ABSTRACT

Objective To observe the effect of ultrafiltrated components of three kinds of serum containing phlegm-resolving prescriptions (Huanglian Wendan Decoction, Gualou Xiebai Banxia Decoction, and Erchen Decoction, respectively) on viability of human leukemic monocytic cell line THP-1 derived macrophages and macrophage differentiation to foam cells. Methods THP-1 cells were induced to differentiate into macrophages with phorbol-12-myristate-13-acetate ( PMA) . And then the achieved macrophages were used for the establishment of the atherosclerotic foam cell model after co-incubation with oxidized low density lipoprotein ( ox-LDL ) . Three kinds of serum containing phlegm-resolving prescriptions and blank serum were prepared and ultrafiltered by 10 KD ultrafiltrator, and then were co-cultured with the foam cell model, respectively. The viability of macrophages and the macrophage differentiation to foam cells were tested by methyl thiazolyl tetrazolium ( MTT) assay and oil red staining. Apoptotic rate of macrophages was determined by flow cytometry. Results After THP-1 cells were induced by PMA (100 nmol/L) for 20 hours and then were co-incubated with ox-LDL (50 μg/mL) for 24 hours, a typical atherosclerotic foam cell model was established successfully. The ultrafiltrated components of serum containing Huanglian Wendan Decoction had obvious effects on increasing the macrophage survival rate, decreasing macrophage apoptotic rate, and inhibiting macrophage differentiation to foam cells. The ultrafiltrated components of serum containing Gualou Xiebai Banxia Decoction or Erchen Decoction had an effect on enhancing the macrophage differentiation to foam cells but had no anti-apoptosis action. Conclusion Huanglian Wendan Decoction has anti-atherosclerosis action, which may be related to resisting macrophage apoptosis and decreasing macrophagic lipid-endocytosing function. Erchen Decoction and Gualou Xiebai Banxia Decoction are effective on increasing macrophagic lipid-endocytosing function.

ABSTRACT

[Objective] To observe the preventive and therapeutic effect of Anxin Granules in the treatment of hyperlipidemia [Methods] Thirty - two New Zealand male rabbits were randomized into 4 groups. Groups A, B, C and D were given with common forage, high fat forage, high fat forage with Zhibituo Tablets, high fat forage with Anxin Granules respectively. After 10 weeks of treatment, serum levels of total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol ( LDL-C), high-density lipoprotein cholesterol (HDL-C) and malondialdehyde ( MDA) and activity of superoxide dismustase (SOD) were detected. [ Results ] Anxin Granules decreased the serum levels of TC, TG, LDL-C and MDA. [Conclusion] Anxin Granules has a good effect in preventing and treating hyperlipidemia and atherosclerosis by regulating blood lipid metabolism.

ABSTRACT

【Objective】To explore the relationship between damp-heat syndrome(DHS)and aortic atheroslerosis(AA),and to observe the effect of heat-clearing,dampness-resolving and stasis-removing therapy on AA.【Methods】DHS rabbit models were established by feeding with fatty diet and intravenous injection of endotoxins.The development of AA were observed under light microscope and electron microscope to evaluate the effect of heat-clearing,dampnessresolving and stasis-removing therapy.【Results】 Compared with the blank control group,an obvious atherosclerotic changes were showed in the model rabbits: injury of arterial intima,appearance of atherosclerotic plaque,formation of foam cells and proliferation of smooth muscle cells.After treatment,the above damages were relieved.【Conclusion】There has histological evidence for the pathogenesis of internal retention of damp-heat causing blood stasis,and heat-clearing,dampness-resolving and stasis-removing therapy is effective for AA.

ABSTRACT

[Objective] To investigate the influence of serum containing Danggui Buxue Decoction (DBD) on chemotaxis of oxidized low-density lipoproteins ( OxLDLs) for monocytes ( MC). [ Methods ] Six New Zealand rabbits were randomized to DBD group, Radix Angelicae Sinensis (RAS) group and Radix Astragali seu Hedysari (RASH) group. The rabbits were treated by gastric infusion of the above drugs respectively, bid for three days. On the third day, the serum containing drugs was obtained by heart blood sampling one hour after administration. Free chemotaxis group (normal control group) and OxLDLs group ( model group) were established for control. The number and figure of OxLDLs - chemotactic MC were observed by micropore filter method with chemotactic chamber. ABSTRACT

[Objective] To observe the preventive effect of Huanglian Jiedu Decoction (HJD), a prescription with the actions of clearing heat and removing toxicity, on rabbits atherosclerosis (AS) induced by Chlamydia pneumoniae (CP) infection. [Methods] Fifty two New Zealand rabbits were fed with forage containing 2. 5g/kg cholesterol and were infected with CP via nasopharynx for three times during 6 weeks. At the end of the sixth week, forty four rabbits with serum CP-IgG positive were randomized into 4 groups: group A treated with HJD 2g?kg-1 ?d-1 by gastric gavage, group B with HJD 1g?kg-1?d-1, group C with azithromycin 20 mg?kg-1?d-1 and model group D with normal saline for 6 weeks. Group E was set up in 8 rabbits fed with fatty forage and served as the control. Serum levels of high-sensitive C reactive protein (hs-CRP) and CP-IgG were detected before infection, after infection and after treatment. At the end of the 18th week, the rabbits were executed and the pathological features of aortic tissue were observed under microscope. Meanwhile, the expression of CP-DNA and tumor necrosis factor ?(TNF-?) in the aortic tissue was detected with polymerase chain reaction (PCR) and immunohistochemical method respectively. [Results] At the end of the experiment, atherosclerotic changes were obvious in groups D and E, particularly in group D. Compared with group D, the atherosclerotic damage was much relieved, and AS indexes such as maximum intimal thickness (MIT), atherosclerotic damage percentage and plaque area index were much improved in groups A and C (P

Objective To explore the possibility of Lipi Huatan Formula (LHF) in inhibit ing the occurrence of early atherosclerosis and its mechanism.
