ABSTRACT

The aim of this work was to measure HMGB1, TNF-alpha, and IL-8 in bronchoalveolar lavage (BAL), serum and TLR2 and TLR4mRNA expression in lung tissue of rabbits with two grades of acute lung injury (ALI). The animals were randomly assigned to groups with severe (S) and mild/moderate (MM) ALI, induced with warm saline, and a control group. HMGB1, TNF-alpha, IL-8, TLR2mRNA and TLR4mRNA were measured after ALI induction. The results showed increased levels of IL-8, TNF-alpha, HMGB1 and TLR4mRNA in the ALI groups. HMGB1, IL-8 and TNF-alpha concentrations in BAL were higher in S compared MM. Increased TLR4mRNA was observed in S and MM versus control. The results suggest an early participation of HMGB1 in ALI together with IL-8 and TNF-alpha and association with severity. TLR4 has early expression and role in ALI pathophysiology but is not associated with severity.(AU)


O objetivo deste trabalho é determinar os níveis de HMGB1, TNF-alfa e IL-8 no lavado broncoalveolar (BAL), bem como quantificar a expressão sérica de TLR2 e TLR4 mRNA em tecido pulmonar de coelhos com dois graus de lesão pulmonar aguda (LPA). Os animais foram distribuídos aleatoriamente em grupos com LPA grave (S) e leve / moderada (MM), induzidas com solução salina morna, e um grupo controle. HMGB1, TNF-alfa, IL-8, TLR2mRNA e TLR4mRNA foram medidos após a indução de LPA e quatro horas de ventilação mecânica. Os resultados mostraram níveis aumentados de IL-8, TNF-alfa, HMGB1 e TLR4mRNA nos grupos com LPA. As concentrações de HMGB1, IL-8 e TNF-alfa no LBA foram maiores no S comparado ao MM. Aumento de TLR4mRNA foi observado em S e MM versus controle. Os resultados sugerem uma participação precoce da HMGB1 na LPA em conjunto com IL-8 e TNF-alfa e associação com a gravidade da LPA. O TLR4 foi expresso na ALI e possivelmente possui papel precoce na fisiopatologia da LPA, mas sem associação com a gravidade.(AU)

Abstract This paper reports the in vitro antiproliferative effects, antiprotozoal, anti-herpes and antimicrobial activities of 32 organic extracts of 14 marine sponges and 14 corals collected in northeast Brazilian coast. The ethanolic extracts of the sponges Amphimedon compressa and Tedania ignis, and the acetone extract of Dysidea sp. showed relevant results concerning the antiproliferative effects against A549, HCT-8, and PC-3 cell lines by sulforhodamine B assay, but also low specificity. Concerning the antiprotozoal screening, the ethanolic extract of Amphimedon compressa and the acetone and ethanolic extracts of Dysidea sp. were the most active against Leishmania amazonensis and Trypanosoma cruzi expressing β-galactosidase in THP-1 cells. In the preliminary anti-HSV-1 (KOS strain) screening, the ethanolic extracts of the sponges Amphimedon compressa, Haliclona sp. and Chondrosia collectrix inhibited viral replication by more than 50%. The most promising anti-herpes results were observed for the ethanolic extract of Haliclona sp. showing high selective indices against HSV-1, KOS and 29R strains (SI> 50 and >79, respectively), and HSV-2, 333 strain (IS>108). The results of the antibacterial screening indicated that only the ethanolic extract of Amphimedon compressa exhibited a weak activity against Enterococcus faecalis, Pseudomonas aeruginosa and Escherichia coli by the disk diffusion method. In view of these results, the extracts of Amphimedon compressa, Tedania ignis and Dysidea sp. were selected for further studies aiming the isolation and identification of the bioactive compounds with antiproliferative and/or antiprotozoal activities. The relevant anti-herpes activity of the ethanolic extract of Haliclona sp. also deserves special attention, and will be further investigated.


Resumo Este artigo reporta as atividades in vitro antiproliferativa, atiprotozoárica, anti-herpética e antimicrobiana de 32 extratos orgânicos provenientes de 14 esponjas marinhas e 14 corais coletados no litoral nordestino brasileiro. Os extratos etanólicos das esponjas Amphimedon compressa e Tedania ignis, e o extrato acetônico de Dysidea sp. demonstraram resultados promissores em relação aos efeitos antiproliferativos frente as linhagens celulares A549, HCT-8, PC-3 pelo método da sulforrodamina B, mas sem especificidade. Em relação à atividade antiprotozárica, os extratos etanólico de Amphimedon compressa e acetônico e etanólico de Dysidea sp. apresentaram atividade contra Leishmania amazonensis e Trypanosoma cruzi através do método de expressão de β-galactosidase em células THP-1. Na investigação preliminar de atividade antiviral frente ao vírus Herpes simplex tipo 1 (cepa KOS), os extratos etanólicos das esponjas Amphimedon compressa, Haliclona sp. e Chondrosia collectrix inibiram mais de 50% da replicação viral. O extrato etanólico da esponja Haliclona sp. demonstrou resultados promissores para atividade anti-herpética com altos índices de seletividade para as cepas KOS (IS >50) e 29R (IS>79) frente ao VHS-1 e cepa 333 (IS>108) frente ao VHS-2. O extrato etanólico da esponja Amphimedon compressa exibiu uma pequena atividade contra Enterococcus faecalis, Pseudomonas aeruginosa and Escherichia coli pelo método de difusão em disco. De acordo com os resultados apresentados, os extratos das esponjas Amphimedon compressa, Tedania ignis e Dysidea sp. serão selecionados para futuros estudos de isolamento e identificação dos compostos bioativos para as atividades antiproliferativa e antiprozoárica. O extrato etanólico de Haliclona sp. será investigado por possuir atividade relevante anti-herpética.

Abstract We aimed in this study utilize environmental indicators as a quantitative method to evaluate and discuss the nitrogen (TN) and phosphorus (TP) flux by a production stage grow-out (termination) of Nile tilapia (Oreochromis niloticus) in fishpond. The TN and TP load, the mass balance, the input of TN and TP via feed and the converted nutrients in fish biomass are the environmental indicators applied in this study. During the production cycle (128 days), the system exported 15,931 g TN and 4,189 g TP that were related to the amount of feed supplied (r Pearson = 0.8825 and r = 0.8523, respectively), corroborated by the feed conversion ratio (1.61:1). The indicators showed that 26% TN and 45% TP were reversed into fish biomass, 62% TN and 40% TP were retained in the fishpond, and 12% TN and 15% TP were exported via effluent. The largest contribution of nutrients generated by the system and exported via effluent was observed in phase III and IV. This result is supported by the feed conversion ratio 2.14 and 2.21:1 obtained at this phase, a fact explained by the amount of feed offered and the fish metabolism. Application of environmental indicators showed to be an efficient tool to quantify flux of TN and TP produced during the grow-out period of Nile tilapia and therefore, guide management practices more sustainable. Concerning the environmental sustainability of the activity the implementation of best management practices such as the better control of the feed amount offered would lead to a smaller loss of TN and TP to the water. Furthermore, the use of better quality feeds would allow greater nutrient assimilation efficiency.


Resumo Nós objetivamos neste estudo, utilizar indicadores ambientais como método quantitativo para avaliar e discutir sobre o fluxo de nitrogênio (TN) e fósforo (TP) na etapa final de crescimento (terminação) de tilápia-do-nilo (Oreochromis niloticus) em viveiro escavado. A carga de TN e TP, o balanço de massa, a entrada de nutrientes via ração e o TN e TP convertido em biomassa de peixe foram os indicadores ambientais utilizados neste estudo. Durante o ciclo produtivo (128 dias), o sistema exportou 15.931 g NT e 4.189 g PT os quais foram relacionadas às quantidades de alimento fornecido (r Pearson = 0,8825 e r = 0,8523, respectivamente), corroborada pela conversão alimentar (1,61:1). Os indicadores evidenciaram que 26% NT e 45% PT foram revertidos em biomassa de peixe, 62% NT e 40% PT ficaram retidos no viveiro e 12% NT e 15% PT foram exportados via efluente. O maior aporte de nutrientes gerado pelo sistema e exportado via efluente foi verificado nas fases III e IV. Este resultado é corroborado pelas taxas de conversão alimentar de 2,14 e 2,21:1 obtida nestas fases, fato explicado pela quantidade de ração ofertada e pelo metabolismo dos peixes. A aplicação dos indicadores ambientais mostrou ser uma ferramenta eficiente para quantificar o fluxo de TN e TP produzidos durante a etapa final de crescimento de tilápia-do-nilo e com isso orientar práticas de manejo mais sustentáveis. Com vistas à sustentabilidade ambiental da atividade, a implantação de boas práticas de manejo tais como o melhor controle da quantidade de alimento ofertado levaria a menor perda de NT e PT para a água. Além disso, o uso de rações de melhor qualidade permitiria maior eficiência de assimilação desses nutrientes.

ABSTRACT

Abstract The phosphorus and nitrogen discharge via effluent of intensive trout farming system was quantified through the use of environmental indicators. The nutrient loads, the mass balance, the estimated amount of nutrients in feed and the amount of nutrients converted in fish biomass were calculated based on the concentrations of phosphorus (P) and nitrogen (N) in the feed and in the water. Of the offered feed, 24.75 kg were available as P and 99.00 kg as N, of these, 9.32 kg P (38%) and 29.12 kg N (25%) were converted into fish biomass and 15.43 kg P (62%) and 69.88 kg N (75%) were exported via effluent. The loads and the mass balance show the excessive discharge of nutrients via effluent, corroborated by the feed conversion ratio (2.12:1) due to the low efficiency of feed utilization, therefore, it is proposed the use of this zootechnical parameter as environmental indicator. In addition, feed management practices are not adequate, highlighting the low frequency of feeding during the day, excessive amount and low quality of feed offered. These results demonstrate the need for adequate feed management and the need for careful monitoring of effluent.


Resumo A descarga de fósforo e nitrogênio via efluente do sistema intensivo de truticultura foi quantificada através da utilização de indicadores ambientais. As cargas de nutrientes, o balanço de massa, a quantidade estimada de nutrientes na ração e a quantidade de nutrientes convertidos em biomassa de peixes foram calculados com base nas concentrações de fósforo (P) e nitrogênio (N) na ração e na água. Da ração oferecida, 24,75 kg estavam disponíveis como P e 99,00 kg como N, destes, 9,32 kg de P (38%) e 29,12 kg de N (25%) foram convertidos em biomassa de peixe e 15,43 kg P (62%) e 69,88 kg N (75%) foram exportados via efluente. As cargas e o balanço de massa mostram a descarga excessiva de nutrientes via efluente, corroborado pela taxa de conversão alimentar (2,12:1), devido à baixa eficiência na utilização da ração, portanto, propõe-se a utilização deste parâmetro zootécnico como indicador ambiental. Além disso, as práticas de manejo alimentar não são adequadas, destacando a baixa frequência de alimentação durante o dia, quantidade excessiva e baixa qualidade da alimentação ofertada. Esses resultados demonstram a necessidade de manejo alimentar adequado e de monitoramento do efluente.

ABSTRACT

Abstract This study aims to evaluate the environmental factors determining of the changes in phytoplankton structure in spatial (upper, middle and lower course) and seasonal (dry and rainy period) scales in a eutrophic river (Almada River, northeastern Brazil). In the study period, total accumulated rainfall was below of the historic average, resulting in flow reduction, mainly in rainy period. High orthophosphate concentration was found at the sampling sites. Phytoplankton chlorophyll a increased from upstream to downstream. Geitlerinema splendidum (S1) and Chlamydomonas sp. (X2) were the most abundant species in the upper course and several species of diatoms (D), Euglenophyceae (W1, W2) and Chlorophyceae (X1) in the middle and lower course. The functional groups were found to be characteristic of lotic ecosystem, shallow, with low light availability, rich in organic matter and eutrophic environments. We conclude that phytoplankton community structure was sensitive to change of the river flow and nutrient availability in spatial and seasonal scale in a tropical river.


Resumo O objetivo deste estudo foi avaliar os fatores ambientais determinantes das mudanças na estrutura do fitoplâncton em escala espacial (alto, médio e baixo curso) e temporal (seca e chuva) em um rio eutrófico tropical (rio Almada, nordeste do Brasil). No período de estudo, a precipitação acumulada mensal foi abaixo da média histórica, resultando na redução da vazão, principalmente no período chuvoso. A concentração de ortofosfato foi elevada nos pontos de amostragem. A clorofila a do fitoplâncton aumentou de montante para jusante. Geitlerinema splendidum (S1) e Chlamydomomas sp. (X2) foram as espécies mais abundantes no alto curso e várias espécies de diatomáceas (D), Euglenophyceae (W1, W2) e Chlorophyceae (X1) no médio e/ou baixo curso. Os grupos funcionais foram característicos de ambientes lóticos, rasos, com baixa disponibilidade de luz, rico em matéria orgânica e eutrófico. Concluímos que a estrutura da comunidade fitoplanctônica foi sensível às mudanças na vazão e na disponibilidade de nutrientes em escala espacial e temporal em um rio tropical.

ABSTRACT

Semen from the first 15mL of the ejaculate (P1) obtained from two boars (30mL) was diluted in glycine-egg yolk extender, cooled at 5°C in a special container and rediluted in standard doses of 3x109 mobile spermatozoa after 12h of storage. Semen was also stored up to 24h after redilution. The physical characteristics of the semen were evaluated at different storage periods (fresh, 0h, 12h, rediluted, 24h, and 36h). The reproductive performance of the boars and their fertility regarding the insemination of primiparous sows were also determined. Two treatments were used: T1-15B sows inseminated with semen originated from hyperconcentrated heterospermic doses (15x109 mobile spermatozoa per dose), rediluted after 12h of storage at 5°C for standard doses of 3x109 mobile spermatozoa per dose and stored at 5°C up to 24h after redilution (n=10); T2-3B sows inseminated with standard heterospermic doses (3x109 mobile spermatozoa per dose), stored at 5°C up to 36h after semen collection (n=10). There was no effect (P>0.05) of treatments on the spermatic motility, even though a pronounced decrease (P>0.05) of their values at 12h of storage was recorded. However, they remained higher than 70% until 36h. There was effect of treatments on spermatic vigour at 0h (P<0.05), when T1-15B vigour was higher. There was also effect of the storage period for both treatments with a progressive decrease throughout 36h of storage, although the differences were not always significant. Pregnancy rates (90%) and the number of total farrowed piglets (15, 11-T1-15B; 13, 44- T2-3B) did not differ (P>0.05) between the treatments. It was concluded that the semen hyperconcentration of 15 billion of mobile spermatozoa per dose, stored at 5°C for 12h, did not result in drawbacks considering the physical characteristics of the semen, maintaining the pregnancy rates and prolificacy of the inseminated sows.


Os primeiros 15mL do ejaculado (P1) de dois varrões foram coletados (30mL) e diluídos em diluidor glicina-gema de ovo, resfriados a 5°C em contêiner especial e rediluídos para doses padrão de 3x109 espermatozoides (sptz) móveis, após 12 horas de armazenamento. Além disso, foram armazenados por até 24 horas após a rediluição, sendo as características físicas avaliadas em diferentes períodos de estocagem (fresco, zero hora, 12h, Red12h, 24h e 36h) e a fertilidade avaliada por meio de fêmeas primíparas inseminadas. Foram realizados dois tratamentos: T1-15B: porcas inseminadas com sêmen de doses heterospérmicas hiperconcentradas (15x109 sptz móveis/dose), rediluídas após 12 horas de armazenamento a 5°C para doses padrão de 3x109 sptz móveis/dose, e armazenadas a 5°C por até 24 horas após a rediluição (n=10); T2-3B: porcas inseminadas com doses heterospérmicas padrão (3x109 sptz móveis/dose), armazenadas a 5°C por até 36 horas após coleta. Não houve efeito (P>0.05) dos tratamentos sobre a motilidade espermática e, embora tenha ocorrido queda (P<0.05) às 12 horas, a motilidade foi superior a 70% durante as 36 horas de armazenamento. Houve efeito (P<0.05) dos tratamentos no tempo zero hora quanto ao vigor espermático, sendo E1T1-15B superior. Além disso, houve efeito do período de estocagem para os dois tratamentos, com queda progressiva do vigor ao longo das 36 horas, embora nem sempre as diferenças tenham sido significativas. As taxas de gestação (90%) e o número total de leitões nascidos (15, 11 - T1-15B; 13, 44 - T2-3B) não diferiram (P>0.05) entre os tratamentos. Concluiu-se que a hiperconcentração do sêmen para 15x109 sptz móveis/dose, armazenado a 5°C por 12 horas não resultou em prejuízos quanto à manutenção das características físicas do sêmen e ao desempenho reprodutivo dos varrões, sendo capaz de manter a taxa de gestação e a prolificidade das fêmeas inseminadas.

ABSTRACT The aim of this study was to characterize components of the EOAz and its hexane (HFEOAz), chloroform (CFEOAz) and methanol (MFEOAz) fractions, and its antihypertensive effect. EOAz was extracted from leaves by hydrodistillation. Aliquot was subjected to selective desorption with silica gel column and eluted with hexane, chloroform and methanol. The components of the EOAz and fractions were analyzed by gas chromatography coupled with mass spectrometry and nuclear magnetic resonance spectroscopy of hydrogen. Experiments of vascular reactivity were performed with isolated aortic rings of male Wistar rats. Antihypertensive effect was evaluated in hypertensive rats submitted to the inhibition of synthesis of nitric oxide. Blood pressure was measured indirectly by tail plethysmography. MFEOAz showed the lowest EC50 (150.45 µg/mL), 1,8-cineole (27.81%) and terpinen-4-ol (57.35%) as main components. Single administration by nasogastric tube of EOAz, fractions and captopril significantly reduced the blood pressure of hypertensive rats, when compared to animals of the negative control group with distilled water. In conclusion, the potency of the MFEOAz was higher than that of EOAz and other fractions. The antihypertensive effect of EOAz and fractions was similar, higher than the negative control and lower than that of captopril.


RESUMO O objetivo deste estudo foi caracterizar os componentes do óleo essencial das folhas de Alpinia zerumbet (OEAz) e suas frações hexânica (FHOEAz), clorofórmica (FCOEAz) e metanólica (FMOEAz), e seu efeito anti-hipertensivo. OEAz foi extraído das folhas por hidrodestilação. Uma alíquota foi submetida à desadsorção seletiva com coluna de gel de sílica e eluída com hexano, clorofórmio e metanol. Os componentes do OEAz e fracções foram analisadas por cromatografia gasosa acoplada à detector de massa e por espectros de ressonância magnética nuclear de hidrogênio. Experimentos de reatividade vascular foram realizados com anéis aórticos isolados de ratos Wistar machos. Efeito anti-hipertensivo foi avaliado em ratos hipertensos submetidos à inibição da síntese de óxido nítrico. A pressão arterial foi medida indiretamente por pletismografia de cauda. FMOEAz mostrou a menor CE50 (150,45 μg/mL), 1,8-cineol (27,81%) e terpinen-4-ol (57,35%) como componentes principais. A administração em dose única por sonda nasogástrica de OEAz, frações e captopril reduziu significativamente a pressão arterial de ratos hipertensos, quando comparados aos animais do grupo controle negativo com água destilada. Em conclusão, a potência da FMOEAz foi maior que a do OEAz e outras frações. O efeito anti-hipertensivo de OEAz e frações foi semelhante, maior do que o controle negativo e menor do que o captopril.

El objetivo de esta investigación es identificar la relación entre la asimetría facial y la pérdida de contacto en línea mediana entre dientes maxilares y mandibulares. Se realizó un estudio descriptivo en 18 sujetos portadores de asimetría facial que consultaron en la División de Cirugía Oral y Maxilofacial de la Universidad de La Frontera por tratamiento quirúrgico de la asimetría. En todos ellos se evaluó la presencia y grado de la asimetría facial y la presencia y desvío desde la línea mediana facial del punto mediano mandibular y maxilar mediante la prueba estadística t de student considerando un valor de p <0,05; todas las evaluaciones fueron realizadas mediante fotografías 2D de rostro y dientes obtenidas en condiciones estandarizadas. Se identificó que todos los sujetos portadores de asimetría facial presentaron asimetría dentaria; los desvíos dentarios fueron de casi 7 mm y existió relación estadísticamente significativa entre desvío facial y desvío dentario. Se concluye que los sujetos con asimetría dentaria pueden presentar asimetría facial de forma que la evaluación esqueletal del rostro es necesaria para realizar un diagnóstico correcto.


The aim of this research was to find the relation between facial asymmetry and loss of continuity in median line of maxillary and mandibular teeth. A descriptive study was carried out in 18 subjects with facial asymmetry with surgical indication and observed at the Division of Oral and Maxillofacial Surgery of the Universidad de La Frontera. In all of them the grade of facial asymmetry and the relation in median line of the maxillary and mandibular teeth was evaluated using the t student test considering p value < 0.05; all the evaluations were executed using 2D photography of the face and tooth obtained with standardized conditions. In all the subjects we observed dental asymmetry with almost 7 mm from the median line; statistical relations between dental asymmetry and facial asymmetry. It is concluded that in subjects asking for dental asymmetry treatment a facial evaluation is necessary in view of the relation with skeletal facial deformity.

Impaired cholinergic neurotransmission can affect memory formation and influence sleep-wake cycles (SWC). In the present study, we describe the SWC in mice with a deficient vesicular acetylcholine transporter (VAChT) system, previously characterized as presenting reduced acetylcholine release and cognitive and behavioral dysfunctions. Continuous, chronic ECoG and EMG recordings were used to evaluate the SWC pattern during light and dark phases in VAChT knockdown heterozygous (VAChT-KDHET, n=7) and wild-type (WT, n=7) mice. SWC were evaluated for sleep efficiency, total amount and mean duration of slow-wave, intermediate and paradoxical sleep, as well as the number of awakenings from sleep. After recording SWC, contextual fear-conditioning tests were used as an acetylcholine-dependent learning paradigm. The results showed that sleep efficiency in VAChT-KDHET animals was similar to that of WT mice, but that the SWC was more fragmented. Fragmentation was characterized by an increase in the number of awakenings, mainly during intermediate sleep. VAChT-KDHET animals performed poorly in the contextual fear-conditioning paradigm (mean freezing time: 34.4±3.1 and 44.5±3.3 s for WT and VAChT-KDHET animals, respectively), which was followed by a 45% reduction in the number of paradoxical sleep episodes after the training session. Taken together, the results show that reduced cholinergic transmission led to sleep fragmentation and learning impairment. We discuss the results on the basis of cholinergic plasticity and its relevance to sleep homeostasis. We suggest that VAChT-KDHET mice could be a useful model to test cholinergic drugs used to treat sleep dysfunction in neurodegenerative disorders.

Métodos alternativos para o controle de doenças e pragas de plantas vêm sendo pesquisados buscando causar menores danos ao ambiente e a saúde humana. Diante disso, o objetivo deste estudo foi identificar extratos de plantas e óleos essenciais com possíveis efeitos sobre Phakopsora pachyrhizi, agente etiológico da ferrugem-asiática da soja. Assim, 61 extratos de plantas da flora nativa da região do Alto Rio Grande, Minas Gerais e cinco óleos essenciais obtidos comercialmente foram utilizados com a finalidade de testar os efeitos na germinação de urediniósporos do referido fungo. Foram utilizadas placas de Petri de 6 cm, com meio ágar-água, adicionado com 150 µL de extrato de planta ou 6 µL do óleo essencial misturado ao meio de cultura. Como testemunhas foram utilizadas 150 µL de Tween 20 a 0,7% e 150 µL de água destilada. Após, foram adicionados 50µL de uma suspensão de urediniósporos, na concentração de 2mg.5mL-1 e espalhados com alça de Drigalsky. As placas foram colocadas em BOD, a 23(±2ºC), por 4 horas. A germinação foi paralisada com adição de lactoglicerol. A avaliação da percentagem de esporos germinados foi realizada em microscópio de luz. Dentre os extratos vegetais testados, dez apresentaram percentual de germinação abaixo de 15%, tendo os extratos obtidos das plantas Pelargonium sp., Salvia officinalis, Lavandula officinalis, Mentha pulegium e M. arvensis apresentado germinação de esporos de 3,0%, 3,5%, 3,0%, 2,0% e 4,0%, respectivamente. Todos os óleos essenciais avaliados foram efetivos na inibição da germinação, com destaque para o óleo essencial de Caryophilus aromaticus, que apresentou percentagem de germinação de 5%, resultando num percentual de inibição de 50,0%.


Alternative methods to control plant diseases and plagues have been studied, in order to cause less damages to the environment and human health. The purpose of this study was to identify plant extracts and essential oils as possible alternative products that affect the urediniospore germination of Phakopsora pachyrhizi, the causal agent of the Asian soybean rust. Plant extracts (61) from the native flora of the Alto Rio Grande region, State of Minas Gerais, Brazil, as well as five commercial essential oils were used with the purpose of testing their effects on the germination of urediniospores of the referred fungus. 6cm Petri dishes were used, with water-agar medium, added with 150 µL of plant extract or 6 µL of essential oil mixed to the culture medium. As control, 150 µL of Tween 20 at 0.7% and 150 µL of distilled water were used. Then, 50 µL of a suspension of urediniospores were added, at a concentration of 2mg5mL-1 and spread with Drigalsky spatula. Then, the plates were put in BOD, at 23(±2ºC), for 4 hours. Germination was paralyzed by adding lactoglicerol. The percentage of spore germination was evaluated in a light microscope. Among the vegetal extracts tested, ten of them presented a germination percentage below 15%. The extracts from the Pelargonium sp., Salvia officinalis, Lavandula officinalis, Mentha pulegium and M. arvensis plants presented 3.0%, 3.5%, 3.0%, 2.0% and 4.0% of urediniospore germination, respectively. All the essential oils evaluated were effective in inhibiting germination, especially the Caryophilus aromaticus essential oil that presented 5.0% of germination, with 50.0% of inhibition.

Implantation of Walker 256 tumor decreases acute systemic inflammation in rats. Inflammatory hyperalgesia is one of the most important events of acute inflammation. The L-arginine/NO/cGMP/K+ATP pathway has been proposed as the mechanism of peripheral antinociception mediated by several drugs and physical exercise. The objective of this study was to investigate a possible involvement of the NO/cGMP/K+ATP pathway in antinociception induced in Walker 256 tumor-bearing male Wistar rats (180-220 g). The groups consisted of 5-6 animals. Mechanical inflammatory hypernociception was evaluated using an electronic version of the von Frey test. Walker tumor (4th and 7th day post-implantation) reduced prostaglandin E2- (PGE2, 400 ng/paw; 50 µL; intraplantar injection) and carrageenan-induced hypernociception (500 µg/paw; 100 µL; intraplantar injection). Walker tumor-induced analgesia was reversed (99.3% for carrageenan and 77.2% for PGE2) by a selective inhibitor of nitric oxide synthase (L-NAME; 90 mg/kg, ip) and L-arginine (200 mg/kg, ip), which prevented (80% for carrageenan and 65% for PGE2) the effect of L-NAME. Treatment with the soluble guanylyl cyclase inhibitor ODQ (100% for carrageenan and 95% for PGE2; 8 µg/paw) and the ATP-sensitive K+ channel (KATP) blocker glibenclamide (87.5% for carrageenan and 100% for PGE2; 160 µg/paw) reversed the antinociceptive effect of tumor bearing in a statistically significant manner (P < 0.05). The present study confirmed an intrinsic peripheral antinociceptive effect of Walker tumor bearing in rats. This antinociceptive effect seemed to be mediated by activation of the NO/cGMP pathway followed by the opening of KATP channels.

Although several alleles of susceptibility to Alzheimer’s disease (AD) have been studied in the last decades, few polymorphisms have been considered as risk factors for the disease. Among them, the APOE-e4 allele appears to be the major genetic risk factor for the onset of the disease. However, it is important to confirm the potential susceptibility of these genetic variants in different populations in order to establish a genetic profile for the disease in specific communities. This study analyzed the APOE polymorphisms regarding susceptibility to AD in a sample of 264 individuals (primarily Caucasians; 82 cases and 182 controls) in the population from Vitória, ES, Brazil, by PCR restriction fragment length polymorphism (PCR-RFLP) methods. The patients were selected according to clinical criteria for probable AD. Whereas the e4 allele showed statistically significant positive association with susceptibility to AD (OR = 3.01, 95 percentCI = 1.96-4.61; P < 0.0001), the e2 allele did not. The results of the e4 allele confirm the role of this polymorphism as a risk factor for AD in the sample studied as observed in other populations. Although the e3 allele has been considered neutral in several studies, our results suggest that it acts as a protective factor against AD in the population studied (OR = 0.46, 95 percentCI = 0.30-0.67; P < 0.0001). This study may provide a new insight into the role of the APOE-e3 allele in the etiology of AD and might help to estabilish a profile of risk for AD in the population from Vitória, ES.

El objetivo de este estudio fue realizar un análisis retrospectivo de la etiología, incidencia, uso de dispositivos de seguridad, tipo y severidad de las fracturas, tratamiento y complicaciones de los traumatismos del tercio medio facial en una población pediátrica e adolescente del Estado de São Paulo. La muestra fue seleccionada a partir del análisis de historias clínicas de 2986 pacientes víctimas de traumatismos faciales atendidos entre 1999 y 2008 por el Área de Cirugía Buco-Maxilo-Facial de la Facultad de Odontología de Piracicaba, Universidad Estatal de Campinas, Estado de São Paulo-Brasil. Durante este periodo, 757 pacientes menores de 18 años fueron víctimas de traumatismos maxilofaciales. De éstos, 138 pacientes presentaron 148 fracturas nasales y de tercio medio. Los adolescentes fueron el grupo etario más afectado, mientras que el género masculino fue el de mayor prevalencia. Los accidentes ciclísticos se constituyeron en la principal etiología (21.7%). El tratamiento conservador fue utilizado en la gran mayoría de los casos (118 pacientes, 85.5%). Veinte pacientes (14.5%) recibieron tratamiento quirúrgico y solo un caso de complicación pos-operatoria fue reportado. La incidencia de traumatismos y fracturas del tercio medio fue alta en la región de estudio. Los accidentes ciclísticos y las agresiones físicas fueron los principales factores etiológicos, siendo el género masculino predominante en todos los grupos etarios evaluados. Las fracturas nasales y del complejo zigomático orbitario fueron las más prevalentes, mientras que el tratamiento conservador fue instituido en la gran mayoría de los casos


This study showed a retrospective analysis of the etiology, incidence, use of safety devices, type and severity of fractures, treatment and complications of midface trauma in a pediatric and adolescent population of São Paulo - Brazil. We analyzed 2986 medical records of facial trauma patients treated by the Division of Oral and Maxillofacial Surgery Piracicaba Dental School, State University of Campinas, São Paulo - Brazil between 1999 and 2008. During this period, 757 patients under 18 were identified and 138 had 148 nasal and midface fractures. The adolescents were the most affected age group, male gender being the most prevalent. The most common injury causes were bicycle accident (21.74%) and violence (19.6%). Conservative treatment was used in the majority of cases (118 patients, 85.5%). Twenty patients (14.5%) received surgical treatment and only one case of postoperative complication was reported. The incidence of trauma and middle third fractures in pediatric and adolescents patients was high in the study area, being the main etiological factors the bicycle accidents and violence. The male were most affected in all age groups tested. Nasal fractures and zygomatic complex were most prevalent, while conservative treatment was instituted in most cases

ABSTRACT

Epilepsy is a neurological disorder associated with excitatory and inhibitory imbalance within the underlying neural network. This study evaluated inhibitory γ-amino-butyric acid (GABA)ergic modulation in the CA1 region of the hippocampus of male Wistar rats and Wistar audiogenic rats (aged 90 ± 3 days), a strain of inbred animals susceptible to audiogenic seizures. Field excitatory postsynaptic potentials and population spike complexes in response to Schaffer collateral fiber stimulation were recorded in hippocampal slices before and during application of picrotoxin (50 µM, 60 min), a GABA A antagonist, and the size of the population spike was quantified by measuring its amplitude and slope. In control audiogenic-resistant Wistar rats (N = 9), picrotoxin significantly increased both the amplitude of the population spike by 51 ± 19 percent and its maximum slope by 73 ± 21 percent. In contrast, in slices from Wistar audiogenic rats (N = 6), picrotoxin caused no statistically significant change in population spike amplitude (33 ± 46 percent) or slope (11 ± 29 percent). Data are reported as means ± SEM. This result indicates a functional reduction of GABAergic neurotransmission in hippocampal slices from Wistar audiogenic rats.

Objetivo: El presente estudio evaluó la microdureza de dos distintas resinas que fueron fotopolimerizadas a cuatro distancias diferentes. Materiales y Métodos: Se obtuvieron cuarenta muestras utilizando una matriz metálica de acero y dos resinas fotopolimerizables (Filtek-A110, Filtek-Z250). Las muestras se prepararon de forma aleatoria constituyendo 8 grupos, 4 por cada resina fotopolimerizable (n igual que 5), las cuales fueron polimerizadas a 0, 2, 4 y 8 mm de distancia de la fuente de luz, usando la misma intensidad de energía. Posteriormente, las medidas de microdureza Vickers fueron obtenidas en la superficie superior e inferior de las muestras. Los datos resultantes fueron sometidos al t test o ANOVA y al test Tukey (alfa igual que 0.05). Resultados: La microdureza de la resina microhíbrida (Filtek-Z250), fue superior en todas las distancias analizadas en relación con la resina de microrelleno (Filtek-A110). Para la resina Filtek-Z250, la distancia de polimerización no influyó en la dureza de la parte superior o inferior, excepto a 8mm de la distancia inferior, donde se encontró el menor valor de microdureza. En la resina Filtek-A110, se observó una disminución progresiva de la microdureza con el incremento de la distancia, en la parte superior e inferior. En general, la microdureza en la parte superior fue significativamente mayor que en la parte inferior de ambas resinas. Conclusiones: El incremento en la distancia de fotopolimerización no modificó el grado de polimerización de la resina microhíbrida. Sin embargo, esta distancia es un factor importante a ser considerado durante la polimerización de las resinas de microrelleno.


Objetive: This study investigated the surface microhardness of two different composite resins light cured at four different distance. Material and methods: Forty specimens were obtained sin composites (Filtek-A110 and Filtek-Z250). The samples were randomly prepared, constituting 8 groups, 4 for each resin composite (n=5), which were light cured at 0,2,4 or 8 mm distances, using the same output power. Afterwards, Vickers microhardness measurements were obtained on top and botton surfaces of the samples. Data were submitted to t test or ANOVA followed by Turkey test (a=0.05). Results: The microhardness of the microhybrid resin, Filtek-Z250, was superior in all distances analyzed in relation to the microfilled resin, Filtek-A110. Filtek-Z250, the distance of polymerization did not influence the hardness at top or bottom, except for 8 mm distance at bottom, where the lowest microharness was found. For Filtek-A110, a progressive decrease in microhardness with the increase of distance was observed at both top and bottom. At general, hardness at top was significantly higher than at bottom for both resins. Conclusions: In the conditions of this study, the increase in the distance of polymerization did no change microhybrid composite resin polymerization. However, this distance is an important factor to be observed in polymerization of microfilled composite resins.

Fifty-four samples were collected from growing and finishing pigs for the molecular diagnosis of enzootic porcine pneumonia. Nineteen lung fragments were obtained from pigs that showed signs of respiratory disease and 35 nasal swabs were obtained from clinically healthy pigs. For the detection of the bacterial genome in the samples, the nested PCR technique was used to amplify a fragment of 706bp. This fragment was subsequently cloned and sequenced. The sequence of obtained nucleotides was compared with six other sequences of Mycoplasma hyopneumoniae and 11 sequences of other bacteria available in the Genbank. To measure the sensitivity of the nested PCR, serial dilutions (10-1 to 10-15) of cloned fragments were conducted based on the concentration of 300ng. Ten lung fragments and eight nasal swabs showed positive for M. hyopneumoniae and the limit of detection was estimated to be 0.3fg DNA cloned. The sequence of nucleotides obtained showed 99.1 percent homology with the other sequences of M. hyopneumoniae, demonstrating that the nested PCR used in this study may provide an important diagnostic tool for the detection of this agent.


Foram coletadas 54 amostras de animais em fase de crescimento e terminação para o diagnóstico molecular da pneumonia enzoótica suína. Dezenove fragmentos de pulmão foram obtidos de suínos que apresentavam sinais de doença respiratória e 35 suabes nasais foram obtidas de suínos clinicamente saudáveis. Para a detecção do genoma bacteriano nas amostras, foi utilizada a técnica de nested PCR que originou um fragmento de 706pb, o qual foi, posteriormente, clonado e sequenciado. A sequência de nucleotídeos obtida foi comparada com outras seis sequências de Mycoplasma hyopneumoniae e 11 sequências de outras bactérias disponíveis no Genbank. Para medir a sensibilidade da nested PCR, foram realizadas diluições seriadas (10-1 a 10-15) do fragmento clonado, partindo da concentração de 300ng. Dez fragmentos de pulmões e oito suabes nasais apresentaram resultado positivo para M. hyopneumoniae e o limite de detecção foi estimado em 0,3fg de DNA clonado. A sequência de nucleotídeos obtida foi de 99.1 por cento de homologia com as outras sequências de M. hyopneumoniae, demonstrando que a nested PCR utilizada neste estudo pode ser uma importante ferramenta de diagnóstico para a detecção desse agente.

ABSTRACT

Avaliou-se o efeito da adição de diferentes tipos e concentrações de soro sobre o desenvolvimento e a sobrevivência de folículos ovarianos pré-antrais (FOPA) caprinos in vitro. Além disso, verificou-se a relação entre as concentrações de nitrito presentes no meio de cultivo e a viabilidade folicular. Cada par ovariano foi dividido em 29 fragmentos, sendo um destinado ao controle. Os fragmentos foram cultivados por um ou sete dias em meio essencial mínimo suplementado (MEM+) ou MEM+ com diferentes concentrações (10 ou 20 por cento) de soro fetal bovino (SFB), soro de cabra em estro (SCE) ou soro de cabra em diestro (SCD). Na análise morfológica após sete dias, apenas o tratamento com 10 por cento de SFB apresentou percentual de FOPA normais similar ao MEM+ (P>0,05). A análise ultra-estrutural dos folículos cultivados por sete dias com MEM+ ou MEM+ com 10 por cento de SFB mostrou danos oocitários, porém células da granulosa normais. A análise do meio de cultivo revelou correlação positiva entre a viabilidade folicular e a produção de nitrito. A suplementação com soro não melhorou a viabilidade de FOPA e a concentração de nitrito no meio de cultivo funcionou como um indicador da viabilidade das células da granulosa de FOPA caprinos cultivados in vitro.


The effect of the addition of different types and concentrations of sera on the viability and development of caprine preantal follicles (PAF) in vitro cultured was analyzed. In addition, it was evaluated the correlation between nitrite concentrations in culture medium and folicular viability. Each ovarian pair was divided in 29 fragments and one was used as control. The fragments were cultured for one or seven days in minimal essential medium (MEM+) or MEM+ with different concentrations of (10 or 20 percent) bovine fetal serum (BFS), estrous goat serum (EGS), or diestrous goat serum (DGS). After seven days, the morphological analysis showed that only the treatment with 10 percent BFS maintained the percentage of normal PAF similar to MEM+ (P>0.05). The ultrastructural analysis of follicles cultured for seven days in MEM+ or MEM+ with 10 percent BFS showed some oocyte damage, although the granulosa cells were normal. Analysis of culture medium revealed a positive correlation between follicular viability and nitrite production. Supplementation with serum did not improve the viability of PAF and nitrite levels in culture medium served as an indicator of viability of granulose cells from caprine PAF in vitro cultured.

ABSTRACT

Third molar extraction is a common procedure frequently accompanied by moderate or severe pain, and involves sufficient numbers of patients to make studies relatively easy to perform. The aim of the present study was to determine the efficacy and safety of the therapeutic combination of 10 mg piroxicam, 1 mg dexamethasone, 35 mg orphenadrine citrate, and 2.5 mg cyanocobalamin (Rheumazin®) when compared with 20 mg piroxicam alone (Feldene®) in mandibular third molar surgery. Eighty patients scheduled for removal of the third molar were included in this randomized and double-blind study. They received (vo) Rheumazin or Feldene 30 min after tooth extraction and once daily for 4 consecutive days. Pain was determined by a visual analogue scale and by the need for escape analgesia (paracetamol). Facial swelling was evaluated with a measuring tape and adverse effects and patient satisfaction were recorded. There was no statistically significant difference in facial swelling between Rheumazin and Feldene (control group). Both drugs were equally effective in the control of pain, with Rheumazin displaying less adverse effects than Feldene. Therefore, Rheumazin appears to provide a better risk/benefit ratio in the mandibular molar surgery. Since the side effects resulting from nonsteroidal anti-inflammatory drug administration are a severe limitation to the routine use of these drugs in clinical practice, our results suggest that Rheumazin can be a good choice for third molar removal treatment.
