ABSTRACT

【Objective】 To investigate the effects of dexmedetomidine (DEX) intervention on the expressions of chondrocytes and related factors in vitro and its possible molecular mechanisms. 【Methods】 C28/I2 normal human chondrocyte lines were cultured in vitro, and dexmedetomidine at the concentration of 1 μmol/L was selected to intervene for 24 h and 48 h, respectively. The morphology and cell density of chondrocytes were observed after DEX culture at different time points. Immunofluorescence technique was used to detect the expression levels of disintegrin and metalloproteinase with thrombospondin motifs 5 (ADAMTS5) in chondrocytes in each group. The expression levels of Adamts5, aggrecan (Acan), versican (Vcan), Furin, proprotein convertase subtilisin/kexin type 6 (Pcsk6), collagen type Ⅰ alpha 1 (Col1a1), collagen type Ⅱ alpha 1 (Col2a1), collagen type X alpha 1 (Col10a1), and SRY2 related high mobility group box gene9 (Sox9) were detected by RT-PCR. Adamts5 gene knockout chondrocytes were constructed by lentivirus transfection technology and treated with DEX; RT-PCR was used to detect the effects of DEX on the expression levels of Acan, Vcan, Furin, Pcsk6 and Sox9 after Adamts5 gene knockout. 【Results】 After 24 and 48 h of intervention with 1 μmol/L DEX, the morphology and size of chondrocytes did not change significantly, but the cell density increased slightly. Immunofluorescence assay showed that the expression of ADAMTS5 increased at first and then decreased after DEX treatment for 24 and 48h, respectively (P=0.032). RT-PCR results showed that with the extension of intervention time, the expression of Adamts5 first increased and then decreased. The expression difference between 48 and 24 h after culture was statistically significant (P=0.032). The change trend of Pcsk6 was the same as that of Adamts5, while the change trend of Acan expression was opposite that of Adamts5. Chondrocytes knocked out Adamts5 gene and intervened with DEX for 24 and 48 h. The results of RT-PCR showed that the expression of Pcsk6 decreased while that of Acan increased and the changes were significant. 【Conclusion】 Dexmedetomidine may activate ADAMTS5 zymogen through Pcsk6, thereby promoting proteoglycan degradation in chondrocytes.

ABSTRACT

Seven cases of acute leukemia treated by fetal liver transplantation (FLT) are reported in this article. Patients received intensive chemoradiotherapy with cycl-ophosphamide and total body irradiation (TBI), followed by an infusion of fetal liver cells.Hematologic reconstitution occurred in all patients, and temporary chimera developed in 3 patients after FLT. Until the time of analysis, 3 patients remained in complete remission (55, 30 and 20 months), one relapsed but was still alive (6 rronths), and 3 died from relapse or bacterial sepsis (3, 7 and 11 months).

ABSTRACT

5.16 log of K562, U937, HIMeg and HL60 cells, respectively, but spare 23.37 ? 25.66% of normal CFU-GM (0.63 log killing). When HL60 cells mixed with normal mononucleated bone marrow cells in 1:9 ratio were studied, the killing of HL60 colonies was more than 4.29 log. The result suggested that photocarcinorin plus light could eliminate leukemic progenitors, and might be used in clinical ABMT. Several factors that might affect the photosensitization effect on cells were discussed.

ABSTRACT

Twenty two cases of acute agranulocytosis were treated with fetal liver infusion (FLI) in this study. The total effective rate was 90.91%. The FLI group markedly increased leukocyte counts and absolute value of granulocytes as compared with the control group. There were no significant side effects. The result shows that FLI would be helpful in the treatment of acute agranulocytosis. The mechanism of treatment of acute agranulocytosis with FLI is also discussed.

ABSTRACT

The present paper reports the clinical observations on 45 cases of multiple myeloma. In 35 patients, the tumors were immunologically typed as type IgG in 16, IgA type in 9, 2 each of ?, ? type light chain disease (LCD) and double light chain disease (D-LCD). 2 of IgD type, 1 each of non-secreting type (n-SMM) and ? heavy chain disease (?HCD). Further immunological subtyping dis-closed that most of the cases belonged to IgG. ? and IgA1 ? types.

ABSTRACT

Peripheral blood mononuclear cells (PBMNC) isolated from patients with acute leukemia (AL) and from normal controls were cultured in medium containing 1000 units/ml of recombinant interleukin 2 (IL-2). Marked LAK activity was induced on the third culture day in the normal controls,, with the highest cytotoxicity appearing between day 3 and 5 whereas induction of LAK activity in the AL patients began on the 5th day of culture, with the peak level appearing at day 15, showing that the peak of LAK activity was significantly delayed in AL. LAK cells surface phenotyping tests showed that CD8+ and CD16+ positive cells began to increase significantly from day 5 and reached the highest level at week 3, whereas CD4+ subclass began to decrease on day 5 and dropped to the nadir at week 3. The proportion of CD8+ and CD16+ cells were positively cor related with LAK activity, but, that of the CD4+ cell was inversely related with the LAK activity.
