ABSTRACT

Objective:To explore the predictive value of peripheral blood CD34-positive cell count for the stem cell mobilization effect of plerixafor in patients with multiple myeloma (MM).Methods:The clinical data of 12 MM patients who used plerixafor for stem cell mobilization in the First Affiliated Hospital of Guangxi Medical University from December 2019 to February 2021 were retrospectively analyzed. The changes of peripheral blood CD34-positive cell count and the collection status of stem cell in all patients before and after the mobilization of plerixafor were analyzed.Results:Twelve patients were included in this study. These patients were in international staging system (ISS) stage Ⅱ-Ⅲ, and the induction therapy was mainly VRD regimen. The CD34-positive cell count was increased after the use of plerixafor in all patients no matter which mobilization strategies were used before plerixafor. The CD34-positive cell count was 3.63/μl (0.72-13.53/μl) and 32.11/μl (8.52-53.68/μl) before and after the use of plerixafor, and the difference was statistically significant ( Z = -0.40, P<0.001); the median increasing time was 11.50 times (1.61-23.71 times). The mobilization failure occurred in 1 patient. The CD34-positive cell count in his blood was less than 1/μl before the use of plerixafor; though increased 11.83 times after the use of plerixafor, the CD34-positive cell count was still less than 10/μl. Pearson analysis showed that among the patients with CD34-positive cell count less than 4/μl before the use of plerixafor, there was a positive correlation in peripheral blood CD34-positive cell count before and after the use of plerixafor ( r = 0.80, P = 0.032). Conclusions:The peripheral blood CD34-positive cell count has a certain predictive value for the stem cell mobilization effect of plerixafor in MM patients.

ABSTRACT

Objective:To analyze and summarize the epidemiological and molecular characteristics of SARS-CoV-2 Delta variant, a variant of concern (VOC), in Henan Province in 2021 in order to provide a basis for epidemic prevention and control.Methods:According to the feedback of sequencing results from Chinese Center for Disease Control and Prevention, 111 patients infected with SARS-CoV-2 Delta VOC were selected from the Henan imported and local cases in 2021. Basic patient information was obtained from the pandemic website. The differences in age, gender, vaccination history, the number of vaccine doses and different clinical types were analyzed. Moreover, the differences in RT-qPCR results of ORF1 ab gene and N gene Ct values between cases of different genders and symptoms were analyzed statistically. Sequencing results of the nucleotide and S protein mutation sites were analyzed. Results:There was no significant difference in the gender distribution of 111 cases between different age groups (χ 2=2.217, P=0.529). There was also no significant difference in clinical types between patients with different vaccination history (χ 2=12.074, P=0.209). The Ct values of most SARS-CoV-2 nucleic acid-positive specimens were distributed in the lower range and the viral loads were higher. The difference in the Ct value of ORF1 ab gene between different gender groups was not statistically significant (χ 2=1.646, P=0.439), but were significantly different among asymptomatic, mild, normal, and severe cases (χ 2=13.257, P=0.039). There was no significant difference in N gene Ct value among cases of different genders or different symptoms (all P>0.05). The 111 patients in this study were mainly found through close-contact screening and full-staff nucleic acid screening and accounted for 62.2% (69 cases) of the total. The sequencing length coverage was basically greater than 99% (accounting for 90.1%, 100/111); the total number of nucleotide mutation sites was mostly in the range of 46-50 (86.4%, 89/103); the total number of S protein mutation sites was mostly 12 (82.5%, 85/103). The 103 Delta mutants all contained nine mutation sites, which were T19R, R158G, L452R, T478K, D614G, P681R, D950N, E156del and F157del, with a mutation rate of 100%. Conclusions:People were highly susceptible to the SARS-CoV-2 Delta in Henan Province in 2021. High viral load and increase in the ORF1 ab gene load would aggravate the clinical symptoms.

ABSTRACT

OBJECTIVE:To establish HPLC fingerprint of Coptis chinensis inflorescence,and study its spectrum-effect relationship with antioxidant and antibacterial effects. METHODS :Taking 14 batches of C. chinensis inflorescence from different producing areas as the object ,HPLC method was adopted. The determination was performed on Supersil C 18 column with mobile phase consisted of acetonitrile- 0.1% phosphoric acid solution(gradient elution )at the flow rate of 1.0 mL/min. The column temperature was set at 25 ℃. The detection wavelength was set at 329 nm,and sample size was 10 μL. The fingerprints of 14 batches of C. chinensis inflorescence were established by Similarity Evaluation System of TCM Fingerprint (2012 A edition ),and the similarity evaluation and common peak identification were carried out. Taking DPPH free radical scavenging rate and hydroxyl radical scavenging rate as antioxidant effects index ,relative antibacterial activity (Escherichia coli )as antibacterial effect index , SPSS 21.0 software was adopted to analyze the Pearson correlation between common peaks of C. chinensis inflorescence and above efficacy indexes ;their spectrum-effect relationship was established and validated. RESULTS :A total of 7 common peaks were obtained in HPLC fingerprint of C. chinensis inflorescence,and the similarity was no less than 0.916. No. 5 peak was identified as berberine hydrochloride. Seven common peaks were positively correlated with DPPH free radical scavenging rate ;No. 1-3,4,6,7 peaks were positively correlated with hydroxyl radical scavenging rate ,while No. 5 peak was negatively correlated with hydroxyl radical scavenging rate. There was a positive correlation between No. 5 peak and antibacterial activity in vitro . After validation , relative error between the predicted values and the measured values of DPPH free radical scavenging rate ,hydroxyl radical scavenging rate and relative antibacterial activity was 0.92%- 14.5% . CONLUSIONS :The established spectrum-effect relationship model can be used to evaluate antioxidant andantibacterial effects of C. chinensis inflorescence. The chemical components represented by No. 1,2,3,4,6,7 peaks are the material basis of antioxidant effect of C. chinensis inflorescence, and berberine hydrochloride is the material basis of antibacterial effect.

ABSTRACT

OBJECTIVE:To evaluate the quality of inflorescence of Coptis chinensis from different altitude with different growth years and drying processing methods ,and to provide reference for its utilization and quality control. METHODS :The contents of total flavonoids and berberine hydrochloride in 24 batches of inflorescence of C. chinensis (S1-S24)from different altitude with different growth years and drying processing methods were determined by UV-Vis spectrophotometry and HPLC. Using the contents of total flavonoids and berberine hydrochloride as indexes ,and taking the short drying time as the best ,the weight ratios of total flavonoids content ,berberine hydrochloride content and standardized value of drying time were 30,40 and 30,respectively;comprehensive score was calculated ,then the quality of 24 batches of samples was evaluated. Using the contents of total flavonoids and berberine hydrochloride as variables ,systematic cluster analysis was performed for 24 batches of samples by using SPSS 19.0 statistical software. RESULTS :For inflorescence of C. chinensis with altitude about 1 200 m and the growing years of 4 years and above ,the higher the comprehensive score (84-94 score)and the better the quality were. The comprehensive score of inflorescence of C. chinensis procesed by gradient drying method was generally higher than samples processed by other methods. Results of cluster analysis showed that S 1-S4,S9,S10,S13-S18 were clustered into one category ,and other 12 batches were clustered into one category , which were basically consistent with the results of comprehensive scoring method. CONCLUSIONS:Different altitude ,different growth years and different drying processing methods have certain effects on the quality of inflorescence of C. chinensis ,among which the samples processed by gradient drying method with growth altitude of 1 200 m,growth years of 4 years and above are the best.

ABSTRACT

Objective To study the effect of recombinant human platelet activating factor acetylhydrolase (rPAF-AH) on candidate plasticity-related gene 15 (CPG15) expression in focal cerebral ischemia rats.Methods Forty-five SD rats were randomly divided into sham operation group,saline group and rPAF-AH group (15 in each group).A middle cerebral artery occlusion model of rats was established for the latter two groups.Each group was divided into 2 d group,7 d group,14 d group.Five rats in each of 2 d group,7 d group,14 d group were used in experiment.The total protein was extracted from coronary sections using homogenate.CPG15 protein expression was detected by Western blot.Results The CPG15 protein expression level was significantly higher in saline group and rPAF-AH group than in sham operation group on days 2,7,14 and reached its peak on day 7,and in rPAF-AH group than in saline group on day 7 (1.48±0.04 vs 1.12±0.05,P<0.01).Conclusion PAF-AH plays a positive role in neuroplasticity of cerebral ischemia by upregulating the CPG-15 expression in focal cerebral ischemia rats.

ABSTRACT

Objective@#To analyze the VP1 region characteristics of coxsakievirus B4 (CoxB4) strain isolated from patients with hand, foot and mouth disease (HFMD) in Henan province and to compare the similarity and difference among these viruses isolated from different diseases.@*Methods@#The 134 samples were tested by reverse-transcription polymerase chain reaction (RT-PCR) and cell-culture-based isolation of CoxB4. The VP1 of 8 CoxB4 isolated was amplified using specific primers. The sequences were spliced, edited, analyzed and mapped into phylogenetic tree by bioinformatics software ATGC, lustalx1.83, BioEdit and MEGA4.0, respectively.@*Results@#The VP1 region length of Henan CoxB4 isolates were 852 bp. Compared with other CoxB4 strains released in GeneBank, the similarities of nucleotide and amino acid is 80.3%-97.0%, 96.1%-100%. The largest difference was 19.7% between Henan isolates and diabetes isolates, and 3.0% was the smallest difference with HFMD isolates. Phylogenetic tree analysis showed that Henan CoxB4 isolates belonged to Genotype V, and the isolates from HFMD were close to those of meningitis, distributed in a cluster. These isolates from diabetic patients were in another cluster.@*Conclusions@#There were Genotype V spreading and circulating of CoxB4 that infected HFMD patients in Henan.

ABSTRACT

<p><b>PURPOSE</b>To explore the impact of subtropical maritime monsoon climate on the frequency of ambulance use for trauma patients in a coastal region in China.</p><p><b>METHOD</b>Statistical analysis of data on ambulance use from the 120 Emergency Command Center in Shantou City, Guangdong Province, from January to December 2012 as well as daily meteorological data from a Shantou observatory was performed to determine how climatic factors (seasons, time, and weather) affect the frequency of ambulance use for trauma patients.</p><p><b>RESULTS</b>The daily ambulance use for trauma patients differed between spring and summer or autumn (p<0.05), between sunny and rainy days (p<0.05), and between cloudy and lightly or moderately rainy days (p<0.05).We found a linear correlation between daily maximum temperature and daily ambulance use for trauma patients (R² =0.103, p<0.05). In addition, there was significant difference in ambulance use between good and bad weather (p<0.05).</p><p><b>CONCLUSION</b>Frequency of ambulance use for trauma patients is affected by the subtropical maritime monsoon climate in the coastal region. Better weather contributes to increased daily frequency of ambulance use, which is the highest in autumn and lowest in spring.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To clone and express the recombinant capsid protein VP2 of enterovirus type 71 (EV71) and to identify the immune activity of expressed protein in order to build a basis for the investigation work of vaccine and diagnostic antigen.</p><p><b>METHODS</b>VP2 gene of EV71 was amplified by PCR, and then was cut by restriction enzyme and inserted into expression vector pMAL-c2X. The positive recombinants were transferred into E.coli TB1, the genetically engineered bacteria including pMAL-c2X-VP2 plasmids were induced by isopropyl thiogalactoside ( IPTG) , and the expression products were analyzed by SDS-PAGE and western blotting method. EV71 IgM antibody detection method by ELISA was set up, and the sensitivity and specificity of this method was assessed; 60 neutralizing antibody positive serum samples from hand foot and mouth disease (HFMD) patients were determined, of which 52 samples were positive and 8 samples were negative; a total of 88 acute phase serum samples of HFMD patients diagnosed in clinical were also detected.</p><p><b>RESULTS</b>VP2 gene of 762 bp was obtained by PCR, the gene segment inserted into the recombinant vector was identified using restriction enzyme digestion. The recombinant vector could express a specific about 71 500 fusion protein in E.coli by SDS-PAGE. The purified recombinant protein of EV71-VP2 can react with the serum of HFMD patients to produce a specific band by western blotting. The sensitivity and specificity of ELISA was 87% and 83%, respectively. Of the 88 acute phase serum samples from children with HFMD, 48 samples (55%) were positive by the ELISA assay.</p><p><b>CONCLUSIONS</b>VP2 gene of EV71 has been cloned and a prokaryotic high expression system for VP2 gene was successfully constructed in the present study. The recombination EV71-VP2 has well antigenicity, which could be useful for developing diagnose reagent or vaccine of EV71.</p>

ABSTRACT

Objective To analyze the evolution and genetic characteristics of Echovirus 11 (Echo11) strains isolated from patients with hand food and mouth disease ( HFMD) in Henan province. Methods Enterovirus strains were isolated from stool samples of patients with HFMD from year 2010 to 2012.The sequences of VP1 region of all positive strains were amplified , sequenced and then analyzed by BLSAT to determine their genotypes .The entire VP1 coding regions of 10 Echo11 strains were amplified by RT-PCR, and the homology analysis was conducted among them and other Echo 11 strains published in Gen-Bank.A phylogenetic tree was constructed to evaluate the evolution of Henan isolates and the prevalence of different genotypes .Results From year 2010 to 2012 , 184 non-Enterovirus 71 ( EV71 ) strains and non-Coxsakievirus A16 (CA16) strains causing HFMD were isolated, 10 (5.43%) of with were Echo11 strains. There were 876 nucleotides ( nt) in the complete VP1 gene sequence , encoding 292 amino acids ( aa) .The 10 Echo11 strains shared 93.1%-100% homologies in nt sequences and 97.3%-100% in aa sequences. The homology analysis indicated that Henan strains and prototype strains were highly similar but different with the homology of 77.8%-78.8% in nt and 90.8%-91.8% in aa, respectively.Conclusion Echo11 was one of the pathogens causing HFMD in He-nan province and genotype A was the dominant genotype .

ABSTRACT

<p><b>OBJECTIVE</b>To study effects of different harvest times and processing methods on the quality of Gardeniae Fructus.</p><p><b>METHOD</b>The content of asminoidin and the similarity of the fingerprint of Gardeniae Fructus were determined and applied for assessment of the quality.</p><p><b>RESULT</b>Gardeniae Fructus harvested in October with yellow-green appearance was the better time point for harvest. Drying in the sun and boiling in the water is the better processing method for Gardeniae Fructus.</p><p><b>CONCLUSION</b>Different harvest times and processing methods can affect the quality of Gardeniae Fructus.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of removing bacterial endotoxin in the key processes of Reduning injection.</p><p><b>METHOD</b>The content of bacterial endotoxins was detected by kenitic-turbidimetry and the removal efficacy was studied before and after using 0.8% of activated carbon and ultrafiltration with molecular weight cut-off of 10 x 10(3).</p><p><b>RESULT</b>The adsorption rate of bacterial endotoxins was 78.7% by using activated carbon, while the removal efficacy of bacterial endotoxins was 99.6% with ultrafiltration membrane at cut-off molecular weight 10 x 10(3).</p><p><b>CONCLUSION</b>The key technology can effectively guarantee the safety of Reduning injection.</p>

ABSTRACT

<p><b>OBJECTIVE</b>Applying for the activity of enzyme in vitro,the research optimized the best preparation procedure for the anticoagulated blood region from Lumbricus.</p><p><b>METHOD</b>All through our experiment, the content of protein and theactivity of enzyme were examined. The extraction process, the refining technology, concentration processes of Lumbricus were optimized with single factor checking and orthogonal design method.</p><p><b>RESULT</b>At 37 degrees C, the coarse powder of Lumbricus soaking with 15 fold of 0.9% sodium chloride and ultrasonic extracting 40 minites for three times was the best ultrasonic extraction. Utrafiltration membrane with molecular weights of 30 x 10(3) for refining and 10 x 10(3) for concentrating were selected.</p><p><b>CONCLUSION</b>Ultrasonic extraction and membrane separation technology, to well improve the effect of purification for the anticoagulant site of Lumbricus, is conducive to further study.</p>

ABSTRACT

Objective To establish the method for the content determination of sinomenine in Qiwei Tongbi oral liquid. Methods Sinomenine ofQiwei Tongbi oral liquid was determined by HPLC, with the column being Waters XTerra column temperature setting at 25℃, flow rate being 1.0 ml/min, and the detective wavelength being 262 nm. Results There was a good linearity of sinomenine in the range of 0.248~2.480 μg, and the average recovery was 98.82% with RSD of 0.13%. Conclusion This method was simple, accurate, reproducible, and thus could be used for quality control of Qiwei Tongbi oral liquid.
