ABSTRACT

Objective To investigate the protective effect of Echinococcus granulosus hydatid cyst fluid protein (HCFP) on ovalbumin (OVA)-induced allergic rhinitis (AR) in mice. Methods Twenty-four BALB/c mice at ages of 8 to 10 weeks, each weighing approximately 20 g, were randomly divided into four groups, including groups A (blank control group), B (blank intervention group), C (AR model group) and D (AR+HCFP intervention group), with 6 mice in each group. On days 0, 2, 4, 6, 8, 10 and 12, mice in groups A, B, C and D were injected with 200 μL sterile phosphate buffered saline (PBS), 200 μL sterile PBS containing 20 μg HCFP, 200 μL sterile PBS containing 50 μg OVA and 5 mg Al(OH)3 gel, and 200 μL sterile PBS containing 50 μg OVA, 5 mg Al(OH)3 gel and 20 μg HCFP, respectively. On days 14 to 20, mice in groups A, B, C and D were administered with 40 μL sterile PBS, 40 μL sterile PBS containing 20 μg HCFP, 40 μL sterile PBS containing 2 mg OVA and 40 μL sterile PBS containing 2 mg OVA and 20 μL HCFP by nasal drop, respectively. Mouse behavioral changes were observed and behavioral scores were estimated. The serum levels of interferon-γ (IFN-γ), interleukin-4 (IL-4), IL-5, IL-10, transforming growth factor-β (TGF-β) and OVA-specific IgE antibody (OVA-sIgE) were measured using enzyme-linked immunosorbent assay (ELISA), and the pathological changes of mouse nasal mucosa were observed by hematoxylin and eosin (HE) staining. Results The mean behavioral score was significantly greater in Group C (6.83 ± 0.50) than in groups A (1.17 ± 0.52) and B (1.33 ± 0.52) (P < 0.05), while a lower mean behavioral score was estimated in Group D (3.50 ± 0.50) than in Group C (P < 0.05). There were significant differences among the groups in terms of serum IFN-γ (F = 4.08, P < 0.05), IL-4 (F = 275.90, P < 0.05), IL-5 (F = 96.82, P < 0.05), IL-10 (F = 77.67, P < 0.05), TGF-β (F = 9.98, P < 0.05) and OVA-sIgE levels (F = 44.69, P < 0.05). The serum IFN-γ level was significantly lower in Group C than in groups A, B and C (P < 0.05), and the serum levels of IL-4, IL-5 and OVA-sIgE were significantly higher in Group C than in groups A, B and C (P < 0.05), while the serum IL-10 and TGF-β levels were significantly greater in Group D than in Group C (P < 0.05). Microscopy showed apparent loss of nasal mucosa cilia, increased number and enlargement of goblet cells, interstitial edema and submucous vascular dilation in Group C, while the pathological changes of nasal mucosa were alleviated in Group D relative to Group C. Conclusions E. granulosus HCFP has a protective activity against OVA-induced allergic rhinitis in mice.

ABSTRACT

Texture sensory attributes are the key items in quality control of Chinese medicinal honeyed pills. The purpose of this study is to develop a quality control method for assessing the texture sensory attributes of Chinese medicinal honeyed pills based on real-world Tongren Niuhuang Qingxin pilular masses and finished products. First, parameters of texture profile analysis(TPA) were optimized through single factor and central composite design(CCD) experiments to establish a detection method for texture sensory attri-butes of Tongren Niuhuang Qingxin Pills. The results showed that the established detection method was stable and reliable, with the optimal parameters set up as follows: deformation percentage of 70%, detection speed at 30 mm·min~(-1), and interval time of 15 s. Furthermore, 540 data points yielded form six texture sensory attributes of pills from 30 batches were subjected to multivariate statistical process control(MSPC) with Hotelling T~2 and squared prediction error(SPE) control charts to establish the quality control method of Tongren Niuhuang Qingxin Pills. This study is expected to provide a reference for improving the quality control system of Chinese medicinal honeyed pills.

ABSTRACT

For the field detection problems of critical quality attribute(CQA) of moisture content in traditional Chinese medicine(TCM) manufacturing process, big brand TCM Tongren Niuhuang Qingxin Pills were used as the carrier, to establish a moisture content NIR field detection model with or without cellophane in real world production with use of near infrared(NIR) spectroscopy combined with stoichiometry. With the moisture content determined by drying method as reference value, the partial least square method(PLS) was used to analyze the correlation between the spectrum and the moisture reference value. Then the spectral pretreatment methods were screened and optimized to further improve the accuracy and stability of the model. The results showed that the best quantitative model was developed by the spectral data pretreatment of standard normal variate(SNV) with the latent variable factor number of 2 and 7 of Tongren Niuhuang Qingxin Pills with or without cellophane samples. The prediction coefficient of determination(R_(pre)~2) and standard deviation of prediction(RMSEP) of the model with cellophane samples were 0.765 7 and 0.157 2%; R_(pre)~2 and RMSEP of the model without cellophane samples were 0.772 2 and 0.207 8%. The NIR quantitative models of moisture content of Tongren Niuhuang Qingxin Pills with and without cellophane both showed good predictive performance to realize the rapid, accurate and non-destructive quantitative analysis of moisture content in such pills, and provide a method for the field quality control of the critical chemical attributes of moisture in the manufacturing of big brand TCM.

ABSTRACT

Objective To analyze the components of proteins from Echinococcus granulosus cyst fluid using the shotgun method, and to identify the active components with potential regulatory effects for immune dysregulation diseases. Methods The E. granulosus cyst fluid was collected aseptically from the hepatic cysts of patients with cystic echinococcosis, and characterized by liquid chromatography (LC) tandem mass spectrometry (MS/MS) following digestion with trypsin. The protein data were searched using the software MaxQuant version 1.6.1.0 and the cellular components, molecular functions, and biological processes of the identified proteins were analyzed using the Gene Ontology (GO) method. Results The E. granulosus cyst fluid separated by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) had a relative molecular mass of 25 to 70 kDa. LS-MS/MS analysis identified 37 proteins, including 32 known proteins and 5 unknown proteins. At least 4 proteins were preliminarily found to exhibit potential regulatory effects for immune dysregulation diseases, including antigen B, glutathione-S-transferase (GST), thioredoxin peroxidase (TPX) and malate dehydrogenase (MDH). GO enrichment analysis showed that the identified proteins had 149 molecular functions and were involved in 341 biological processes. Conclusions E. granulosus cyst fluid has a variety of protein components, and four known proteins are preliminarily identified to be associated with immune dysregulation diseases.

ABSTRACT

Trichinellosis is a worldwide zoonotic parasitic disease, which may infect humans and a wide range of animals, including carnivores, herbivores and even birds. Humans get infections through ingestion of raw or semi-cooked meat or meat products containing live Trichinella larvae. Currently, this zoonosis is predominantly prevalent in southwestern, northeastern and central parts of China, which poses a great threat to human health and socioeconomic development. In addition, the emergent public health hazards caused by trichinellosis outbreaks greatly threaten human health and social stability. This review describes the current status, diagnosis, treatment and control measures of trichinellosis in China, so as to provide insights into trichinellosis control in the country.

ABSTRACT

@#AIM: To explore the factors affecting visual acuity recovery after phacoemulsification for high myopia complicated with cataract.<p>METHODS: From March 2015 to October 2017, 86 eyes of 70 patients with high myopia complicated with cataract were treated by phacoemulsification, and the factors affecting the visual acuity recovery after phacoemulsification for high myopia complicated with cataract were analyzed.<p>RESULTS: There was no correlation between posterior vitreous detachment and postoperative visual acuity recovery. Axial length, corneal astigmatism and macular disease were correlated with visual acuity recovery. Axis length, corneal astigmatism and macular disease were the risk factors affecting the outcome of surgery.<p>CONCLUSION: Phacoemulsification can effectively improve the visual acuity of patients with high myopia complicated with cataract. Macular lesion, excessive axial length and corneal astigmatism are the three main factors affecting the poor outcome after operation.

ABSTRACT

<p><b>OBJECTIVE</b>To study the role of PI3K/Akt pathway in the neuroprotective effect of paeoniflorin on PC12 cells.</p><p><b>METHOD</b>The paeoniflorin group (5, 10, 20 μmol · L(-1)) was pretreated for 30 min, and then added with Aβ25-35 (20 μmol · L(-1)) for interaction for 24 h. Inhibitor LY294002 (10 μmol · L(-1)) was pretreated for 30 min before the action of paeoniflorin (10 μmol · L(-1)). The MTT colorimetric method was used to detect the cell viability. The apoptosis rate was tested by the FITC-Annexin V/PI staining. The protein expression of p-AKT, Bax, Bcl-2 and cleaved caspase-3 protein were detected by Western blot analysis.</p><p><b>RESULT</b>Paeoniflorin could significantly inhibit the Aβ25-35-induced PC12 cell toxicity and apoptosis. Its protection effect may be achieved by up- regulating AKT phosphorylation level, increasing Bcl-2 protein expression, reducing Bax protein expression, inhibiting the activation of caspase-3. Inhibitor LY294002 could weaken the above protective effects of paeoniflorin.</p><p><b>CONCLUSION</b>Paeoniflorin could activate PI3K/Akt signaling pathway to protect the PC12 cell injury induced by Aβ25-35.</p>

ABSTRACT

This study was purposed to explore whether the blood transfusion of surgical patients can increase the severity of postoperative infection by a retrospective analysis of patients with postoperative infection in Chinese PLA General Hospital. By using a software "clinical transfusion database" developed by our department, 150 infected surgical cases were retrieved and divided into deep infection group and superficial infection group according to the infected location. These two groups were compared in term of the patient's age, duration of hospitalization, red blood cell transfusion volume, none-red cell transfusion volume, transfusion frequency and average transfusion volume. The results showed that red blood cell transfusion volume or none-red cells transfusion volume of patients with superficial infection was 4.50 (0 - 59) U or 2.95 (0 - 119.6) U, and that of deep infection was 9.00 (0 - 153) U and 8.05 (0 - 136.6) U, the differences was significant (P < 0.05). Between two groups, the transfusion frequency showed the most significant difference, median in the patients with superficial infection was about 2 (1 - 31) times, less than the deep infection group about 4 (1 - 49) times (P < 0.001). There was no significant difference between two groups in the average transfusion volume. It is concluded that perioperative blood transfusion volume and frequency of surgical patients seems to display a positive correlation with the degree of postoperative infection.

ABSTRACT

This study was purposed to explore the influence of S-nitrosoglutathione (GSNO) on membrane glycoprotein of frozen platelet. The levels of membrane glycoprotein on fresh liquid platelets, frozen platelets and frozen platelets with GSNO were measured by flow cytometry. The results showed that the GSNO obviously decreased platelet aggregation, the PAC-1 change in the three groups was not significant. The changes of CD42b and CD62P in fresh liquid platelet group, frozen platelet group and frozen platelets with GSNO were significant different. The change of membrane glycoprotein in above-mentioned three group was not significant. It is concluded that the GSNO inhibits platelet aggregation, maintains the function of platelets and may be used as a cryoprotectant. When frozen platelets were added with GSNO, the molecular rearrangement, structure change and other mechanism may occur in platelets.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the neuroprotective effects and mechanism of paeoniflorin on Abeta25-35-induced oxidative stress in PC12 cells.</p><p><b>METHOD</b>The proliferation of induced PC12 cells were investigated by the MTT method. The leakage rate of lactate dehydrogenase (LDH), the intracellular content of malondialdehyde (MDA), superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were also measured. The changes of the intracellular reactive oxygen species (ROS) level and the mitochondrial membrane potential (MMP) were investigated by flow cytometry using DCFH-DA and Rh123 staining. The protein expression of HO-1, Cyt C and cleaved Caspase-3 was detected by Western blot.</p><p><b>RESULT</b>The results showed that paeoniflorin at different doses (5, 10, 20 micromol . L-1) could increase the cell viability and activities of antioxidant enzyme (GSH-Px, SOD and HO-1), prevent LDH release and lipid peroxidation (MDA) production, decrease the level of intracellular ROS, increase MMP, inhibit the release of cytochrome c from the mitochondria and attenuate activation of Caspase-3.</p><p><b>CONCLUSION</b>Paeoniflorin can protect PC12 cells from Abeta25-35 oxidative stress injuries. The mechanism may be related to its antioxidant action and inhibition of mitochondria-mediated caspase signaling pathways.</p>

ABSTRACT

The aim of this study was to investigate the influence of S-nitrosoglutathione (GSNO) on agglutination and nitric oxide (NO) concentration in frozen platelets. The agglutination of platelets was detected by using platelet agglutination apparatus, the level of NO in platelets was detected by the nitrate enzyme reduction method. The results showed that the rates of agglutination in freeze platelets and frozen platelets treated with GSNO were (35.47 ± 2.93) and (24.43 ± 3.07), which were significantly lower than that in fresh liquid platelets (63.44 ± 2.96). The level of NO concentration in frozen platelets was (22.16 ± 6.38), which was significantly lower than that in fresh liquid platelets (31.59 ± 16.88). The level of NO concentration in frozen platelets treated with GSNO was (45.64 ± 6.31), which was significantly higher than that in fresh liquid platelets (P < 0.01). It is concluded that GSNO increases the concentration of NO in frozen platelets, inhibits platelet activation and maintains platelet function, thus GSNO can be used as a frozen protective agent.

ABSTRACT

This study was aimed to explore the correlation of hemogram changes during pregnancy of healthy women with postpartum blood transfusion. The outpatient and inpatient information of expectant lying-in women in our hospitals was collected, the route blood test, lever and kidney function and blood coagulation function tests were performed from the 4th to the 10th month of pregnancy. The pregnant women without underlying diseases and non-elderly pregnant women with single fetus were selected as the subjects of study. They were divided into postpartum blood transfusion group and non-blood transfusion group. The white blood cell (WBC) count, hemoglobin (Hb) level, platelet (Plt) count, plateletocrit (PCT), mean platelet volume (MPV), platelet distribution width (PDW) were compared in 2 groups. The results showed that 68 cases out of 450 expectant lying-in women received blood transfusion, among them 30 cases with complete data of puerperal transfusion were taken as blood transfusion group, the 28 cases of non transfusion puerperal as control group. There was no significant difference of hemogram changes between the two groups. However, there was a slight decline in Plt count and Hb level of late pregnant women. What is more, there was no correlation between Plt count change and the PCT, MPV and PDW. It is concluded that the changes of hemogram during pregnancy has no correlation with postpartum hemorrhage and blood transfusion in healthy pregnant women, the Plt count and Hb level of pregnant women slightly decline. Nevertheless, PCT, MPV and PDW are within the normal range.

ABSTRACT

<p><b>OBJECTIVE</b>To compare the anti-fertility effects of the four extracts from the roots of Rhynchosia volubilis Lour on male mice, that is, ethanolic extract, ethyl acetate extract, n-butanol extract and aqueous extract.</p><p><b>METHODS</b>Four extracts from the roots of Rhynchosia volubilis Lour (1%, 0.1 ml/10 g), were administered orally for 11 weeks to adult male mice. The fertility and testicular function of the mice were assessed by mating tests and analyses of sperm motility in cauda epididymides and biochemical and histological indexes in the blood samples and reproductive organs.</p><p><b>RESULTS</b>The four extracts, especially aqueous extract, gradually decreased the pregnancy rate of the experimental mice from the 77th day of the treatment, with an obvious reduction in the number of spermatozoa. Morphological observation of the reproductive organs by light microscopy showed that the numbers of the secondary spermatocytes and spermatids were decreased in varied degrees, and the seminiferous tubules were disarranged, while the numbers and shapes of and spermatids were decreased in varied degrees, and the seminiferous tubules were disarranged, while the numbers and shapes of spermatogonia, Sertoli cells and Leydig cells remained unchanged.</p><p><b>CONCLUSION</b>The four extracts from the roots of Rhynchosia volubilis Lour all have anti-fertility effects on male mice, and that of the aqueous extract is more obvious.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the stress distribution on cast retentive clasp arms in dislodging denture, and to discuss the deepest undercuts of the second mandibular premolar (abutment) for cobalt-chromium alloy cast clasps with different widths.</p><p><b>METHODS</b>Three-dimensional finite element models of the abutment with different depths of undercuts and retentive arms with different widths were set up. Dynamic displacement load (3 mm/s) was exerted on the middle of the retentive arms to analyze the stress in retentive arms while they were being removed from the abutment.</p><p><b>RESULTS</b>The peak stress in retentive arms was positively correlated to the undercuts displaced by clasp tips, and those were not obviously related to the undercuts displaced by the middle of retentive arms. When width/thickness of retentive arms was 3, the increase of peak stress of retentive arms with similar locations of clasp tips was significantly related to the increase of the arm width. The deepest undercuts of the second mandibular premolar for cobalt-chromium alloy cast retentive arms with different widths of 1.8 mm, 1.6 mm, and 1.4 mm were 0.25 mm, 0.30 mm, and 0.35 mm respectively.</p><p><b>CONCLUSIONS</b>When width/thickness of the retentive clasp arm is fixed, the wider the arm is, the smaller depth it should be placed on the undercut of abutment. Retentive clasp arms with different widths should be placed on different depths of undercuts in order to prevent their permanent deformation.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the effect of Rhynchosia volubilis Lour ethyl acetate extract (RVLEAE) on male mouse procreation and analyse their chemical composition.</p><p><b>METHOD</b>With the method of solvent extraction, RVLEAE was extracted and concentrated. In the experiment of mice, 80 male mice were randomly and equally divided into four groups: Normal Saline control, positive control with 0.1% triperygium wilfordii glycoside, 100 mg x kg(-1) x d(-1) RVLEAE and 400 mg x kg(-1) x d(-1) RVLEAE. Every mouse was fed with drug 0.01 mL x g(-1), once a day, ig, for eleven consecutive weeks. After two and 10 weeks, male and female mouse naturally mated for one week. The pregnancy rate, number of fetus and nonviable fetus, the viability of spermatozoon in the epididymis cauda, pathological change of testis and epididymis were observed in this experiment. In the analysis of chemical composition, RVLEAE were separated with column chromatography, and chemical compositions were identified with thinlayer chromatography, infrared chromatography and nuclear magnetic resonance.</p><p><b>RESULT</b>The pregnancy rate of mice was markedly decreased. The number and viability of spermatozoon were slightly reduced in I and II after two and 10 weeks, but the pathological changes of testis and epididymis were markedly occurred. Main chemical compositions were identified as saccharide, glycosides especially analog of fucose, alcohols, and phenols.</p><p><b>CONCLUSION</b>RVLEAE can inhibit the procreation of male mice, and inhibitory target tissue may be the epididymis. Active mechanism of RVLEAE may be that glycosides interfere the maturation of spermatozoon in the epididymis cauda.</p>
