ABSTRACT

@#Objective    To explore the association between radiotherapy and the risk of subsequent primary lung cancer in female patients with breast cancer. Methods    The EMbase, Web of Science, PubMed, CNKI, VIP and WanFang databases were searched from the establishment date to July 1, 2021. The relative risk (RR) with 95% confidence interval (CI) were combined and all statistical analyses were performed by STATA 12.0 software. Results    A total of 10 publications including 466 510 participates from 14 studies were included. The meta-analysis indicated that radiotherapy was a risk factor for the occurrence of second primary lung cancer among female breast cancer patients [RR=1.45, 95%CI (1.18, 1.78), P<0.001]. Subgroup analysis based on the relative position of breast cancer and lung cancer was conducted and the results demonstrated that radiotherapy only increased the incidence rate of ipsilateral lung cancer [RR=1.27, 95%CI (1.10, 1.45), P=0.001], without significant change of the risk of contralateral lung cancer [RR=1.16, 95%CI (0.77, 1.74), P=0.487]. Conclusion    Radiotherapy is one of the risk factors for subsequent primary pulmonary carcinoma among female breast cancer patients, especially for ipsilateral lung carcinoma. However, more high-quality studies are still needed to verify above findings.

ABSTRACT

Objective:To investigate the effect and mechanism of bone marrow mesenchymal stem cells (BMMSCs) on liver transplantation with 50% reduced-size in rat models.Methods:For 40 normal male Brown Norway(BN) and Lewis rats weighing 210-250 g were included respectively to generate the acute rejection models following 50% reduced-size liver transplantation in rats. The recipients were divided into BMMSCs group ( n=20) and normal saline group ( n=20). Healthy male BN rats were used to prepare BMMSCs. Transplanted liver tissues were collected at 0h, 1d, 3d, 7d post the transplantation for further analysis. Pathological changes and the extent of rejection were evaluated under the light microscope. The levels of microtubule-associated protein 1 light 3 (LC3) and autophagy regulator Beclin-1 proteins were detected by immunohistochemical analysis and Western blotting. Results:Rejection activity indices of the normal saline group at 0d, 3d, 7d after the surgery were (2.33±0.58), (4.00±0.00), (6.33±0.58). The BMMSCs group were (2.10±0.58), (3.73±0.58), (5.67±1.15), which was decreased comparing with normal saline group, difference was statistically significant ( P<0.05). On the 1d, 3d, 7d after the transplantation, compared with normal saline group, expression of autophagy-related proteins LC3 and Beclin-1 in BMMSCs group was increased ( P<0.05). Conclusions:It showed that autophagy has an effect on the protection of BMMSCs liver graft of rats.

ABSTRACT

Objective To investigate the efficacy of rosuvastatin combined with silibinin capsule in treatment of hyperlipidemic patients with nonalcoholic fatty liver disease.Methods One hundred and nineteen hyperlipidemic patients with non-alcoholic fatty liver disease admitted in hospital from January 2016 to March 2017 were randomly assigned into two groups:60 patients received rosuvastatin(10 mg,q.d) and silibinin capsules(70 mg,t.i.d.,study group) and 59 patients received starch tablets(10 mg,q.d) and silibinin capsules(70 mg,t.i.d.,control group).The blood lipid,liver function,inflammatory factors,liver CT value,the specific CT value of liver and spleen were compared at baseline and 6 months after intervention.Results Compared to baseline values,the levels of total cholesterol (TC),triglyceride (TG),alanine aminotransferase (ALT),γ-glutamyltranspeptidase (GGT),TNF-α,IL-6,and CRP were decreased (t=2.44,2.04,2.48,6.95,2.38,2.78,3.97,all P<0.05);and the levels of HDL-C,liver CT value,liver/spleen CT value ratio were increased (t=5.44,2.26,2.08,all P<0.05) after treatment in study group.The improvement of TG,LDL-C,ALT,GGT,TNF-α,CRP,liver CT value,liver/spleen CT value ratio were more marked than those of the control group (P<0.05) (t=1.92,2.65,2.02,5.62,2.11,2.20,2.22,2.08,all P<0.05).The effective rate of study group was higher than that of the control group[98.2%(55/56) vs.85.7%(48/56),x2=2.05,P=0.04];the adverse reactions were similar in two groups.Conclusion Compared with silibinin alone,rosuvastatin combined with silibinin capsules is more effective in treatment of hyperlipidemic patients with nonalcoholic fatty liver disease,which can significantly improve the blood lipids,liver function,inflammatory response and aliphatic degree without increase of adverse reaction.

ABSTRACT

Objective: To investigate the role of p38 MAPK pathway in IL-1β induced inflammatory secretory properties mesenchymal stem cells(SFMSCs) from human synovial fluid of temporomandibular joint. Methods: In vitro cultured hSFMSCs were divided into control group and IL-1β treated group(10 ng/ml IL-1β for 2 h), the activation of p38 MAPK was examined with western blot. Then, the gene expression and the secretion of IL-6 and IL-8 in control group, IL-1β treated group and p38 MAPK inhibitor pre-treated group(pretreatment with SB-203580 10 μmol/L prior to the addition of 10 ng/ml IL-1β) were examined by RT-PCR and cytometric bead array(CBA). Results: It was observed that the protein level of p-p38 MAPK, the secretion of IL-8 and IL-6 were increased in IL-1β treated group, and suppressed in p38 MAPK inhibitor pre-treated group. Conclusion: p38 MAPK pathway of SFMSCs might play a role in IL-6 and IL-8 secretion by SFMSCs in inflammatory environment.

ABSTRACT

Objective To explore the role and mechanism of thymosin β4 (Tβ4) in the treatment of non-alcoholic fatty liver disease (NAFLD).Methods Forty male C57BL/J6 mice were divided into normal group,NAFLD group,low dose Tβ4 group and high dose Tβ4 group with 10 mice in each group.NAFLD mice model was established by feeding with high fat and high sugar diet for 16 weeks.The mice in low-dose Tβ4 group and high dose Tβ4 group were intraperitonealy injected with Tβ4 at 0.05 mg · kg-1 · d-1 and 0.20 mg · kg-1 · d-1,respectively,for eight weeks.The liver function indexes and serum tumor necrosis factor-α (TNF-α) level were detected;the pathological changes of liver tissue were observed under optical microscope and non-alcoholic fatty liver disease activity score (NAS) was evaluated.The protein expression levels of nuclear factor-κB p65 (NF-κB p65) and nuclear factor κB inhibit protein a (IκBa) at the protein level in liver tissue were measured by Western blotting method.The expression of TNF-α in liver tissue was detected by immunohistochemistry.Mean integral absorbance (MIA) was calculated.T test was performed for groups comparison.Results The levels of alanine aminotransferase (ALT),γ-glutamine transferase (GGT) and serum TNF-α levels of high dose Tβ4 group were all lower than those of NAFLD group ((28±17) U/L vs.(76±29) U/L,(61±39) U/L vs.(102±56) U/L,(144.1± 48.2) ng/L vs.(187.3±58.8) ng/L,respectively),and the differences were statistically significant (t=4.52,2.78 and 2.30,all P<0.05).The NAS of low dose Tβ4 group and high dose Tβ4 group were both lower than that of NAFLD group (3.7±40.4,2.3±0.3 vs.4.6±0.3),and the differences were statistically significant (t=5.69 and 17.14,both P<0.01).The relative expression level of Tβ4 protein of NAFLD group was lower than that of normal group (0.2±0.1 vs.1.4±0.6),and the difference was statistically significant (t=6.24,P<0.01).The relative expression levels of Tβ4 and IκBa of high dose Tβ4 group were higher than those of NAFLD group (1.0±0.3,0.5±0.3 vs.0.2±0.1),and the differences were statistically significant (t=8.00 and 3.00,both P<0.01).The relative expression level of NF-κB p65 in liver tissue of high dose Tβ4 group was lower than that of NAFLD group (0.6±0.3 vs.1.5±0.7),and the difference was statistically significant (t=3.74,P<0.01).The MIA of high dose Tβ4 group was lower than that of NAFLD group (0.4±0.2 vs.0.7±0.3),and the difference was statistically significant (t=2.63,P< 0.01).Conclusion Tβ4 can effectively treat NAFLD probably through inhibiting the NF-κB pathway.

Objective@#To analyze related factors on the number of mesenchymal stem cells in the synovial fluid of the temporomandibular joint (TMJ) and provide an research basis for understanding of the source and biological role of mesenchymal stem cells derived from synovial fluid in TMJ.@*Methods@#One hundred and twenty-two synovial fluid samples from 91 temporomandibular disorders (TMD) patients who visited in Department of TMJ Center, Guanghua School of Stomatology, Hospital of Stomatology, Sun Yat-sen University from March 2013 to December 2013 were collected in this study, and 6 TMJ synovial fluid samples from 6 normal volunteers who were studying in the North Campus of Sun Yat-sen University were also collected, so did their clinical information. Then the relation between the number of mesenchymal stem cells derived from synovial fluid and the health status of the joints, age of donor, disc perforation, condylar bony destruction, blood containing and visual analogue scale score of pain were investigated using Mann-Whitney U test and Spearman rank correlation test.@*Results@#The number of mesenchymal stem cells derived from synovial fluid had no significant relation with visual analogue scale score of pain (r=0.041, P=0.672), blood containing (P=0.063), condylar bony destruction (P= 0.371). Linear correlation between the number of mesenchymal stem cells derived from synovial fluid and age of donor was very week (r=0.186, P=0.043). The number of mesenchymal stem cells up-regulated when the joint was in a disease state (P=0.001). The disc perforation group had more mesenchymal stem cells in synovial fluid than without disc perforation group (P=0.042).@*Conclusions@#The number of mesenchymal stem cells derived from synovial fluid in TMJ has no correlation with peripheral blood circulation and condylar bony destruction, while has close relation with soft tissue structure damage of the joint.

ABSTRACT

Objective To evaluate the significance of serum 8-hydroxy-deoxyguanosine acid ( 8-OHdG) in the diagnosis of nonalcoholic steatohepatitis ( NASH).Methods Patients or healthy subjects were enrolled at the Second Hospital of Tianjin Medical University and the Second People ′s Hospital of Tianjin from May 2013 to December 2015.A total of 41 patients with nonalcoholic fatty liver disease were enrolled in the study , including 20 nonalcoholic simple fatty liver ( NAFL) patients and 21 NASH patients whose diagnosis were proven by liver biopsy.The other 32 healthy subjects were studied as controls.Serum 8-OHdG, ALT, AST and GGT were tested.Nonalcoholic fatty liver disease activity score ( NAS ) and expression of 8-OHdG in liver was investigated between NAFL patients and NASH patients.The correlations between serum 8-OHdG and serum ALT , AST, GGT, and 8-OHdG in liver tissue in NASH group were investigated.In addition , the receiver operating characteristic ( ROC) curve analyses for ALT and 8-OHdG levels were performed in NAFL patients and NASH patients , and the cut-off value was determined.Results Serum 8-OHdG values in healthy controls , NAFL and NASH patients were (0.19 ±0.16) μg/L, (0.22 ±0.16) μg/L, (0.42 ±0.21) μg/L respectively.The serum 8-OHdG and serum ALT, GGT and 8-OHdG in liver tissue were all positively correlated in NASH group with respective correlation coefficient r values as 0.454 7, 0.382 9, and 0.497 6.AUC of 8-OHdG was 0.901 with cut-off value 0.39 μg/L.Its sensitivity was 88.3%and specificity was 81.5%, which were higher than those of ALT.Conclusion The value of serum 8-OHdG would be used as a marker for the diagnosis of NASH.

Objective To explore the effect of anoxic microenvironment and hypoxia related microRNA-210 (miR-210) on the epithelial-mesenchymal transition (EMT) in gastric cancer cell line 803.Methods The mRNA relative expressions of E-cadherin and Twist1 were detected by qRT-PCR in gastric cancer cell line after being induced hypoxia by CoCl2 (100,200 and 300 μ mol/L),which was compared with normoxic group (0 mol/L).The cell proliferation of gastric cancer cell line was observed by cell proliferation assay.The relative expressions of E-cadherin and Twist 1 were detected in gastric cancer cell line after transfecting miR-210 mimics.Results With the increase of the concentrations of CoCl2,the relative expression of miR-210 and the proliferation rate of gastric cancer cells showed a tendency of increase first and then decrease (P < 0.05).The relative expression of E-cadherin mRNA was lower in hypoxia group than that of normoxic group.The relative expression of Twist1 mRNA was higher in hypoxia group than that of normoxic group.The relative expressions of Ecadherin mRNA were significantly higher in 200 and 300 μ mol/L CoCl2 groups than that of 100 μmol/L CoCl2 group (P <0.05).There was no significant difference in the relative expression of Twist1 mRNA between the three CoCl2 groups.The relative expression of Twist1 mRNA was higher in the transfection group than that of control group.The relative expression of E-cadherin mRNA was significantly lower in the transfection group than that of control group (P < 0.05).Conclusion Hypoxia can promote the proliferation and EMT of gastric cancer cells.The up-regulation of miR-210 can also promote the procession of EMT,which may be the intermediate link in the process of EMT induced by hypoxia.

ABSTRACT

Objective To explore the expression and significance of CD44 gastric cancer stem cell biomarker positive circulating tumor cells (CTC ) in peripheral blood of patients with gastric cancer . Methods From March 2010 to December 2012 ,45 patients with gastric cancer and 20 healthy individuals were enrolled .Peripheral blood samples of all the subjects were collected .The expression of CTC and CD44 postive CTC were detected by density gradient centrifugation method and double immunofluorescent staining .Chi‐square test and t‐test were performed for statistical analysis .Results CTC in peripheral blood of healthy controls were not detected .Among 45 patients with gastric cancer ,CTC were detected in 27 patients and 19 of them were CD44‐positive .The difference in positive rate of CTC expression was statistically significant between two groups (χ2 =10 .986 , P<0 .05) .Among 27 CTC‐positive patients , 19 were proximal gastric cancer and 16 of them were CD44‐positive;16 patients had lymph node metastasis and 14 of them were CD44‐positive;13 patients had distant metastasis and 12 of them were CD44‐positive;14 patients had recurrence and 13 of them were CD44‐positive .The CD44 positive CTC was correlated with tumor location ,lymph node metastasis ,distant metastasis and recurrence (χ2=5 .891 ,5 .527 ,5 .787 and 7 .052 ,respectively ,all P<0 .05) .Furthermore ,in 13 patients with recurrence and CD44 positive CTC ,the recurrence time was (10 .54 ± 5 .55) months which was shorter than that of one patients with CD44 negative CTC (19 months) ,and the difference was statistically significant (t= -3 .654 , P=0 .004) .Conclusions CTC which express cancer stem cell marker may be associated with recurrence and metastasis of gastric cancer .It has more important clinical significance to detect this subtype of CTC .

ABSTRACT

<p><b>OBJECTIVE</b>To compare the temporomandibular joint changes in adolescent Class II division 1 malocclusion patients with mandibular retrusion treated with Twin- block and Class II elastics.</p><p><b>METHODS</b>Sixty Class II division 1 malocclusion patients with mandibular retrusion were divided into 3 groups (Twin- block group, Class II elastic group and control group, 20 patients in each group) randomly. Differences of condylar process morphology were compared using cone- beam CT (CBCT) before and after treatment.</p><p><b>RESULTS</b>Condylar largest anteroposterior diameter and upper condylar height increased after treatment in both Twin-block group and Class II elastic group. The condylar largest anteroposterior diameter and upper condylar height were longer in both Twin- block group [(8.51 ± 1.48) and (7.39 ± 1.42) mm] and Class II elastic group [(8.44 ± 1.03) and (7.24 ± 0.89) mm] than those in the control group [(7.47 ± 1.27) and (6.47 ± 1.56) mm] (P<0.05). No significant difference was found in other measurements (P>0.05).</p><p><b>CONCLUSIONS</b>Condylar largest anteroposterior diameter and upper condylar height in Class II division 1 malocclusion patients increased after treatment using Twin- block and Class II elastics. There was no significant difference between two groups.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To design and fabricate the custom-made titanium mandibular condyle by the reverse engineering technology combined with selective laser melting (SLM) technology and to explore the mechanical properties of the SLM-processed samples and the application of the custom-made condyle in the temporomandibular joint (TMJ) reconstruction.</p><p><b>METHODS</b>The three-dimensional model of the mandibular condyle was obtained from a series of CT databases. The custom-made condyle model was designed by the reverse engineering software. The mandibular condyle was made of titanium powder with a particle size of 20-65 µm as the basic material and the processing was carried out in an argon atmosphere by the SLM machine. The yield strength, ultimate strength, bending strength, hardness, surface morphology and roughness were tested and analyzed. The finite element analysis (FEA) was used to analyze the stress distribution.</p><p><b>RESULTS</b>The complex geometry and the surface of the custom-made condyle can be reproduced precisely by the SLM. The mechanical results showed that the yield strength, ultimate strength, bending strength and hardness were (559±14) MPa, (659±32) MPa, (1 067±42) MPa, and (212±4)HV, respectively. The surface roughness was reduced by sandblast treatment.</p><p><b>CONCLUSIONS</b>The custom-made titanium condyle can be fabricated by SLM technology which is time-saving and highly digitized. The mechanical properties of the SLM sample can meet the requirements of surgical implant material in the clinic. The possibility of fabricating custom-made titanium mandibular condyle combined with the FEA opens new interesting perspectives for TMJ reconstruction.</p>

ABSTRACT

Objective To explore the correlation between the change of hypoxic in microenvironment and the genesis of circulating tumor cells (CTCs) in gastric adenocarcinoma.Methods After human gastric adenocarcinomn BGC-823 cells treated with different concentration of CoCl2 (0,100,200,300,400 μmol/L)for four hours,the expression levels of hypoxia inducible factor 1 alpha (HIF-1α)were detected by Western blot to select the optimal concentration.A total of 40 BALB/c mice were divided into two equal groups,named hypoxic group and normoxic group.Optimal concentration of CoCl2 treated BGC-823 gastric cancer cells and non hypoxic induced BGC 823 gastric cancer cells were injected through the tail vein.After three days blood samples were collected through the angular vein.CTCs were enriched by density gradient centrifugation,identified with cytokeratin19 (CK19) antibody marker by cell immunofluorescence and cell number was counted.Statistical significace test were performed by analysis of variance and Dunnett-T test.Results The expression of HIF-1α reached a maximun of 1.40±-0.16 when BGC-823 cells were treated with 200 μmol/L CoCl2 for four hours.The number of CTCs in hypoxic group was higher than that of normoxic group ((4.70± 1.72)/high power field (HPF) vs (1.15±0.56)/HPF,Z=-4.824,P<0.01).Conclusion Hypoxic microenvironment might promote the genesis of CTC in gastric adenocarcinoma.

Background:To date,clinical studies on intravenous rabeprazole sodium for treatment of duodenobulbar ulcer bleeding are still lacking.Aims:To evaluate the efficacy and safety of intravenous rabeprazole sodium with different doses and times of administration in treating patients with duodenobulbar ulcer bleeding.Methods:A multicenter,randomized, double-blind,positive drug parallel-group controlled trial was performed.One hundred and five patients with duodenobulbar ulcer bleeding proved by gastroscopy were randomly divided into four groups.Patients in group A,B and C were treated with intravenous rabeprazole sodium 20 mg qd,40 mg qd and 20 mg bid for 5 days,respectively.Patients in control group received intravenous omeprazole sodium 40 mg bid for 5 days.Hemostatic rate was the primary endpoint,hemostatic time and amount of blood transfusion were the secondary endpoints.Results:Hemostatic rates in group A,B,C and control group were 96.2% (25 /26),92.6% (25 /27),100.0% (26 /26)and 100.0% (26 /26),respectively,no significant difference was seen between the four groups (P >0.05).Median hemostatic time in group A,B,C and control group were 24 (24,72)h,24 (24,72)h,24 (24,48)h and 24 (24,48)h,respectively,no significant difference was seen between the four groups (P >0.05).No patient need blood transfusion during the treatment course.Slight leucopenia was the exclusive adverse effect seen in one case in group C after accomplishment of treatment.Conclusions:Three intravenous rabeprazole sodium regimens with different doses and times of administration were all effective and safe for treatment of mild to moderate duodenobulbar ulcer bleeding.Administration with 20 mg bid seems more effective among the three regimens.

ABSTRACT

Objective: To detect circulating tumor cells (CTCs) in patients with gastric cancer and evaluate the relationship among CTCs, clinico-pathological characteristics, and prognosis of gastric cancer. Methods: Peripheral blood samples (10 mL in EDTA) were obtained from 45 patients with gastric cancer. CTCs were detected using density-gradient centrifugation and immunofluo-rescence staining. The clinical significance of the two methods were also compared and investigated. Results:CTC-positive case was defined by the presence of at least one CK19 (+)-CTC per 10 mL of the sample. CTCs were found in 27 of the 45 patients with gastric cancer. The presence of CTCs was significantly correlated with lymph node metastasis, distant metastasis, and recurrence (P=0.007, 0.035, 0.035, respectively). However, CTCs were not significantly correlated with sex, age, tumor location, TNM staging, and tumor differentiation (P>0.05). Conclusion:CTCs were associated with poor prognosis of gastric cancer.

ABSTRACT

Objective To investigate the effect of dual antiplatelet drugs on the intestinal damage in rats. Meth-ods Eighty SD rats were randomly allocated into four groups:control group (normal saline, n=20), aspirin group (10.41 mg/kg, n=20), clopidogrel group (7.81 mg/kg, n=20) and clopidogrel combined aspirin group (n=20). Each group was given intra-gastric administration of drugs once per day for 14 days. All rats received operation after the final intragastric administration. The intestinal injury was observed. The jejunal fluid was taken for bacterial culture. The intestinal permeability was detected by ethidium bromide (EB) method. The small intestinal mucosal injury was estimated by Chiu method. Results The differ-ent degrees of small intestinal mucosal injury were found in four groups. The scores of pathological lesions were significantly higher in aspirin group, clopidogrel group and clopidogrel combined aspirin group than those of control group (P<0.05). The dual antiplatelet group showed the highest score (3.450±1.356). The small intestinal permeability was significantly increased in experiment groups compared with that of control group (54.012±3.513μg/g, P<0.05). There was a significantly higher small intestinal permeability in dual antiplatelet group than that of aspirin group and clopidogrel group (μg/g:130.533 ± 29.631 vs 90.965±3.765 vs 66.800±4.853, P<0.001). The total jejunal bacteria was significantly increased in dual antiplate-let group than that of control group (CFU/mL:61924.805±1751.159 vs 18154.280±1153.376, P<0.001). The ratio of entero-bacterium and enterococcus was significantly decreased in dual antiplatelet group compared with that of control group (0.220±0.089 vs 1.007±0.148, P<0.001). Conclusion The routine dose of dual antiplatelet drug aggravates the small intes-tinal injury in rats compared with that of single drug. The manifestations of intestinal mucosal injury include increased intes-tinal bacteria, dysbacteriosis, and increased small intestinal permeability.

ABSTRACT

Objective To investigate the risk factors of dual anti-platelet treatment in upper gastrointestinal hemor-rhage after percutaneous coronary intervention (PCI). Methods Clinical data of 2004 PCI patients with dual anti-platelet treatment were collected,including gender,age,body mass index (BMI),history of drug therapy, history of digestive disease, history of smoking and drinking,history of hypertension,history of diabetes,history of cerebrovascular disease and history of combined treatment with a proton pump inhibitor (PPI). The different characteristics of gastrointestinal bleeding were com-pared. The risk factors of upper gastrointestinal bleeding were analyzed. Results The risk factors for PCI patients with up-per gastrointestinal bleeding were age>65 years of old, history of smoking, history of hypertension,abnormal platelet count, creatinine and hemoglobin. The combined treatment with a proton pump inhibitor was a protective factor associated with gas-trointestinal bleeding. Conclusion We should pay attention to the upper gastrointestinal bleeding induced by dual anti-platelet treatment, and take appropriate measures to reduce the incidence of gastrointestinal bleeding in PCI patients.

ABSTRACT

The clinical data of 186 patients with acute gout attack during upper gastrointestinal bleeding were analyzed retrospectively.The ratio of male to female was 1.7∶1, smokers and alcohol drinkers accounted for 66.1% ( 123/186 ) and 62.4% ( 116/186 ) of patients were had underlying diseases.All patients had different degree of joint painful and fever , the blood uric acid levels were higher.Gastroscopic examinations were performed in 166 patients , of whom 88 cases received endoscopic intervention.The symptoms of gout were improved after treatment with dexamethasone and celecoxib ; and the medication did not induce or increase gastrointestinal bleeding.

Objective To observe selective cyclooxygenase-2 (COX-2) inhibitor celecoxib on the cell migration, MMP-9 and VEGF expression of human gastric cancer SGC-7901. Methods In vitro culture of SGC-7901 cells, the cells were divided into blank control group and celecoxib treatment group (different concentrations of celecoxib 25, 50 and 100μmol/L). SGC-7901 cells were treated with celecoxib for 24 h. The effect of celecoxib on cell migration of SGC-7901 was detected by the damage repair experiment. RT-PCR assay was used to detect the effect of celecoxib on MMP-9 mRNA and VEGF mRNA expressions in SGC-7901 cells. Results The migration distance of SGC-7901 cells was decreased with the increased concentration of celecoxib. There were significant differences between medium-dose and high-dose celecoxib groups and the control group. RT-PCR assay showed that MMP-9 mRNA and VEGF mRNA expressions in gastric cancer SGC-7901 were significantly decreased with the increased concentration of celecoxib, and there was a significant difference compared with those of control group (P<0.05). Conclusion Celecoxib can inhibit migration of human gastric cancer SGC-7901 cells, which may be related to the inhibition of COX-2 downstream of MMP-9 and VEGF expressions.

Objective To investigate the expression of humanβ-defensin 2 (HBD-2) in gastric mucosa of Helico-bacter pylori (H. pylori) associated gastric mucosa-associated lymphoid tissue (MALT) lymphoma, and the role of HBD-2 in gastric MALT lymphoma. Methods Forty gastric mucosa specimens from patients with H. pylori associated gastric MALT lymphoma were collected. And 36 gastric mucosa specimens from chronic superficial gastritis without H. pylori infection were included as control group. The expression of HBD-2 was detected by immunohistochemistry staining. Results The ex-pression of HBD-2 was significantly higher in H. pylori associated gastric MALT lymphoma than that of control group. (P<0.01). The expression of HBD-2 was significantly decreased after the eradication of H. pylori (P<0.01). The expression of HBD-2 was significantly higher in H. pylori associated gastric MALT lymphoma than that of lymphoma cells (P<0.01). There was no expression of HBD-2 in lymphoma cells. Conclusion HBD-2 is possibly involved in the pathogenesis of H. pylori associated gastric MALT lymphoma. But whether it has anti-tumor effect is not clear.

ABSTRACT

Objective To investigate the impact and clinical significance of helicobacter pylori (Hp) elimination on ghrelin. Methods Forty patients with chronic superficial gastritis (CSG), 42 patients with chronic atrophic gastritis (CAG), 41 patients with peptic ulcer (PU) and 17 patients with gastric adenocarcinoma (CA) were included in this study. All of pa-tients in four groups were Hp-positive. Forty patients with Hp-negative were used as control. The Hp elimination were only performed in CAG,CSG and PU groups. The serum ghrelin and pepsinogen (PG) levels before and after Hp elimination were detected with ELISA assay in CSG, CAG and PU groups. The correlation between PG and glrelin was also detected. The ex-pression of ghrelin in gastric mucosa was detected by RT-PCR. Results Comparing with control group (30.41 ± 8.97), the ghrelin level was increased in PU group (35.42±9.87), but which were decreased in CAG group (18.59±8.19) and CA group (18.33±6.88). There was no significant difference in ghrelin level between CSG group (26.08±9.14) and control group. After Hp elimination, the serum and gastric mucosa ghrelin levels were significantly increased in CSG group (P<0.01), but both serum and gastric mucosa ghrelin levels were significantly decreased in PU group (P<0.01). And no significant difference in the level of ghrelin after Hp elimination in CAG group (P>0.05). A positive correlation was found between serum PGⅠ/PGⅡand serum ghrelin level in CSG, CAG and CA groups (r=0.668,P<0.01). Conclusion Hp elimination has an impact on ghrelin level in patients with upper gastrointestinal diseases. The changes of ghrelin level related to PGⅠ/PGⅡ. Ghrelin can be used as one of the indexes of diagnostic and prognostic evaluation in Hp related upper gastrointestinal diseases.
