ABSTRACT
Effects of nifedipine and cimetidine were studied in isolated rat stomach fundus strip preparation at four dose levels. The contractions were induced by 8 micro g/mg acetylcholine [Ach]. There was dose dependent inhibition of contractions by nifedipine were as dose dependent potentiation by cimetidine. The 0.02 micro g/ml nifedipine significantly [P<0.001] reduced the acetylcholine - induced contractions whereas 60 micro g/ml cimetidine significantly [P<0.01] potentiated the acetylcholine - induced contractions. The spontaneous contractions of rat stomach strip were completely abolished by 0.8 micro g/ml nifedipine whereas 1000 micro g/ml cimetidine had no effect. It is concluded that nifedipine may prevent stress - induced gastric motility and therefore ulcer formation whereas cimetidine may aggravate it as a side effect

Effects of nifedipine and cimetidine on gastric acidity were studied in pylorus ligated rats in doses of 0.25 mg, 0.5 mg and 1.0 per Kg given intraperitoneallv. Both drugs inhibited gastric secretions dose dependently. Nifedipine was found to be slightly more effective than cimetidine in raising gastric pH and reducing acidity titratable to 7.0 pH at all dose levels. However, it was less effective in reducing voume of gastric secretions. It is likely that nifedipine has greater anti-ulcer effect than cimetidine in rats
