ABSTRACT

Six compounds were isolated from the roots of Ephedra sinica Stapf using various chromatographic techniques such as silica gel column chromatography, thin layer chromatography and semi-preparative HPLC. Their chemical structures were identified by analysis of physicochemical properties and spectral data, and determined as (Z)-docosanylferulate (1), (E)-docosanylferulate (2), bis (2-ethylheptyl) phythalate (3), 2,2′-oxybis (1,4-di-tert-butylbenzene) (4), diisobutyl phthalate (5), bis (2-ethylhexyl) phthalate (6). Among them, compound 1 is a new compound, compounds 2-4 were first isolated from Ephedra. A corticosterone-induced PC-12 cell injury model was used for compound activity screening. The results showed that compounds 1 and 5 significantly improved corticosterone-induced PC-12 cell injury and significantly increased 5-HT7 receptor protein expression in the cells, indicating potential antidepressant activity.

ABSTRACT

ObjectiveTo describe the epidemiological characteristics of confirmed COVID-19 cases in Shangrao City, and provide scientific evidence for the development of prevention and control strategies. MethodsEpidemiological characteristics of 123 confirmed cases, such as time, sex, age, occupations, area, and source of infection, were analyzed by combining epidemiological investigation with laboratory testing. ResultsOf the 123 confirmed cases, 34 were imported cases and 89 were local cases. The sex ratio was 1.32∶1. Average time interval of the COVID-19 was (18.4±4.4) days, with a median time interval between the onset and final diagnosis of 5 days. There were 19 clustering cases, of which 89.5% were family clusters and 77.3% occurred in families. Majority of the cases were common type. The main source of infection was travelers from the epidemic areas. ConclusionHealth monitoring of travelers from epidemic areas and strict quarantine of close contacts are the main measures to control the spread of the COVID-19 epidemic.

ABSTRACT

The purpose of the study was to investigate the effect of sophoridine on the proliferation and apoptosis of human gastric cancer MKN45 cells and the possible mechanism. MKN45 cells were randomly divided into control and sophoridine (including 6 subgroups) groups. Cell proliferation was detected by methyl thiazolyl tetrazolium (MTT) colorimetric method. The protein expression of high mobility group-box 3 (HMGB3) was observed by immunocytochemical staining and Western blot. Hoechst 33342 staining method was used to observe the morphological changes of cells treated with sophoridine. Apoptosis was detected by flow cytometry. The results showed that the proliferation of cells was inhibited by 48-hour treatment of sophoridine in a dose-dependent manner. Compared with control group, sophoridine group showed decreased HMGB3 protein expression and increased apoptotic rate. These results suggest that sophoridine can inhibit the proliferation of MKN45 cells and promote their apoptosis, which may be related to down-regulation of HMGB3 protein expression.

ABSTRACT

The chemical constituents of Dioscorea opposite Thunb. were isolated and purified by Diaion HP-20, Sephadex LH-20, Toyopearl HW-40, MCI Gel CHP-20, ODS, silica gel column chromatography and semi-preparative-HPLC. Nine compounds were found and their structures were elucidated by spectral data and physicochemical properties, which were identified as 2-(1',2',3'-trihydroxybutyl-4'-O-α-D-glucopyranoside)-6-(2",3",4"-trihydroxybutyl)-pyrazine (1), uracil (2), xanthine (3), hypoxanthine (4), thymine (5), adenosine (6), uridine (7), inosine (8), and 5'-deoxy-5'-methylsulphinyladenosine (9). The compound 1 is a new pyrazine derivative, and the compound 3-5, 8, 9 are found in this plant for the first time.

ABSTRACT

The chemical constituents of the seeds of Lepidium apetalum Willd. were investigated using chromatographic methods, including Diaion HP-20, Toyopearl HW-40, MCI Gel CHP-20, ODS, silica gel chromatography and semi-preparative-HPLC. Three compounds were isolated and their structures were elucidated by spectral data and physicochemical properties, which were identified as lepidiumamide A (1), cis-desulfoglucotropaeolin (2), trans-desulfoglucotropaeolin (3). Among those, compound 1 is a new phenylacetamide, compound 2 and 3 were isolated from this plant for the first time, and their configurations were also identified for the first time.

AIM@#To investigate the chemical constituents of Selaginella sinensis (Desv.) Spring.@*METHODS@#Chromatographic separations on Diaion HP-20, silica gel, and Sephadex LH-20 were used. The structures of the isolates were elucidated on the basis of spectroscopic analysis, as well as chemical methods.@*RESULTS@#Eight compounds were obtained and their structures were identified as sinensioside A (1), syringaresinol-4- O-β-D-glucopyranoside (2), (+)-medioresinol-4-O-β-D-glucopyranoside (3), pinoresinol-4, 4'-di-O-β-D-glucopyranoside (4), quercetin (5), eucomic acid (6), shikimic acid (7), and 2, 3-dihydroamentoflavone (8).@*CONCLUSION@#Compound 1 is a new dihydrobenzofuran sesquilignan glycoside from Selaginella sinensis.

ABSTRACT

The osmotic pressure of ammonium sulfate solutions has been measured by the well-established freezing point osmometry in dilute solutions and we recently reported air humidity osmometry in a much wider range of concentration. Air humidity osmometry cross-validated the theoretical calculations of osmotic pressure based on the Pitzer model at high concentrations by two one-sided test (TOST) of equivalence with multiple testing corrections, where no other experimental method could serve as a reference for comparison. Although more strict equivalence criteria were established between the measurements of freezing point osmometry and the calculations based on the Pitzer model at low concentration, air humidity osmometry is the only currently available osmometry applicable to high concentration, serves as an economic addition to standard osmometry.

ABSTRACT

This study is to observe the protection effect of amentoflavone (AMT) in Selaginella tamariscina against TNF-alpha-induced vascular inflammation injury of endothelial cells. On the basis of TNF-alpha induced human umbilical vein endothelial cell, observe the influence of AMT on endothelial active factor, the contents of SOD and MDA, the protein expression of vascular endothelial adhesion molecules and inflammatory factor; study the effect of its common related signal pathways such as NF-kappaB; research the effect of AMT against TNF-a induced human umbilical vein endothelial cell injury by means of MTT, ELISA, Western blotting and the cell immunofluorescence. The results showed that AMT could increase the content of NO and decrease the levels of VCAM-1, E-selectin, IL-6, IL-8 and ET-1; enhance the activity of SOD, reduce the content of MDA; downregulate the protein expressions of VCAM-1, E-selectin, NF-kappaBp65 and up-regulate IkappaBalpha, attenuate the NF-kappaBp65 transfer to cell nucleus. AMT has the effect of protect vascular endothelial and maybe via the signal pathway of NF-kappaB to down-regulate the inflammation factor and oxidative damage factor of downstream.

ABSTRACT

<p><b>OBJECTIVE</b>To study the chemical constituents in the aerial part of Acalypha australis.</p><p><b>METHOD</b>The aerial part of Acalypha australis were extracted with EtOH at room temperature. The compounds were separated by various kinds of chromatographic methods and their structures were identified on the basis of spectroscopic techniques.</p><p><b>RESULT</b>Eleven compounds have been isolated and identified: emodin (1), beta-sitosterol (2), loliolide (3), 2,6-dimethoxy-1,4-benzoquinone (4), nicotinic acid (5), protocatechuic acid (6), daucosterol (7), gallic acid (8), rutin (9), succinic acid (10), brevifolin (11).</p><p><b>CONCLUSION</b>Compounds 1, 3-5, 10 and 11 were obtained from A. australis for the first time.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study genotype distribution and the characteristics of hepatitis B virus (HBV) in Uighur patients with chronic hepatitis B (CHB) in Xinjiang, China.</p><p><b>METHODS</b>Type specific primers and PCR were used to detect the HBV genotypes of 127 Uighur CHB patients in Xinjiang. Genotyping results were confirmed by PCR product sequencing.</p><p><b>RESULTS</b>Among the 127 patients, the proportions of genotype D, B, C and B/D, C/D, B/C/D were 39.4% (50/127), 22.0% (28/127), 16.5% (21/127) and 9.4% (12/127), 8.7% (11/127) and 3.9% (5/127), respectively. The distribution of the HBV genotypes showed no significant differences between male and female patients (x2 = 8.058, P > 0.05), between HBeAg positive and negative patients (x2 = 6.033, P > 0.05), and between patients of different ages (x2 = 3.137, P > 0.05).</p><p><b>CONCLUSION</b>Genotype D HBV is predominant in Uighur patients with chronic hepatitis B in Xinjiang. The distribution of various HBV genotypes shows no significant differences between these Uighur patients with different HBeAg positivity, sex and age.</p>

ABSTRACT

<p><b>AIM</b>To study the effects of ferulic acid (FA) on E-selectin expression in human umbilical vein endothelial cells (HUVECs) activated by lipopolysaccharide and leukocyte-endothelial cell adhesion.</p><p><b>METHODS</b>The effects of FA on E-selectin and E-selectin mRNA expression were determined by flow cytometry and reverse transcription polymerase chain reaction. The effect of FA on HL60-HUVEC adhesion was evaluated with the method of staining the cells by Rose Bengal.</p><p><b>RESULTS</b>The expression of E-selectin and E-selectin mRNA were down regulated by FA (0.62 and 0.41 mmol x L(-1), respectively). HL60 cells adhered to activated HUVECs were also reduced by FA (0.62 and 0.41 mmol x L(-1), respectively).</p><p><b>CONCLUSION</b>FA can inhibit the expression of E-selectin and E-selectin mRNA and HL60-HUVEC adhesion. This may contribute to its protective effect against ischemia-reperfusion injury.</p>

ABSTRACT

Objective To study the prevalence of transfusion transmitted virus (TTV) in normal children and hepatitis B virus (HBV) carrying children. Methods Anested polymerase chain reaction(nPCR) assay was established using two - set of primers designed from the first open read frame of TTV genome. Serum from the children who received physical examination was tested by nPCR assay for the presence of TTV- DNA. Results The total detectable rate of TTV - DNA in those 230 objects was 17.4% , in which the positive rate of TTV - DNA was 13.7% in normal children and was 25.7% in children carrying HBV. There was significant difference in the positive rate of TTV - DNA between normal children and the children carrying HBV. Conclusion There is TTV infection in normal children,and there is higher positive rate of TTV-DNA in children carrying HBV in Xinjiang.
