ABSTRACT

Objective:To investigate the risk factors of diabetes mellitus complicated by pulmonary tuberculosis.Methods:The clinical data of 83 patients with diabetes mellitus complicated by pulmonary tuberculosis who received treatment in Taiyuan Fourth People's Hospital from March 2020 to March 2022 were collected. These patients were divided into sensitive group ( n = 45) and resistant group ( n = 38 ) according to the results of drug sensitivity test. Univariate and multivariate non-conditional logistic regression was performed to analyze the influential factors of drug resistance. Results:Univariate logistic regression results revealed that there were significant differences in blood CD4 +T lymphocyte count ( χ2 = 11.73, P = 0.001) and diabetic complications ( χ2 = 4.94, P = 0.026). Multivariate non-conditional logistic regression analysis was performed taking whether blood CD4 +T lymphocyte count was lower than the average level and whether patients with diabetes mellitus had complications as independent variables, and taking whether drug resistance was a dependent variable. The results showed that the OR (95% CI) value of the decreased blood CD4 +T lymphocyte count was 4.909 (1.926-12.514). It is a risk factor for drug resistance of diabetes mellitus complicated by pulmonary tuberculosis. Conclusion:The decrease of blood CD4 +T lymphocyte count is a risk factor of drug resistance in diabetes mellitus complicated by pulmonary tuberculosis, and it should be intervened early in the clinic.

ABSTRACT

Objective To analyze the correlation of hepatic steatosis with blood lipids and uric acid metabolism in elderly patients with chronic hepatitis B (CHB). Methods The clinical data of 120 patients with CHB admitted to the hospital from January to December 2021 were retrospectively analyzed. According to the presence or absence of hepatic steatosis, the patients were divided into steatosis group (n=35) and non-steatosis group (n=85). The general clinical data, serological indicators of hepatitis B virus, blood lipid and uric acid levels were compared between the two groups. The correlation of hepatic steatosis grading with blood lipids and uric acid metabolism was analyzed. Results The inflammation and fibrosis degree of liver tissues were significantly different in the two groups (P0.05). Pearson correlation analysis found that the grade of hepatic steatosis in patients with CHB was negatively correlated with liver tissue inflammation, fibrosis degree and HDL-C level (P<0.05), and positively correlated with TG and TC levels (P<0.05). Conclusion Elderly patients with CHB and hepatic steatosis have abnormal blood lipid metabolism. Hepatic steatosis will exacerbate abnormal blood lipid metabolism but not liver tissue inflammation or fibrosis degree. Clinically, attention should be paid to blood lipid monitoring of elderly patients with CHB.

ABSTRACT

Objective:To explore the profile and gene functional changes of gut microbiota (GM) in women with postmenopausal osteoporosis (PMOP) in Northwest China, and the correlations between GM and bone mineral density (BMD).Methods:From November 2018 to October 2019, postmenopausal women were screened on their initial visits to our hospital, and 24 new osteoporosis (OP) patients, 30 new osteopenia patients and nine negative controls were recruited. Fecal samples were collected for GM DNA extraction, and Illumina platforms were used for high-throughput sequencing of 16S rRNA and metagenome. Species annotation, GM profile and gene functions were viewed and analyzed.Results:GM profiles were significatly different in different groups, and the LDA scores of Peptostreptococcaceae, Romboutsia, unidentified Clostridiales, Megamonas, Erysipelatoclostridium, Klebsiella and Erysipelatoclostridium ramosum were more than 3 in OP group. Metagenomic sequencing analysis indicated that gene numbers were positively correlated with BMD, and metabolism, carbohydrate metabolism, starch and sucrose metabolism and oxidative phosphorylation were negatively correlated with BMD. Receiver operating characteristic curve(ROC) showed that carbohydrate metabolism, starch and sucrose metabolism, amino sugar and nucleotide sugar metabolism, oxidative phosphorylation, respectively, could identify OP with preferable sensitivity and specificity (areas under curve were 0.70, 0.72, 0.73 and 0.75, respectively). Conclusions:High-throughput sequencing had great potential for GM analysis of postmenopausal women with OP, providing evidence of the correlations between GM and BMD.

ABSTRACT

OBJECTIVE To investigate the effect and mechanisms of doxorubicin (DOX) on isolated thoracic aorta and carotid artery of rats. METHODS The cumulative dosing method was used to add DOX every 10 min to reach the final concentrations of 1, 3, 10, 30 and 100 μmol·L-1, and the tension of isolated thoracic aorta and carotid artery in basal tension, precontracted by phenylephrine (PE) 1 μmol ·L-1 and KCI 60 mmol ·L-1, was respectively recorded in vitro vascular ring perfusion apparatus. Furthermore, No-nitro-L-arginine methyl ester (L-NAME), tetraethylammonium (TEA), 4-aminopyridine (4-AP), BaCI2, glibenclamide (Gli), indometacin (Indo) and proparanolol were used to explore the mechnisms of the vasodilating effect of DOX. RESULTS DOX had no effect on the thoracic aorta and carotid artery in basal tension and precontracted by KCI. However, DOX induced concentration-dependent relaxation in both the thoracic aorta and carotid artery precontracted by PE (P<0.05), and it had stronger vasodilating effect on the endothelium-intact thoracic aorta than on the endothelium-denuded thoracic aorta (P<0.05). After the treatment with nitric oxidase inhibitor, L-NAME 0.1 mmol·L-1, calcium activated potassium channel (KCa) inhibitor, TEA 1 mmol·L-1, voltage-dependent potassium channel (KV) inhibitor, 4-AP 1 mmol·L-1 and inward rectifier potassium channel (KIR) inhibitor, BaCI2 1 mmol·L-1, the vasodilating effect of DOX was obviously decreased (P<0.05), but the application of Indo 0.01 mmol ·L-1 Gli 0.01 mmol ·L-1 and β-adrenregic receptor blocker propranolol 0.01 mmol·L-1 had no effect on the vasodilating effect of DOX. Additionally, DOX 1, 10 and 100 μmol·L-1 significantly reduced PE-induced contraction of the thoracic aorta in Ca2+-free solution, and the concentration-effect curve of CaCI2 could be shifted to the right in the presence of DOX (P<0.05). CONCLUSION These results have demonstrated that DOX vasodilating effects on the thoracic aorta and carotid artery are concentration-dependent. The mechanisms are likely to be related to nitric oxide/cGMP pathway, KV, KCa, K1R, intracellular calcium release and extracellular calcium influx.

ABSTRACT

Accumulative evidence has shown the adverse effects of a geomagnetic field shielded condition, so called a hypomagnetic field (HMF), on the metabolic processes and oxidative stress in animals and cells. However, the underlying mechanism remains unclear. In this study, we evaluate the role of HMF on the regulation of cellular reactive oxygen species (ROS) in human neuroblastoma SH-SY5Y cells. We found that HMF exposure led to ROS decrease, and that restoring the decrease by additional HO rescued the HMF-enhanced cell proliferation. The measurements on ROS related indexes, including total anti-oxidant capacity, HO and superoxide anion levels, and superoxide dismutase (SOD) activity and expression, indicated that the HMF reduced HO production and inhibited the activity of CuZn-SOD. Moreover, the HMF accelerated the denaturation of CuZn-SOD as well as enhanced aggregation of CuZn-SOD protein, in vitro. Our findings indicate that CuZn-SOD is able to response to the HMF stress and suggest it a mediator of the HMF effect.

ABSTRACT

Objective To investigate the social support of clinical nurses in second-level hospitals in remote areas of Tibet, and to explore the influencing factors, so as to provide the basis for nursing managers to develop targeted interventions. Methods A total of 212 nurses were selected by convenient holistic sampling method and investigated by self-designed questionnaire and social support rating scale. Results The total score of social support of clinical nurses was (30.72 ± 6.78) points. The main influencing factors of social support were educational background(F=4.602),ages(F=2.694),working years (F=2.387), budgeted posts(t=2.391), income(F=3.112) and marital status (F=2.636). Difference was statistically significant(P<0.05). Conclusions The social support of clinical nurses in second-level hospitals in remote areas of Tibet is low. It is important to establish a relatively perfect social support system according to the difference of nurses working conditions and geographical to improve their work enthusiasm and satisfaction rate.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the neuroprotective effects of icariin on formaldehyde (FA)-treated human neuroblastoma SH-SY5Y cells and the possible mechanisms involved.</p><p><b>METHODS</b>SH-SY5Y cells were divided into FA treatment group, FA treatment group with icariin, and the control group. Cell viability, apoptosis, and morphological changes were determined by cell counting kit-8 (CCK 8), flow cytometry, and confocal microscopy, respectively. The phosphorylation of Tau protein was examined by western blotting.</p><p><b>RESULTS</b>FA showed a half lethal dose (LD50) of 0.3 mmol/L in SH-SY5Y cells under the experimental conditions. Icariin (1-10 µmol/L) prevented FA-induced cell death in SH-SY5Y cells in a dose-dependent manner, with the optimal effect observed at 5 µmol/L. After FA treatment, the absorbance in FA group was 1.31±0.05, while in the group of icariin (5 µmol/L) was 1.63±0.05. Examination of cell morphology by confocal microscopy demonstrated that 5 µmol/L icariin significantly attenuated FA-induced cell injury (P <0.05). Additionally, Icariin inhibited FA-induced cell apoptosis in SH-SY5Y cells. Results from western blotting showed that icariin suppressed FA-induced phosphorylation at Thr 181 and Ser 396 of Tau protein, while having no effect on the expression of the total Tau protein level. Furthermore, FA activated Tau kinase glycogen synthase kinase 3 beta (GSK-3β) by enhancement of Y216 phosphorylation, but icariin reduced Y216 phosphorylation and increased Ser 9 phosphorylation.</p><p><b>CONCLUSION</b>Icariin protects SH-SY5Y cells from FA-induced injury poßsibly through the inhibition of GSK-3β-mediated Tau phosphorylation.</p>

ABSTRACT

Living organisms are exposed to the geomagnetic field (GMF) throughout their lifespan. Elimination of the GMF, resulting in a hypogeomagnetic field (HMF), leads to central nervous system dysfunction and abnormal development in animals. However, the cellular mechanisms underlying these effects have not been identified so far. Here, we show that exposure to an HMF (<200 nT), produced by a magnetic field shielding chamber, promotes the proliferation of neural progenitor/stem cells (NPCs/NSCs) from C57BL/6 mice. Following seven-day HMF-exposure, the primary neurospheres (NSs) were significantly larger in size, and twice more NPCs/NSCs were harvested from neonatal NSs, when compared to the GMF controls. The self-renewal capacity and multipotency of the NSs were maintained, as HMF-exposed NSs were positive for NSC markers (Nestin and Sox2), and could differentiate into neurons and astrocyte/glial cells and be passaged continuously. In addition, adult mice exposed to the HMF for one month were observed to have a greater number of proliferative cells in the subventricular zone. These findings indicate that continuous HMF-exposure increases the proliferation of NPCs/NSCs, in vitro and in vivo. HMF-disturbed NPCs/NSCs production probably affects brain development and function, which provides a novel clue for elucidating the cellular mechanisms of the bio-HMF response.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the dynamic changes that occur in T cell subsets, particularly involving the surface expression of programmed death 1 (PD-1), in response to pegylated (Peg)-interferon (IFN) a-2a therapy in patients with chronic hepatitis C virus (HCV) infection.</p><p><b>METHODS</b>Twenty-five patients with HCV genotype 1b chronic infection and 10 healthy controls were enrolled in the study. All the HCV patients received combination antiviral therapy of Peg-IFNa-2a (180 mug/week) plus ribavirin. At treatment weeks 0 (baseline), 4, 12, 24 and 48, the level of PD-1 protein expression on the surface of total peripheral CD8+ and CD4+ T cells was determined by flow cytometry and the level of PD-1 mRNA expression in peripheral blood mononuclear cells (PBMCs) was determined by reverse transcription-polymerase chain reaction. Independent student's t-test were used to compare mean values between the two groups, repeat measure variance analysis was used to compare mean values among multiple groups, and Pearson's correlation coefficient was used to assess correlation significance.</p><p><b>RESULTS</b>Over the course of antiviral therapy, the proportions of CD4+ T cells and CD8+ T cells, as well as the CD4+/CD8+ ratio, increased (F = 81.23, 39.28, and 7.01 respectively; all P less than 0.01). In contrast, the PD-1 protein expression frequency on CD4+ T cells and CD8+ T cells significantly declined (F = 100.11 and 158.40 respectively; all P less than 0.01). The PD-1-mRNA expression level in PBMCs was: 1.40+/-0.26 at baseline, 1.30+/-0.27 at week-4, 1.14+/-0.18 at week-12, 1.06+/-0.26 at week-24, and 0.83+/-0.25 at week-48 (F = 20.09; P less than 0.01). A positive correlation existed between the PD-1 protein expression frequencies on CD4+ T cells and CD8+ T cells and the HCV RNA load detected at baseline (r = 0.82 and 0.75 respectively; all P less than 0.01).</p><p><b>CONCLUSION</b>The ability of Peg-IFN-a-2a-based antiviral therapy to suppress HCV replication may involve reduction of PD-1 protein expression on the surface of CD8+ T cells and CD4+ T cells.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the epidemic characteristics of etiological agents and analyze the clinic differences in children with hand, foot and mouth diseases in Shanghai.</p><p><b>METHODS</b>Collection 551 specimens including vesicle fluid, stool and throat swabs to detect the pathogens. The positive rates were tested by RT-PCR assay with Enterovirus consensus primer and specific primer for Enterovirus 71 (EV71) and Coxsackie A16 (CoxA16), respectively.</p><p><b>RESULTS</b>Out of 357 patients, the overall positive rate was 79.7%. CoxA16 infection was mainly popular in 2009, EV71 was mainly popular in 2011. The epidemic characteristics of enterovirus infection within 2009-2011 had significant differences (P = 0.005). The positive rate of vesicle fluid was highest. The concurrent rate of fever and tendency to be frightened had significant differences in three patients' groups. The count of the peripheral white blood cells and the ratio of the neutrophil in the patients with EV71 infection was higher than that in the patients with CoxA16 or PE infection.</p><p><b>CONCLUSION</b>HFMD showed alternant epidemic characteristics at different times of enterovirus infection in Shanghai. Analyzing the differences of clinic characteristic of EV71 and CoxA16 infection is helpful to prevent and control EV71 infection.</p>

ABSTRACT

Fibrinogen is a key protein involved in coagulation and its deposition on blood vessel walls plays an important role in the pathology of atherosclerosis. Although the causes of fibrinogen (fibrin) deposition have been studied in depth, little is known about the relationship between fibrinogen deposition and reactive carbonyl compounds (RCCs), compounds which are produced and released into the blood and react with plasma protein especially under conditions of oxidative stress and inflammation. Here, we investigated the effect of glycolaldehyde on the activity and deposition of fibrinogen compared with the common RCCs acrolein, methylglyoxal, glyoxal and malondialdehyde. At the same concentration (1 mmol/L), glycolaldehyde and acrolein had a stronger suppressive effect on fibrinogen activation than the other three RCCs. Fibrinogen aggregated when it was respectively incubated with glycolaldehyde and the other RCCs, as demonstrated by SDS-PAGE, electron microscopy and intrinsic fluorescence intensity measurements. Staining with Congo Red showed that glycolaldehyde- and acrolein-fibrinogen distinctly formed amyloid-like aggregations. Furthermore, the five RCCs, particularly glycolaldehyde and acrolein, delayed human plasma coagulation. Only glycolaldehyde showed a markedly suppressive effect on fibrinogenesis, none did the other four RCCs when their physiological blood concentrations were employyed, respectively. Taken together, it is glycolaldehyde that suppresses fibrinogenesis and induces protein aggregation most effectively, suggesting a putative pathological process for fibrinogen (fibrin) deposition in the blood.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate relations between milk consumption and lactose intolerance (LI) in adults and to explore the effect of milk consumption on lactase activity.</p><p><b>METHODS</b>Total of 182 subjects aged 20-70 years were recruited and interviewed by questionnaires, and their accumulative cow's milk intake (AMI) was calculated. LI was evaluated by hydrogen breath test (HBT).</p><p><b>RESULTS</b>A negative correlation was found between AMI and severity of observed LI symptom (r=-0.2884; P<0.05). Binary logistic regression analysis showed a negative correlation between LI and duration and frequency of milk consumption (OR, 0.317 and 0.465, respectively; both P<0.05) and a positive correlation between LI and amount of milk consumed per sitting (OR, 6.337; P<0.05).</p><p><b>CONCLUSION</b>LI is related to various milk consumption behaviors. Most Chinese adults with LI may tolerate moderate milk consumption <160 mL.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To find out the diet measurements for alleviating symptoms of lactose intolerance when people had had a drink of milk.</p><p><b>METHODS</b>Eight lactose intolerance (LI) subjects suffering from watery diarrhea were selected from 38 subjects by using hydrogen breath test (HBT) joined the test of drinking cow's milk along with various other foods designed by orthogonal trial. The 100 healthy college students were selected to observe symptoms after they ingested 25 g milk powder in different manners.</p><p><b>RESULTS</b>The breath-hydrogen concentration in groups with ingesting milk and various other foods was lower than those with ingesting milk only. Egg was better than mantou in alleviating symptoms. (F vaule of stool lactose and symptom score were 7.5, 9.0, respectively, P < 0.05). Whole wheat bread might aggravate flatus. The prevalence of the symptom in 100 college students by drinking milk in fasting state (27/98) was significant in comparing with those drinking milk in regular meal (13/98), chi2 = 6. 156, P < 0. 05.</p><p><b>CONCLUSION</b>The study demonstrated that different kinds of food, and different milk drinking manners should affect the intolerance symptoms. We suggested: should not drink milk in fasting state, and drinking milk together with egg and staple food such as steamed bread, bread or gomuti should be the best choice for breakfast. Drinking milk with regular manner might alleviate the symptoms of intolerance. For avoiding lactose intolerance, whole wheat bread should not be taken along with milk.</p>

ABSTRACT

XYL1 gene,which encodes xylose reductase with dual coenzyme activity from Candida tropicalis,was transformed into Pichia pastoris X-33 by expression vector pGAPZB.The recombination strain was immobilized in Ca-alginate beads and fermentation characterization is studied using corn cob hydrolysates.Fermentation conditions were as follow:initial pH value 6.0,30℃,initial cell concentration of 20%,the Liquid volume of 28%,rotation speed 130r/min.The average xylitol yield was 37.5% on the optimum condition.This result is expected to provide a new alternative method for producing xylitol on a large scale by bioconversion.

ABSTRACT

<p><b>OBJECTIVE</b>To transfect pcDNA3.1/hTM plasmids containing human thrombomodulin (hTM) gene into human umbilical vein endothelial cells (HUVECs), and investigate the expression of hTM and anticoagulating function of transfected HUVECs.</p><p><b>METHODS</b>HUVECs were transfected with pcDNA3.1/hTM by lipofectin. Expression of hTM mRNA was determined by semi-quantitative RT-PCR, hTM antigen on HUVECs membrane by immunohistochemistry, and activated protein C (PC) in HUVECs by chronometry. By using a semiautomatic coagulator, the effect of the reacting liquid from transfected HUVECs mixed with PC from normal peripheral blood was assayed.</p><p><b>RESULTS</b>About 10% HUVECs were transfected by pcDNA3.1/hTM with high-level hTM mRNA and protein expression. Activated PC produced by pcDNA3.1/hTM group, pcDNA3.1(+)/neo group and untransfected group was (2.80 +/- 0.43) microg/ml, (0.75 +/- 0.08) microg/ml and (0.85 +/- 0.11) microg/ml, respectively. APTT was (51.68 +/- 2.73) s, (38.38 +/- 2.44) s, (39.65 +/- 2.39) s, (33.93 +/- 1.73) s and (34.60 +/- 1.86) s and PT was (21.89 +/- 1.66) s, (20.56 +/- 1.74) s, (20.42 +/- 2.04) s, (19.57 +/- 1.36) s and (20.16 +/- 1.35) s in pcDNA3.1/hTM group, pcDNA3.1(+)/neo group, untransfected group, inactivating PC group and control, respectively.</p><p><b>CONCLUSIONS</b>The pcDNA3.1/hTM plasmid could be transfected into endothelial cells and expressed biologically functioning hTM protein on HUVECs membrane. Activated PC could inhibit intrinsic coagulation pathway obviously with slight effect on extrinsic pathway.</p>

ABSTRACT

<p><b>AIM</b>To establish a method for screening active substance with scavenging effects on superoxide anion in vitro by designed superoxide dismutase biosensor.</p><p><b>METHODS</b>The enzyme sensor was built by connecting the immobilized CuZnSOD with optical oxygen sensor through a special way. Superoxide anions were generated by auto-oxidation of pyrogallol. The auto-oxidation speed was examined before and after adding samples into the system, and the Vit C having the scavenging radical activities was served as a positive control.</p><p><b>RESULTS</b>The limit of biosensor detection was 7.0 U in activity, and lifetime of the immobilized enzyme in the reaction-cell was above 2 weeks. The scavenging effects on superoxide radicals of fifteen active substance were studied in vitro by the sensor, and some of them presented scavenging activities.</p><p><b>CONCLUSION</b>The signal from biosensor is stable, easy to be determined, and the kinetic information on scavenging superoxide radicals could be obtained directly. The biosensor system can be used for screening drugs simply and rapidly.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the surgical methods and the outcome of management for traumatic arterial aneurysm (TAA) and traumatic arteriovenous fistula (TAVF).</p><p><b>METHODS</b>A total of 121 patients with TAA or TAVF were treated by surgery. Clinical, operative and postoperative data were collected and analyzed retrospectively.</p><p><b>RESULTS</b>The surgical techniques included aneurysmectomy and arterial end-to-end anastomosis or vascular grafting or artery ligation, aneurysm ligation and bypass, vascular repair, fistula excision and vascular ligation or vascular grafting or repair and so on. One patient died (0.83%). The follow-up rates of TAA and TAVF were 65.7% and 60% respectively.</p><p><b>CONCLUSIONS</b>Complicated TAA and TAVF in different sites should be treated with different methods.</p>

ABSTRACT

The purpose of this review is to confirm the reason resulted the gene silence and explore the countermeasure avoiding the gene silence in transgene plant. The method is to divide the gene silencing into transcriptional gene silencing(TGS) and posttranscriptional gene silencing(PTGS). Several models resulted PTGS were analyzed by RNA threshold model, ectopic pairing and aberrant RNA model and ds-RNA model. The results showed that it was important to decide the phenomena of restraining transgene silencing and the mechanism of PTGS. The strategies of identification of gene function and prevention of virus were presented by RNAi and gene silencing respectively, etc.
