritable cough,and one patient complained of mild chest pain,these symptoms were relieved after symptomatic treatment.During operation,no events that caused the patients unable to cooperate with the treatment,such as severe cough,dysphoria,dyspnea,etc.,occurred.Conclusion For the treatment of malignant airway stenosis,125I seed stent implantation with assistance of airway tube under local anesthesia is safe and feasible.

ABSTRACT

Objective To evaluate the clinical effect of reduced glutathione in protecting the liver function for patients with hepatocellular carcinoma (HCC) after receiving international therapy.Methods The clinical data of 110 HCC patients,who were admitted to authors' hospital during the period from January 2010 to June 2013,were retrospectively analyzed.According to the random number table method the patients were divided into the control group (n=50) and the treatment group (n=60),and transcatheter arterial chemoembolzation (TACE) and transcatheter arterial infusion (TAI) were carried out in all patients.After the treatment,intravenous infusion of 2.0 g vitamin C,0.2 g vitamin B6,2.0 g inosine,and 250 ml branched chain amino acid was employed per day for one week for the patients of both groups,and additional intravenous dripping of 1.8 g reduced glutathione (added in 250 ml of 5% glucose solution) was adopted for the patients of the treatment group.Various liver function indexes were tested before and one week after the treatment,the results were compared between the two groups.Results Before the treatment,no obvious differences in the hepatic functions and in the clinical condition existed between the two groups.One week after the treatment,in both groups the aminotransferase (ALT and AST) and total bilirubin (TBIL) were significantly increased when compared with the preoperative data (P<0.05),while the serum pre-albumin (PA) was remarkably decreased when compared with the preoperative data (P<0.05);these changes of various indexes in the treatment group were apparently better than those in the control group (P<0.05).Conclusion The use of reduced glutathione can effectively relieve the liver function damage caused by interventional therapy in HCC patients,therefore,reduced glutathione has a very good clinical application value.

ABSTRACT

A quantitative method for clopidogrel using online-SPE tandem LC-MS/MS was developed and fully validated according to the well-established FDA guidelines. The method achieves adequate sensitivity for pharmacokinetic studies, with lower limit of quantifications (LLOQs) as low as 10 pg/mL. Chromatographic separations were performed on reversed phase columns Kromasil Eternity-2.5-C18-UHPLC for both methods. Positive electrospray ionization in multiple reaction monitoring (MRM) mode was employed for signal detection and a deuterated analogue (clopidogrel-d 4) was used as internal standard (IS). Adjustments in sample preparation, including introduction of an online-SPE system proved to be the most effective method to solve the analyte back-conversion in clinical samples. Pooled clinical samples (two levels) were prepared and successfully used as real-sample quality control (QC) in the validation of back-conversion testing under different conditions. The result showed that the real samples were stable in room temperature for 24 h. Linearity, precision, extraction recovery, matrix effect on spiked QC samples and stability tests on both spiked QCs and real sample QCs stored in different conditions met the acceptance criteria. This online-SPE method was successfully applied to a bioequivalence study of 75 mg single dose clopidogrel tablets in 48 healthy male subjects.

ABSTRACT

Objective:To investigate the effects of Rad50-targeted inhibition on the radiosensitivity of head and neck squamous cell carcinoma cells.Methods:Rad50 in Tca8113 and OSCC-15 cells was down-regulated by siRNA.Then the cells were treated by a small dose of radiation(2 Gy),Western blot was used to detect the expression of Rad50 protein.Comet assay was used to evaluate the DNA double strand breaks(DSBs).Colony survival assay was performed to detect the survival rate of the cells.Telomere FISH was performed to assess the telomere length in the cells.Results:Target siRNA inhibited Rad50 protein expression in Tca8113 and OSCC-15 cells. siRNA combined with the radiation of 2 Gy produced more DSBs,decreased the proliferation and shortened the nucleus telomere length of the cells more than radiation treatment alone.Conclusion:The targeted inhibition of Rad50 may increase the radiosensitivity of Tca8113 and OSCC-15 cells.

ABSTRACT

Objective To investigate and analyze the data of continuing medical education in order to understand the real need of the learners.Methods Reviewing and making statistic analysis of the data of Renji hospital's continuing medical education courses from 2007 to 2012,including the amount,title,discipline,geographical distribution,training content and the questionnaire survey.Results It shows that the learners from coastal and developed areas have a higher demand for cuttingedge technology and the new progress in medicine(49.1%),while the learners from inland and western regions prefer to learn the basic medical theory and technology (41.9%).Senior doctors tend to study cutting-edge technology and new progress medical courses (51.7%) while the junior persons prefer the basic medical knowledge (46.6%).These mean that the needs of content of continuing medical education is associated with the regions and the level of the doctors (P<0.05).Conclusion The conclusions prompt that medical development between different regions of China may be unbalanced.Chinese government should pay more attention in terms of allocation of health resources and personnel training.It also provides advice for future courses so that it may improve the efficiency of the medical education resources.
