ABSTRACT

Objective To study the protective effect of Shengxian decoction and the single herb decoction against myocardial injury induced by hypoxia/reoxygenation. Methods The H9c2 cells were cultured to establish hypoxia/reoxygenation model. Rats were divided into 8 groups: normal control group, hypoxia/reoxygenation group (model group) and treated groups (Shengxian decoction and the single herb decoction). The apoptotic rate of cardiomyocytes, the activity of reactive oxygen species (ROS) and intracellular calcium concentration (Ca2+) were measured. Results Compared with hypoxia/reoxygenation group, the apoptosis rate, ROS activity and intracellular Ca2+ concentration were significantly lower in all treated groups (P<0.05). The ROS activity and intracellular Ca2+ concentration was decreased by 41.37% and 15.20% in Shengxian decoction group compared to the model group. Conclusion Shengxian decoction and the single herb decoction had protective effect on myocardial injury induced by hypoxia/reoxygenation.

ABSTRACT

Objective To analyze the clinical key points of minimally invasive tracheostomy with percutaneous horn expansion applicated in ICU,and to discuss the problems encountered in minimally invasive surgery and the solutions.Methods The clinical data of 25 patients of minimally invasive tracheostomy with percutaneous horn expansion in ICU were analyzed retrospectively.Results Twenty-one patients with minimally invasive tracheostomy with percutaneous horn expansion were successfully performed in 25 patients of ICU (no incision sputum,subcutaneous emphysema,pneumothorax and other complications),2 patients had more postoperative bleeding,2 patients were difficult to imbedding catheter,of which 1 case abandoned the catheter and received conventional tracheotomy.Conclusion Percutaneous horn expansion minimally invasive tracheostomy has the advantages of small incision,less injury,less operative time,less blood loss and low incidence of complications.However,ICU patients are in severe condition and easy to change.Adequate preoperative preparation,controlling the key steps in the operation can find a solution to the problme.

ABSTRACT

Objective To introduce the structure of blood-brain barrier(BBB)and drug evaluation models,and provide ba?sic theory and experimental techniques for the evaluation of novel central nervous system(CNS)drugs. Methods The literature in re?cent 10 years on the BBB definition,structural characteristics and drug screening BBB models in vivo and in vitro were reviewed. Re?sults The composition and structural characteristics of the BBB were described,and the advantages and disadvantages of each one of these models were evaluated. Conclusion So far,there is nogold standardmodel for estimating the permeability of a compound on BBB,the optimized cell models should be combined with the BBB permeability models in vivo to predict the extent of BBB permeabili?ty of a compound.

ABSTRACT

In vitro transcription systems with T7 RNA polymerase (T7 RNAP) were widely used in preparation of RNA because of their simplicity and high efficiency. The transcripts would have additional 5' sequence since T7 promoter spans the transcription start site, while deletion of the transcription start site would severely reduce the T7 RNAP transcriptional activity. We successfully developed an in vitro transcription by combining of T7 RNAP high efficient transcription system and highly specific self-splicing technology of ribozymes, in this system, ribozyme self-splices at the designed specific site and releases the aim RNA without affecting transcription efficiency of T7 RNAP, the aminoacylation activity of human mitochondrial tRNA(Trp) (HmtRNA(Trp) (UCA)) is 113.6 pmol/microg. This method with its high efficiency on transcription and good repeatability is very suitable for preparation of accurate RNA in large scale.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the gene expression of 4-1BB in peripheral blood mononuclear cells (PBMCs) and the possible significance of the 4-1BB pathway after clinical orthotopic liver transplantation (OLT).</p><p><b>METHODS</b>4-1BB mRNA levels in PBMCs from 22 OLT patients were analyzed by RT-PCR. 4-1BB protein expressed on the surface of CD(4)(+) and CD(8)(+) T cells were detected by flow cytometry, and visualized with direct immunofluorescence and confocal fluorescence microscopy. Patients with primary liver cancer (PLC) and healthy volunteers served as controls. Six cases of recently performed liver transplantation were also observed in this study.</p><p><b>RESULTS</b>4-1BB mRNA was detected in PBMCs from both liver transplant patients with long-term graft acceptance (22 cases) and from transplant patients on day 1 to day 3 post-transplantation (6 cases), but was not found in PBMCs from transplant patients on day 7 to day 30 post-transplantation (6 cases). 4-1BB mRNA was also not found in samples from 8 of the healthy controls and 7 of the PLC patients, though very low expression was detected in the other 4 healthy volunteers and 6 PLC patients. Simultaneously, 4-1BB protein was expressed at nearly undetectable levels on CD(4)(+) and CD(8)(+) T cells from healthy controls, PLC patients, as well as OLT patients within the first month post-transplantation (6 cases). However, 4-1BB expression was found on the surface of CD(4)(+) and CD(8)(+) T cells from liver transplant patients with long-term graft acceptance. Direct immunofluorescent staining and confocal fluorescence microscopy clearly revealed evidence of 4-1BB protein on cell membranes of CD(4)(+) and CD(8)(+) T cells from liver transplant patients with long-term graft acceptance. Simultaneously, a significantly higher percentage of CD(3)(+) CD(25)(+) T cells were found in liver transplant patients with long-term graft acceptance group as compared with the healthy control group (P < 0.05). The expression of 4-1BB protein on T cells did not correlate with the survival time of OLT patients postoperation.</p><p><b>CONCLUSIONS</b>This study demonstrates that although patients remain in stable condition after liver transplantation under the treatment of immunosuppressants, activated T cells are present to some extent and 4-1BB protein may be involved in this process. Effector T-cells can exert permanent immunoresponses against grafts under these circumstances. Therefore, we conclude that a new immune response balance is established under the combination of both treatment with immunosuppressants and natural immune responses against alloantigens. Manipulation of the 4-1BB/4-1BBL pathway may provide a therapeutic technique for prolonging graft survival.</p>

Objective To study the activity of N acetylglucosaminyltransferase Ⅲ, Ⅳ and V in extrahepatic cholangiocarcinoma and its significance.Methods The activities of N acetylglucosaminyltransferase(GnT) Ⅲ, Ⅳ and Ⅴ in extrahepatic bile duct carcinoma (EBDC) of 15 cases were determined by reverse phase HPLC (high performance liquid chromatography), and results were compared with benign biliary diseases in 15 cases. ABSTRACT

AIM: To investigate the transfection efficiency of mouse liver with non-viral vector containing manganese superoxide dismutase (Mn-SOD) gene. METHODS: The eukaryotic expression vector, gWiz/Mn-SOD, encoding human manganese superoxide dismutase was constructed. The plasmids of gWiz/Mn-SOD were mixed with cationic lipids, followed by injection into mice via branch of superior mesenteric vein, to induce Mn-SOD over-expression in murine liver detected by RT-PCR, Western blotting, SOD activity and immunohistochemical staining. RESULTS: gWiz/Mn-SOD transfection resulted in the obvious expression of exogenous Mn-SOD mRNA and protein in hepatic tissues at 8 hours after injection, and elevated mitochondria SOD activity 8.4 times in transfected hepatocytes than that in non-transfected cells at 72 hours after injection. It was showed that nearly 70% of mouse hepatocytes was obviously Mn-SOD positive after transfection. CONCLUSION: High expression efficiency of Mn-SOD gene in mouse liver is achieved safely by injection of gWiz/Mn-SOD and cationic lipid mixture into branch of superior mesenteric vein. [

ABSTRACT

ObjectiveTo explore the clinical significance and characteristics about terminal sialyl residues linked to N-glycans in the tissue and bile glycoproteins from bile duct carcinoma. MethodsThe mRNA expression of sialyltransferases, including ST3Gal-Ⅲ and ST6Gal-Ⅰ, was detected in the tissue extracts of 35 cases with bile duct carcinoma(BDC) and 35 cases of benign biliary disease (BBD) by semiquantitative reverse transcription-polymerase chain reaction(RT-PCR). With the lectin of WGA(Triticum valgaris agglutinin), which can recognise the terminal sialyl residues in N-glycans, all the specimens of BDC and BBD were subject to lectin histochemical staining, followed by computer-aided digital analysis. Lectin dot-blotting with horseradish peroxidase-labeled WGA was performed for the bile samples from each of the 35 cases of BDC and BBD. ABSTRACT

ve To explore the hygienic safty of cosmetics for special use. Methods The detection and analysis on the hygienic safety of 432 domestic cosmetics for special use saled in Guangdong provincial markets were carried out in 2000. Results The orders of qualified rates of cosmetics for special use from high to low were as fol-lows: deodorant cosmetics (100%), hair-removing cosmetics(88.89% ), breast massage (80.00% ), antisunburn lotion (77.88%), fading cream(75.68% ). hair nourishment(64.29% ), beautifying cosmetics(50.00% ), hair dye (40.74%) and hair pem(28.57%). The highest unqualified rate of hygienic safety test on cosmetics was 17.28% for Ames test, next was 14.81% for chromosome aberration test on mammalian cells in vitro, 14.13% for eye irritation test and 10.88% for skin allergic reaction. Conclusion The management on the hygienic safety of cosmetics for special use should be strengthened further.
