ABSTRACT

BACKGROUND Dendritic cells (DCs) specific intercellular adhesion molecule (ICAM)-3-grabbing non integrin receptor (DC-SIGN) binds to subgenera Leishmania promastigotes mediating its interaction with DC and neutrophils, potentially influencing the infection outcome. OBJECTIVES In this work, we investigated whether DC-SIGN receptor is expressed in cells from cutaneous leishmaniasis (CL) lesions as well as the in vitro binding pattern of Leishmania (Viannia) braziliensis (Lb) and L. (L.) amazonensis (La) promastigotes. METHODS DC-SIGN receptor was labeled by immunohistochemistry in cryopreserved CL tissue fragments. In vitro binding assay with CFSE-labeled Lb or La promastigotes and RAJI-transfecting cells expressing DC-SIGN (DC-SIGNPOS) or mock-transfected (DC-SIGNNEG) were monitored by flow cytometry at 2 h, 24 h and 48 h in co-culture. RESULTS In CL lesion infiltrate, DC-SIGNPOS cells were present in the dermis and near the epidermis. Both Lb and La bind to DC-SIGNPOS cells, while binding to DC-SIGNNEG was low. La showed precocious and higher affinity to DC-SIGNhi population than to DC-SIGNlow, while Lb binding was similar in these populations. CONCLUSION Our results demonstrate that DC-SIGN receptor is present in L. braziliensis CL lesions and interact with Lb promastigotes. Moreover, the differences in the binding pattern to Lb and La suggest DC-SIGN can influence in a difference way the intake of the parasites at the first hours after Leishmania infection. These results raise the hypothesis that DC-SIGN receptor could participate in the immunopathogenesis of American tegumentary leishmaniasis accounting for the differences in the outcome of the Leishmania spp. infection.

ABSTRACT

Abstract The aim of this study was to investigate the occurrence, type and severity of anemia at the time of diagnosis of oral cancer, and its potential association with the degree of tumor cell differentiation. This case-control study used 366 medical records of patients treated at two referral centers for oral cancer diagnosis, specifically: cases (n=70) with a histopathological diagnosis of oral squamous cell carcinoma (OSCC) of the oral cavity, and controls (n=296) with benign oral lesions. Sociodemographic, behavioral, and clinical variables of both groups, as well as complete blood count values, were analyzed by descriptive statistics and crude/adjusted logistic regression. Anemia was detected in 15.7% of the cases and 11.8% of the controls. The presence of anemia had an OR=1.64 (odds ratio) (95%CI 0.54-5.00) for OSCC, with no significantly statistical association. Normocytic anemia was the most prevalent form of anemia when oral cancer was diagnosed (91.4% of the controls and 72.7% of the cases), and moderate to severely low hemoglobin levels were associated with OSCC diagnosis (OR 6.49; 95%CI 1.18-35.24), albeit data on hematological examinations were missing.

ABSTRACT

Salmonella Infantis is frequently associated with human infections worldwide and is transmitted by consumption of contaminated foods, particularly those of animal origin, especially the chicken meat. We aimed to evaluate virulence characteristics, antimicrobial resistance and the genetic similarity of 51 strains of S. Infantis isolated from samples of poultry origin. The strains were isolated from 2009 to 2010 in a company with full cycle of broiler's production in the state of São Paulo, Brazil. The antimicrobial susceptibility test was performed and, by PCR, we evaluated the presence of the genes lpfA (hem-adhesion), agfA (hem-biofilm) and sefA (hem-adhesion) and resistance genes to beta-lactams (blaTEM, blaSHV, bla CTX-M and blaAmpC ). The phylogenetic relationship was determined by RAPD-PCR method. Among the drugs tested, the highest percentages of resistance were to amoxicillin (35.3%) and to sulfonamide (15.7%). Eleven antimicrobial resistance patterns were identified (A1 to A11), none of them presented a multiresistance profile (> 3 antimicrobials classes). There was 100% of positivity for the agfA gene, 92.2% for the lpfA gene, and no strain presented the sefA gene. Most of the isolates showed similarities in virulence potential, since they were simultaneously positive for two studied genes, agfA and lpfA (92.2%, 47/51). Of the 18 (35.3%) strains resistant to antimicrobials of the β-lactam class, 10 (55.5%) were positive to blaAmpC gene, five (27.8%) for blaCTX-M , two (11.1%) to blaSHV and no strain presented the blaTEM gene.

ABSTRACT

ABSTRACT Objective to analyze the surface roughness and color stability of the composite resin after surface treatment with the aluminum oxide discs and subsequent exposure to the Advanced Teeth Whitening Strips. Methods 20 specimens of the Filtek Z350XT restorer (3M®) were prepared and daily subjected to home bleaching for 30 minutes during 14 days. A precision rugosimeter was used in order to verify the roughness by means of 3 random readings in the same direction in each sample. For color stability the CIELAB system (L*, a*, b* values) was used through the spectrophotometer (MINOLTA CR -321, Japan). The means of the test specimens as well as the mean of each group were calculated using the random readings. Data statistical analysis were performed by ANOVA- analysis of variance. The level of significance was set at 5% (p ≤ 0.05). Results Roughness did not present great numerical variations. No statistically significant difference between the means obtained concerning the surface roughness of the composite resin with p = 0.44 was observed. However, it was found that there was a statistically significant difference between the means obtained in relation to the color stability of the composite resin, p=0. 007. Conclusion It was concluded that pre-contoured strips containing 6% hydrogen peroxide do not have a significant adverse effect on the roughness of Z350XT (3M®) resin. However, it was also concluded that according to the color stability analysis performed, there may be an indication of the restoration replacement after the bleaching treatment, due to their color change.

ABSTRACT

The importance of helminthoses in different species of butchers is due to the economic damages generated by the condemnation of their carcasses and viscera. The present study evaluated the condemnations for larval migration in livers and lungs in a equidae slaughterhouse under federal inspection located in Minas Gerais from January to December 2016. The 13,636 animals studied were from the States of Goiás, Pernambuco, Bahia and Piauí , the data being obtained through the records of the convictions carried out in the post-mortem inspection according to the criteria of the federal inspection service. Of the condemned livers and lungs, 12% and 8%, respectively, corresponded to the larval migration conviction, and it can be concluded that the parasitic injury is evident in the slaughter of horses, mainly with involvement in a higher percentage of livers, and with the occurrence of parasites also in the lungs.

ABSTRACT

Oral and maxillofacial (OMF) injuries in children may lead to developmental and functional disorders. In this cross-sectional study, we analyzed the pattern of OMF injuries in preschool children admitted to the emergency department of a hospital in southern Brazil. Subjects and method: We retrospectively reviewed the medical records of 1,533 patients admitted from January to December 2011 to identify children aged 0 to 5 years with a diagnosis of OMF trauma. Data were collected on demographic, seasonal and clinical variables. Results: The sample consisted of 288 preschool children (aged 0-5 years) with OMF injuries. Of these, 61.1% were male and 53.2% were <3 years of age. Most children (84.8%) received first medical care from an dentist. Isolated facial injuries were the most common type of injury (85.1%) in this age group. Soft tissue injuries were the most common injury outcome (77.9%), occurring mainly in the nose and lips. Random accidents were the most common cause of OMF trauma (87.7%), and injuries due to falls occurred in 85.9% of these cases. The most common dental trauma was tooth displacement (69.6%). Conclusion: In conclusion, our findings indicate a higher prevalence of OMF injuries in soft tissues, especially in boys in the first three years of life. Falls were the leading cause of injury. Information related to OMF injuries in children are essential for the control of accidents and development of public health policies.

ABSTRACT

Cross-sectional study of 36 children with acute lymphoblastic leukemia (ALL) or acute myeloid leukemia (AML). Intraoral examination and oral hygiene assessment were performed in all patients. Information was also obtained from interviews of patients' mothers and a review of medical records. Results: The sample was largely composed of boys (58.3%), ages 59 years. The most commonly used chemotherapy regimen was the BFM-95 protocol. Most children (83.3%) had a record of some oral manifestation during treatment. On intraoral examination, 17 children (50%) were found to have at least one oral manifestation. The most frequent manifestation was also mucositis (26.5%), followed by gingival bleeding (23.4%). Lower maternal educational level was associated with increased frequency of mucositis, as were induction chemotherapy and maintenance chemotherapy after relapse. Conclusions: Oral abnormalities were common in children receiving chemotherapy and mucositis was the most prevalent manifestation. Oral lesions were associated with the induction phase of chemotherapy. Mucositis was not associated with oral health status.

ABSTRACT

During its life cycle Leishmania spp. face several stress conditions that can cause DNA damages. Base Excision Repair plays an important role in DNA maintenance and it is one of the most conserved mechanisms in all living organisms. DNA repair in trypanosomatids has been reported only for Old World Leishmania species. Here the AP endonuclease from Leishmania (L.) amazonensis was cloned, expressed in Escherichia coli mutants defective on the DNA repair machinery, that were submitted to different stress conditions, showing ability to survive in comparison to the triple null mutant parental strain BW535. Phylogenetic and multiple sequence analyses also confirmed that LAMAP belongs to the AP endonuclease class of proteins.

ABSTRACT

Objective: To identify and analyze cases of OMF treated in an emergency hospital in southern Brazil. Material and Methods: The investigators conducted a retrospective, cross-sectional study. The sample was composed of 1,533 patient medical records. Demographic, seasonal, and clinical variables were collected. Additionally, the medical records also included data on etiological factors, injury site, and tissues involved. A descriptive analysis was conducted based on 1,179 (76.9%) records of OMF injuries. Results: Most patients were male children and adolescents. Accidents (59.7%), such as falls, and interpersonal violence (18.7%), such as physical aggression, were the main etiological factors. Most injuries involved soft tissue (66.4%), especially in the lip region; whereas fractures affected the nasal bones. Transfers and hospitalizations accounted for 8.1% of clinical outcomes and dentists participated in the treatment of 85% of cases. Conclusion: The results of this study suggest a higher prevalence of OMF injuries in men, affecting young people more frequently and showing falls as the main etiological factor. Dentists participated in the majority of the medical care. Future studies will focus on determining risk groups, developing preventive programs, and improving health care services, with the participation of dentists in the multidisciplinary team.

ABSTRACT

Actinic cheilitis is a potentially premalignant condition that is primarily found on the lower lip. Etiology is related to chronic exposure to ultraviolet light and the condition exhibits a strong predilection for people whose work involves intense exposure to the sun. Clinically observable signs include white or reddened areas of maculation or whitish plaques caused by hyperkeratinization of the lip epithelium, which can progress to a point at which the demarcation between the skin, vermillion border of the lip and the mucosa is lost. These lesions are generally asymptomatic but may progress to form areas of erosion and/or ulceration. There are a number of treatments available, one of which, vermilionectomy, is a surgical procedure in which the vermillion border of the lip is partially or totally removed and which, in addition to being inexpensive, offers the advantage of providing material for pathological analysis. The objective of this study is to describe the vermilionectomy technique as used to treat a patient diagnosed with actinic cheilitis...

ABSTRACT

Introduction: There are described in the literature several techniques for shaping the face mask in order to develop models for fabrication of maxillofacial prostheses. Objective: This article aims to describe two techniques used for facial moulage, the conventional technique and conventional technique with modification, highlighting their advantages and disadvantages. Case Report: In the first technique, the impression material is deposited over the region of interest and then a reinforcement layer of plaster type II is putting to cover the impression material. The modification of the conventional technique begins with a relief with cotton over the facial tissue, applying a layer of plaster type II in order to construct the reinforcement layer and, after its crystallization, the application of impression material over the area to be molded, followed by its plaster mask contention. Conclusion: The choice of impression technique and materials used will depend on the ability of the professional. The important thing is to make a moulage in order to get an accurate facial cast for the prosthesis' success.

ABSTRACT

Introduction: The influence of psychological factors on temporomandibular disorders (TMD), such as depression, anxiety and stress has been very discussed in literature. However, there is no consensus about their influence on the clinical manifestation of TMD. Objective: To evaluate the evolution of minor psychiatric disorders and pain symptoms in patients with temporomandibular disorders (TMD) treated with occlusal splints and rehabilitated with dental prosthesis. Material and methods: Sixty volunteers, both genders, aging from 20 to 65 years, diagnosed with TMD, were randomly selected within the university's patient databank. The volunteers were divided into two groups: G1 - 30 males and G2 - 30 females. The volunteers underwent a standard clinical evaluation for TMD diagnosis. Psychological evaluations were performed through Goldberg's General Health Questionnaire (GHQ), before and after 24 months of treatment. TMD treatment comprised occlusal splints and rehabilitated with dental prosthesis. The results were evaluated by Mantel-Haezel, Wilcoxon and Mann-Whitney statistical tests. Results: According to the criteria established by GHQ, the interpretation of the symptom scores should be applied based on gender, because the scores have different values for male and female, consequently no comparisons were made between the groups. There were significant statistical difference in G2 when the variables psychic stress (p = 0.002) and psychosomatic disorders (p = 0.007) were observed. In G1, the variables for psychosomatic disorders (p = 0.002) and general health (p = 0.021) were statistically significant. Significant differences were found in both groups for all the evaluated symptoms (p < 0.005). Conclusion: The used therapy positively interfered in the remission of symptoms and in the incidence of minor psychic disorders of TMD patients.

ABSTRACT

ABSTRACT

ABSTRACT

The objective of the present study was to determine the prevalence of Cervical Spine Disfunction (CSD) in volunteers with Temporomandibular Disfunction (TMD). 200 volunteers were selected from the screening service of Dentistry School of Piracicaba, State University of Campinas, which were evaluated by records prepared by the Center for Research and Treatment of Functional Alterations of the Stomatognathic System (CETASE) for the diagnosis of TMD. Volunteers who presented positive signs and symptoms of TMD underwent a new clinical examination for C5D. The results showed a prevalence of 69% of CSD in volunteers with TMD. The most frequently signs and symptoms observed for the two diseases were the presence of joint noise, temporomandibular joint pain, pain on palpation in the masseter and trapezius muscles. In conclusion, due to the significant association between prevalence of CSD and TMD becomes important clinical research of neck structures during the physical examination for the diagnosis ofTMD.

ABSTRACT

This in vitro study evaluated the effectiveness of clinical methods (visual), radiographic and histological diagnosis of incipient lesions in order to determine the most conservative treatment plan.Methodology: 20 human permanent molar teeth which had apparently intact occlusal surface, darkened but without cavitation, were evaluated. These teeth were subjected to X-ray, using periapical radiographs and visual examination by three calibrated examiners with the aid of reflectors dentistry. The occlusal surfaces of the teeth were photographed and then they were serially sectioned on a diamond cutting machine, resulting in cuts to 0.375 mm thick. Slides were obtained for histological assessment of demineralization in enamel and dentin of the teeth by light microscopy. Scores were used to classificate accessed diagnostic methods and they were evaluated by statistical Gamma, with a significance of 0.05.Results: both clinical and radiographic diagnosis failed to determine the presence, extent and location of tooth decay and there was no correlation between the three methods (p>0.05).Conclusion: it is essential that the Dental Surgeon conects information derived from complementary methods of diagnosis with their prior knowledge in order to choose between the restorative or conservative treatment of these lesions...

ABSTRACT

This study evaluated the marginal microleakage in temporary filling materials used in Endodontics, in different time intervals (3 and 7 days). One hundred and twenty uniradicular human teeth comprised the sample of this study. After endodontic treatment they were randomly divided into 6 groups (n=20) according to the temporary restoring material used: Group 1: Vidrion R, Group 2: IRM, Group 3: Provisory, Group 4: Dentalville, Group 5: Gutta percha and Super Bonder (positive control), and Group 6: cotton balls (negative control). After restoration the teeth were were impermeabilized with a nail varnish and submitted to thermocycling with 500 cycles (0+/- 5°C e 55 +/- 5°C). Subsenquently, the teeth were immersed in 2% blue methylene solution for 3 and 7 days, longitudinally sectioned and examined by means of a stereoscopic lens with magnification of 40x. The Mann Whitney and Kruskal Wallis tests were applied for statistical analysis. The results were statistically significant for the Dentalville material at the time interval of 3 days (p<0.001). After 7 days only group 5 (positive control) did not show marginal microleakage, and there was no significant differences between the other groups. It may be concluded that Dentalville temporary restoring material showed the smallest indexes of microleakage at time interval of 3 days when compared to the other materials evaluated.

ABSTRACT

Obective: evaluate the bacterial contamination in composite resins used daily at graduation clinical of FO/UFJF. Methods: for detection of bacterial growth on culture of samples from 50 tubes of composite resin, and 25 Charisma ® (Heraeus-Kulzer) and 25 Durafill ® (Heraeus-Kulzer). Two new tubes of each brand served as the control group and the other tubes of composite resin used in the study were in use in clinical studies, forming the test group. The collection of samples was performed with aseptic technique in the chapel of laminar flow, using previously sterilized toothpicks. After collecting the material and inoculum in the culture medium, the evaluation systems were maintained in bacteriological incubator at 37º C. Results: after 24 hours of incubation, observed the turbidity of the culture medium in only 2 test tubes of test group. The results were submitted to Pearson's Chi Square, not showing statistically significant results (p>0.005). Conclusion: the composite resins used in the FO/UFJF were not contaminated and it is believed that the technical maintenance of the aseptic chain employed in clinical studies of the institution were effective, but may not be the only ones responsible for not contamination.
