ABSTRACT

Sanggenon C is a kind of flavonoid compound mainly extracted from the traditional Chinese medicine Morus alba. The pharmacological effects and mechanisms of sanggenon C are systematically reviewed and summarized, and it is found that this component can improve pulmonary fibrosis by regulating transforming growth factor-β1 and nuclear factor-κB; it can exert anti- tumor effects by inhibiting the proliferation of tumor cells and inducing the apoptosis of tumor cells; it can exert cardioprotective, neuroprotective and hepatoprotective effects by regulating multiple signaling pathways, such as calcineurin/nuclear factor of activated T cells 2, peroxisome proliferators-activated receptor α, and Ras homolog gene family member A/Rho-associated coiled- coil containing protein kinase, enhancing autophagy, reducing inflammatory response and reducing the level of oxidative stress; it can treat osteoporosis by inhibiting osteoclast uptake and promoting osteoblast formation; it has certain inhibitory effect on many enzymes; it can exert anti-inflammatory effects by regulating nuclear factor-κB signaling pathway; it can exert antioxidant effects by scavenging free radicals. However, researches on the pharmacological effects of sanggenon C mostly focus on the cellular and animal field, and the specific mechanism of action is not yet clear. In the future, basic research and clinical trials are still needed to explore and verify.

ABSTRACT

As a promising candidate seed cell type in regenerative medicine, mesenchymal stem cells (MSCs) have attracted considerable attention. The unique capacity of MSCs to exert a regulatory effect on immunity in an autologous/allergenic manner makes them an attractive therapeutic cell type for immune disorders. In this review, we discussed the current knowledge of and advances in MSCs, including its basic biological properties, i.e., multilineage differentiation, secretome, and immunomodulation. Specifically, on the basis of our previous work, we proposed three new concepts of MSCs, i.e., "subtotipotent stem cell" hypothesis, MSC system, and "Yin and Yang" balance of MSC regulation, which may bring new insights into our understanding of MSCs. Furthermore, we analyzed data from the Clinical Trials database ( http://clinicaltrials.gov ) on registered clinical trials using MSCs to treat a variety of immune diseases, such as graft-versus-host disease, systemic lupus erythematosus, and multiple sclerosis. In addition, we highlighted MSC clinical trials in China and discussed the challenges and future directions in the field of MSC clinical application.

ABSTRACT

Objective To investigate the association of interleukin-6 (IL-6) with depression using meta analysis. Methods A systematic literature search was conducted to examine depression and interleukin-6 in Chinese patients us-ing“depression”and“interleukin-6”in Databases including PubMed, CBM, VIP, CNKI and WanFang Data. A me-ta-analysis was performed to analyze correlation between depression and interleukin-6 using RevMan 5.2.8 software. Results A total of 9 studies were recruited, involving 432 depression patients and 277 healthy controls. The results of meta-analysis showed that concentration of IL-6 was higher in depression patients than in healthy controls(SMD=1.48, 95%CI:0.78~2.18,P<0.05). Subgroup analysis in the first-episode subgroup and the non first-episode subgroup showed that concentration of interleukin-6 was higher in depression patients than that in controls [the first-episode subgroup (SMD=1.94,95%CI:0.07~3.81,P<0.05), the non first-episode subgroup (SMD=1.26,95%CI:0.60~1.93,P<0.05)]. Conclusion There is a good correlation between depression and the serum IL-6 concentration. Due to the small number of studies and heterogeneity in different studies, the present meta-analysis has not yet confirmed the quantitative correla-tion between IL-6 and depression.

ABSTRACT

BACKGROUND:The differentiation of mesenchymal stem cells(MSCs)is strictly regulated by both genetic and epigenetic Factors .Emerging evidences have demonstrated that microRNA also plays an important role in this process.OBJECTIVE:To explore the differential expression of microRNA-125b during neuronal dliferentiation of Flk1~+ adipose-derived MSCs(AD-MSCs).MATERIALS:The fat samples were provided bv healthy female volunteers aged 15-35 years and recruited from the Plastic Surgery Hospital affiliated to the Chinese Academy of Medical Sciences.METHODS:Adult adipose tissues were obtained from healthy voluntears with age of 15-35 years .Using adherence method,Flk1~+ MSCs were obtained and the 3~(rd) passage cells were taken in the experiment.Cultured in neuronalinduction medium.these MSC were induced to differentiate towards neuronal lineage. The expression of microRNA-125b was examined at days 0,4,8 and 12.To explore its role in neuronal differentiation,we need to change its expression.RT-PCR and Taqman real-time PCR were carried out to explore the difierentially expression of microRNA-125b during neuronal differentiation of AD-MSCs.The effect of inhibitor on the expression of microRNA-125b was detected.RESULTS AND CONCLUTION:①The Flk-1~+MSCs were successfully induced into neuronal differentiation and displayed typical morphological changes 12 days after induction:Most cells retracted their cytoplasm ,fomling spherical cell body and emitted cellular protrusions.RT-PCR and immunocytochemistry studies confirmed their phenotype with expression of known neuronal cell markers including neurofilament, glial fibrillary acidic protein.②The expression of microRNA-125b was significant up-regulated during neuronal differentiation .Results of RT-PCR and Taqman real-time PCR were concordarlce with that of microRNA chip technology.③Inhibitor could down-regulate microRNA-125b.The results implied that microRNA-125b may play an important role in neuronal differentiation.

ABSTRACT

OBJECTIVETo explore the management of cerebral ischemia caused by Takayasu's arteritis.

METHODSNinety-three cases treated from June 1984 to September 1999 at the General Post & Telecom Hospital, the Sir Run Run Shaw Hospital, the First Affiliated Hospital of Zhejiang University, the Second Medical College of Beijing University, Beijing An Zhen Hospital, and the Beijing Union Medical College Hospital, including 10 men and 83 women, were reviewed. Of the 93 cases, bypasses from the ascending aorta to the axillary or subclavian artery and from graft to the carotid artery were performed in 47 cases. Subclavian to carotid bypass was performed in six cases. Percutaneous transluminal angioplasty (PTA) was used in five cases and stenting in one.

RESULTSMarked improvement was achieved in 30.3%, fair in 34.9%, improvement in 21.2%, unchanged in 4.6%, and death in 9.0% before discharge; 30.6%, 38.8%, 16.3%, 4.1%, and 2.0% respectively during a mean follow-up of 48 months, and recurrence requiring revision in 8.2%.

CONCLUSIONPatients with occlusive lesions of all four cervical arteries always have severe cerebral ischemia and their distal runoff is always unvisualised by angiography. However, we found by exploration that the internal carotid artery is patent in all but one patient. Therefore, an ascending aorta to carotid bypass is feasible in most instances, and this can and should be done when the cerebral perfusion is jeopardized at a time when the patient is in a stable or relatively stable condition. Unfortunately, the cerebral re-perfusion syndrome is still a serious and not completely solved problem.

ABSTRACT

Objective To investigate the efficacy of cavo-meso-atrial shunt for the treatment of Budd-Chiari syndrome (BCS) with occlusion of both inferior vena cava (IVC) and hepatic veins (HVs) lesions. Method 14 cases with BCS due to combined occlusion of IVC and HVs were treated from 1996. 8 cases were male and 6 cases female. The age ranged from 20 to 36 years old. HVs occlusion with complete IVC occlusion was found in 12 cases and stenosis of IVC in 2 cases. Cavo-meso-atrial shunt was performed. Result There was no perioperative death. Thrombosis of graft occurred in two cases, one died of upper digestive tract bleeding and hepatic coma half year post-operation. The mortality rate at follow-up period was 7%, the total patency of graft was 86% and the patency of cavo-atrial graft was 93%. The decrease in portal vein pressure was between 5 to 26 cmH 2O with an average of 15.5 cmH 2O. No serious complications occurred. Conclusion The cavo-meso-atrial graft shunt is an alternative to treat patients with occlusion of both IVC and HVs due to IVC thrombosis. The long term patency remains to be evaluated.
