ABSTRACT

OBJECTIVE@#The aim of this study is to explore the potential modulatory role of quercetin against Endotoxin or lipopolysaccharide (LPS) induced septic cardiac dysfunction.@*METHODS@#Specific pathogen-free chicken embryos ( n = 120) were allocated untreated control, phosphate buffer solution (PBS) vehicle, PBS with ethanol vehicle, LPS (500 ng/egg), LPS with quercetin treatment (10, 20, or 40 nmol/egg, respectively), Quercetin groups (10, 20, or 40 nmol/egg). Fifteen-day-old embryonated eggs were inoculated with abovementioned solutions via the allantoic cavity. At embryonic day 19, the hearts of the embryos were collected for histopathological examination, RNA extraction, real-time polymerase chain reaction, immunohistochemical investigations, and Western blotting.@*RESULTS@#They demonstrated that the heart presented inflammatory responses after LPS induction. The LPS-induced higher mRNA expressions of inflammation-related factors (TLR4, TNFα, MYD88, NF-κB1, IFNγ, IL-1β, IL-8, IL-6, IL-10, p38, MMP3, and MMP9) were blocked by quercetin with three dosages. Quercetin significantly decreased immunopositivity to TLR4 and MMP9 in the treatment group when compared with the LPS group. Quercetin significantly decreased protein expressions of TLR4, IFNγ, MMP3, and MMP9 when compared with the LPS group. Quercetin treatment prevented LPS-induced increase in the mRNA expression of Claudin 1 and ZO-1, and significantly decreased protein expression of claudin 1 when compared with the LPS group. Quercetin significantly downregulated autophagy-related gene expressions (PPARα, SGLT1, APOA4, AMPKα1, AMPKα2, ATG5, ATG7, Beclin-1, and LC3B) and programmed cell death (Fas, Bcl-2, CASP1, CASP12, CASP3, and RIPK1) after LPS induction. Quercetin significantly decreased immunopositivity to APOA4, AMPKα2, and LC3-II/LC3-I in the treatment group when compared with the LPS group. Quercetin significantly decreased protein expressions of AMPKα1, LC3-I, and LC3-II. Quercetin significantly decreased the protein expression to CASP1 and CASP3 by immunohistochemical investigation or Western blotting in treatment group when compared with LPS group.@*CONCLUSION@#Quercetin alleviates cardiac inflammation induced by LPS through modulating autophagy, programmed cell death, and myocardiocytes permeability.

ABSTRACT

Objective:To investigate patients' compliance of endoscopic surveillance after endoscopic submucosal dissection (ESD) and the influencing factors.Methods:This study was a cross-sectional survey. The data of patients who underwent ESD in Xijing Digestive Hospital of Air Force Military Medical University from January 2014 to December 2015 were collected through questionnaire and telephone call. The questionnaire survey was conducted from May 1, 2019 to July 31, 2020. Patients' compliance for endoscopic surveillance and influencing factors were evaluated. Logistic regression model was used for multivariate analysis to determine independent risk factors affecting compliance of endoscopic surveillance.Results:A total of 413 questionnaires were collected, of which 331 (80.1%) patients were well complied with endoscopic surveillance, and 82 (19.9%) were not. Univariate analysis showed that gender ( χ2=4.68, P=0.034), lesion type ( χ2=10.10, P=0.002), educational background ( χ2=4.00, P=0.049), disposable income ( χ2=7.00, P=0.009), employment ( χ2=11.29, P=0.004) and medical insurance ( χ2=13.59, P=0.001) affected patients' endoscopic surveillance compliance. Logistic regression analysis indicated that patients with submucosal lesions were less likely to be compliant than patients with mucosal lesions with the same type of medical insurance ( P<0.001, OR=0.383, 95% CI: 0.227-0.644); and with the same lesion type, patients with urban medical insurance were more likely to be compliant than those with rural cooperative medical insurance ( P<0.001, OR=2.938, 95% CI: 1.696-5.090). Conclusion:The endoscopic surveillance compliance for post-ESD patients is related to the lesion type, the type of medical insurance, and the awareness of the disease. More attention should be paid to those with submucosal lesions and rural cooperative medical insurance due to their poor compliance. And it is necessary to improve health education on the disease for better surveillance compliance.

ABSTRACT

Chronic obstructive pulmonary disease (COPD) is a heterogeneous disease characteristic of small airway inflammation, obstruction, and emphysema. It is well known that spirometry alone cannot differentiate each separate component. Computed tomography (CT) is widely used to determine the extent of emphysema and small airway involvement in COPD. Compared with the pulmonary function test, small airway CT phenotypes can accurately reflect disease severity in patients with COPD, which is conducive to improving the prognosis of this disease. CT measurement of central airway morphology has been applied in clinical, epidemiologic, and genetic investigations as an inference of the presence and severity of small airway disease. This review will focus on presenting the current knowledge and methodologies in chest CT that aid in identifying discrete COPD phenotypes.

ABSTRACT

To investigate the influences of zwitterionic polymer chain length on mucus permeability and cellular uptake, the nanoparticles (NPs) were coated with poly(sulfobetaine methacrylate) (pSBMA) with different chain lengths. The di-block polymer poly(ε-caprolactone)-block-poly(sulfobetaine methacrylate) (PCL-pSBMA) with different chain lengths were synthesized via atom transfer radical polymerization (ATRP) combining with ring-opening polymerization of ε-caprolactone, and corresponding nanoparticles (pSBMAn NPs) were prepared by nanoprecipitation method. The sizes of different pSBMAn NPs were around 100 nm, and zeta potential were about -7 mV. Mucin interaction or mucus penetration study based on transwell systems were employed to evaluate mucus permeability of NPs. Caco-2 cells and mucus-producing HT-MTX-E12 cells were employed to illustrate the endocytosis efficiency of pSBMAn NPs. The results showed that the permeability coefficient of NPs coated with shorter chain length of pSBMA (pSBMA10 NPs) was only 42.83% of that coated with longer pSBMA (pSBMA80 NPs). On the contrary, the cellular uptake of pSBMA10 NPs was 2.44 fold higher compared to pSBMA80 NPs. Although the cellular uptake of pSBMAn NPs was reduced in the presence of mucus, pSBMA10 NPs still presented the highest cellular uptake. However, the in vivo results indicated that the oral bioavailability of pSBMA20 NPs was higher than that of pSBMA10 NPs. All animal procedures were performed in accordance with the Guidelines of the Sichuan University Animal Care and Use Committee and were approved by the Animal Ethics Committee of Sichuan University. This study provides a reference for oral delivery of zwitterionic nanoparticles.

This article reviews and analyzes the current status of the development of CDSS and MDT in China and foreign countries and summarizes their features and shortcomings, and with the background of the new era of “medical big data resources” and “big data technology”, this article looks into the future of developing an intelligentized diagnostic and therapeutic mode (a combination of CDSS and MDT) for hepatobiliary tumors suitable for the national conditions of China.

ABSTRACT

MnO2nanowires-reduced graphene oxide composite (MnO2-RGO) was used to modify glassy carbon electrodes (GCE) and applied for the electrochemical determination of dopamine (DA). The microstructure of MnO2nanowires and MnO2-RGO nanocomposite material was characterized by scanning microscope and X-ray powder diffraction. Then the electrochemical reduction condition for preparing MnO2-RGO/GCE and experimental conditions for determining DA were optimized systematically. The electrochemical behavior of DA on the bare electrode and RGO or MnO2-RGO modified electrodes was also investigated in pH 3.5 phosphate buffer solution (PBS) by cyclic voltammetry. The results shows that the oxidation peaks of ascorbic acid (AA), dopamine (DA) and uric acid (UA) can be well separated and the peak to peak separations were 268 mV (AA-DA) and 128 mV (DA-UA), respectively. Moreover, the linear response ranges for the determination of DA were 0.06-1.0 μmol/L and 1.0-80 μmol/L with the detection limit of 1.0 nmol/L (S/N=3). The proposed method has been applied to the determination of dopamine in human blood serum sample with satisfactory results.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the correlation between dynamic change of IL-32 level and disease development in the patients with acute leukemia(AL) and to explore its clinical significance.</p><p><b>METHODS</b>The serum IL-32 levels and IL-32 mRNA expression in 82 cases of AL and 30 healthy persons were measured by ELISA and real-time PCR.</p><p><b>RESULTS</b>Compared with healthy persons, the serum IL-32 protein level and IL-32 mRNA expression in AL, acute lymphoblastic leukemia(ALL) and acute non-lymphocytic leukemia(ANLL) groups all were significantly higher(P<0.05). The serum level of IL-32 protein and mRNA expression in newly diagnosed, PR and relapsed ALL and ANLL groups were all higher than those in the control group(P<0.05), and the serum protein level of IL-32 in relapsed ALL and ANLL groups were higher than that in other stage group(P<0.05). The serum levels of IL-32 protein and mRNA were not significantly different between CR and control group(P>0.05).</p><p><b>CONCLUSION</b>The IL-32 in the peripheral blood of patients with AL has been found to be closely related with the occurrence and development of disease, therefore, monitoring the dynamic changes of serum IL-32 level would contribute to the clinical judgment of the severity, the IL-32 levels can be used as indicators for the therapeutic efficacy for AL.</p>

ABSTRACT

BACKGROUND: There are various treatment methods for osteonecrosis of the femoral head (ONFH) and collapse, but conservative treatment is invalid. Once femoral head collapse occurs, the development is irreversible. Our previous research has shown that local administration of zoledronic acid can prevent necrotic femoral head collapse. Moreover, bone marrow mononuclear cells obtain satisfactory short-term efficacy in the treatment of ONFH. OBJECTIVE: To investigate the curative efficacy of local administration of mononuclear cell, platelet-rich plasma and zoledronic acid for the prevention and treatment of early ONFH and collapse. METHODS: This prospective, single-center, randomized, parallel, controlled clinical trial was conducted at the Chinese PLA General Hospital, Beijing, China. One hundred patients with ONFH (stages I-II by Ficat and Arlet classification) were enrolled and randomly assigned into either the treatment group or control group (n=50 per group). Patients were given an injection of mononuclear cell, platelet-rich plasma and zoledronic acid into the necrotic femoral head, or drilling decompression at the necrotic area. Patients in both groups were then followed up for 4, 8, 12, and 18 months. The primary outcome measures were the blood supply, osteogenesis and appearance of the necrotic femoral head observed on hip perfusion by dynamic MRI, CT restruction of the hip joint and radiography of the hip joint, as well as Harris hip scores and numerical rating scale scores. Secondary outcome measures included SF-36 Health Survey and Activities of Daily Living scores. DISCUSSION: The outcomes of this trial have provided quantitative data for analyzing the effectiveness of local administration of mononuclear cell, platelet-rich plasma and zoledronic acid on ONFH and collapse. Written approval for this protocol was obtained from the Ethics Committee of the Chinese PLA General Hospital in China (approval No. S2015-082-01). Participants and their families are informed of the study protocol and procedures, and signed an informed consent. The study was in accordance with the guidelines of the Declaration of Helsinki, formulated by the World Medical Association. Trial began in January 2015 and will be completed in December 2017. Trial results will be published in scientific reports, or in peer-reviewed journals. This trial was registered with the ClinicalTrials.gov identifier: NCT02721940. Patient recruitment is ongoing.

ABSTRACT

The role of autophagy is known to be highly complex and context-dependent, and may be characterized as both tumor suppression and tumor promotion in some tumors, such as breast cancer and prostate cancer. This review outlines recent advances in the studies of the involvement of autophagy in the development, progression and treatment of prostate cancer, focusing on autophagy modulation during androgen deprivation, with a special discussion on the regulatory effect of androgens on the autophagy of prostate cancer cells. A critical evaluation and analysis of the studies suggests that autophagy inhibition combined with androgen deprivation therapy is a promising approach to the treatment of prostate cancer.

ABSTRACT

Recent studies have demonstrated the importance of the non-protein coding part of human genome in carcinogenesis and metastasis of prostate cancer. Long non-coding RNAs (lncRNAs) play a key regulatory role in prostate cancer biology. LncRNAs are dysregulated in prostate cancer and the expression levels of certain lncRNAs are associated with the recurrence, metastasis and prognosis of cancer. It is also proved that lncRNAs, as oncogenes, can promote carcinogenesis and development of prostate cancer. This review focuses on the progress in the studies of lncRNAs in prostate cancer.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate Epstein-Barr virus(EBV) infection in Hodgkin's lymphoma (HL) of Uygur patients and related clinicopathological characteristics.</p><p><b>METHODS</b>EBV-encoded small RNA (EBER) was detected in 40 cases of HL and 20 cases of lymphoid reactive hyperplasia by in-situ hybridization. Expression of LMP2A in HL was investigated by immunohistochemistry.</p><p><b>RESULTS</b>EBV was detected in 26/40 (65.0%) of HL and 5/20 of lymphoid reactive hyperplasia (P < 0.05). The expression level of EBER showed significant difference among various histological subtypes of HL (P < 0.05) and between patients with and without B symptom (P = 0.02). However, no difference was found in relation to gender, clinical stage and tumor burden. The expression of LMP2A in the mixed cellularity and nodular sclerosis classical HL associated with EBV infection was 57.7% (15/26). Expression of LMP2A was not detected in lymphoid reactive hyperplasia cases.</p><p><b>CONCLUSION</b>Uyghur patients with Hodgkin's lymphoma have a high infection rate of EBV and distinct clinicopathologic characteristics.</p>

ABSTRACT

Objective To observe the effect of anti-pyretic and anti-inflammatory of Shufeng Jiebiao decoction.Methods Intraperitoneal injection of Lipopolysaccharides (LPS) was made to cause fever in rats and then to observe the anti-pyretic effect of Shufeng Jiebiao decoction.Intraperitoneal injection of glacial acetic acid was made to led inflammatory exudate in rats and then to observe the anti-inflammatory effect of Shufeng Jiebiao decoction.Smearing cylene in auricles was done to cause inflammatory swelling in rats and then to observe the effect of the alleviation of the inflammatory swelling of Shufeng Jiebiao decoction.Results ①The temperature of rats in the group of the aspirin and the Shufeng Jiebiao decoction were become lower at each time.The basic temperature of the model control group was (37.14±0.39) ℃,the temperature in the first hour was (40.31±0.34) ℃,the second hour was (40.44±0.44) ℃,the fourth hour was (40.24±0.34) ℃,the sixth hour was (40.05 ±0.44)℃,and the eighth hour was (39.85 ±0.37)℃.The basic temperature of the aspirin group was (37.13±0.33)℃,the temperature in the first hour was (38.74±0.42)℃,the second hour was (38.86±0.33) ℃,the fourth hour was (39.05±0.36)℃,the sixth hour was (38.74±0.37)℃,and the eighth hour was 38.64±0.24) ℃.The basic temperature of the Shufeng Jiebiao decoction group was (37.03±0.46) ℃,the temperature in the first hour was (39.02±0.49) ℃,the second hour was (38.82±0.49) ℃,the fourth hour was (38.63±0.46)℃,the sixth hour was (38.62±0.52)℃,and the eighth hour was (38.42±0.44)℃.The differences were statistical significance compared with the model control group (P<0.01),the onset of anti-pyretic of the Shufeng Jiebiao decoction group was slower than the aspirin group,but it had a longer lasting effect.Moreover,the rats' temperature decrease of the Shufeng Jiebiao decoction group in the fourth hour had a statistical significance compared with the aspirin group.(P<0.05).② After the intevention of aspirin and the Shufeng Jiebiao decoction,the optical density of evans blue:the model control group was (0.221 ±0.045),the aspirin group was (0.162±0.053),the Shufeng Jiebiao decoction group was (0.176±0.049),the permeability of the abdominal capillary of the rats reduced significantly (P<0.01).The intervention of the aspirin and the Shufeng Jiebiao decoction had almost no difference.③ After the intervention of the dexamethasone and the Shufeng Jiebiao decoction,the weight of the auricals:the model control group was (1.94±0.55)mg,dexamethasone group was (1.18±0.40)mg,Shufeng Jiebiao decoction group was (1.04±0.41)mg,showing the degree of the swelling of auricals decreased obviously (P<0.01).The intervention of the dexamethasone and the Shufeng Jiebiao decoction had almost no difference.Conclusion Shufeng Jiebiao Decoction had anti-pyretic and anti-inflammatory effects.

ABSTRACT

<p><b>OBJECTIVE</b>To study the value of high-frequency electrocautery combined with high-pressure balloon expansion in the treatment of benign airway obstruction.</p><p><b>METHODS</b>Twelve patients with complete airway obstruction received treatment with high-frequency electrocautery and high-pressure balloon expansion, and 6 months of follow-up was performed.</p><p><b>RESULTS</b>After the initial treatment, the rate of airway obstruction relief exceeded 50% in 8 patients, who showed lung recruitment and an increased dyspnea index, suggesting total effectiveness; one patient showed mild improvement with airway stenosis relief by less than 50% (the upper-left lobe remained obstructed). Stents were implanted eventually in 7 patients for airway maintenance. Two patients showed relief of the airway obstruction by over 50% after the initial treatment, but lung recruitment or dyspnea relief failed to be achieved, suggesting non-response to the treatment. The airway failed to be found in 1 case (who received 5 previous operations of ablation and dilatation), which was considered a non-response case.</p><p><b>CONCLUSION</b>High-frequency electrocautery combined with high pressure balloon expansion is clinically effective with few complications in some cases of benign complete airway stenosis. The possibility of airway stenosis and even obstruction should be considered in patients with tuberculosis during the treatment.</p>

ABSTRACT

Objective To observe the improving effect in joint function of moderate skeletal fluorosis treated by traditional Chinese medication(main ingredient was Strychnine).MethodsFrom December 2007 to July 2009,120 moderate skeletal fluorosis patients met the inclusion criteria were divided into the treatment group(60 cases)and the control group(60 cases)in the skeletal fluorosis hospital of Xinzhou,the treatment group was given basic treatment and traditional Chinese medication,the control group wa8 given basic treatment and placebo.The treatment lasted 12 weeks,follow up 24 weeks.Before treatment,after treatment 4 weeks,8 weeks,12 weeks,36 weeks,a third party evaluate comprehensive function of both upper and lower limb and joint dysfunction.Results The main effect of both drugs was statistically significance in the scores of the upper forearm in the finger by touching the posterior contralateral ear, upper arm touched by the finger back in the opposite corner subscapularis function, lower limb function and single-joint dysfunction(F values were 4.08,14.32,35.81,13.02, all P<0.05), the main effect of time also was significant (F values were 82.63,72.82,277.33,328.16, all P<0.05),①the upper forearm in the finger by touching the posterior contralateral ear functions:At the time of 8,12 weeks,scores of the treatment group were lower than those of before treatment and control group (all P<0.05);At the time of 36 weeks,scores of the treatment group were lower than that 12 weeks(all P<0.05);At the time of 8,12,36 weeks, scores of the control group were lower than those of before treatment(all P < 0.05);②upper arm function, namely fingers touching the opposite corner subscapularis:At the time of 4,8,12 weeks, scores of the treatment group were lower than those of before treatment(all P<0.05); At the time of 36 weeks, scores of the treatment group were lower than that 12 weeks(all P<0.05); At the time of 8,12,36 weeks, scores of the treatment group were lower than those of the control group (all P<0.05);③Lower extremity functions: At the time of 8,12 weeks,scores of the treatment group were lower than those of before treatment and control group (all P<0.05);At the time of 36 weeks, scores of the treatment group were lower than that 12 weeks(all P<0.05) ; At the time of 8,12,36 weeks, scores of the control group were lower than those of before treatment (all P<0.05);④single joint functions:At the time of 4,8,12 weeks,scores of the treatment group were lower than those of before treatment(all P<0.05); At the time of 36 weeks,scores of the treatment group were lower than that 12 weeks(all P<0.05) ; At the time of 8,12,36 weeks, scores of the control group were lower than those of before treatment(all P<0.05);At the time of 4,8,12,36 weeks, scores of the treatment group were lower than those of control group(all P<0.05);⑤At the end of treatment and follow-up,the improvement rate in joint functions in the treatment group were 88.33% (53/60),93.33% (56/60); the control group were 28.07%(16/57),40.35%(23/57), (Fisher test, P<0.01,X2=56.21, P<0.01). ConclusionTraditional Chinese medication(its main ingredient is Strychnine), an effective drug for improving joint dysfunction in patients suffering from moderate skeletal fluorosis, is simple and effective.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the therapeutic effects of mitral valve repair for the treatment of the anterior leaflet prolapse of mitral valve.</p><p><b>METHODS</b>From November 1998 to October 2007, 210 patients with severe anterior leaflet prolapse of mitral valve underwent valve repair. The condition of valve was preoperative, intraoperative, and postoperative assessed with echocardiography.</p><p><b>RESULTS</b>Edge-to-edge repair technique was used in 134 cases (63.8%). The cardiac function was NYHA class I in 168 cases and class II in 40 cases after operation. Patients were followup for 1 - 150 (25.7 +/- 29.0) months, two patients (0.95%) died of postoperative low cardiac output syndrome. Echocardiography examination indicated that the mean JP2 postoperative left atrial diameter was (37.7 +/- 9.2) mm against the preoperative value of (47.5 +/- 12.7) mm (P < 0.05), the mean postoperative left ventricular end-diastolic diameter was (51.7 +/- 7.9) mm against the preoperative value of (67.7 +/- 10.3) mm (P < 0.05), the mean postoperative left ventricular ejection fraction was (62.2 +/- 3.2)% against the preoperative value of (52.2 +/- 6.4)% (P < 0.05), and the mean preoperative regurgitation area was (10.4 +/- 4.1) cm(2) against the postoperative value of (4.1 +/- 1.7) cm(2) (P < 0.01).</p><p><b>CONCLUSIONS</b>Optimal outcome was achieved by appropriate edge-to-edge technique or other mitral valve repair techniques for anterior leaflet prolapse of mitral valve. Edge-to-edge technique is a reliable and efficient surgical technique.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To establish an quick, sensitive and specific assay for effective inactivatian test of inactivated hepatitis A vaccine.</p><p><b>METHODS</b>effective inactivatian test of inactivated hepatitis A vaccine were carried out using integrated cell culture/strand-specific RT-PCR (ICC/strand-specific RT-PCR) assay compared with traditional ELISA and nest RT-PCR assay.</p><p><b>RESULTS</b>all the samples were infectious negative detecting by both ICC/ strand-specific RT-PCR and ELISA assay,while some samples appeared false positive detecting by nest RT-PCR.</p><p><b>CONCLUSION</b>ICC/strand-specific RT-PCR assay is a novel, rapid, sensitive and reliable method for effective inactivatian test of inactivated hepatitis A vaccine. Shorting detection period largely, this assay may be used as an alternative method for routine inactivated hepatitis A vaccines test.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the association of the polymorphism in manganese superoxide dismutase (Mn-SOD) gene in Chinese type 2 diabetic patients with diabetic retinopathy.</p><p><b>METHODS</b>The Ala(-9)Val polymorphism of the Mn-SOD gene was determined by polymerase chain reaction and direct sequencing in 198 normal control subjects and 264 patients with type 2 diabetes mellitus, among them there were 139 non-diabetic retinopathy (NDR) subjects and 125 subjects with diabetic retinopathy (DR).</p><p><b>RESULTS</b>There was no statistic difference in the frequencies of VV genotype and V allele between the type 2 diabetic group and the control group. However, the frequencies of VV genotype and V allele were significantly higher in the DR group than that in the NDR group (chi-square (2)=5.015, P=0.025; chi(2)=10.253, P=0.001),but there was no statistic difference in the NDR group compared with the control group (P > 0.05). The presence of V allele was shown to be associated with diabetic retinopathy (OR=1.96, 95%CI: 1.29-2.97). Furthermore, the subjects carrying the VV genotype had lower serum Mn-SOD level (P=0.025) and had a tendency of higher total serum SOD activity, but this tendency had no statistic significance.</p><p><b>CONCLUSION</b>The Ala(-9)Val polymorphism in the Mn-SOD gene may not be related to the etiology of type 2 diabetes, but it seems to contribute to the development of diabetic retinopathy in Chinese type 2 diabetic patients.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the clinical effects of prunrllae oral solution in treating hyperthyrea.</p><p><b>METHOD</b>56 cases with hyperthyrea were randomized into two groups; group A1 was treated by classic method, B1 was treated by classic method combined with prunrllae oral solution. The size, vessel caliber of thyroidea, volume of blood flow and blood flow rate pre-and post-treatment were measured by color supersonic, meanwhile, 20 normal thyroidea were measured as control group.</p><p><b>RESULT</b>The size and vessel caliber of thyroidea of the two groups pre-treatment were obviously bigger than those of the control group, the volume of blood flow and blood flow rate were obviously slower than those of the control group (P < 0.001), the sizes of thyroidea of the two groups became smaller, especially the group B1 was more obvious, and there was no significant difference in the size of thyroidea between group B1 and control group.</p><p><b>CONCLUSION</b>It is indicated that combined treatment of traditional Chinese medicine (prunrllae oral solution) and western medicine is superior to western medicine in treating hyperthyrea.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To produce specific monoclonal antibody (mAb) against recombinant human erythropoietin (rHuEPO) for development of highly efficient methods for erythropoietin detection in biological fluids.</p><p><b>METHODS</b>rHuEPO was covalently coupled with bovine serum albumin (BSA) and the conjugate was used to immunize mice to produce specific mAb against rHuEPO based on hybridoma technology. The obtained F3-mAb was characterized by enzyme-linked immunosorbent assay (ELISA), SDS-PAGE and Western blot.</p><p><b>RESULTS</b>The isotype of F3-mAb was found to be IgM with an affinity constant of 2.1 x 10(8) L/mol. The competitive ELISA using the obtained IgM showed a broader linear range and lower detection limit compared with previous work.</p><p><b>CONCLUSIONS</b>The modification of rHuEPO was proved to be successful in generating required specific mAb with high avidity to rHuEPO.</p>
