ABSTRACT

O livro que temos em mãos nasce de um momento de participação social que foi a Primeira Mostra de experiências dos trabalhadores e trabalhadoras de saúde do município de Manicoré. O título do livro foi inspirado na 17ª Conferência Nacional de Saúde, realizada em 2023, que teve como tema "Garantir direitos, defender o SUS, a vida e a democracia ­ Amanhã vai ser outro dia!". Portanto, o livro inicia na perspectiva de uma escrita coletiva e participativa, que também podemos denominar de Educação Permanente em Saúde porque houve uma reflexão e uma sistematização sobre o fazer e o saber no contexto do trabalho, percorrendo diferentes caminhos da gestão até as comunidades mais distantes. A caminhada foi coletiva, com muitas mãos, vozes, corpos, rostos, imagens e relatos, ou seja, foi um grande mutirão de produção do conhecimento a partir do lugar do trabalho e da gestão nos mais distintos territórios de Manicoré.

ABSTRACT

Lecanemab is a new drug used to treat early Alzheimer's disease (AD) with mild cognitive impairment or mild dementia. It is a human anti-Aβ fibril monoclonal IgG1 antibody, which is injected intravenously into the patient, through the blood-brain barrier into the brain, clearing amyloid plaque, thereby slowing the rate of cognitive decline in patients and delaying disease progression. This article reviews the pharmacological studies, clinical studies, safety and limitations of lecanemab, in order to help clinical understand the current research status and existing achievements of this drug.

ABSTRACT

Objective To assess the retention of 241Am in the whole-body bone of worker by measuring the retention of 241Am in the skull. Methods A whole-body counter with high-purity germanium detector was used to measure the 59.5 keV full-energy peak count of 241Am in the skull. The efficiency of the measurement was calibrated by the digital skull phantom combined with the geometric model of the detector. The retention of 241Am in the worker’s skull was calculated. Results The proportion of skull dry weight relative to the whole-body bone dry weight was used as the coefficient to calculate the 241Am deposition in the whole-body bone from the measured activity of skull 241Am. Conclusion With the retention of 241Am in the skull and the proportion of skull dry weight, the retention of 241Am in the whole-body bone of the worker was calculated to be about 806 Bq.

ABSTRACT

Abstract Simple, precise, accurate and specific spectrophotometric methods are progressed and validated for concurrent analysis of Furosemide (FUR) and Spironolactone (SPR) in their combined dosage form depend on spectral analysis procedures. Furosemide (FUR) in the binary mixture could be analyzed at its λmax 274 nm using its recovered zero order absorption spectrum using constant multiplication method (CM). Spironolactone (SPR) in the mixture could be analyzed at its λmax 238 nm by ratio subtraction method (RS). Concurrent determination for FUR and SPR in their mixture could be applied by amplitude modulation method (AM), absorbance subtraction method (AS) and ratio difference (RD). Linearity ranges of FUR and SPR were (2.0µg/mL-22.0 µg/mL) and (3.0µg/mL-30.0 µg/mL), respectively. Specificity of the proposed spectrophotometric methods was examined by analyzing the prepared mixtures in laboratory and was applied successfully for pharmaceutical dosage form analysis which have the cited drugs without additives contribution. The proposed spectrophotometric methods were also validated as per as the guidelines of ICH. Statistical comparison was performed between the obtained results with those from the official methods of the cited drugs, using one-way ANOVA, F-test and student t-test. The results are exhibiting insignificant difference concerning precision and accuracy

ABSTRACT

The use of small interfering RNAs (siRNAs) has been under investigation for the treatment of several unmet medical needs, including acute lung injury/acute respiratory distress syndrome (ALI/ARDS) wherein siRNA may be implemented to modify the expression of pro-inflammatory cytokines and chemokines at the mRNA level. The properties such as clear anatomy, accessibility, and relatively low enzyme activity make the lung a good target for local siRNA therapy. However, the translation of siRNA is restricted by the inefficient delivery of siRNA therapeutics to the target cells due to the properties of naked siRNA. Thus, this review will focus on the various delivery systems that can be used and the different barriers that need to be surmounted for the development of stable inhalable siRNA formulations for human use before siRNA therapeutics for ALI/ARDS become available in the clinic.

@#<b>Objective</b> To preliminarily study and establish a method for measurement of the transuranic nuclide <sup>241</sup>Am in fecal samples, and to provide technical support for internal radiation monitoring of staff. <b>Methods</b> Fecal samples were collected with a self-made stool sampler and treated with a self-made carbonization and ashing furnace. DGA resin was used to separate and purify <sup>241</sup>Am from fecal samples. With <sup>243</sup>Am as the tracer, the orthogonal method was used for condition optimization. <b>Results</b> The optimum conditions for separation and purification were: the acidity of HNO<sub>3</sub> added into the column, 6 mol/L; column flow rate, 0.6 mL/min; and the volume of analytical solution,12 mL. The method based on inductively coupled plasma mass spectrometry showed a detection limit of 9.79×10<sup>−4</sup> Bq for <sup>241</sup>Am in fecal samples, which was satisfactory and feasible. <b>Conclusion</b> This method fills the vacancy of <sup>241</sup>Am measurement in fecal samples to some extent, which is of practical significance for internal radiation monitoring and protection for analysts.

ABSTRACT

Objective To investigate the radiation dose to operators in the process of 241Am-Be neutron source logging, and discuss neutron source management and protective measures for operators in well logging. Methods Through on-site observation and measurement of 241Am-Be neutron source logging in a company, we obtained the surface γ dose rate and neutron dose rate of the neutron source, as well as the operating time and distance of various processes including source taking, transfer, and loading, calculated the radiation dose to operators in various processes, and analyzed the source and proportion of the personal effective dose to operators. Results The effective doses of neutron irradiation and γ irradiation were 94.17 μSv and 2.72 μSv, respectively, for the combined processes of source tank inspection, transfer, and detection; 36.66 μSv and 24.08 μSv, respectively, for source loading and unloading; and 130.83 μSv and 26.80 μSv, respectively, for the whole neutron source logging process. The total annual effective dose of neutron source logging was 15.78 mSv, as estimated by logging 100 times per year. Conclusion In the process of 241Am-Be neutron source logging in the company, the effective dose to operators mainly arises from neutron irradiation. Therefore, it is necessary to strengthen neutron source management and take effective protective measures against neutron radiation.

O objetivo do presente estudo foi analisar as oportunidades de estimulação motora em ambientes domiciliares e fatores associados em crianças de Parintins/Amazonas. Participaram 300 pais/responsáveis por crianças da cidade de Parintins/Amazonas. Affordances in the Home Environment for Motor Development foi utilizado para avaliar oportunidades de estimulação motora. A prevalência de baixa estimulação moto-ra foi 78% maior nas casas em que moravam quatro ou mais adultos (RP: 1,78; IC95%: 1,07-2,87); mães com o ensino fundamental (RP: 15,93; IC95%:2,45-17,50) e ensino médio (RP: 10,07; IC95%: 1,46-54,01) forne-cem baixa estimulação motora; e crianças do sexo masculino apresenta-ram maior probabilidade de baixa estimulação motora (RP: 1,56; IC95%: 1,09-1,94). Identificamos que lares nessa região não ofertam oportuni-dades de estimulação motora adequada para as crianças.


The objective was to analyze the opportunities for motor stimulation of home environments and associated factors in children from Parintins/Amazonas. Participants included 300 parents/guardians of children from the city of Parintins/ Amazonas participated. Affordances in the Home Environment for Motor Development was used to assess opportunities for motor stimulation. The prevalence of low motor stimulation was 78% higher in the homes where four or more adults lived (PR: 1.78; 95% CI: 1.07-2.87); mothers with elementary school (PR: 15.93; 95% CI: 2.45-17.50) and high school (PR: 10.07; 95% CI: 1.46-54.01) provide low motor stimulation; and male children were more likely to have low motor stimulation (PR: 1.56; 95% CI: 1.09-1.94). We identified that homes in this region do not offer opportunities for adequate motor stimulation for children.


El objetivo fue analizar las oportunidades de estimulación motora de los ambientes domésticos y factores asociados en niños de Parintins/Amazonas. Los participantes incluyeron a 300 padres/tutores de niños de la ciudad de Parintins/Amazonas. Se utilizó Affordances in the Home Environment for Motor Development para evaluar las oportunidades de estimulación motora. La prevalencia de estimulación motora baja fue 78% mayor en los hogares donde vivían cuatro o más adultos (RP: 1,78; IC 95%: 1,07-2,87); las madres con escuela primaria (RP: 15,93; IC del 95%: 2,45-17,50) y secundaria (RP: 10,07; IC del 95%: 1,46-54,01) proporcionan una estimulación motora baja; y los niños varones tenían más probabilidades de tener una estimulación motora baja (PR: 1,56; IC del 95%: 1,09-1,94). Identificamos que los hogares en esta región no ofrecen oportunidades para la estimulación motora adecuada para los niños.

ABSTRACT

Acute respiratory distress syndrome (ARDS) is characterized by the severe inflammation and destruction of the lung air-blood barrier, leading to irreversible and substantial respiratory function damage. Patients with coronavirus disease 2019 (COVID-19) have been encountered with a high risk of ARDS, underscoring the urgency for exploiting effective therapy. However, proper medications for ARDS are still lacking due to poor pharmacokinetics, non-specific side effects, inability to surmount pulmonary barrier, and inadequate management of heterogeneity. The increased lung permeability in the pathological environment of ARDS may contribute to nanoparticle-mediated passive targeting delivery. Nanomedicine has demonstrated unique advantages in solving the dilemma of ARDS drug therapy, which can address the shortcomings and limitations of traditional anti-inflammatory or antioxidant drug treatment. Through passive, active, or physicochemical targeting, nanocarriers can interact with lung epithelium/endothelium and inflammatory cells to reverse abnormal changes and restore homeostasis of the pulmonary environment, thereby showing good therapeutic activity and reduced toxicity. This article reviews the latest applications of nanomedicine in pre-clinical ARDS therapy, highlights the strategies for targeted treatment of lung inflammation, presents the innovative drug delivery systems, and provides inspiration for strengthening the therapeutic effect of nanomedicine-based treatment.

ABSTRACT

Objective To design and build a set of experimental equipment for neutron radiation irradiation by using the 241Am-Be neutron source. Methods In the preliminary work, the spatial distribution data of the neutron energy spectrum and the gamma energy spectrum inside and outside the device were simulated by Monte Carlo method, and the changes of radiation fluence rate with spatial distribution was studied. The model of the 241Am-Be neutron device was established, and the neutron transport process in the irradiation field was studied using the method of shadow cone, inverse square law and other data analysis methods. Results Based on the simulation results, the normalized effective does of fast neutron fluence at the measurement point is about 72.9 pSv/n, and the one of photon fluence is about 3.04 pSv/γ. The ratio of effective dose of photon fluence to neuteon is about 4.17%. Conclusion Using Monte Carlo method, a standard model of 241Am-Be neutron source was constructed, the shadow cone design was optimized, and the feasibility of using the shadow cone conversion method to establish a standard neutron source radiation device was discussed.

ABSTRACT

The 18 kDa translocator protein (TSPO), previously known as the peripheral benzodiazepine receptor, is predominately localized to the outer mitochondrial membrane in steroidogenic cells. Brain TSPO expression is relatively low under physiological conditions, but is upregulated in response to glial cell activation. As the primary index of neuroinflammation, TSPO is implicated in the pathogenesis and progression of numerous neuropsychiatric disorders and neurodegenerative diseases, including Alzheimer's disease (AD), amyotrophic lateral sclerosis (ALS), Parkinson's disease (PD), multiple sclerosis (MS), major depressive disorder (MDD) and obsessive compulsive disorder (OCD). In this context, numerous TSPO-targeted positron emission tomography (PET) tracers have been developed. Among them, several radioligands have advanced to clinical research studies. In this review, we will overview the recent development of TSPO PET tracers, focusing on the radioligand design, radioisotope labeling, pharmacokinetics, and PET imaging evaluation. Additionally, we will consider current limitations, as well as translational potential for future application of TSPO radiopharmaceuticals. This review aims to not only present the challenges in current TSPO PET imaging, but to also provide a new perspective on TSPO targeted PET tracer discovery efforts. Addressing these challenges will facilitate the translation of TSPO in clinical studies of neuroinflammation associated with central nervous system diseases.

ABSTRACT

Los hongos micorrízicos arbusculares (HMA) son biotrofos obligados que viven en asociación simbiótica con las raíces de las plantas. Se encuentran entre los microorganismos del suelo más extendidos que proporcionan a la planta huésped nutrientes y protección contra patógenos. Las prácticas agrícolas modernas, como la labranza frecuente, el alto empleo de fertilización inorgánica pesticidas junto con condiciones climáticas cambiantes debido al calentamiento global, tienen enormes impactos en la colonización de los HMA, la interacción con las plantas y la productividad de los cultivos. Los HMA afectan positivamente la tolerancia de las plantas al estrés biótico y abiótico, a los ecosistemas severos y sus patógenos al alterar la estructura de las raíces, la exudación, la microflora de la rizosfera, la producción de antifúngicos y antibacterianos, y al competir con los patógenos por la absorción de nutrientes. Por lo tanto, juegan un papel importante en el crecimiento, la productividad y la calidad de las plantas. Además, el efecto de un fungicida varía según su modo de acción y las especies de HMA asociadas, lo que sugiere que estos hongos tienen un gran potencial como herramienta para la agricultura sostenible ecológica en el actual escenario de calentamiento global.


Arbuscular Mycorrhizal Fungi (AMF) are obligate biotrophs living in symbiotic association with roots of plants. They are among the most widespread soil microorganisms that provide the host plant with nutrients and pathogen protection. Modern farming practices like frequent tillage, high input inorganic fertilization and pesticide along changing climatic conditions due to global warming, have huge impacts on AMF colonization, interaction with plants and on crop productivity. AMF positively affect the plant tolerance to biotic and abiotic stresses, harsh ecosystems and plant pathogens by altering root structure, exudation, rhizosphere microflora, production of antifungals, antibacterials, and competing with pathogens for nutrient uptake. Thus, it plays a significant role in plant growth, productivity and quality. Further, the effect of a fungicide is varied depending on its mode of action and the associated AMF species, suggesting that these fungi have a strong potential as a tool for eco-friendly sustainable farming in the present scenario of global warming.

ABSTRACT

OBJECTIVE@#To compare the infuences on circadian rhythm of blood pressure in the patients with non-dipper essential hypertension between the combined treatment of time acupuncture and western medication and the simple western medication.@*METHODS@#A total of 70 patients with non-dipper essential hypertension were randomized into an acupuncture plus western medication group (35 cases, 2 cases dropped out) and a western medication group (35 cases). In the western medication group, levamlodipine maleate tablets were taken orally, 2.5 mg each time, once daily. In the acupuncture plus western medication group, on the base of the treatment as the western medication group, acupuncture was applied specially in the period of the day from 7:00 am to 9:00 am. The acupoints included Fengchi (GB 20), Zhongwan (CV 12), Tianshu (ST 25), Hegu (LI 4), Quchi (LI 11), Zusanli (ST 36), etc. Acupuncture was given once daily, 5 treatments a week. The duration of treatment in the two groups was 4 weeks. The clinic blood pressure before and after treatment, 24 h ambulatory blood pressure and the levels of serum melatonin (MT) and 5-serotonin (5-HT) were observed in the two groups.@*RESULTS@#The total effective rate of anti-hypertension was 75.8% (25/33) in the acupuncture plus western medication group, better than 54.3% (19/35) in the western medication group (<0.05). The 24 h average systolic blood pressure, the daytime average systolic blood pressure, the daytime average diastolic pressure, and the nighttime average systolic blood pressure were all reduced after treatment in the two groups (<0.05). The reduction effect of the aforementioned 4 indexes in the acupuncture plus western medication group was much more obvious as compared with the western medication group (<0.05). After treatment, the serum level of MT was increased and 5-HT decreased in the patients of two groups (<0.05). The serum level of MT in the acupuncture plus western medication group was higher than that in the western medication group and the level of 5-HT was lower than the western medication group (<0.05).@*CONCLUSION@#Time acupuncture therapy in the period of the day from 7:00 am to 9:00 am, combined with western medication effectively reduce blood pressure and regulate the levels of serum MT and 5-HT so as to maintain the circadian rhythm of blood pressure in patients with non-dipper essential hypertension. The therapeutic effect of this combined treatment is superior to simple western medication.

ABSTRACT

The trend toward designing large hydrophobic molecules for lead optimization is often associated with poor drug-likeness and high attrition rates in drug discovery and development. Structural simplification is a powerful strategy for improving the efficiency and success rate of drug design by avoiding "molecular obesity". The structural simplification of large or complex lead compounds by truncating unnecessary groups can not only improve their synthetic accessibility but also improve their pharmacokinetic profiles, reduce side effects and so on. This review will summarize the application of structural simplification in lead optimization. Numerous case studies, particularly those involving successful examples leading to marketed drugs or drug-like candidates, will be introduced and analyzed to illustrate the design strategies and guidelines for structural simplification.

ABSTRACT

Objective To observe the effect of varying intensities of water-jet force on autologous fat graft viability.Methods Lipoaspirate was taken from 12 female patients undergoing waterjet assisted abdominal liposuction at our department.According to the intensity of water-jet force,the experimental group was divided into four subgroups:R1 (pressure,30 bar),R2 (pressure,50 bar),R3 (pressure,70 bar) and R4 (pressure,90 bar).Hand-held suction was taken as the control group C.Adipose tissue was filtered with cotton cushion and centrifuged at low speed,and the composition ratio of water and fat tissue from each group was observed.Calcein-AM/Hoechst 33342 staining was used to detect the viability of adipocytes.Results Fat aspirates was divided into four layers:oil layer,pure fat tissue,liquid and bottom sediment.Oil ratios of R1,R2,R3,R4 and C were (8.9 ± 2.3) %,(9.6±2.1)%,(10.3±1.3)%,(14.2±1.6)% and (9.5±1.8)%,respectively.There was no statistically significant difference between R1,R2,R3 and C (P>0.05).Statistically significant difference was found between R4 and other groups (P<0.001).Viability of adipocytes from R1,R2,R3,R4 and C groups were (88.1±2.8)%,(89.9±1.9)%,(84.8±2.3)%,(78.0±1.7)% and (91.1±2.9)% respectively.There was no statistically significant difference between R1,R2 and C (P> 0.05).Statistically significant difference was found between R3,R4 and C (P < 0.05).Conclusions Viability of fat graft harvested under lower intensity of water-jet force (R1,R2) is higher than that harvested under higher intensity of water-jet force (R3,R4).

Objective: To study the effects of different inoculation periods on seedling growth and steroidal saponin content of Paris polyphylla var. yunnanensis, in order to lay a foundation for cultivating and domesticating high-quality Paris polyphylla var. yunnanensis. Methods: The content of steroidal saponins I, II, VI, and VII in the different vaccination periods of Paris polyphylla var. yunnanensis was determined by HPLC. Meanwhile, trypan blue, UV spectrophotometry and so on were adopted to explore colonization rate, colonization intensity, physiological and biochemical indexes, rhizome biomass of the roots of Paris polyphylla var. yunnanensis. Results: Infection rate of AM fungi was higher in different inoculation periods, activities of protective enzymes, photosynthetic pigments and soluble sugars were increased, soluble protein was not changed, content of MDA was decreased, stress resistance of the seedlings of Paris polyphylla var. yunnanensis was improved, and the growth and development were good. Quality of the cultivar one-year seedlings (recovered in August 2015, T7) was relatively low, and cultivar one-year seedlings (harvested in June or July 2015, T5 and T6) and cultivar two years seedlings (recovered in August 2015, T8) were the best. S2, S3 and S6 treatment groups had better effect in different AM fungi mixed groups. Conclusion: Seedling growth and steroidal saponin content of Paris polyphylla var. yunnanensis inoculated by different inoculation periods had certain effects.

ABSTRACT

Objective To screen preponderant arbuscular mycorrhizal (AM) fungi species from Paris polyphylla var. yunnanensis seedlings based on mycorrhiza viability, rhizome biomass, and active components. Methods After 28 exogenous species of AM fungi were inoculated in sterilized soil, the effects of the mycorrhiza viability of seedling, the biomass in rhizome, the total amount of polyphyllins in single whole seedling, the amount of polyphyllins in rhizome, and the amount of nucleosides in rhizome of P. polyphylla var. yunnanensis were observed by inoculation test in pot at room temperature. Results All the 28 AM groups formed well mycorrhizal structure, the mycorrhiza viability of P. polyphylla var. yunnanensis seedling was enhanced, but it showed preference. Most of the AM treatment groups could improve the biomass in rhizome, the contents and yields of polyphyllins increased, which implied that the accumulation of polyphyllins and nucleosides in P. polyphylla var. yunnanensis seedling were enhanced after inoculation. AM also changed polyphyllins and nucleosides allocation in different organs of P. polyphylla var. yunnanensis seedling in the secondary metablism, the total amount of four kinds of polyphyllins in rootlet were higher than those in rhizome, but the total amount of nine kinds of nucleosides in rhizome were higher than rootlet. Conclusion The performance of different AM strains in this study is of great reference for further improving the biological quantity and quality of the screening of preponderant AM fungi species from P. polyphylla var. yunnanensis seedlings. Gigaspora gigantean, Scutellospora calospora, Diversispora spurca, Dentiscutata heterogama, Septoglomus deserticola, and Ambispora leptoticha can use as the preponderant AM fungi species for the cultivation of mycorrhizal seedlings of P. polyphylla var. yunnanensis.

[Objective]To investigate the effects of ghrelin on inflammatory signaling protein kinase B(Akt),nuclear factor-κB(NF-κB)and inducible nitric oxide synthase(iNOS)in alveolar macrophage(AM).[Methods]24 Male SD rats were randomly divided into Sham,CLP,CLP+ghrelin,and Sham+ghrelin groups. Cecal ligation and puncture(CLP)was used to induce sepsis. Ghrelin(20 nmol/kg)was administered by intraperitoneal injection at 3 h and 15 h post-operation. Histopathological changes of lungs were observed and scored.AM were extracted from bronchoalveolar lavage fluid(BALF). Interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)in BALF were detected by ELISA. IL-1β,TNF-α,and IL-6 mRNA in AM were detected by qPCR.NF-κB p65,IκBα,p-IκBα,Akt,p-Akt and iNOS in AM were detected by immunofluorescence(IF)and Western blotting.[Results]The histologic score(6.7±0.8),BALF IL-1β[(146±12)pg/mL]and IL-6[(182±10)pg/mL]from CLP+ghrelin group were respectively 35.4%,44.5% and 46.42% lower than those from CLP group[(10.3±0.7),(263±17)pg/mL,and(273±5)pg/mL],P<0.05.No significant difference was found in BALF TNF-α between CLP group and CLP+ghrelin group.The IL-1β,TNF-α and IL-6 mRNA in AM from CLP+ghrelin group were respectively 54.38%,53.6% and 46.42% lower than those from CLP group,P<0.05. The nuclear NF-κB p65 and cytoplasmic p-IκBα,p-Akt and iNOS from CLP+ghrelin group were respectively 32.58%,45.42%,27.6% and 48.33% lower than those from CLP group,P<0.05. There was no significant difference in all data between Sham group and Sham+ghrelin group.[Conclusion]Ghrelin can decrease the activity of inflammatory signaling proteins Akt,NF-κB and iNOS in AM,therefore restricts AM expressing pro-inflammatory cytokines IL-1β,TNF-α,and IL-6,thus alleviates sep-sis-induced acute lung injury(ALI).

ABSTRACT

Objective To develop a HPLC-MS/MS methodfor determination of cyclosporin A(CSA)and AM1 in bone marrow transplant patient,and explore the relationship of CSA and its main metabolite AM1 within individual and between individuals,and provide reliable basis for clinical rational use of monitoring in CSA blood drug concentration.Methods CSD was used as internal standard,and whole blood samples were treated with internal standard methanol precipitated protein.The column was Ultimate XB-C18 with a column temperature of 65 ℃ and the mobile phase was eluted with 0.1% of formic acid and 2 mmol/L ammonium acetate in water and methanol containing 0.1% formic acid.The mass spectrometry was detected by electrospray ion trap positive ion mode,MRM scanning,monitoring CSA 1219.9 to 1203.1 m/z,AM1 1236.1 to 1219.1 m/z,CSD 1234.0 to 1217.0 m/z.Results The concentration of CSA was linear in the range of 16 to 1600 ng/mL,Y=0.0143X+0.0213(r=0.9976).The concentration of AM1 was linear in the range of 10~1000 ng/mL,Y=0.00363X-0.00528(r=0.9973).The ratio of CSA to AM1 (AM1/CSA) between individual ranged from 32% to 356%.The ratio of CSA to AM1 within indiviolual(AM1/CSA) ranged from 27 % to 147 %.Conclusion HPLC-MS/MS method for the simultaneous detection of CSA and AM1 was established.The variation of CSA exists in the bone marrow transplant patient between individuals and within individual;the HPLC-MS/MS method can be used for monitoring of whole blood concentration of CSA in bone marrow transplant patients.

ABSTRACT

Objective T o explore the effectiveness of direct am plification for the ST R analysis of carti-lage, and to accelerate the effectiveness of disaster victim identification. Methods E ighty-eight cartilage sam ples w ere directly am plified by Pow erPlex誖21 kit, and the results of genotyping w ere com pared w ith that obtained by the m agnetic beads m ethod. Results In 88 cartilage sam ples, the ST R genotypes w ere successfully detected from 84 sam ples by direct am plification and m agnetic beads m ethod, and both the results of genotyping by tw o m ethod w ere consistent. Conclusion D irect am plification w ith Pow er-Plex誖21 kit can be used for ST R genotyping of cartilages. T his m ethod is operated easily and prom ptly, w hich has a potential application in the individual identification of m ass disasters.
