ABSTRACT

Objective:To study the effect of Sanjie Xiaoliu Recipe on transplanted tumors in breast cancer mice through in vivo experiments, in order to explore the efficacy and mechanism of Sanjie Xiaoliu Recipe on breast cancer patients. Methods:45 BaL B/c female mice were selected to establish the transplanted tumor model of breast cancer. All the transplanted tumor models of breast cancer mice were randomly divided into five groups: the control group (intragastric administration of normal saline), the low, medium and high dose group of Sanjie Xiaoliu Recipe (intragastric administration of different doses of Sanjie Xiaoliu Decoction), and the paclitaxel group (intraperitoneal injection of paclitaxel). After 24 days of continuous administration, the diet and activities of mice were observed; the body weight, weight and volume changes of transplanted tumor mice were recorded, and the tumor inhibition rate was calculated. Western blot was used to detect the expression of epithelial mesenchymal transformation related proteins (E-cadherin, N-cadherin, Vimentin) in the transplanted tumor tissues of mice in each group.Results:(1) The food intake and activity status of mice treated with Sanjie Xiaoliu Recipe were less affected by the transplanted tumor of breast cancer. (2) The volume and weight of transplanted tumor in the treat groups were smaller than those in the control group (all P<0.01), and the volume of transplanted tumor in the middle dose group of Sanjie Xiaoliu Recipe was smaller than that in the low dose group ( P<0.05). The tumor inhibition rates among the treatment groups were: Sanjie Xiaoliu Recipe medium dose group 52.4%, paclitaxel group 40.3%, Sanjie Xiaoliu Recipe low dose group 39.5%, Sanjie Xiaoliu Recipe high dose group 34.1%. (3) The results of Western blot showed that the expression level of E-cadherin in the transplanted tumor tissue of the treat groups was higher than that in the control group, and the expression levels of N-cadherin and Vimentin were lower than that in the control group, with statistically significant difference (all P<0.05). Conclusions:The Sanjie Xiaoliu Recipe can improve the weakness and reduced consumption of breast cancer mice, which can inhibit the tumor mass growth in mice to a certain extent. Its mechanism may be that Sanjie Xiaoliu Recipe can inhibit the epithelial mesenchymal transformation of breast cancer and the invasion and metastasis of breast cancer.

ABSTRACT

BACKGROUND@#Immune checkpoint inhibitor monotherapy is reported to have little effect in advanced non-small cell lung cancer (NSCLC) patients with driver oncogenes. However, recent studies have shown that some patients with driver genes are still benefit from combination immunotherapy after tyrosine kinase inhibitors (TKIs) drug resistance. The purpose of this study was to analyze the efficacy of posterior line immunotherapy in NSCLC patients with epidermal growth factor (EGFR) sensitive mutation, and to evaluate the value of immunotherapy in posterior line therapy in patients with advanced EGFR mutation.@*METHODS@#A total of 27 patients with EGFR mutation diagnosed in Beijing Chest Hospital, Capital Medical University from June 2018 to November 2020 were collected. After the progress of targeted therapy, they had received programmed cell death protein 1 (PD-1) checkpoint inhibitor combined with chemotherapy and anti-angiogenic drug therapy.@*RESULTS@#Of the 27 advanced NSCLC patients, 19 cases (70.4%) did not have T790M mutation. There were 8 cases (29.6%) with T790M point mutation. The total objective response rate (ORR) was 40.7%. Kaplan-Meier survival analysis showed that there was no statistically significant difference among different EGFR mutations (χ²=4.15, P=0.230). But progression-free survival (PFS) was significantly longer in patients without T790M mutation than in patients with T790M mutation (9.2 mon vs 3.3 mon, χ²=2.808, P=0.041), and the same trend was observed in patients with overall survival treated with the PD-1 inhibitor (12.2 mon vs 7.3 mon, χ²=3.22, P=0.062). ORR of patients without T790M was significantly better than that with T790M (52.63% vs 12.5%, P=0.045).@*CONCLUSIONS@#Patients with EGFR mutation can benefit from later-line combined immunotherapy. The patients with T790M mutation in the population of EGFR mutation had the worst effect of immunotherapy in the later line. Therefore, the follow-up treatment and whole-course management of these patients need to explore better treatment strategies to improve the benefit.

ABSTRACT

BACKGROUND@#Immunotherapy represented by immune checkpoint inhibitors (ICIs) has been widely used in the treatment of lung cancer. There are controversies in clinical practice for patients with advanced non-small cell lung cancer (NSCLC) and high programmed cell death-ligand 1 (PD-L1) expression receiving ICIs monotherapy or combination chemotherapy.@*METHODS@#This study retrospectively analyzed the clinical data of 49 patients with advanced NSCLC and high PD-L1 expression. Immunohistochemistry was performed with 22C3 antibody, and the expression level of PD-L1 was evaluated according to tumor proportion score (TPS). Objective response rate (ORR) and progression free survival (PFS) were compared by groups of different clinical characteristics.@*RESULTS@#ORR of monotherapy and combination therapy group was 47.1% (8/17) and 43.8% (14/32), respectively, without statistical difference (P=0.825). The median PFS of monotherapy and combination therapy group was 8.0 months and 6.8 months, respectively, without statistical difference (P=0.502). Statistical analysis of predictors of immunotherapy for the patients showed first-line immunotherapy had better ORR than subsequent immunotherapy (12/19, 63.2% vs 10/30, 33.3%, P=0.041), however no difference in PFS. And there were no differences in ORR or PFS among groups of age, gender, smoking status, performance status (PS), pathological type, tumor size and tumor-node-metastasis (TNM) stage.@*CONCLUSIONS@#The therapeutic effect is similar between ICIs monotherapy and combination chemotherapy for patients with advanced NSCLC and high PD-L1 expression. ORR of first-line immunotherapy was better in patients with advanced NSCLC and high PD-L1 expression. The optimal treatment for this population remains further prospective clinical studies.

ABSTRACT

BACKGROUND@#Targeted therapy for patients with driver genes positive and immunotherapy for patients with driver gene-negative but high programmed death ligand 1 (PD-L1) expression are the standards of first-line treatment for patients with advanced non-small cell lung cancer (NSCLC). The treatment options for patients with driver gene positive and high PD-L1 expression are still worth exploring.@*METHODS@#The characteristics of 315 patients with NSCLC were identified to analyze the clinicopathological characteristics of patients with driver gene positive and high PD-L1 expression, and the efficacy of targeted therapy.@*RESULTS@#Among the 315 patients, the total positive rate of driver genes was 62.2%, and the high PD-L1 expression rate (≥50.0%) was 11.2%. The proportion of patients with driver gene positive and high PD-L1 expression was 10.7%. PD-L1 was highly expressed in patients with epidermal growth factor receptor (EGFR) mutation, KRAS mutation, ALK fusion, BRAF mutation, and MET 14 exon skip mutation, the proportions were 7.8% (11/141), 18.2% (4/22), and 23.1%, (3/13), 50.0% (2/4) and 100.0% (1/1) respectively. EGFR mutation positive with PD-L1 high expression was mainly in patients with stage IV lung adenocarcinoma. KRAS mutation positive with PD-L1 high expression was mainly in patients with a history of smoking. Among them, two patients were followed in detail for targeted therapy, who with ALK fusion-positive and PD-L1 high expression (90.0%), EGFR L858R mutation and PD-L1 high expression (70.0%) respectively. The total OS of the patients was 5 months, 2 months.@*CONCLUSIONS@#The high PD-L1 expression rate in NSCLC patients with different driver gene mutations was variable, which maybe correlated with distinct clinicopathological characteristics. Patients with sensitive mutations and high PD-L1 expression may be less benefit from targeted therapy and have poor prognosis.

ABSTRACT

Objective To compare effect of swaddle bathing and tub bathing on neonatal behavior state, and improve the safety and comfort of newborn infants. Methods This was a prospective non-randomized control study. From February 2016 to September 2017, a total of 110 neonates were divided into swaddle bathing group or tub bathing group, each group including 55 cases. Neonates in swaddle bathing group received swaddle bathing after discharge, while neonates in tub bathing group received tub bathing. Behavior state in the process of bathing and temperature before and after bathing were compared. Results In swaddle bathing group, 85.5% (47/55) of neonates kept quiet during the bathing process, which was significantly higher than 56.4%(31/55) in the tub bathing group (χ2=11.282, P<0.01). 30.9%(17/55) of neonates in swaddle group were crying, significantly lower than 50.9% (28/55) in tub bathing group (χ2=4.550, P<0.05). Duration of neonatal grasp reflex, crying time (seconds), and neonatal body temperature after bathing was (2.3±0.4) points,2(0,7)s and (36.49±0.02)℃in swaddle bathing group, and (7.9±0.5) points, 6 (5,10) s and (36.38±0.03)℃in tub bathing group, and the difference was significant(t=4.366, 2.910, Z=3.534, P<0.01). Conclusions Swaddle bathing can promote neonatal behavior state in the process of bathing and reduce neonatal the feeling of fear. Moreover, it could help keep warm more effectively.

ABSTRACT

Objective To explore the influential factors of dangerous placenta previa pathogenesis,and to analyze their peri natal period outcomes.Methods The clinical data in 143 pregnant women and parturients of dangerous placenta previa(research group) and 286 pregnant women and parturients of non-dangerous placenta previa(control group) in our hospital from January 2011 to December 2015 were retrospectively analyzed.Results The average age of the research group was higher than that of the control group (t =2.92,P =0.004);the previous pregnancy frequency,previous parity,previous abortion frequency,previous cesarean delivery frequency and incidence of placenta previa in the research group were higher than those in the control group(P<0.01);the incidence of placenta previa in the research group was higher than that in the control group(x2 =7.60,P=0.006);the prenatal gestational weeks of the research group were lower than those of the control group(t=-12.33,P<0.01).The multi-factor analysis showed that the age,previous abortion frequency and previous cesarean delivery frequency were the influence factors of placenta previa occurrence.Among them,the previous cesarean delivery frequency was the main risk factor.The occurrence rates of antepartum hemorrhage,postpartum hemorrhage,uterine removal and premature infants in the research group were higher than those in the control group(P<0.01);The intraoperative blood loss of the research group was higher than that of the control group(P<0.05);the neonatal birth weight and the 1 min Apgar score of the research group were lower than those in the control group(P<0.05).Conclusion The age,previous abortion frequency and previous cesarean section frequency are related to the occurrence of dangerous placenta previa.

ABSTRACT

Objective To test the inhabiting effect of Lactobacillus acidophilus on E.coli O157: H7 in intestinal colonization and explore its mechanism. Methods The suppressive effects of Lactobacillus acidophilus against E.coli O157:H7 adhering to Ht29 cells were carried out by competition , exclusion and replacement as-says. Furthermore, we evaluated the cytokine levels of IL-4, IL-12, and INF-γ in serum of mice. In addition, E.coli O157:H7 fecal shedding was monitored and the pathological changes of intestines were observed in mice. Results The competition, exclusion and replacement assays showed Lactobacillus acidophilus inhibited E.coli O157:H7 adhering to Ht29 cells. In vivo, the mice of treatment group were induced significantly higher level of IL-4, IL-12, and INF-γ, though prevention group induced IL-12 only. Fifteen days after E.coli O157:H7 infec-tion, there were 8 mice (80%) in prevention group and 5 mice (50%) in treatment group stopped shedding. Moreover, the pathological changes of intestines of both prevention group and treatment group appeared normal , but control groups showed seriously damaged in intestinal villus. Conclusion Lactobacillus acidophilus inhibits E.coli O157:H7 in intestinal colonization and the preventative effect was better than treatment effect. Thus , Lac-tobacillus acidophilus can be used for E.coli O157:H7 in prevention and treatment infection as probiotics.

Objective Single voxel proton spectroscopy technique (1H-MRS)was performed in the healthy newborn brain tissue to investigate the metabolic ratio in the right basal ganglia and the right frontal lobe changes with gestational age, and to improve the understanding of the neonatal brain development. Methods In this study, 69 newborns were selected, including twenty-five premature and forty-four term neonates. By using the Gesell developmental scale to evaluate brain development , they all had normal neurologic outcomes at 12-months corrected age. A point-resolved spectroscopy sequence was used to study the metabolic data obtained from each region of interest. Results N-acetylaspartate: creatine and N-acetylaspartate:choline were positively correlated with gestational age (the right basal ganglia r = 0.27, 0.63, the right frontal lobe r = 0.44, 0.64 respectively, P < 0.05) and choline:creatine was negative (the right basal ganglia r = -0.51,the right frontal lobe r = -0.41, P < 0.05), with a linear regression relationship among variables. In the same gestational age group , the N-acetylaspartate: choline of the right basal ganglia was higher than that of the right frontal lobe , and the metabolic ratios among the anatomic locations were significantly different (P < 0.05). Conclusion Neonatal brain metabolite levels are associated with gestational age , and MRS can monitor newborn brain maturation quantitatively.

ABSTRACT

Objective To explore the correlation between apparent diffusion coefficient(ADC)and neonatal behavioral neurological assessment(NBNA) of newborns with hypoxic ischemic encephalopathy (HIE),and to evaluate ADC in the early diagnosis of HIE from the imaging perspective. Method One hundred and nine neonates aged 0—7 days with HIE underwent conventional MRI and DWI. According to HIE grading standards, there were 43 neonates in the mild group, 38 in the moderate group, 28 in the severe group, and meanwhile 24 normal neonates with the same ages were selected as the control group. All cases were assessed with NBNA, and ADC values of bilateral frontal white matter, parietal white matter, periventricular white matter, posterior limb of the internal capsule(PLIC), ventral lateral nucleus of thalamus, lenticular nucleus, splenium of the corpus callosum, brainstem were measured. ADC values of different groups were compared by analysis of variance, and the correlations between ADC values of all ROIs and NBNA were analyzed by Pearson correlation. Results Except lenticular nucleus and the brainstem, ADC values of frontal white matter, parietal white matter, periventricular white matter and PLIC, ventral lateral nucleus of thalamus, splenium of the corpus callosum were decreased in the mild, moderate, severe group. In the mild, moderate, and severe group, the ADC values of frontal deep white matter were(1.82± 0.33)× 10-3,(1.77 ± 0.34)× 10-3 and(1.62 ± 0.31)× 10-3 mm2/s,while they were(1.81 ± 0.34)× 10-3,(1.79 ± 0.27)× 10-3 and(1.72 ± 0.31)× 10-3 mm2/s for the parietal deep white matter,(1.27 ± 0.15)× 10-3,(1.23 ± 0.12)× 10-3 and(1.15 ± 0.17)× 10-3 mm2/s for the periventricular white matter,(1.08 ± 0.09)× 10-3,(0.97 ± 0.07)×10-3 and(0.84±0.06)×10-3 mm2/s for the PLIC,(1.13±0.07)×10-3 ,(1.08±0.13)×10-3 and(1.00± 0.13)× 10-3 mm2/s for the ventral lateral nucleus of thalamus,(1.27 ± 0.22)× 10-3,(1.18 ± 0.16)× 10-3 and (1.00 ± 0.23)× 10-3 mm2/s for the splenium of the corpus callosum. There were statistically significant differences between the 3 groups (F=61.27,16.27, 23.26, 72.70, 26.73, 66.09,all P<0.05). In the mild, modreate and severe group, NBNA were(36.8 ± 1.4) in the mild group,(33.5 ± 1.6) in the moderate,and (29.3 ± 2.6) in the severe group. There was positive correlation between ADC values of frontal white matter, parietal white matter, periventricular white matter and PLIC, ventral lateral nucleus of thalamus, splenium of the corpus callosum and NBNA scores(r=0.60,0.49,0.54,0.67,0.56,0.65,all P<0.05). Conclusions There are correlations between ADC values of the related ROIs of HIE and NBNA scores. Combining two aspects might diagnose the brain injury of HIE more accurately and objectively.

ABSTRACT

Objective To investigate the killing effect of allitride on fungus inside Candida albicans biofilm (BF) .Methods The polyascchride components of BF was labeled by FITC-conA .The fluorescence microscopic was adopted to observe the effect of al-litride on the BF forming process of Candida albicans in morphology .The specimens were divided into 3 groups ,the control group (allicin concentration of 0) ,0 .25 mg/mL allicin group ,0 .5 mg/mL allicin group ,1 .0 mg/mL allicin group ,2 .0 mg/mL allicin group .The XTT reduction assay was performed to quantitatively detect the killing effect of allitride on fungus inside Candida albi-cans BF .Results After the treatment by different concentrations of allitride ,the BF structure was observed by fluorescence micros-copy .The fluorescence intensity of high concentration group was weakened compared with the low concentration group and the con-trol group .The ODA450 values of the XTT reduction assay after acting on Candida albicans BF for 5 h by different concentrations of allitride (0 .5 ,1 .0 ,2 .0 mg/mL) were 1 .87 ± 0 .31 ,1 .64 ± 0 .25 and 1 .30 ± 0 .29 ,respectively ,and the OD450 value of the control group was 2 .11 ± 0 .26 ,there was statistically significant difference between the allitride groups and control group (P0 .05) .Conclusion Allitride can kill the fungus inside BF of Candida albicans and has the inhibiting effect on biofilm .Moreover the inhibiting effect on biofilm is increased with its concentration increase within certain concentration range .

ABSTRACT

Objective To study the Doppler ultrasound measurement of fetal middle cerebral artery peak ve -locity in the prediction of fetal thalassemia clinical value .Methods 90 cases of singleton pregnancies who were diag-nosesed for light or thalassemia intermedia patients ,also including her spouse were selected as the observation group , while in the same period in our hospital ,pregnant women of singleton pregnancy detected by Doppler ultrasound who had had no thalassemia gene ,also including her spouse were selected as control group .Gestation fetal brain hemody-namics related indicators of (22.0 ±3.0)weeks were analyzed for both groups .Results The thalassemia fetal middle cerebral artery peak velocity averaged (52.06 ±11.03)cm/s,compared with the control group ,the difference was sig-nificant(χ2 =348.20,P0.05);Light with thalassemia intermedia fetal middle cerebral artery peak ve-locity had no significant difference compared with control group (χ2 =1.63, 5.79, P >0.05 ).Conclusion Doppler ultrasound used to detect fetal middle cerebral artery peak systolic velocity prediction thalassemia has the ad -vantages of valid,reliable,non-invasive,repeatable,which has broad application prospects .

ABSTRACT

Seventy patients with advanced non-small-cell lung cancer (NSCLC) aged 65 or above were treated with epidermal growth factor receptor tyrosine kinase inhibitor (EGFR-TKI) erlotinib or gefitinib from February 2006 to September 2010. The efficacy and toxicities of treatment were retrospectively analyzed.The overall response rate and disease control rate were 31.4% and 84.3%,respectively. Themedian progression-free survival time and median survival time were 8.0 months and 13.5 months,respectively(P < 0.05 ). One-year survival rate was 54.3%. Response rate ( CR + PR) ( 42.9% ) anddisease control rate (94.3% )in female patients were superior to males (20.0% and 74.3% ) (P < 0.05 ).Non-smoking and PS score < 2 were good predictors for survival.The side effects were generally mild and mainly were skin rash and diarrhea.

ABSTRACT

<p><b>BACKGROUND</b>Uroacitides is a group of cell differentiation inducers, which is purified from fresh human urine. Preclinical studies of Uroacitides have showed that cancer cells could be induced to differentiate, and the growth of cancer cells could be inhibited by Uroacitides. The aim of this study is to compare the efficacy and toxicity between Uroacitides combined with NP regimen and NP alone in treatment of advanced non-small cell lung cancer (NSCLC).</p><p><b>METHODS</b>Forty-two cases of advanced NSCLC were randomized into Uroacitides+NP and NP groups. NP group: NVB 25mg/m² on days 1 and 8, DDP 75mg/m² on day 1. Uroacitides combined with NP group: Uroacitides of 300mL was given through subclavian catheter daily for 7 days prior to the NP chemotherapy, then concurrently with NP regimen for 2 cycles, except the days of administration of chemotherapy.</p><p><b>RESULTS</b>In the Uroacitides+NP group, the overall response rate was 44.4%, and 20.0% in the NP group (P > 0.05). The median survival time was 9 months in the Uroacitides+NP group and 6 months in the NP group (P=0.0287). The main toxicities were myelosuppression, gastrointestinal reaction and alopecia, and there was no significant difference in incidences of toxicities between the two groups (P > 0.05).</p><p><b>CONCLUSIONS</b>Uroacitides combined with NP regimen shows a good curative effect and low toxicity, and may significantly prolong the median survival time for advanced NSCLC.</p>

ABSTRACT

<p><b>BACKGROUND</b>To evaluate the efficacy, side-effects, median survival duration and survival rate of vinorelbine (NVB) combined with cisplatin (DDP) in the treatment of non-small cell lung cancer (NSCLC).</p><p><b>METHODS</b>A total of 220 patients with inoperable NSCLC received NVB and DDP combined chemotherapy: NVB 25-30 mg/(m²*d) on days 1 and 5 (or 8), DDP 60-80 mg/(m²*d) on day 2. The schedule was repeated every 28 days. The efficacy and side-effects were analysed and followed-up after at least two cycles of chemotherapy.</p><p><b>RESULTS</b>The overall response rate (CR+PR) was 30.9% (68/220). The response rate was 31.3% (51/163) in initial treatment group, and 29.8% (17/57) in retreatment group. The median survival duration was 8.3 months. The 1-, 2- and 3-year survival rates were 39.23%, 19.31% and 6.32%, respectively. The main side-effects were myelosuppression and digestive tract reactions.</p><p><b>CONCLUSIONS</b>Vinorelbine plus cisplatin is an effective and well-tolerated regimen for non small cell lung cancer and myelosuppression is its dose-limiting toxicity.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To find out the relationship between the opacity in lens and the contents of 2,6-dinitro-4-amino-toluene(DNAT) in the urine of exposed workers.</p><p><b>METHODS</b>Testing the exposed worker's lens and measuring the contents of DNAT in the urine after work.</p><p><b>RESULTS</b>When the opacity of the lens occurred, the contents of DNAT in the urine(2.38 mg/L) of workers exposed to TNT were significantly higher than those without opacity in lens(1.44 mg/L) (P < 0.05).</p><p><b>CONCLUSION</b>The severity of opacity of lens increased with the contents of DNAT raised in the urine. The threshold value suggested by ILO is not applicable to Chinese occupational population, which recommends the contents of DNAT(30 mg/L) in the urine for the workers exposed to TNT as biological occupational exposed limits.</p>
