ABSTRACT

Danshensu [3-(3, 4-dihydroxyphenyl) lactic acid, DSS], one of the significant cardioprotective components, is extracted from the root of Salvia miltiorrhiza. In the present study, an ester prodrug of Danshensu (DSS), palmitoyl Danshensu (PDSS), was synthesized with the aim to improve its oral bioavailability and prolong its half-life. The in vitro experiments were carried out to evaluate the physicochemical properties and stability of PDSS. Although the solubility of PDSS in water was only 0.055 mg·mL, its solubility in FaSSIF and FeSSIF reached 4.68 and 9.08 mg·mL, respectively. Octanol-water partition coefficient (log P) was increased from -2.48 of DSS to 1.90 of PDSS. PDSS was relatively stable in the aqueous solution in pH range from 5.6 to 7.4. Furthermore, the pharmacokinetics in rats was evaluated after oral administration of PDSS and DSS. AUC and t of PDSS were enhanced up to 9.8-fold and 2.2-fold, respectively, compared to that of DSS. C was 1.67 ± 0.11 μg·mL for PDSS and 0.81 ± 0.06 μg·mL for DSS. Thus, these results demonstrated that PDSS had much higher oral bioavailability and longer circulation time than its parent drug.

ABSTRACT

This study aimed to establish a method for the detection and identification of H7N9 avian influenza viruses based on the NA gene by pyrosequencing. According to the published NA gene sequences of the avian influenza A (H7N9) virus, a 15-nt deletion was found in the NA gene of H7N9 avian influenza viruses. The 15-nt deletion of the NA gene was targeted as the molecular marker for the rapid detection and identification of H7N9 avian influenza viruses by pyrosequencing. Three H7N9 avian influenza virus isolates underwent pyrosequencing using the same assay, and were proven to have the same 15-nt deletion. Pyrosequencing technology based on the NA gene molecular marker can be used to identify H7N9 avian influenza viruses.

ABSTRACT

Schmallenberg virus (SBV), a novel orthobunyavirus, was first isolated in 2011. SBV preferentially infects the central nervous system of cattle and sheep and causes fever, diarrhea, a drop in milk yields, congenital malformations and stillbirths. Until June 2014, more than 200 scientific publications regarding SBV have been published. Although more than 20 articles on SVB were published in China, most of these articles provided only a brief introduction of the disease without fully discussing the associated disease characteristics. As a new disease, it has been made a focus of the National Research Center for Exotic Animal Diseases at the China Animal Health and Epidemiology Center. In this review, in order to provide a reference for research into SBV in China, we have reviewed the state of current research progress on the etiology, diagnosis and epidemiology of SBV, and vaccine development.

ABSTRACT

Virus-like particles (VLPs) are composed of multiple copies of one or more expressed recombinant viral structural proteins which spontaneously assemble into particles upon expression. VLPs are non infectious because they assemble without incorporating genetic material. VLPs have structural characteristics and antigenicity similar to the parental virus because they mimick the wild-type virus structure. Hence, they are recognized readily by the immune system which induces strong anti-viral immune responses to stop virus infection. VLPs have therefore shown dramatic effectiveness as candidate vaccines and diagnostic reagent for virus. Here, in order to provide reference to the research of influenza VLPs, we reviewed the current research progress of influenza VLPs, and discussed the characteristics associated with producing VLPs for influenza virus.

ABSTRACT

OBJECTIVETo investigate the aesthetic effect of wound repair with flaps.

METHODSOne thousand nine hundred and ninety-six patients with 2082 wounds hospitalized from January 2004 to December 2011. These wounds included 503 deep burn wounds, 268 pressure sores, 392 soft tissue defects caused by trauma, 479 soft tissue defects due to resection of skin cancer and mole removal, 314 soft tissue defects caused by scar excision, and 126 other wounds. Wound area ranged from 1.5 cm x 1.0 cm to 30.0 cm x 22.0 cm. Sliding flaps, expanded flaps, pedicle flaps, and free flaps were used to repair the wounds in accordance with the principle and timing of wound repair with flaps.

RESULTSFive flaps showed venous congestion within 48 hours post-operation, 2 flaps of them improved after local massage. One flap survived after local heparin wet packing and venous bloodletting. One flap survived after emergency surgical embolectomy and bridging with saphenous vein graft. One flap showed partial necrosis and healed after skin grafting. The other flaps survived well. One thousand three hundred and twenty-one patients were followed up for 3 months to 2 years, and flaps of them were satisfactory in shape, color, and elasticity, similar to that of normal skin. Some patients underwent scar revision later with good results.

CONCLUSIONSApplication of suitable flaps in wound repair will result in quick wound healing, good function recovery, and satisfactory aesthetic effect.

ABSTRACT

The permeability of cell membrane was enhanced by exogenous SA in the culture of Taxus cuspidata. Nuclei condense and fragments were observed in some cells by using fluorescent microscope, and the degraded chromosomal DNA was observed by using agarose gel electrophoresis. The changes in soluble proteins of the suspension cultures of Taxus cuspidata induced by salicylic acids were analyzed by using two-dimensional polyacrylamide gel electrophoresis. Comparing with the control, seven new protein spots were found and six protein spots were not found in the cultures grown with SA at 48h. The results obtained showed that SA could induce the expression of some special proteins that might be related with the action of SA in the suspension cultures of Taxus cuspidata.

ABSTRACT

The authors have studied the changes in the concentration of glutathione of the red blood cell (RBC-GSH) and plasma malondialdehyde (MDA) in 54 patients with acute brain stroke. The results indicated that the concentration of RBC-GSH reduced obviously and that of plasma MDA increased markedly within 72 hours after illness. However, there is no obvious difference between the cases of hemorrhagic cerebrovascular diseases and cerebral infarction group. The RBC-GSH level rised again in the 7th day, while the plasma MDA still remained higher level, they were normal during the convalescent period. The pathological basis of the change may be the response of the free radicals in human body.
