ABSTRACT

To compare the coronary atherosclerotic plaque 64-slice spiral CT characteristics and the risk factors of Han [in Inner Mongolia] and Mongolian coronary artery disease patients. The plaques of 126 Mongolian and 269 Han patients were analyzed by 64-slice spiral CT coronary angiography. Their gender, age, height, body mass, the history of hypertension, diabetes, smoking and family diseases, the levels of triglycerides [TG], total cholesterol [TC], high density lipoprotein cholesterol [HDL-C] and low density lipoprotein cholesterol [LDL-C] were compared. The incidence of plaques [P < 0.05], the proportion of plaques in the circumflex branch [P < 0.05], the proportion of medium-severe lumen stenosis induced by plaques [P < 0.05], and the proportion of obstructive plaque involved multi-branch [P < 0.05] of the Mongolian patients were higher. The plaque compositions of the two groups did not differ significantly [P > 0.05]. The body mass index of the Mongolian patients was higher [P < 0.05]. The hypertension, diabetes, smoking history, TG, TC, HDL-C and LDL-C of the two groups did not differ significantly [P > 0.05]. The higher incidence of coronary atherosclerotic plaques and the more severe lesions of the Mongolian patients may be related to their higher body mass index

ABSTRACT

MicroRNA (miRNA) is a class of micro,endogenously-initiated non-coding RNA that negatively regulate gene expression at the post-transcriptional level and involved in tumorigenesis.Polymorphisms in miRNA sequences (miRSNP) alter mature miRNA levels and target mRNA selection.Recent findings have demonstrated that miRSNP mediated epigenetic alteration of miRNA can be one of the mechanism in tumor progression and may play critical role of hallmarks in cancer susceptibility.This review described the most recent advances in the field of hot miRSNP and susceptibility of tumour.

ABSTRACT

ObjectiveTo detect the expressions of orotate phosphoribosyl transferase (OPRT) and dihydropyrimidine dchydrogenase (DPD) mRNA in gastric cancer tissues,and investigate their relationship with the clinicopathological factors.MethodsThe gastric cancer tissues and adjacent normal tissues were collected from 53 patients with gastric cancer at the Affiliated Hospital of Inner Mongolia Medical College from June 2007 to November 2008.The mRNA expressions of OPRT and DPD were detected by reverse transcriptional-polymerase chain reaction,and the correlation between the mRNA expressions of OPRT and DPD and the clinicopathological factors was analyzed.All data were analyzed by using the t test and the one-way analysis of variance.Results The mRNA expression of OPRT in the gastric cancer tissue was 1.15 ± 0.56,which was significantly higher than 0.88 ± 0.31 in the adjacent normal tissues ( t =3.66,P < 0.05 ).The mRNA expressions of DPD in gastric cancer tissues and the adjacent normal tissues were 0.95 ± 0.50 and 0.90 ± 0.41,respectively,with no significant difference between the 2 groups (t =0.68,P > 0.05 ).The mRNA cxprcssions of OPRT in the gastric cancer tissues with no lymph node metastasis was 1.42 ± 0.54,which was significantly higher than 1.00 ± 0.52 in the gastric cancer tissues with lymph node metastasis (t =7.94,P < 0.05 ).Poorly differentiated adenocarcinoma (0.93 ± 0.24) and mucinous adenocarcinoma (1.58 ± 0.38) showed a significantly higher DPD mRNA expression than moderately differentiated or well differentiated adenocarcinoma ( 0.67 ± 0.36 ) ( F =27.71,P < 0.05 ).Conclusions The mRNA expre ssion of OPRT in gastric cancer tissue is higher than that in the adjacent normal tissues,and its expression in patients with lvmph node metastasis is lower than that in patients without lymph node metastasis.Poorly differentiated adenocarcinoma showed higher DPD mRNA expression than moderately or well differentiated adenocarcinoma,and its expression is highest in mucinous adenocarcinoma.

ABSTRACT

Objective To investigate the relationship between the fluorouracil pathway gene and effect of chemotherapy in advanced gastric cancer after surgery. Methods 52 postoperative patients with advanced gastric cancer using FOLFOX4 6-cycles combined chemotherapy were collected to set up the database. The expression of thymidine synthase (TS), dihydropyrimidine dehydrogenase (DPD) and orotate phosphoribosyltransferase(OPRT) in tumor tissue and adjacent non-tumor tissue of 52 patients with advanced gastric cancer were measured by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). The influence of fluorouracil pathway gene on chemotherapy was investigated. Results The TS-mRNA level in tumor tissue was significantly higher than non-tumor tissue (0.92±0.28 vs 0.71±0.30) (t = 3.730, P =0.001).OPRT-mRNA level in tumor tissue was positively correlated with the non-tumor tissue (r =0.45, P =0.001). No correlations were observed among other gene expressions. Patients whose high OPRT-mRNA gene expression in their tumors and non-tumor tissue showed an obviously better survival (t = 3.036, P =0.003;t = 2.713, P =0.009). Patients with low DPD-mRNA gene expression survived longer than those with high DPD-mRNA gene expression in tumor tissue with statistical significance(t = 2.708, P =0.009), whereas prolonged survival was observed in patients with high DPD-mRNA gene expression in non-tumor tissue (t = 2.616, P =0.012).Conclusion There is close relationships between chemotherapy in advanced gastric cancer and the expression of DPD-mRNA, OPRT-mRNA;while the expression of TS-mRNA have no relation with the survival time.

ABSTRACT

Objective To investigate the feasibility and cosmetic results of endoscopic thyroidectomy via breast areola approach.Methods From January 2004 to January 2006,endoscopic thyroidectomy was performed through the breast areola approach in 18 patients with benign thyroid mass.Results The surgery was accomplished successfully in all the 18 cases.The operation time ranged 45-120 min(mean,62 min) for unilateral resection and 90-180 min(mean,98 min) for bilateral resection.The intraoperative blood loss ranged 6-20 ml(mean,11 ml) for unilateral resection,and 15-70 ml(mean,29.5 ml) for bilateral resection.Postoperative hospital stay was 3-5 days.No complication occurred after the operation.The patients were followed up for 6-12 months(mean,9 months).The wound healed well in all the patients,inconspicuous scars or no recurrence was observed during the follow-up.Conclusions Endoscopic thyroidectomy via breast areola approach is a safe and feasible procedure with satisfying neck cosmetic outcome.

ABSTRACT

Objective:To investigate the expression of metabolic enzymes of fluoropyrimidines in primary colorectal cancer. Methods: Sixty-one cases of pathological confirmed primary colorectal cancer were collected in Beijing Cancer Hospital from August 2003 to February 2004. The expressions of thymidylate synthase (TS), dihydropyrimidine dehydrogenase (DPD) and orotate phosphoribosyl transferase (OPRT) were detected in paired tumor tissues and non-cancerous tissues by RT-PCR. Results: Expression of each enzyme in tumor tissues was positively correlated with that in paired non-cancerous tissues respectively. Compared with the expression of each enzyme in cancer with that in non-cancer, OPRT was higher in cancer while that of DPD and TS was comparable. The expression of OPRT was approximately 4.38-folds higher in colorectal cancer tissues than that in non-cancerous tissues (P0.000). The lowest expression of OPRT was found in mucinous carcinoma while the highest levels in poorly-differentiated adenocarcinoma. There were correlations between the ratio of tumor tissues / non-cancerous tissues OPRT expression (T/N) and metastasis to the lymph nodes (r0.36; P0.005), small vessel invasion (r0.26; P0.041) and differentiation (r-0.33; P0.009). Conclusion: The detection of three enzymes by means of RT-PCR is practical and can be used in clinical application. Our results also suggest that OPRT is a potential biomarker in predicating prognosis of colorectal cancer.

ABSTRACT

Objective To explore the relationship between cholecystoadenomyomatosis and gallbladder cancer.Method The expression of p53,bcl-2,EGFR oncoproteins were examined by method of immunohistochemistry in benign lesions and carcinomas in gallbladder.Results The study revealed that overexpressions of p53,bcl-2,EGFR oncoproteins were detected in 0,4,3 cases of 21 patients with chronic cholecystitis;0,5,3 cases of 20 patients with cholecysto-adenomyomatosis and 16,14,11 cases of 25 patients with adenocarcinoma of the gallbladder respectively.There were significantly differences in single or multiple oncogene expression rates among groups,but not in chronic cholecystitis to adenomyomatosis.Conclusions These results suggests that oncogenic changes of p53,bcl-2,EGFR expression may play a role in gallbladder oncogenesis,cholecystoadenomyomaltosis should not be considered as an important precancerous lesion of gallbladder.
