ABSTRACT

Objective: This study systematically explore the efficacy and safety of fourth-generation chimeric antigen receptor T-cells (CAR-T), which express interleukin 7 (IL7) and chemokine C-C motif ligand 19 (CCL19) and target CD19, in relapsed or refractory large B-cell lymphoma. Methods: Our center applied autologous 7×19 CAR-T combined with tirelizumab to treat 11 patients with relapsed or refractory large B-cell lymphoma. The efficacy and adverse effects were explored. Results: All 11 enrolled patients completed autologous 7×19 CAR-T preparation and infusion. Nine patients completed the scheduled six sessions of tirolizumab treatment, one completed four sessions, and one completed one session. Furthermore, five cases (45.5%) achieved complete remission, and three cases (27.3%) achieved partial remission with an objective remission rate of 72.7%. Two cases were evaluated for disease progression, and one died two months after reinfusion because of uncontrollable disease. The median follow-up time was 31 (2-34) months, with a median overall survival not achieved and a median progression-free survival of 28 (1-34) months. Two patients with partial remission achieved complete remission at the 9th and 12th months of follow-up. Therefore, the best complete remission rate was 63.6%. Cytokine-release syndrome and immune effector cell-associated neurotoxicity syndrome were controllable, and no immune-related adverse reactions occurred. Conclusion: Autologous 7×19 CAR-T combined with tirelizumab for treating relapsed or refractory large B-cell lymphoma achieved good efficacy with controllable adverse reactions.

ABSTRACT

Objective: To further elucidate the clinical efficacy and safety of a combination regimen based on the BTK inhibitor zebutanil bridging CD19 Chimeric antigen receptor T cells (CAR-T cells) in the treatment of relapsed/refractory diffuse large B-cell lymphoma (r/r DLBCL) . Methods: Twenty-one patients with high-risk r/r DLBCL were treated with a zanubrutinib-based regimen bridging CAR-T between June 2020 and June 2023 at the Department of Hematology, Tongji Hospital, Tongji University and the Second Affiliated Hospital of Zhejiang University, and the efficacy and safety were retrospectively analyzed. Results: All 21 patients were enrolled, and the median age was 57 years (range: 38-76). Fourteen patients (66.7%) had an eastern cooperative oncology group performance status score (ECOG score) of ≥2. Eighteen patients (85.7%) had an international prognostic index (IPI) score of ≥3. Three patients (14.3%) had an IPI score of 2 but had extranodal infiltration. Fourteen patients (66.7%) had double-expression of DLBCL and seven (33.3%) had TP53 mutations. With a median follow-up of 24.8 (95% CI 17.0-31.6) months, the objective response rate was 81.0%, and 11 patients (52.4%) achieved complete remission. The median progression-free survival (PFS) was 12.8 months, and the median overall survival (OS) was not reached. The 1-year PFS rate was 52.4% (95% CI 29.8% -74.3%), and the 1-year OS rate was 80.1% (95% CI 58.1% -94.6%). Moreover, 18 patients (85.7%) had grade 1-2 cytokine-release syndrome, and two patients (9.5%) had grade 1 immune effector cell-associated neurotoxicity syndrome. Conclusion: Zanubrutinib-based combination bridging regimen of CAR-T therapy for r/r DLBCL has high efficacy and demonstrated a good safety profile.

ABSTRACT

@#AIM: To compare the efficacy of conbercept and ranibizumab on neovascular age-related macular degeneration(nARMD)of type 1 macular neovascularization(MNV)with fibrovascular pigment epithelial detachment(fPED).<p>METHODS: Retrospective clinical study. From January 2019 to December 2020, 48 patients(48 eyes)of nARMD type 1 MNV patients with fPED diagnosed in our hospital were included and divided into conbercept group with 26 patients(26 eyes)and ranibizumab group with 22 patients(22 eyes)according to the drugs they received. All patients received treatment of 3+PRN. Followed up for 12mo, the best corrected visual acuity(BCVA)of the two groups was observed, and optical coherence tomography(OCT)was used to measure the macula foveal thickness(CFT)and the regression degree(height, area, volume)of retinal pigment epithelial detachment(PED).<p>RESULTS: There was no significant difference between two groups in BCVA, CFT and PED height, area and volume before treatment(<i>P</i> >0.05). The PED height of the two groups was significantly improved at 3, 6 and 12mo after the first intravitreal injection treatment compared with those before treatment(<i>P</i><0.05). But the PED area and volume were not significantly improved(<i>P</i>>0.05). There was no significant improvement in BCVA between the two groups after treatment compared with those before treatment(<i>P</i>>0.05). The CFT of the conbercept group was significantly improved at 3, 6 and 12mo after treatment compared with those before treatment(<i>P</i><0.05), and the ranibizumab group improved significantly only 3mo after treatment(<i>P</i><0.05). There were no significant differences in BCVA, CFT, and PED height, area and volume between the two groups at 3, 6 and 12mo after treatment(<i>P</i> >0.05).<p>CONCLUSION: The conbercept and ranibizumab have good effects on type 1 MNV with fPED in nARMD, which can reduce the PED height and CFT, and stabilize the visual acuity, PED area and volume. However, conbercept can achieve longer reduction of macular edema.

ABSTRACT

Objective To study the clinical efficacy of bridge baton fixation with arthroscopic surgery in treatment of calcaneal fractures. Method From May 2013 to November 2014, 18 cases suffered from calcaneal fracture with arthroscopic surgical bridge baton fixation treatment in our hospital served as the object of the study. They were retrospectively analyzed during preoperative, postoperative the joint surface, the Bohler angle and Gissane angle , mean follow-up of 12 months, Maryland Foot Score were compared by the analysis. Results All cases were followed up for 12 months averagely. Joint surface flatness of 18 cases were significantly improved, Bohler angle, Gissane angle were also significant differences (P < 0.05). The excellent rate of Maryland score was 86.8%. Conclusion The bridge baton fixation with arthroscopic surgery for treatment of calcaneal fractures is certain effective and minimally invasive. Arthroscopy can restore the articular surface. The foot function recovery of the patients is satisfied.

Objective:To investigate the policy changes of medical insurance for urban and rural residents in Urumqi before and after the integration,so as to provide suggestion for the development of urban and rural medical insurance whole as plan.Methods:The health care financing after integration standards,reimbursement ratio and the changes of management methods were analyzed.Results:The personal financing standard and the starting line for patients who participated New Rural Cooperative Medical Insurance in tertiary hospitals raised after medical insurance,which effected the medical choice of patients participated the medical insurance to a certain extent.The gap between urban and rural areas affected the integration of urban and rural residents health care work.Conclusion:The level of individual pay cost should be consistent with the level of treatment in the design of ginseng protect personnel It needed to increase the government support,payment standard and treatment standard reference or innovation.

ABSTRACT

Objective To investigate the clinical efficacy of anti-inflammatory therapy (intranasal corticosteroids combined with oral leukotriene receptor antagonist) in pediatric mild to moderate obstructive sleep apnea hypopnea syndrome (OSAHS),and analyze the relationship between OSAHS and inflammation factors.Methods Fifty patients with mild to moderate OSAHS,diagnosed by polysomnography (PSG) during Jan.to Nov.2016,were enrolled in present study.The patients' medical history,data of special physical examination,paryngorhinoscopy,PSG and OSA-18 questionnaire were collected.Patients received the therapy of intranasal corticosteroids combined with oral leukotriene receptor antagonist for 12 weeks.Special physical examination,paryngorhinoscopy,PSG and OSA-18 questionnaire were reviewed and the data before and after treatment were compared.Results Of the 50 subjects,37 were with mild OSAHS and 13 with moderate OSAHS.A total of 19 cases (38%) were cured including 17 mild OSAHS and 2 moderate cases.Other 19 cases (38%) got therapeutic effect but not be cured.Twelve cases (24%) were invalid or aggravated.There were 10 patients (20%) who received surgical treatment after drug treatment.The average values of obstructive apnea index (OAI) and mixed apnea index (MAI) decreased significantly in mild group and only OAI decreased in moderate group.After treatment,the average volumes of adenoids and tonsils were significantly reduced in mild OSAHS children but not in moderate OSAHS children.The OSA-18 questionnaire score declined only in mild group.No obvious correlation existed between the change of tonsil volume and the parameters of PSG.Conclusion Anti-inflammatory therapy of intranasal corticosteroids combined with oral leukotriene receptor antagonist may obviously reduce the volumes of adenoids and tonsils,improve the PSG indexes and the life quality of OSAHS patients,especially for those children with mild OSAHS.

ABSTRACT

On account of the dense cuticles of the fresh stem and the light, hard and pliable texture of the dried stem, Dendrobii Caulis is difficult to dry or pulverize. So, it is very important to the ancient doctors that Dendrobii Caulis should be properly treated and applied to keep or evoke its medicinal effects. The current textual research results about the preliminary processing, processing and usage methods of Dendrobii Caulis showed that: (1) In history the clinical use of fresh or processed Dendrobii Caulis as teas and tinctures were very common. (2) Its roots and rhizomes would be removed before using. (3) Some ancillary approaches were applied to shorten drying times, such as rinsing with boiling mulberry-ash soup, washing or soaking with liquor, mixing with rice pulp and then basking, etc. (4) According to the ancients knowledge, the sufficient pulverization, by means of slicing, rasping, hitting or pestling techniques, was necessary for Dendrobii Caulis to take its effects. (5) The heat processing methods for Dendrobii Caulis included stir-baking, stir-frying, steaming, decocting and stewing techniques, usually with liquor as an auxiliary material. Among above mentioned, steaming by pretreating with liquor was most commonly used, and this scheme was colorfully drawn in Bu Yi Lei Gong Pao Zhi Bian Lan (Ming Dynasty, 1591 CE) ; moreover, decocting in advance or long-time simmering so as to prepare paste products were recommended in the Qing Dynasty. (6) Some different processing programs involving stir-baking with grit, air-tightly baking with ondol (Kangs), fumigating with sulfur, which appeared in modern times and brought attractive outward appearance of the drug, went against ancients original intentions of ensuring drug efficacy.

ABSTRACT

Objective To explore the influence factors of the compliance in colorectal cancer( CRC)screening. Methods A total of 140 096 local people were selected for fecal occult blood test( FOBT)in Songjiang District Shanghai City. They were over 40 years old and thought to be high risk groups of colorectal cancer. The participants with FOBT positive were asked to following colonoscopy. All the participants were interviewed by a questionnaire. The influencing factors of compliance on colorectal cancer screening were analyzed by multivariate regression. Results Of all the residents with high risk of colorectal cancer,The FOBT positive rate was 16. 83%. Of the FOBT positive residents,8 566 cases took the examination of colonoscopy,and the screening rate was 36. 33%. A total of 142 cases were diagnosed as colorectal cancer, with the detection rate of 1. 66%. Compared with the positive rate of FOBT in 2012 and 2013,the rate was significantly lower(18. 16%,15. 78%,p<0. 05),and the rate of colonoscopy examination increased(45. 66%,26. 04%,p<0. 05). Multivariate logistic regression showed that fear of pain( OR=3. 01 ),the thought of uselessness( OR=1. 89 )were risk factors for compliance in colorectal cancer screening( p <0. 05 ). Conclusion The compliance of colorectal cancer screening in Songjiang District is relatively low. High-risk groups with regular physical examination,high level education were prone to have good compliance,and fear of pain,think it useless were the main reason for incompliance.

ABSTRACT

<p><b>OBJECTIVE</b>To compare iron bioavailability (Fe BV) from ten selected kinds of Chinese wheat flours in order to provide scientific basis for further human trials and enable plant breeding programs to screen biofortified wheat cultivars.</p><p><b>METHODS</b>An in vitro digestion/Caco-2 cell model was used to assess Fe BV of ten flour samples from six leading Chinese wheat cultivars and the stability of Fe BV in one cultivar was studied across three growing environments.</p><p><b>RESULTS</b>Significant differences were observed in both Fe BV and Fe bioavailability per gram of food (Fe BVPG) among cultivars (P<0.01) grown at the same location with the same flour extraction rate. Zhongyou 9507 and Jingdong 8 had Fe BV 37%-54% and Fe BVPG 103%-154% higher than the reference control. In the Anyang environment, Zhongyou 9507 had a higher wheat flour-Fe level and Fe BVPG. Differences in Fe BV were detected in cultivars with different flour extraction rates.</p><p><b>CONCLUSION</b>Zhongyou 9507 and Jingdong 8 were identified as the most promising cultivars for further evaluation of efficacy by using human subjects. The growing environments had no effect on Fe BV, but did have a significant effect on Fe BVPG. Fe bioavailabilities in low-extraction (40%) flours were higher than those in high-extraction (78%) flours.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the need and utilization for health services in patients with allergic rhinitis in Qingdao area.</p><p><b>METHODS</b>An investigation was conducted by the method of combining multi-stage sampling and cluster sampling. SPSS 13.0 software was used to analyse the data.</p><p><b>RESULTS</b>Among Two thousand and four hundred questionnaires, two thousand and fifty-two questionnaires were available. The effective rate was 85.5%. The total two-week prevalence in male was 6.5%, while that in female was 6.2%. There was no significant difference between male and female (χ(2) = 0.103, P > 0.05). The total ratio of those who visited doctors was 1.7%. The ratio of those who visited doctors in the first two weeks was 26.7% (35/131). 49.0% of non-users of health service thought it was unnecessary to consult a doctor because of mild symptoms. In the investigation about the allergic rhinitis, 60.2% patients (1235/2052) confessed that they heard of allergic rhinitis, 30.0% patients (616/2052) thought of allergic rhinitis was common cold.</p><p><b>CONCLUSIONS</b>The number of patients treated for allergic rhinitis was less than its prevalence. The primary reason for low attendance was that the majority of patients with mild symptoms thought it was no need to see a doctor. In order to improve the residents' health services utilization, medical education for allergic rhinitis is necessary.</p>

ABSTRACT

Background Researches demonstrated that avastin-assisted vitrectomy for serious proliferative diabetic retinopathy can decrease intra-operation complication and bring down difficulty of surgery.It is speculated that this is associated with suppression of avastin on neovascularization.However,the evidence of its pathology is lack.Objective The aim of this study was to evaluate the application and mechanism of avastin in vitrectomy for severe proliferative diabetic retinopathy. Methods Twenty-four eyes from consecutive 24 patients with vitrectomy for severe proliferative diabetic retinopathy were enrolled in this study.Fourteen eyes received all intravitreal injection of 0.06 ml avastin(1.5 mg)14 days prior to vitrectomy。And the other 10 eyes underwent only vitrectomy without avastin injection as controls,Preretinal membranes were collected during vitrectomy for histopathologic examination by hemotoxylin and eosin staining.The differences in the density of the neovessels and micro-neovessels between the two groups were observed by detecting the expression of CD34 in vesse]endothelial cells using immunohistochemistry.Written informed consent was obtained from each patient before surgery. Results No statistically significant differences were found in the demography and eye manifestations between only vitreetomy group and avastin+vitrectomy group(P>0.05).The neovessels with grade three was in 10 eyes in only vitrectomy group and 1 eye in avastin+vitrectomy group(P<0.01).More capillary-like neovascularization with single vascular endothelial cells,obvious hemorrhage and inflammatory cells infiltration were observed on preretinal membranes in vitrectomy group.However,there were less hemorrhage,ceUular components and few capillary-like neovascularization but more hyaline degeneration of fibrous tissue were observed in avastin+vitrectomy group under the light microscope.Immunochemistry revealed that CD34 was positively expressed in vascular endothelial cells on preretinal membrane in both two groups.The neovessels density and miero-neovessels density were 15.40±7.42/field and 1.88±1.70/field in avastin+vitrectomy group and those in vitreetomy group were 18.00±3.80/field and 0.45±0.56/field respectively,showing significant differences between these two groups(neovessels density:Z=-4.102,P<0.01;micro-neovessels density:Z=-4.137,P<0.01).Conclusion As an adjunct drug during the vitrectomy for proliferative diabetic retinopathy, avastin can improve the successful rate of surgery by inhibiting the neovascular formation and alleviating retinal edema.

ABSTRACT

<p><b>BACKGROUND</b>Neovascularization can cause vision loss in proliferative diabetic retinopathy (PDR) and may be affected by many factors. Stromal cell-derived factor-1 (SDF-1) is a potent stimulator of angiogenesis. The study was aimed to investigate the expression of SDF-1 and its correlation with vascular endothelial growth factor (VEGF) in the eyes with diabetic retinopathy.</p><p><b>METHODS</b>The levels of SDF-1 and VEGF were measured by enzyme-linked immunosorbent assay in the vitreous of 41 eyes of 41 patients with PDR and 12 eyes of 12 patients with idiopathic macular hole (IMH). Vitreous fluid samples and fibrovascular preretinal membranes were obtained at vitrectomy. SDF-1 and VEGF were localized using immunohistochemistry.</p><p><b>RESULTS</b>The vitreous concentration of VEGF was significantly higher in eyes with PDR ((2143.7 +/- 1685.21) pg/ml) than in eyes with IMH ((142.42 +/- 72.83) pg/ml, P < 0.001). The vitreous level of SDF-1 was also significantly higher in eyes with PDR ((306.37 +/- 134.25) pg/ml) than in eyes with IMH ((86.91 +/- 55.05) pg/ml, P < 0.001). The concentrations of both VEGF and SDF-1 were higher in eyes with active PDR than in eyes with inactive PDR. Panretinal photocoagulation (PRP) could decrease the SDF-1 levels in the vitreous of PDR patients. The vitreous concentration of SDF-1 correlated with that of VEGF in eyes with PDR (r = 0.61, P < 0.001). The costaining of SDF-1 and VEGF was confined to the vascular components in preretinal membranes.</p><p><b>CONCLUSIONS</b>SDF-1 protein is highly expressed in both the vitreous and preretinal membranes of PDR patients; SDF-1 may be correlated with VEGF in angiogenesis in PDR.</p>

ABSTRACT

Endometriosis (EM) is one of the common and frequently encountered gynecological diseases that seriously influences women's health. Its morbidity reaches 10%-15% in women at reproductive ages, and shows an evident rising tendency. In recent years, the Chinese medicine treatment of EM has won favorable therapeutic effects with few adverse reactions. A brief review on this topic has been made through analyzing and summarizing recent pertinent literatures in terms of treatment depending on syndrome differentiation, cycle treatment, external treatment, integrative medicinal treatment, so as to try to know the status quo of Chinese medicine treatment on EM, and to provide some instructive views for clinical treatment and research.

ABSTRACT

<p><b>AIM</b>To determine whether the cardioprotection of ischemic postconditioning and heptanol in ischemic heart against ischemia/reperfusion (I/R) is mediated by gap junction.</p><p><b>METHODS</b>The effect of ischemic postconditioning, heptanol at different doses (0.03, 0.06, 0.30, and 0.60 mg/kg) and AAP10 (10 mg/kg) on the intact rat heart during 30 min ischemia and 2 h of reperfusion was observed. Ischemic postconditioning was achieved by 3 cycles of 10 s reperfusion/10 s regional ischemia starting at the beginning of the reperfusion. The infarct size and the arrhythmia scores were measured. The effect of ischemic postconditioning, heptanol at different doses (0.05, 0.10, 0.50 and 1.00 mmol/L) and AAP10 (1 x 10(-7)mol/L) on the isolated heart during 30 min ischemia and 2 h of reperfusion was observed. Ischemic postconditioning was achieved by 6 cycles of 10 s reperfusion/10 s global ischemia starting at the beginning of the reperfusion. The arrhythmia scores and conduction velocity of ventricle muscle were measured.</p><p><b>RESULTS</b>In the intact rat heart model, ischemic postconditioning and heptanol reduced infarct size and arrhythmia scores. In the Langendorff perfused rat heart model, ischemic postconditioning and heptanol reduced arrhythmia scores and conduction velocity of ventricle muscle. Administration of AAP10, an opener of gap junction attenuated the cardioprotection of ischemic postconditioning and heptanol.</p><p><b>CONCLUSION</b>The cardioprotection of ischemic postconditioning and heptanol may be related to the attenuation of gap junction communication on myocardiac ischemia/reperfusion injury.</p>

ABSTRACT

Objective: To investigate the laboratory,clinical and economic results of early postoperative enteral nutrition(EEN) in patients with gastrointestinal cancer.Methods: 120 patients with gastrointestinal cancer were randomly divided into three groups: EEN,early postoperative parenteral nutrition(PN), and traditional nutrition(CON)groups.The 1/3 and 2/3 target volumes were administered on the first and second postoperative days(1POD,2POD),and the full volume was given from 3POD to 8POD in EEN and PN groups.The routine formula was offerd from 1POD to 8POD in CON group.The nutritional and immune parameters were detected one day before surgery and 9POD, and the clinical and economic results were collected in three groups.Results: The levels of ALB,TF,PA,CD3,CD4,CD4/CD8 were significantly higher in EEN and PN groups than those in CON group(both P<0.01),and the CD3,CD4,CD4/CD8 were better in EEN group than those in PN group(P<0.05).The side effects of EEN group were similar to those of PN group(P>0.05). The recovery of bowel function was quicker in EEN group than that in PN and CON groups(both P<0.05).The complication rate, cost of nutrition support,and cost of complication treatment were lower in EEN group than those in PN group(P<0.05, P<0.01, P<0.01).Although the cost of nutrition support was lowest but the complication rate and cost of complication treatment were all highest in CON group.Conclusion: EEN is better in immune function, recovery of bowel function,complication rate, and cost-effectiveness than those in PN, and from our multiple angular viewpoints EEN is the first choice for patients with gastrointestinal cancer.

ABSTRACT

<p><b>BACKGROUND</b>Neural apoptosis is generally believed to be mediated by two distinct pathways, caspase-dependant and caspase-independent pathways. This study investigated the apoptotic pathways involved in retinal ganglion cells in acute diabetes in rats.</p><p><b>METHODS</b>Diabetes was induced in male Wistar rats by a peritoneal injection of streptozotocin (STZ). Expression and localization of caspase-3 and apoptosis-inducing factor (AIF) proteins in the retina of diabetic rats was examined by Western blotting and immunohistochemistry analyses. Terminal transferase dUTP nick end labeling (TUNEL) assay and immunofluorescent staining specific for caspase-3 and AIF were applied to analyze for apoptosis of retinal ganglion cells. In addition, a caspase-3 inhibitor DEVD-CHO was injected intravitreally to further determine the apoptotic pathways of retinal ganglion cells triggered in acute diabetes.</p><p><b>RESULTS</b>Two weeks after induction of diabetes, a significant increase in caspase-3 protein expression and localization occurred in the nerve fiber layer, ganglion cell layer, and inner plexiform layer of the retina. Four weeks after the onset of diabetes, the increase in caspase-3 expression was profound eight weeks postinduction of diabetes (P < 0.05). Meanwhile, no AIF protein expression was detected in this study. In addition, intravitreal administration of the caspase-3 inhibitor DEVD-CHO reduced apoptosis of retinal ganglion cells by its direct inhibitory action on caspase-3.</p><p><b>CONCLUSION</b>Caspase-dependent apoptotic pathways may be the main stimulant of STZ-induced retinal ganglion cell apoptosis in acute diabetes.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To discuss the value of pre-operative semen analysis of patients with varicocele as a predictive restore index of sperm motility and fertilizing capacity after varicocelectomy.</p><p><b>METHODS</b>Semen analysis was carried out with computer-aided sperm analyzer in 107 patients with varicocele and all patients were referred to the clinic with diagnosis of male infertility. Stratification of patients as group A (n = 32), B ( n = 36) and C (n = 39) was based on pre-operative total motile sperm count (TMSC). Follow-up included semen analysis and pregnancy data after three months following left or bilateral varicocelectomy.</p><p><b>RESULTS</b>The average post-operative TMSC increased significantly when compared with the pre-operative. However, a mean absolute increase in group A and B was better than that in group C (P < 0.05). Of the 68 patients in groups A and B based on pre-operative TMSC, 56 patients' TMSC (82.4%) was > or =20 x 10(6) after varicocelectomy, and that of only 8 (20.5%) patients in group C was > or =20 x 10(6) following varicocelectomy. Of the 98 patients wives, 36 had natural conception. Pregnancy rates in groups A and B were higher than that in group C (P < 0.05).</p><p><b>CONCLUSION</b>Varicocelectomy may be the most effective method to patients with varicocele with pre-operative TMSC > or = 5 x 10(6), but it may be not the best method for patients with severe oligoasthenospermia (pre-operative TMSC < 5 x 10(6)).</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effects of Bisphenol A in adult rats and its possible mechanisms.</p><p><b>METHODS</b>BPA (in corn oil) was administered orally to 9-week-old male Sprague-Dawley rats for 14 days (0, 1 and 5 g/kg bw), and incubated primary Sertoli cells from pubertal SD rats with 0, 10(-7), 10(-6), 10(-5), 10(-4) mol/L BPA.</p><p><b>RESULTS</b>After oral administration, a significant decrease in right testis weight was observed in 5 g/kg dose group, but not in the 1 g/kg bw dose group. Germ cells were detached from basement membrane of seminiferous tubules and Sertoli cells in BPA-treated groups. Administration of BPA at 1 g/kg bw and 5 g/kg bw produced both nucleus pycnosis and vacuolized nucleus in germ cells and Sertoli cells. A marked loss in vimentin staining in Sertoli cells from testis of BPA-treated rats was detected. No change in levels of serum estradiol and testosterone was observed after two-week exposure to BPA. In Sertoli cell primary culture, BPA destroyed the cytoskeleton and cell-cell junctions, and elongated Sertoli cells.</p><p><b>CONCLUSION</b>These results suggest that BPA may injure reproductive function of male rats by destroying the cytoskeleton and changing the form of Sertoli cells.</p>

ABSTRACT

Objective To explore the mechanism of effect of estrogic bisphenol_A(BPA) on vimentin in Sertoli cells.Methods Immunohistochemical methods and situ hybridization technique were applied to analyze the effets of BPA on expressions of vimentin and protein kinase C(PKC) gene in primary cultured Sertoli cells.Results The results showed that BPA lengthened the Sertoli cells,the vimentin gene in Sertoli cells could be expressed completely in vitro,but BPA could prohibited translation of vimentin gene,however there was no direct relationship between BPA and vimentin protein degradation.PKC gene couldn't be expressed in Sertoli cells. Conclusion The results suggested that BPA prohibited translation of vimentin gene and therefore blocked the formation of cytoskeleton of Sertoli cells,and finally caused deformation of these cells,and that protein kinase C didn't disturb degradative metabolism of vimentin in Sertoli cells.
