ABSTRACT

Objective To study miRNA expression patterns in endometrial carcinoma(EC)combined with the Metabolic Syn-drome(MS)and non-combined with MS in order to explore the role of the microRNAs in EC combined with the MS.Methods Three fresh-frozen samples of endometrial carcinoma Combined with MS and 3 cases of non-Combined with MS and 1 case of nor - mal endometrium were collected.Total RNA was extracted from samples and miRNA microarray was used to evaluate the differenceof expression spectrum of miRNA.Results There were different expression of miRNA among the two groups of endometrium andnormal endometrium.There were 51 miRNA different expressions between EC combined with the MS and the normal endometri - um,and among them there are 15 upregulated miRNA and 36 downregulated miRNA.There were 9 miRNA different expressionsbetween EC non-combined with the MS and the normal endometrium ,and there are 3 upregulated miRNA and 6 downregulatedmiRNA among them.10 different miRNA were revealed between the two type of EC,and there are 4 upregulated miRNA and 6downregulated miRNA among them.Conclusion Different expressions of miRNA between EC Combined and non-Combined withMS may provide insights into their roles in the progression of EC .The study of miRNA contributes to elucidate the molecular mech - anismof endometrial carcinoma.

ABSTRACT

<p><b>OBJECTIVE</b>To establish a one-step four multiplex reverse transcription polymerase chain reaction (RT-PCR) method based on Homo-Tag Assisted Non-Dimer System (HAND) system for simultaneous detection of 4 diarrhea viruses of rotavirus, astrovirus, norovirus and sapovirus.</p><p><b>METHODS</b>Primers were designed according to the conserved genome sequence of the 4 viruses and the homologous tail sequences were added to the 5' end. The multiplex RT-PCR system was constructed by optimizing the PCR parameters such as the concentration of universal tag primer and genome-specific Homo-Tailed primers. The specificity, stability and sensitivity of the method were evaluated systematically.</p><p><b>RESULTS</b>The 4 multiplex RT-PCR methods based on HAND system was established successfully. Specificity analysis showed no cross reaction between the 4 diarrhea viruses. The sensitivity analysis showed detection limits for rotavirus, astrovirus, norovirus and sapovirus of 48, 1.92, 9.6 and 48 pg per reaction, respectively.</p><p><b>CONCLUSION</b>The established HAND system-based multiplex RT-PCR assay allows simple, rapid, specific, sensitive, and stable for detection of the 4 common diarrhea viruses at low costs and is suitable for application in general medical laboratories.</p>

ABSTRACT

The manufacturing process, quality Control and Stability test of "901" anti-influenza complex granule were presented. The granule tallied with requirements of Chinese Pharmacopiea (1990th edition). It s main components (Scutellaria baicallensis Georgi. and Bupleurum chinese DC.) were identified with TLC. First derivative spectrometry was used to determine the content of baicalin in this preparation. As a result of stability test, the shelf life at 250℃ is estimated to be 1.97 years.
