ABSTRACT

【Objective】 To investigate the feasibility of allogeneic platelet-rich plasma (PRP) for the treatment of herpes zoster wounds secondary to systemic lupus erythematosus (SLE), especially large ulcer wounds. 【Methods】 The treatment process of a patient with massive herpes zoster wounds in perineum and hip accompanied by extensive soft tissue necrosis secondary to SLE was retrospectively analyzed. The clinical efficacy of allogeneic PRP was explored combined with treatment key points and literature review. 【Results】 The patient′s wound bed was prepared until the wound was fresh, then treated externally with allogeneic PRP 3 times a week. The wound was healed completely after 42 days. 【Conclusion】 In the case of autologous PRP unavailable or unsuitable, allogeneic PRP is a safe alternative, which can effectively promote tissue regeneration, and this patient achieved curative effect in a short period of time.

ABSTRACT

【Objective】 To investigate the effect of leukocyte filter in removal of tumor cells in blood and the viability of using leukocyte filter in intraoperative cell salvage in tumor patients. 【Methods】 The leucocyte-depleted suspended RBCs prepared from 200 mL human whole blood were spiked with HCT116 cells cultured in vitro to simulate the autologous blood recovered from tumor patients during operation. Conventional filter (group A, n=6), filter with small pore size (group B, n=6) and filter with thicker membrane and small pore size (group C, n=6)) were used. The CD45-EpCAM+ tumor cells were detected by anti-CD45-PE and anti-CD326 (EpCAM)-FITC flow cytometry, and the absolute count of tumor cells were calculated using Flow Count fluorescence microspheres. The activity of tumor cells was detected by cell culture and trypan blue staining. Furthermore, blood biochemical indexes (LDH, K+ ) and red cell recovery rate were detected. 【Results】 Tumor cell counts before and after the application of leukocyte filter A, B, C were(1.52±0.48)×107/mL vs(2.73±1.74)×104/mL, (1.48±0.55)×107/ml vs(2.96±1.85)×104/mL and (1.44±0.46)×107/mL vs(3.08±2.33)×104/mL, respectively (P >0.05), which was significantly reduced after filtration (P < 0.01). No viable cells were found in the filtered blood cultured for 7 days. There were no significant differences in K+ and LDH value before and after filtration among the three groups(P >0.05). The blood recovery rates of group A, B, C were 85.22%, 84.97% and 82.86%, respectively. 【Conclusion】 The conventional filter can significantly reduce the number of circulating tumor cells, and it is feasible for intraoperative cell salvage in tumor patients.

ABSTRACT

Objective To explore the relationship between case mix index ( CMI ) and the bed allocation in clinical departments, and to evaluate the performance of bed scale of departments at tertiary hospitals based on CMI. Methods Based on the HIS system of a tertiary hospital, the authors collected the disease treatment information and bed allocation information of each clinical department in 2015. The CMI value was calculated by means of diagnosis-related group( DRG) , while the proportion of cases and average daily beds occupancy corresponding to the quartile of the CMI value of each department was also counted. Then the bed occupancy and distribution of various patients were analyzed. Results The hospital′s overall CMI value, DRG groups, DRG cases at the departments, days of stay, DRGs, CMI values and its quartiles were calculated. It was found that the medical and surgical CMIs had a linear relationship with the critical proportion (r=0. 998, 0. 996, P<0. 001). The main influencing factors of average daily bed occupancy were ranged from high to low as followed: the number of cases that were allocated in the group, employee numbers and CMI (F=87. 656, P<0. 001). Conclusions CMI is an ideal indicator for the difficulty of medical services at different departments and for evaluating the bed scale performance. Hence it can be used as an important reference for bed number adjustment in hospitals.

ABSTRACT

At the present,systemic lupus erythematosus is a heterogeneous disease in which diagnosis,especially early diagnosis,is always rather difficult.Because of this,the systems of classification criteria for clinical diagnosis,which include recommendations of the American college of rheumatology and the systemic lupus international collaborating clinics,have been utilized since 1971.However,there is still no single biomarker that is pathognomonic for the disease.Currently,the diagnosis still requiresold-fashioned clinical acumen with the assistance of standardized clinical and laboratory criteria.In the light of the commentary,future research may lead us to define subtypes of lupus from a genomic and proteomic standpoint.

ABSTRACT

Objective Irregular antibody screening and identification are very important to reduce hemolytic reaction.The dis-tribution of irregular antibodies of blood group was analyzed in patients, to provide evidence for using Micro column gel test to detect and identify irregular antibodies transfusion safety. Methods Micro column gel anti-globulin technique was used to screen the irregular an-tibodies and cross-matching in 29770 patients.If the serum had irregular antibodies, the spectrum cells were used to identify the specific-ity of irregular antibodies. Results Among 29 770 patients, 120 cases had irregular antibodies of blood group.The positive rate was 0.4%.The irregular antibodies include Rh system(n=67),MNS system (n=7), Lewis system(n=5),cases of Kidd system(n=3) and undefined specified antibodies(n=38).Among the 120 antibody-positive patients, 70 were female patients and 50 were male patients. Further study showed that the pregnancy history was to the main factor causing irregular antibodies in women (46 positive cases among 70 female patients) . Conclusion The study showed that some patients had irregular antibodies of blood group, especially female patients with pregnancy history.These results suggested that irregular antibody screening and cross-matching by micro column gel method in pa-tients before transfusion had great significance for ensuring the blood transfusion safety.

ABSTRACT

Objective:To evaluate the effectiveness of using two specimens for ABO grouping on the prevention of ABO-incompatible transfusions caused by sample errors. Methods A two specimen policy for blood samples of pre-transfusion testing was implemented from July 2007. This policy was defined as that for patients requiring transfusion or likely to require transfusion,ABO group was determined twice on 2 separate-samples. The rate of mislabeled samples and miscollected samples prior to blood transfusion were recorded and analyzed.Results From Jul 2007 to Jun 2009,79 115 samples for pre-transfusion compatibility testing were accepted. A total of 273 samples were identified as mislabeled. The rate of mislabeled samples was 1 in 290. The rate of miscollected samples with correction for silent errors was 1 in 1485 (23 miscollected samples of 47 507 repeat samples,with a correction factor of 1.391 2). Of the 23 miscollected samples detected by double-step typing policy,9 cases (39%) could have caused ABO-incompatible transfusions.Conclusion The risk of ABO-incompatible transfusions caused by mislabeled and miscollected samples can be reduced by two specimen policy.

ABSTRACT

Objective To study the effects of phase change material (cooling agent) on the quality of blood after long-distant transportation.Methods A total of 12 bags, packed with pre-chilled phase change materials, were put into regular blood transportation kit, and the new blood transportation kit, which was developed by our hospital. In each kit there were 54 U of red blood cell suspensions. The temperatures in the kit were recorded during the transportation at different times.Results The temperature in the kit was kept between 1—7℃ during the 72h transportation. The temperature met the needs of red blood cell transportation.Conclusion Phase change material could be used as an effective cooling agent for long distance blood transportation.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the effects of zinc and selenium on the sperm quality of oligoasthenozoospermic men.</p><p><b>METHODS</b>Thirty-four cases of oligoasthenozoospermia were treated with Xinxibao (Zinc & Selenium Tablet) three times a day and five tablets at a time for ninety days in succession. The seminal routine analysis was performed by computer-assisted sperm analysis (CASA) at the end of each month.</p><p><b>RESULTS</b>The sperm quality was improved significantly sixty days and ninety days after treatment. Five cases (14.7%) showed remarkable effect, 25 (73.5%) improved, and 4 (11.8%) did not respond.</p><p><b>CONCLUSION</b>Zinc and selenium can significantly improve the sperm quality of oligoasthenozoospermic men.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the segregation of sex chromosomes in the spermatozoa of a 46, XY/47, XXY patient with oligozoospermia.</p><p><b>METHODS</b>The number of X and Y chromosomes of the ejaculated spermatozoa from the patient with mosaic 46, XY/47, XXY was analysed by X/Y dual fluorescence in-situ hybridization (FISH).</p><p><b>RESULTS</b>Of the 100 spermatozoa analysed, 97 showed either one X chromosome-specific green signal or one Y-chromosome-specific red Y signal in each spermatozoon and only 3 showed no signal. The frequencies of X- and Y-bearing spermatozoa were 49% and 48% respectively. The ratio of X- to Y-bearing spermatozoa was about 1:1 as expected. There was no statistical difference between the chromosome XX and XY frequencies in each spermatozoon from the patient in comparison with those estimated in the control.</p><p><b>CONCLUSION</b>The spermatozoa of 46, XX/47, XXY mosaic patients have a normal gonosomal complement, which allows infertility treatment to be carried out by ICSI.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To measure uric acid of seminal plasma in fertile and infertile males.</p><p><b>METHODS</b>One hundred and sixty-three infertile males were divided into an obstructive azoospermic group (15 cases), a non-obstructive azoospermic group (36 cases), an oligozoospermic group (43 cases), and an asthenozoospermic group (69 cases). Twenty fertile males were included in the control group. Uric acid concentrations of seminal plasma in the fertile and infertile men were assessed by spectrophotometer, and sperm parameters were analyzed by computer-assisted semen analysis (CASA) system.</p><p><b>RESULTS</b>Uric acid concentration of seminal plasma in the control group was significantly higher than all the infertile groups (P < 0.01), and that of the obstructive azoospermic group significantly lower than the other infertile groups (P < 0.1), but no significant difference was observed among the other infertile groups (P > 0.05).</p><p><b>CONCLUSION</b>Uric acid may play an important role in male reproduction because of its antioxidative property.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To report a true hermaphroditism due to a teragametic chimerism and to discuss the pathogenesis of tetragametic chimerism.</p><p><b>METHODS</b>Chromosomal analysis and fluorescence in situ hybridization(FISH) were carried out on the lymphocytes from the blood and on the fibroblasts from the cultured skin and on fibroblasts from two different kinds of gonadal tissues of the patient with ambiguous genitalia respectively. Blood groups, human leukocyte antigen (HLA) haplotyping and 77 short tandem repeat (STR) microsatellite markers were tested. The two kinds of tissues in the gonad were detected by histopathological examination. Blood groups, HLA haplotying and 77 STR microsatellite markers parents of the patient's were also analyzed.</p><p><b>RESULTS</b>Either 46,XX or 46,XY karyotype was found in the lymphocytes of the blood and in the fibroblasts of the cultured skin and of the two different kinds of gonadal tissues. Two X chromosome-specific signals or one X and one Y signal were detected in each interphase nucleus by FISH from the lymphocytes of the blood and the fibroblasts of three different tissue cultures. The karyotype of the 46,XY cell line predominated in all cultures except the cultured-fibroblasts from yellow gonadal tissues. STR marker analysis, ABO grouping and HLA study from the patient were identified a single haplotype in the patient from the mother and two different haplotypes from the father. Two kinds of tissues in the gonad were observed by histopathological examination. The yellow tissue was ovary and the white one was testis.</p><p><b>CONCLUSIONS</b>Histopathological examination and chromosomal analysis combined with FISH are very useful methods for the diagnosis of true hermaphroditism. Blood typing, HLA and short tandem repeat microsatellite markers afford strong evidence for confirming tetragametic chimerism. The mechanism of tetragametic chimerism in true hermaphroditism can be explained by a parthenogenetic division of a haploid nucleu into two identical gametes, followed by fertilization with both X and Y spermatozoa and then developed into an organism.</p>

ABSTRACT

The sperm DNA integrity is important for the success of natural or assisted fertilization, as well as normal development of the embryo, fetus and child. Some investigations have demonstrated that when the damaged spermatozoa DNA of the husband exceeds than 30%, the couple has very low potential of natural fertility. Several methods are available for the assessment of sperm DNA integrity, which is considered to be a better marker of male fertility potential than conventional semen parameters. Furthermore, monitering sperm DNA integrity may become an important part of the evaluation for couples seeking assisted reproductive techniques such as intracytoplasmic sperm injection (ICSI). Finally, the effects of cryopreservation and preparation techniques on sperm DNA integrity are summarized.

ABSTRACT

Objective:To observe whether nitric oxide (NO) could enhance the damaging effect of cyclophosphamide on L1210 cells cultured in vitro, and to investigate the mechanism of this action. Methods:L1210 cells were co-cultured with 3T3 cells in DMEM medium supplemented with cyclophosphamide (400 ?g/ml). The L1210 cells were divided into three groups based on different transfected 3T3 cells: pcDNA3.0-iNOS plasmid transfected 3T3 cells (Group 1), pcDNA3.0 plasmid transfected 3T3 cells (Group 2), pcDNA3.0-iNOS plasmid transfected 3T3 cells plus DEVD-CHO(Group 3). The viability and apoptosis rate of L1210 cells at different culture periods were determined by trypan blue exclusion and TUNNEL method, respectively. And the cell cycles at G_1 and S phase were detected by flow cytometry. ABSTRACT

Objective To test the performance of field blood transportation vehicle from the aspects of heat preservation,refrigeration and oil consumption.Methods Two vehicles without power supply were stowed with maximum load,one with red cell suspension,and the other with frozen plasma.With the environmental temperature higher than 35 ℃ or lower than 25 ℃,related time lengths were detected respectively.Results The holding time of the vehicles was from 1.5 h to 7 h,refrigerating time was from 15 min to 138 min,and a full tank of gasoline was exhausted around 5~7 hours.Conclusion During line-haul,field blood transportation vehicle has to be equipped with additional cooling agents,in case the refrigeration equipment out of work for the reason of gasoline exhaustion.

ABSTRACT

Objective:Temperature keeping is vital in the long distance transportation of red blood cell suspensions in military affairs and emergency.This study aimed to develop a composite phase change material to be used for preserving blood in long time transporation without electricity.Methods: The n-tetradecane was adsorbed to the pore structure of silica aerogel through the capillarity and hydrophobicity of silica aerogel.The efficiency of the phase change material was detected by differential scanning calorimetry.Results: The specific surface area and the adsorption capacity were 863.59 m2/g and 9.16,respectively.The phase change temperature was 5.47℃ and phase change latent heat was 180J/g.Conclusion: The n-tetradecane /silica aerogel composite phase change material could be used for blood preservation.

ABSTRACT

Objective: To investigate the temperature control methods for long-distance transportation of red blood cell suspensions in military affairs or emergence.Methods: We loaded blood transport cars with red blood cell suspensions to full capacity and kept the temperature by combined cooling methods.Results: The temperature in the container was kept between 2-6.2℃ during the 72 h transportation,even when the refrigeration system of the car was out of work.Conclusion: The required temperature for long-distance transportation of red blood cell suspensions could be attained by combined cooling methods in case of insecurity of transportation time and the effect of single cooling method.

ABSTRACT

Objective To explore the effects of ultraviolet irradiation on platelet. Methods Three assays were been developed.These include platelet count,release of alpha granule membrane protein 140(GMP-140)and morphology of platelet by electron micrograph in those samples before and after ultraviolet irradiation.Results Compared with the nonirradiated controls,the irradiated units showed significant changes including decrease of platelet count,increase of release of GMP-140 and platelet lesion in morphology.Conclusion Ultraviolet irradiation may have activation and lesion effect in platelet.
