ABSTRACT

Abstract Acute diarrhea is the second leading cause of morbidity and mortality attributed to infections in children under five years of age worldwide, with 1.7 million annual estimated cases and more than 500,000 deaths. Although hydroelectrolytic replacement is the gold standard in treating diarrhea, it does not interfere with the restoration of the intestinal microbiota. Several studies have searched for an adequate alternative in restructuring intestinal homeostasis, finding that treatments based on probiotics, prebiotics, and synbiotics are effective, which made such treatments increasingly present in clinical practice by reducing illness duration with minimal side effects. However, there are still controversies regarding some unwanted reactions in patients. The diversity of strains and the peculiarities of the pathogens that cause diarrhea require further studies to develop effective protocols for prevention and treatment. Here, we provide a descriptive review of childhood diarrhea, emphasizing treatment with probiotics, prebiotics, and synbiotics.

ABSTRACT

American visceral leishmaniasis (AVL) is a vector-borne zoonosis in which the dog has an important role in the epidemiology of the disease. In Brazil, a high prevalence of canine infection is directly correlated with an increased risk of occurrence of AVL. The aim of this study was to investigate the phlebotomine fauna and seroprevalence of canine visceral leishmaniasis in Pedra 90 region of Cuiabá municipality. For the entomological survey, CDC traps were used from August 2014 to July 2015. In the serological evaluation of dogs, the immunochromatographic test DPP LVC was employed for screening the samples while enzyme-linked immunosorbent assay (Bio-Manguinhos) was used as a confirmatory assay. The previously unreported phlebotomine species Lu. andersoni, Lu. braziliensis, Lu. bourrouli, and Lu. scaffi were added to the phlebotomine fauna of Cuiabá. In addition, the medically important phlebotomine species Lu. cruzi, Lu. flaviscutellata, and Lu. whitmani were identified. The canine survey revealed the prevalence of 1.14% for canine visceral leishmaniasis in the Pedra 90 region, the region being considered a transmission area.(AU)

ABSTRACT

The aim of this study was to evaluate the pharmacological semen collection method with urethral catheterization (CT) in captive cougar (Puma concolor) males. The pharmacological method (CT; N= 3) was compared to the electroejaculation technique (EE; N= 4). For CT collection, medetomidine was administrated to induce semen release using a tomcat catheter inserted into the urethra to collect by capillarity. The proposed method was efficacious on all animals used. Through the CT method, semen collected yielded smaller volume (106,7±30,5aµL) and higher concentration (524,1±54,3b x 106sperm/mL) compared to EE (450,0±0,1bµL and 205,0±141,8a x 106 sperm /mL). Evaluations of vigor, motility and sperm pathology demonstrated that CT does not affect semen quality when compared to EE (P> 0.05). Thus, the proposed method consists of a more practical and efficient technique for semen collection with good quality, eliminating the need for eletroejaculation.(AU)

ABSTRACT

The aim of this study was to determine the occurrence of EGUS and to quantify serum gastrin levels in jumping horses during competition season and interseason period. Forty jumping horses, competing at high level were randomly allocated into two groups, the Training Group: twenty jumping horses undergoing intense training and participating in competitions, and the Rest Group: twenty jumping horses in the interseason (resting period). The gastroscopic examinations and blood samples of the horses in the training group were performed 1-2 days following the competition while in the horses of the rest group, following 4 weeks of rest. The serum gastrin levels were measured at two different times: pre-feeding and two hours after feeding the horses (postprandial) by ELISA kit. Gastric lesion score data were compared by the Mann-Whitney U test (α= 0.05) and the mean gastrin values were compared between the groups and between the two moments by the paired tet tests, respectively (α= 0, 05). Squamous gastric ulcers were detected in 42.5% of all jumping horses examined independent of the period, competition season or interseason. Serum gastrin levels were significantly higher in the Training Group with no difference between pre-feeding and postprandial values.(AU)

ABSTRACT

We report a case of secondary skin lesionby infection with Corynebacterium pseudotuberculosis in bovine. Abscesses and multiple nodular lesions, ranging from 8x10 to 20x25 cm in diameter, firm, sensitive to touch, and without mobility, some with ulcerated surface, surrounded by reddish halo and draining piosanguinolenta secretion, and other surfaces with alopecia, were identified in the skin of the animal`s thoracic area. Histopathology and isolation of the agent from fragments obtained after surgical removal of the lesions confirmed the cutaneous infection by C. pseudotuberculosis. Microscopic lesions were characterized by lumpy skin disease and ulcerative pyogranulomatous. Evaluation of in vitro susceptibility to antimicrobial demonstrated that the agent was resistant to amikacin, kanamycin, neomycin and penicillin G, and sensitive to ampicillin + subactam, amoxicilia with clavulonic acid, cephalexin, cephalothin, cefotaxime, enrofloxacin, gentamicin, and tetracycline. The surgical removal of the lesions, and treatment with enrofloxacin resulted in animal cure. Skin lesions observed in case of ptiose, squamous cell carcinoma and atypical form of Actinobacillosis should be considered in the differential diagnosis of cutaneous form of C. pseudotuberculosis infection in cattle. The data presented demonstrate that infection with C. pseudotuberculosis should be considered in the differential diagnosis of skin lesions in cattle in Brazil.(AU)

ABSTRACT

In the canine species, the precise mechanisms of pregnancy maintenance and the initiation of parturition are not completely understood. The expression of genes encoding the receptors for estrogen (ERα mRNA) and oxytocin (OTR mRNA) was studied in the endometrium and myometrium during pregnancy and parturition in dogs. Real-time PCR was performed to quantify the levels of ERα mRNA and OTR mRNA in the uterus of bitches during early (up to 20 days of gestation), mid (20 to 40 days) and late pregnancy (41 to 60 days), and parturition (first stage of labor). All tissues expressed ERα and OTR mRNA, and are thus possibly able to respond to eventual estrogen and oxytocin hormonal stimuli. No statistically significant differences in the expression of ERα mRNA were verified in the endometrium and myometrium throughout pregnancy and parturition, but expression of OTR mRNA increased at both parturition and late pregnancy. We concluded that the increase of endometrial and myometrial OTR mRNA expression in dogs is not an event dependent on estrogenic stimulation. Moreover, the contractility response of the canine uterus to oxytocin begins during pregnancy and maintains myometrial activity. The expression of OTR mRNA in canine uterine tissues varied over time, which supports an interpretation that the sensitivity and response to hormone therapy varies during the course of pregnancy and labor. Further studies are needed to elucidate the factors underlying the synthesis of uterine oxytocin receptors and the possible role of ERβ rather than ERα in the uterine tissues during pregnancy and parturition in dogs.

ABSTRACT

We aimed to study the territorial behavior of a male cougar, captured in the Parque Estadual da Serra do Brigadeiro (PESB), using a GPS collar and the Globalstar Satellite. Monitoring the animal for nine months we obtained 328 locations. The data suggest that the area used by this feline is approximately 610km2, the largest area described for this specie. It was also possible to observe that only 20% of the locations of the animal occurred within the PESB area. The territorial behavior observed in this study shows the vulnerability of the animal to human activities such as hunting, road kill and contact with pathogens from domestic animals. It was also observed that two roads constitute artificial barriers for animal movement, possibly preventing gene flow between the PESB and two other parks. The creation of an effective connectivity between the PESB, the Parque Estadual do Rio Doce and the Parque Nacional do Caparaó is critical to ensure genetic exchange between the animal populations and thereby promote their conservation.

ABSTRACT

Aims: Develop an anti-tuberculosis (TB) Fixed Dose Combination (FDC) tablet containing an immediate release layer (IRL) composed of both rifampicin (RIF) and pyrazinamide (PYR) and a retarded release layer (RRL) comprised of isoniazid (INH) which would allow segregated delivery of RIF and INH. Study Design: Trials were conducted on the pre-formulations and formulations to assess the compatibility of excipients and obtain a modified release profile, for an IRL consisting of both RIF and PYR and a RRL containing INH. Place and Duration of Study: This study was performed at the Laboratory of Pharmaceutical Industrial Technology, Drug and Pharmaceutical Department, Faculty of Pharmacy, between March 2008 and April 2010. Methodology: Preformulation studies were performed on RIF and PYR, alone and in combination with excipients. The pharmacopeic attributes of the distinct layers and the FDC tablets were evaluated for hardness, friability and disintegration time. The FDC bilayer tablets were analyzed for their drug content and cumulative dissolution of the drug. Results: Fourier transform infrared, x-ray diffraction and differential scanning calorimetry results revealed the presence of amorphous and crystalline RIF forms and no potentially relevant incompatibilities were identified in the kneaded system containing RIF, PYR and excipients. In vitro drug release from the FDC tablets revealed that the intragranular addition of croscarmellose sodium into the IRL rendered a cumulative dissolution of RIF and PYR within the limits of simulated gastric fluid. And, for RRL, the most effective retardant matrix excipient was found to be hydroxypropyl methylcellulose. Conclusion: Segregated delivery of RIF and INH from bilayer tablets is an option for the development of immediate release FDC tablets and the retarded release of INH, this strategy proved suitable for preventing contact of these two drugs under acidic conditions. This finding suggested that RIF had a high in vivo bioavailability which qualifies this FDC for future bioavailability studies.

ABSTRACT

Considering the venereal transmission of visceral leishmaniasis from dogs to bitches, the aim of this study was to verify if the penile surface and smegma from infected dogs can be the source of parasites in bitches. Twelve Leishmania infantum infected dogs had semen and smegma samples collected for submission to PCR identification of the DNA of the parasite. Semen (41.7 percent) and smegma (50.0 percent) have similar positive incidence (P>0.05; Fisher's exact test), with 58.3 percent of the dogs positive for semen and/or smegma samples. The proportion of positivity for both semen and smegma was 33.3 percent, but 8.3 percent was positive only for semen, and 16.7 percent only for smegma, revealing a moderate agreement between tests (K=0.5; Kappa index). It was concluded that Leishmania infantum is present in the smegma of contaminated dogs and it can be a source of parasites for the semen and the bitch...

ABSTRACT

The aim of this work was to adequate raw refrigerated milk quality of five dairy farms to microbiological and somatic cell count (SCC) standards established by Paraná state legislation. The dairy farms were monitored during seven months after the implementation of good milking practices and recommendations regarding mastitis treatment, and milking equipment maintenance and hygiene. Before the adoption of recommendations, the average total bacterial count (TBC) was 1.36 x 10(6)CFU/mL and average SCC was 1.87 x 10(6)/mL. All farms reached established standards for TBC, originating an average reduction of 93.4%. Average SCC reduction was 74.3%. Only three farms reached established values for SCC. The remaining reached values near the limits. The suitability of TBC and SCC produced an increase in the monthly income due to payment quality reward of the dairy product, between R$120.00 and R$828.00, according to each farm production.

ABSTRACT

This paper is the first report of unilateral cryptorchidism in an adult wildlife ocelot, captured in Parque Estadual do Rio Doce. Cage traps were used to capture the animal, using bovine offal as bait. The animal was anesthetized with anesthetic darts and kept under anesthesia through a combination of ketamine and xylazine. The andrologycal examination showed that the left testicle was located subcutaneously near the inguinal region. In this case of unilateral cryptorchidism, the testis was soft and had a volume of 2.57mL, while the contralateral testis had a firm consistency and volume of 11.50mL. The length of the region occupied by the penile spikes was similar to other breeding animals. Cryptorchidism is an inherited condition linked to low genetic variability previously reported in consanguineous wild cats. Due to the increasing isolation of wild population in forest fragments, this finding is concerning because it can be indicative of inbreeding in wild ocelot populations.

ABSTRACT

Nonsteroidal anti-inflammatory drugs (NSAIDs) have been used extensively to control inflammatory pain. Several peripheral antinociceptive mechanisms have been described, such as opioid system and NO/cGMP/KATP pathway activation. There is evidence that the cannabinoid system can also contribute to the in vivo pharmacological effects of ibuprofen and indomethacin. However, there is no evidence of the involvement of the endocannabinoid system in the peripheral antinociception induced by NSAIDs. Thus, the aim of this study was to investigate the participation of the endocannabinoid system in the peripheral antinociceptive effect of NSAIDs. All experiments were performed on male Wistar rats (160-200 g; N = 4 per group). Hyperalgesia was induced by a subcutaneous intraplantar (ipl) injection of prostaglandin E2 (PGE2, 2 μg/paw) in the rat’s hindpaw and measured by the paw pressure test 3 h after injection. The weight in grams required to elicit a nociceptive response, paw flexion, was determined as the nociceptive threshold. The hyperalgesia was calculated as the difference between the measurements made before and after PGE2, which induced hyperalgesia (mean = 83.3 ± 4.505 g). AM-251 (80 μg/paw) and AM-630 (100 μg/paw) were used as CB1 and CB2 cannabinoid receptor antagonists, respectively. Ipl injection of 40 μg dipyrone (mean = 5.825 ± 2.842 g), 20 μg diclofenac (mean = 4.825 ± 3.850 g) and 40 μg indomethacin (mean = 6.650 ± 3.611 g) elicited a local peripheral antinociceptive effect. This effect was not antagonized by ipl CB1 cannabinoid antagonist to dipyrone (mean = 5.00 ± 0.9815 g), diclofenac (mean = 2.50 ± 0.8337 g) and indomethacin (mean = 6.650 ± 4.069 g) or CB2 cannabinoid antagonist to dipyrone (mean = 1.050 ± 6.436 g), diclofenac (mean = 6.675 ± 1.368 g) and indomethacin (mean = 2.85 ± 5.01 g). Thus, cannabinoid receptors do not seem to be involved in the peripheral antinociceptive mechanism of the NSAIDs dipyrone, diclofenac and indomethacin.

ABSTRACT

The myelography procedure is reported in an adult of the Sphiggurus villosus (hairy-tree-porcupine) species coming from the wild, in the region of Curitiba/PR. After proceeding with the examination, while the animal was under general anesthesia, there was a dorsal deviation of the contrast in the height of the seventh thoracic vertebra (T7) caused by compression in the ventral region of the vertebra. This is suggestive of edema caused by intramedullary hemorrhage secondary to trauma suffered by the animal in the wild. Therefore, the use of myelography in Sphiggurus villosus (hairy-tree-porcupine) proved to be quite valid and efficient, presenting the advantages of greater precision to delimit and locate the lesion present in the vertebral column.

ABSTRACT

Helicobacter pylori, a gram-negative bacterium, possesses two important virulence factors: the vacuolating toxin (vacA), and the cytotoxin-associated gene product (cagA). The aim of the present study was to evaluate the presence of H. pylori in the stomach and oral cavity of humans and compare the cagA and vacA genotypes of H. pylori found in different samples (stomach, saliva and dental plaque) from the same patient. Gastric biopsies, saliva and dental plaques were obtained from 62 dyspeptic adults. DNA was extracted and evaluated for the presence of H. pylori and the alleles cagA and vacA. Persons with gastritis had a higher frequency of H. pylori -positive samples in the stomach while positive samples from gastric biopsies were significantly correlated with those from the oral cavity. There was a high H. pylori frequency in patients while the cagA gene was associated with vacA s1 alleles in gastric biopsies. Our results suggest a reservoir of the species in the oral cavity and that, in one patient, more than one H. pylori strain may exist in the saliva, dental plaque and stomach. We found a relationship between gastric infection and the bacterium in the oral cavity, with the cytotoxin genotype varying between saliva and dental plaque.(AU)

ABSTRACT

In this study, a dorsocaudal portion resection of the left caudal lung lobe was performed using an endoscopic stapler by thoracoscopic procedure. Ten healthy horses, eight males and two females were evaluated by clinical examination and submitted to the surgical procedure. The animals were sedated and maintained in standing position. The procedure was done in the left hemitorax using three access previously studied: the first portal was done at the 12th intercostal space (IS); the second one at the 14th, 15th, or 16th IS; and the third one at the 14th or 15th IS, in accordance with the necessity of positioning the instruments. The employed surgical technique seemed to be adequate in all animals, since it helped controlling the existing pneumothorax; furthermore, clinical recovery during the postoperative period was satisfactory, except for one animal. These results suggest that thoracoscopic may successfully be employed for partial lung resection in horses, using endoscopic staplers, and its practical use may contribute to the knowledge and resolution of thoracic diseases.

ABSTRACT

A 9-year-old horse presented serosanguineous nasal discharge, absence of breath out through the right nostril, and respiratory noise. Endoscopic and radiographic exams revealed a six centimeter diameter mass, covered by yellowish mucosa, which was obstructing the entire right nasal cavity and part of the left one. The mass was excised through a right frontal sinusotomy. The microscopic exam and the culture revealed a fungic granulomatous rhinitis. Antibiotic and anti-inflammatory drugs were postoperatively administered; moreover, camomile tea and distilled water were flushed in a drain placed above the bone flap.

ABSTRACT

OBJECTIVE: To compare the results from fingertip-to-floor and straight leg raise tests by means of photogrammetry, investigate the concordance of hip angle findings between the two tests and evaluate the intra and inter-examiner reliability of the photogrammetric analysis procedure. METHOD: The tests were applied to 35 healthy male subjects (mean age 23 ± 3.4 years, height 170 ± 4 cm and weight 68 ± 11 kg). The subjects were divided into two groups on the basis of the test classifications. Surface markers were placed on bone references in order to calculate the hip, thoracolumbar, leg, knee and tibiotarsal angles. The reliability of the photogrammetric analysis procedure on the angles measured by two examiners was tested. RESULTS: For the hip angle, there were significant differences between the flexibility groups in both tests, and subjects with reduced flexibility had lower angles. There was a significant difference (22.8 percent) between the classifications designated by the two tests. However, the hip angle showed high levels of concordance between the tests (ICC: 0.89 and mean difference of -0.85º). The photogrammetric procedures presented high reliability rates, both for intra and inter-examiner (ICC ranging from 0.94 to 0.99). CONCLUSION: The tests presented differences in the classification of the subjects' flexibility. However, considering the hip angle, there was high concordance between the test results. The photogrammetric analysis procedures were reproducible, both for intra and for inter-examiner measurements, thus making this a useful analysis tool for the tests in question.
