ABSTRACT

The aim of the present study was to determine and compare the fatty acids, lipid soluble vitamins, sterols, phenolics, and antioxidant capacities of three endemic Salvia L. taxa (S. euphratica var. Montbret & Aucher ex Bentham leiocalycina (Rech. Fil.) Hedge, S. euphratica var. Montbret & Aucher ex Bentham euphratica (Rech. Fil.) Hedge and S. pseudoeuphratica Rech.Fil.) and to evaluate these results systematically. The fatty acid compositions were determined by using gas chromatography, while the lipid soluble vitamins, sterols, and phenolics were determined by using HPLC. Also, the antioxidant capacities of three Salvia taxa were measured in vitro. Palmitic acid was found as major saturated fatty acid while oleic acid, linoleic acid, α-linolenic acid, and erucic acid were found as major unsaturated fatty acids in the present study. It was found that S. euphratica var. euphratica had lower palmitic acid (8.94 ± 0.71%), total saturated fatty acid (19.16 ± 0.15%), and higher unsaturated fatty acid content (82.08 ± 0.52%) than other studied taxa. Furthermore, it was shown that S. euphratica var. euphratica had different 18.3/18:2 (0.36) unsaturated/saturated fatty acid (4.28) ratios. However, this study demonstrated that Salvia taxa had low lipid soluble vitamins, sterol contents. On the other hand, it was shown that Salvia taxa had similar catechin (509.2 ± 4.21 µg/g and 552.2 ± 9.21 µg/g) and vanillic acid amounts (351.2 ± 2.17 µg/g and 396.8 ± 4.1 µg/g) in this study. And also, it was found that Salvia taxa had high rosmarinic acid content while S. euphratica var. leiocalycina had the highest rosmarinic acid content (1480 ± 7.57 µg/g). On the other hand, it was shown that the two ferulic acid contents of S. euphratica varieties were higher (1175 ±/5.21 µgmg-1740.2 ± 4.82 µg/mg) than the ferulic acid content of S. pseudoeuphratica of which was the lowest (19.2 ± 0.97 µg/mg). O objetivo do presente estudo foi determinar e comparar os ácidos graxos, vitaminas lipossolúveis, esteróis, fenóis e capacidades antioxidantes de três espécies endêmicas de Salvia L. Além disso, foi demonstrado que a S. euphratica var. euphratica apresentou diferentes proporções 18:3/18:2 (0.36) de ácidos graxos insaturados/saturados (4.28). No entanto, este estudo demonstrou que o gênero Salvia tinha baixo teor de vitaminas lipossolúveis e baixo conteúdo de esteróis. Por outro lado, foi demonstrado que as espécies do gênero Salvia contém quantidades de catequinas (509.2 ± 4,21 µg/mg-552.2 ± 9.21 µg/mg) e ácido vanílico semelhantes (351.2 ± 2.17 µg/mg 396,8 ± 4,1 µg/mg). Descobriu-se também que o gênero Salvia tinha alto conteúdo de ácido rosmarínico enquanto a espécie S. euphratica var. leiocalycina apresentou o maior teor desse ácido (1.480 ± 7.57 µg/g). ABSTRACT

Abstract In the current study, nanocapsules (NC) formulations containing a co-load of clotrimazole (C), a highly prescribed antifungal drug, and diphenyl diselenide [(PhSe)2], an organoselenium compound with a promising scope of pharmacological actions, were prepared. Formulations were characterized as well as the potential toxicity, antioxidant action, and antifungal effect were assessed using in vitro techniques. The NCs were prepared employing Eudragit® RS 100 as polymeric wall and medium chain triglycerides or virgin coconut oil (CO) as core. All NC suspensions had pH around acid range, compound content close to theoretical value (1 mg/mL/drug), average diameter in nanometric range, positive values of zeta potential as well as high encapsulation efficacy and mucoadhesive property. Physicochemical stability was performed over a 30-day period and showed no modification in the aforementioned parameters to all samples. Preliminary screening of toxicological potential performed by the hen's egg test chorioallantoic membrane technique classified the formulations as non-irritant. The DPPH radical assay revealed that nanoencapsulated compounds had superior antioxidant action in comparison to their free forms (concentration range tested 1.0-25.0 µg/mL). Importantly, the formulation composed of CO and containing C and (PhSe)2 showed the highest antioxidant potential and was selected for further investigation regarding antifungal effect against some Candida spp strains. Results of in vitro antifungal assay demonstrated that the C and (PhSe)2 co-encapsulation had a minimum inhibitory concentration (MIC) values around 60. Thus, our study supplies additional data about advantages achieved by encapsulating active compounds.

ABSTRACT

OBJECTIVE: To examine the antioxidant properties of the total flavonoids from Lithocarpus polystachyus Rehd (TFL) in vitro and in vivo. METHODS: The TFL was obtained by ultrasound-assisted extraction with 70% ethanol and purified with D-101 macroreticular resin. DPPH, ABTS, reducing power and ORAC of TFL were analyzed for antioxidant activity in vitro, with VC and rutin as control. Acute liver injury mice model was induced with carbon tetrachloride (CCl4), the effect of TFL on the superoxide dismutase (SOD), catalase (CAT), glutathione (GSH) and malondialdehyde (MDA) levels were examined. RESULT: At 1 mg·mL-1 of TFL, the DPPH and ABTS radicals-scavenging activities was 81.25% and 85.54% respectively, while the reducing power was slightly lower than that of VC but higher than that of rutin. In the concentration of 1%, 2% and 3%, the value of ORAC was 47.86, 65.43 and 81.9 μmol TE·g-1 respectively. In mice with CCl4-induced actute liver injury model, TFL at the concentration of 100, 200 and 400 mg·mL-1 significantly decreased the MDA content, and increased the activities of SOD, CAT and GSH. CONCLUSION: The total flavonoids from Lithocarpus polystachyus Rehd. have a strong antioxidant effect.

ABSTRACT

Carotenoids from some of the coloured yeasts like Rhodotorula, Phaffia rhodozyma have attracted commercial interest as a natural pigment for foods. Red yeast isolated from contaminated Potato dextrose agar plate (PDA), designated as Rhodotorula glutinis DFR-PDY has been found to produce carotenoids. In the present study toxicological evaluation of carotenoid pigment has been reported. Experiment was conducted on 3 groups of albino rats. One group with vehicle control (palm oil) and 2 groups with two different doses of red yeast pigment (lower and higher dose) were fed to rats (both male and female) by gavages for 13 weeks. Gain in body weight of rats and food consumption were monitored at regular intervals. Hematological studies revealed that there is no much difference in erythrocytes, packed cell volume, Mean corpuscular volume (MCV), Mean corpuscular haemoglobin concentration (MCHC), platelets and differential counts. Total leucocyte count (TLC) is less in case of higher dose group than the lower and control groups. Whereas, hemoglobin is more in case of higher dose than the lower dose group and least in control group. Even clinico-chemical parameters and urine analysis of vehicle control group and pigment fed rats revealed that there were no major differences between them as well as between two different genders of rats and also interaction between different doses and the genders. Histopathology of these experimental animals revealed that there are no major histological changes found between the groups. It may be concluded that the whole pigment extract from R. glutinis DFR-PDY may be used safely in food preparations as food colourant with an added benefit of antioxidant activity .

ABSTRACT

Six pentacyclic triterpenes were isolated from hexane extract of stems of Maytenus salicifolia Reissek, Celastraceae: 30-hydroxyfriedelan-3-one (1), 3,16-dioxofriedelane (2), friedeline (3), lupeol (4), betuline (5) and lup-20(29)-en-3,30-diol (6). The structure each one was established on the basis of detailed ¹H and 13C NMR spectral investigation and by comparison with the respective literature values. For compound 1, the complete 2D NMR (HMBC, HMQC and NOESY) spectral data were herein reported for the first time. Compounds 1, 2, 5 and 6 were isolated for the first time from this plant. Antioxidant activity is described for some extracts from species of the Celastraceae family, then, the extracts from aerial parts of M. salicifolia were evaluated in relation to antioxidant potential using the DPPH method. Compared to quecertin, the AcEt extract (EAF) from leaves, AcEt (EAPF) and MeOH (EMPF) from pulp fruit and AcEt (EAT) and MeOH (EMT) from stems showed significant antioxidant property.

ABSTRACT

Periodontitis is a bacterially induced chronic inflammatory disease that destroys the connective tissue and bone that support teeth. Bacteria initiates periodontitis and destruction of the alveolar bone and periodontal connective tissue is clearly observed. But, the events occuring between these two points of time remain obscure and this study focusses on these aspects. The proanthocyanidins (PC) have variable pharmacological and nutraceutical benefits including improvement of ischemic cardiovascular disease, prevention of atherosclerosis and antiarthritic, anticancer and antimicrobial activities. The benefits associated with the antioxidant activity of PC have been evaluated both in vivo and in vitro. But, reports on the ameliorative effects of PC on oral diseases and specifically on periodontitis are very few. Hence, a novel attempt is made to review the possible protective effects of PC and its mechanism of action in periodontitis and also to show whether PC could be developed as a therapeutic agent for periodontitis.

ABSTRACT

<p><b>OBJECTIVE</b>To identify the hepatoprotective and in vitro antioxidant activity of Lumnitzera racemosa (L. racemosa) leaf extract.</p><p><b>METHODS</b>Animals in Group 1 served as vehicle control, Group 2 served as hepatotoxin (CCL4 treated) group, Group 3 served as positive control (Silymarin) group, and Group 4, 5 and 6 served as (75, 150 and 300 mg/kg bw p.o.) L. racemosa leaf extract treated groups. Moreover, in vitro antioxidant DPPH, hydroxyl radical scavenging activity (HRSA), NO, ferric reducing antioxidant power (FRAP), lipid hydroperoxide (LPO) and super oxide dismutase (SOD) were also analyzed for the leaf extract.</p><p><b>RESULTS</b>The levels of the serum parameters such as serum glutamic oxaloacetic transaminase (SGOT), serum glutamic pyruvic transaminase (SGPT), alkaline phosphatase (ALP), bilirubin, cholesterol (CHL), sugar and lactate dehydrogenase (LDH) were significantly increased in CCL4 treated rats when compared with the control group (P<0.05). But the L. racemosa leaf extract treated rats showed maximum reduction of SGOT [(210.36±19.63) IU/L], SGPT [(82.37±13.87) IU/L], ALP [(197.63±23.43) IU/L], bilurubin [(2.15±0.84) mg/dL], cholesterol [(163.83±15.63) mg/dL], sugar [(93.00±7.65) mg/dL] and LDH [(1134.00±285.00) IU/L] were observed with the high dose (300 mg/kg bw) of leaf extract treated rats. Histopathological scores showed that, no visible changes were observed with high dose (300 mg/kg bw) of leaf extract treated rats except few mild necrosis. The IC50 values were observed as (56.37±4.87) µg/mL, (57.68±1.98) µg/mL, (64.15±2.90) µg/mL, (61.94±3.98) µg/mL, (94.53±1.68) µg/mL and (69.7±2.65) µg/mL for DPPH, HRSA, NO, FRAP, LPO and SOD radical scavenging activities, respectively.</p><p><b>CONCLUSIONS</b>In conclusion, the hepatoprotective effect of the L. racemosa leaf extract might be due to the presence of phenolic groups, terpenoids and alkaloids and in vitro antioxidant properties.</p>
