ABSTRACT

ObjectiveTo explore the underlying mechanism of bran-fried Atractylodis Rhizoma (AR) in improving gastrointestinal function by comparing the effects of raw AR and bran-fried AR on the small intestine tissue structure and transport-related protein carriers in rats with spleen deficiency syndrome. MethodSeventy male SD rats were randomly divided into a normal group, a model group, high- and low-dose raw AR groups (10, 2.5 g·kg-1), high- and low-dose bran-fried AR groups (10, 2.5 g·kg-1), and a compound glutamine group (9 mg·kg-1), with 10 rats in each group. Except for the normal group, the other six groups were subjected to the spleen deficiency model induced by the method of bitter and cold breaking stagnated Qi and abnormal hunger and fullness for 21 days. After modeling, each treatment group was given medication orally according to the corresponding doses every day for a total of 14 days, and the normal group and the model group were given an equal volume of normal saline orally. During the treatment period, the general survival status, macroscopic syndrome score, daily increase in body weight and food intake, and rectal temperature of the spleen deficiency rats were evaluated, and after the treatment, the rats were sacrificed. The pathological changes in the small intestine tissues of each group were observed by hematoxylin-eosin (HE) staining. The content of serum 5-hydroxytryptamine (5-HT) was detected by enzyme-linked immunosorbent assay (ELISA), and the content of serum D-xylose, lactate, and amylase was detected by colorimetry. The levels of free fatty acid receptor 3 (FFA3) and peptide transporter 1 (PepT1) in small intestinal tissues were detected by the Bradford method, and the protein expression of sodium-dependent glucose transporter 1 (SGLT1) and glucose transporter 1 (GLUT1) in small intestinal tissue was detected by immunohistochemistry. Real-time fluorescence-based quantitative PCR was used to detect the mRNA expression of glucose transporter 2 (GLUT2), sodium/hydrogen exchanger 3 (NHE3), and 5-hydroxytryptamine receptor 4 (5-HT4R). ResultCompared with the normal group, the model group exhibited symptoms of spleen deficiency, such as sluggishness, squint, reduced food intake, and lethargy at the end of modelling, damaged basic structure of the small intestinal mucosal epithelium and lamina propria, increased serum lactate and 5-HT content, and decreased serum amylase and D-xylose (P<0.01). Compared with the model group, all treatment groups showed varying degrees of improvement, with the small intestinal microstructure repaired to different degrees. The daily weight gain, anal temperature, and macroscopic syndrome score of spleen deficiency improved to varying degrees (P<0.05, P<0.01). The serum lactate and 5-HT content decreased to varying degrees, while the serum amylase and D-xylose content increased to varying degrees (P<0.05, P<0.01). The PepT1 content in the small intestinal tissues increased, while the FFA3 content decreased to varying degrees. The protein expression of SGLT1 and GLUT1 increased, while the mRNA expression of GLUT2 and NHE3 increased to varying degrees. The mRNA expression of 5-HT4R decreased to varying degrees (P<0.05, P<0.01). Compared with the high- and low-dose raw AR groups, the high- and low-dose bran-fried AR groups showed significant improvement in general conditions and histopathological improvement of the small intestinal tissues. The daily weight gain, anal temperature, and macroscopic syndrome score of spleen deficiency also improved (P<0.05, P<0.01). The serum lactate and 5-HT content decreased, while the serum amylase and D-xylose content increased (P<0.05, P<0.01). The PepT1 content in the small intestinal tissues increased, while the FFA3 content decreased. The protein expression of SGLT1 and GLUT1 increased, while the mRNA expression of GLUT2 and NHE3 increased. The mRNA expression of 5-HT4R decreased significantly (P<0.05, P<0.01). ConclusionBran-fried AR can improve the spleen deficiency-related symptoms and histopathology of the small intestinal tissues in spleen deficiency model rats. Its mechanism may be related to the regulation of the expression of various transport-related protein carriers and the secretion of various digestive enzymes after stir-frying of AR, thus restoring the absorption and transport function of the small intestine.

ABSTRACT

ObjectiveTo explore the regulatory effect of polysaccharides and n-butanol fractions of Atractylodis Rhizoma stir-fried with bran on the plasma metabolites of spleen-deficient rats, and then to elucidate their mechanisms of spleen-enhancing effects. MethodForty male SD rats were randomly divided into the blank group, model group, polysaccharide group (FD group, 0.075 6 g·mL-1·d-1), n-butanol fractions group (FZ group, 0.012 1 g·mL-1·d-1), with 10 rats in each group. Except the blank group, the other three groups used the compound factors of overwork, dietary disorders and intragastric administration of Sennae Folium decoction to replicate the rat model of spleen deficiency. After the end of modeling, the FD group and FZ group were given the corresponding medicinal solution by gavage for 7 d, meanwhile, the blank group and model group were given an equal volume of saline. The plasma samples from rats in the blank, model, FZ and FD groups were analyzed by ultra-high performance liquid chromatography-quadrupole/electrostatic field orbitrap high-resolution mass spectrometry (UHPLC-Q-Orbitrap-MS), multivariate statistical methods were used to process the data and screen differential metabolites, and metabolic pathway enrichment analysis of the screened differential metabolites was performed using the Kyoto Encyclopedia of Genes and Genomes (KEGG))database and MetaboAnalyst 5.0. ResultThe results of multivariate statistical analysis showed that there were significant differences in plasma metabolites between the model group and blank group, FZ group and model group, FD group and model group. There were 380 differential metabolites between the blank group and the model group, of which 78 and 57 were called back by polysaccharides and n-butanol fractions of Atractylodis Rhizoma stir-fried with bran, respectively. Metabolic pathway enrichment results showed that the n-butanol fractions mainly affected glycine, serine and threonine metabolism, alanine, aspartate and glutamate metabolism, D-arginine and D-ornithine metabolism, which were summarized as amino acid metabolism, while the polysaccharides mainly affected glycine, serine and threonine metabolism, alanine, aspartate and glutamate metabolism, tricarboxylic acid cycle, biotin metabolism and thiamine metabolism. ConclusionBoth of polysaccharides and n-butanol fractions of Atractylodis Rhizoma stir-fried with bran have significant regulating effects on the metabolic abnormalities in spleen-deficient rats, in which the n-butanol fractions is mainly involved in amino acid metabolism, and the polysaccharides are involved in energy metabolism and cofactor and vitamin metabolism in addition to regulating amino acid metabolism.

ABSTRACT

OBJECTIVE To optimize the processing technology of honey bran-fried Atractylodis Rhizoma, and to compare the anti-gastric ulcer effect before and after processing. METHODS Combing with entropy-weight and technique for order preference by similarity to ideal solution model, L(9 34) orthogonal experiment design was adopted to optimize the processing technology of honey bran-fried Atractylodis Rhizoma using the comprehensive score of the contents of atractylone, β-cineole, atractylenolide Ⅲ and atractylodine as evaluation index, using the ratio of excipients to medicine, frying temperature and frying time as factors. The validation tests were conducted. The gastric ulcer model of mice was induced by intragastrical administration of anhydrous ethanol; using Compound aluminum hydroxide tablet as positive control, anti-gastric ulcer effect of Atractylodis Rhizoma was compared with that of honey bran-fried Atractylodis Rhizoma using the contents of serum inflammatory factors [interleukin-2 (IL-2), IL-6, tumor necrosis factor-α (TNF-α)], ulcer index and inhibitory rate of gastric ulcer as evaluation indexes. RESULTS The optimal processing technology of honey bran-fried Atractylodis Rhizoma was as follows:ratio of adjuvant and medicinal materials of 3∶10 (g/g), frying temperature at 140 ℃ and frying time of 4 min. Results of 3 validation tests showed that the contents of 4 components (including atractylone), in honey bran-fried Atractylodis Rhizoma processed by the optimal technology kept stable (RSDs were 3.47%-5.80%, n=3); the comprehensive scores were 95.53%-95.89% (RSD=0.21%, n=3). Atractylodis Rhizoma and honey bran-fried Atractylodis Rhizoma could increase the serum content of IL-2 in mice, but reduce serum contents of IL-6 and TNF-α to varying degrees; honey bran-fried Atractylodis Rhizoma could significantly decrease its ulcer indexes (P<0.05 or P< 0.01); the improvement effect of honey bran-fried Atractylodis Rhizoma on the above indicators was generally better than that of the same dosage of Atractylodis Rhizoma (P<0.05 or P< 0.01). The inhibitory rates of low-dose, medium-dose and high-dose Atractylodis Rhizoma and honey bran-fried Atractylodis Rhizoma to gastric ulcer in mice were 9.18%, 19.30%, 30.70%, and 50.32%, 61.39%, 53.16%, respectively. CONCLUSIONS The optimal processing technology of honey bran-fried Atractylodis Rhizoma is stable and feasible, and the anti-gastric ulcer effect of Atractylodis Rhizoma has been enhanced after being fried with honey bran.

ABSTRACT

Atractylodis Rhizoma is a kind of commonly used clinical Chinese medicine (TCM), which was first recorded in Shennong Bencaojing (《神农本草经》). At that time, it was called "Zhu", which is the general name of Atractylodis Rhizoma and Atractylodis Macrocephalae Rhizoma. After Song dynasty, Atractylodis Rhizoma and Atractylodis Macrocephalae Rhizoma were separated. Atractylodis Rhizoma can be divided into Atractylodes lancea and A. chinensis. In history, A. lancea as authentic, that its quality is better than A. chinensis. However, the quality of Atractylodis Rhizoma was evaluated by the index component atractylodin in the 2020 edition of Chinese Pharmacopoeia. The general results showed that the content of atractylodin in A. lancea was low, even failed to meet the specified standard, and its content in A. chinensis was significantly higher than that in A. lancea. The results were inconsistent with the records of ancient books and documents, and the quality theory of "genuine medicine is the best". It could not reflect the quality advantage of genuine Atractylodis Rhizoma, and may even affect the clinical application and development momentum of genuine medicine. In short, the quality standard of TCM should not only conform to the historical experience, but also have the connotation of modern science and technology, which can stand the test of practice. Based on this, the author intends to sort out relevant laws and regulations, sort out the literature related to the authenticity, composition and efficacy of Atractylodis Rhizoma, and analyze the rationality of the current standard of Atractylodis Rhizoma by integrating the relevant records of historical classics and modern research results, so as to provide a basis for the improvement of the quality standard of Atractylodis Rhizoma.

ABSTRACT

ObjectiveBy comparing the composition and content changes of the volatile components in Atractylodis Rhizoma before and after processing with rice-washed water, the effect of rice-washed water processing on volatile components in Atractylodis Rhizoma was investigated. MethodHeadspace-gas chromatography-mass spectrometry (HS-GC-MS) was used to detect the volatile components in rhizomes of Atractylodes chinensis and A. lancea, and their processed products of rice-washed water. Chromatographic conditions were programmed temperature (starting temperature of 50 ℃ for 2 min, rising to 120 ℃ with the speed of 10 ℃·min-1, then rising to 170 ℃ at 2.5 ℃·min-1, and rising to 240 ℃ at 10 ℃·min-1 for 3 min), the inlet temperature was 280 ℃, the split ratio was 10∶1, and the solvent delay time was 3 min. The conditions of mass spectrometry were electron bombardment ionization (EI) with ionization temperature at 230 ℃ and detection range of m/z 20-650. Then the relative content of each component was determined by the peak area normalization method. SIMCA 14.1 software was used to perform unsupervised principal component analysis (PCA) and supervised orthogonal partial least squares-discriminant analysis (OPLS-DA) on each sample data, the differential components of Atractylodis Rhizoma and its processed products were screened by the principle of variable importance in the projection (VIP) value>1. ResultA total of 60 components were identified, among which 40 were rhizomes of A. chinensis and 38 were its processed products, 46 were rhizomes of A. lancea and 47 were its processed products. PCA and OPLS-DA showed that the 4 kinds of Atractylodis Rhizoma samples were clustered into one category respectively, indicating that the volatile components of the two kinds of Atractylodis Rhizoma were significantly changed after processing with rice-washed water, and there were also significant differences in the volatile components of rhizomes of A. lancea and A. chinensis. The compound composition of Atractylodis Rhizoma and its processed products was basically the same, but the content of the compounds was significantly different. The differential components were mainly concentrated in monoterpenoids and sesquiterpenoids, and the content of monoterpenoids mostly showed a decreasing trend. ConclusionAfter processing with rice-washed water, the contents of volatile components in rhizomes of A. lancea and A. chinensis are significantly changed, and pinene, 3-carene, p-cymene, ocimene, terpinolene, atractylon, acetic acid and furfural can be used as difference markers before and after processing.

ObjectiveTo explore the correlation between dryness and energy metabolism of Atractylodis Rhizoma, and to analyze the difference of medicinal properties between Atractylodes lancea and A. chinensis. MethodA total of 110 healthy SD rats were randomly divided into 11 groups, including normal group, volatile oil of A. lancea 1-5 group (S1-S5 group, doses of 447, 473, 442, 489, 496 mg·kg-1) and volatile oil of A. chinensis 1-5 group (N1-N5 group, doses of 197, 118, 281, 222, 185 mg·kg-1), the administration volume was 0.01 mL·g-1 with intragastric administration for 21 days. Dryness effect of A. lancea and A. chinensis on rats was evaluated by comparing the body weight, drinking water volume, urine volume, whole blood viscosity and pathological sections of submandibular gland stained with hematoxylin-eosin (HE). The expression of aquaporin 2 (AQP2) in rat kidney was measured by immunohistochemistry, the mRNA expressions of cytochrome C oxidase subunit 7A2 (COX7A2) and succinate dehydrogenase (SDH) complex subunit D (SDHD) in liver tissue were detected by real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). The contents of SDH, lactate dehydrogenase (LDH) and sodium ion-potassium ion-adenosine triphosphatase (Na+-K+-ATPase) in rat plasma were determined by colorimetry. The quality of A. lancea and A. chinensis was evaluated by coefficient of variation method, and Pearson correlation coefficient was used to analyze the correlation between dryness and energy metabolism. ResultCompared with the normal group, the amounts of drinking water and urine in volatile oil of A. lancea group and volatile oil of A. chinensis group increased, and the submandibular gland acini atrophied, the whole blood viscosity of rats in the volatile oil of A. lancea group increased significantly (P<0.01), the expression levels of COX7A2 and SDHD mRNA, the activities of SDH, LDH and Na+-K+-ATPase increased significantly (P<0.01), and the expression of AQP2 in kidney decreased significantly (P<0.01). Compared with the normal group, the expression level of COX7A2 mRNA, SDH activity and whole blood viscosity in the volatile oil of A. chinensis group increased (P<0.05, P<0.01), the AQP2 and SDH mRNA expression levels, LDH and Na+-K+-ATPase activities had no significant difference. The comprehensive score analysis of each index showed that the effect of volatile oil of A. lancea on dryness and energy metabolism was stronger than that of volatile oil of A. chinensis, and there was a positive correlation between dryness index and energy metabolism index. ConclusionThe two indexes show that medicinal properties of A. lancea is stronger than that of A. chinensis, and energy metabolism is closely related to the dryness of Atractylodis Rhizoma. It is suggested that it is reasonable to evaluate the dryness effect of Atractylodis Rhizoma from the perspective of energy metabolism, which can further enrich the evaluation indexes of medicinal properties.

ABSTRACT

Objective:To observe the difference in drying dampness between Scutellariae Radix and Atractylodis Rhizoma in model rats with spleen-stomach dampness-heat syndrome and clarify their property-efficacy relationship. Method:Sixty-four healthy male SD rats were randomized into the blank group, model group, high-, medium-, and low-dose Scutellariae Radix groups, as well as high-, medium-, and low-dose Atractylodis Rhizoma groups. The rats were exposed to high-fat and high-sugar diet and external dampness-heat environment for 20 days for inducing the spleen-stomach dampness-heat syndrome. The macroscopic manifestations of rats were observed and the morphological changes in stomach and colon were detected under a light microscope after hematoxylin-eosin (HE) staining, followed by the calculation of pathological scores. The serum tumor necrosis factor-<italic>α</italic> (TNF-<italic>α</italic>), interleukin-4 (IL-4), and interferon-<italic>γ</italic> (IFN-<italic>γ</italic>) levels were determined by enzyme-linked immunosorbent assay (ELISA). The protein and mRNA expression levels of aquaporin-4 (AQP4) in the gastric tissue were measured by Western blot and Real-time polymerase chain reaction (Real-time PCR), respectively. Result:Rats in the model group presented with the manifestations of dampness-heat syndrome. The inflammatory reaction in stomach and colon was obvious, and the pathological score was significantly increased (<italic>P</italic><0.01). The serum IFN-<italic>γ</italic>, IL-4, and TNF-<italic>α</italic> levels were elevated (<italic>P</italic><0.05), and so were the AQP4 protein and mRNA expression levels in the gastric tissue except that there was no statistical difference. The clinical symptoms of rats in the medication groups were alleviated. Scutellariae Radix significantly relieved the gastric and colonic inflammation in model rats. Atractylodis Rhizoma inhibited the colonic inflammation in model rats to a certain extent, but it had no obvious effect on gastric inflammation. The pathological score of each Scutellariae Radix group was decreased. In terms of the pathological score of gastric tissue, only the high-dose Scutellariae Radix produced a significant difference (<italic>P</italic><0.01), and the pathological scores of the three Atractylodis Rhizoma groups were not significantly different from that in the model group. As for the pathological score of colonic tissue, all the medication groups except for the low-dose Atractylodis Rhizoma group exhibited a significant difference in comparison with that of the model group (<italic>P</italic><0.01). Scutellariae Radix and Atractylodis Rhizoma at each dose reduced not only the serum IFN-<italic>γ</italic>, IL-4, and TNF-<italic>α</italic> levels (<italic>P</italic><0.05, <italic>P</italic><0.01), but also the AQP4 protein expression in gastric tissue of model rats (<italic>P</italic><0.01). The AQP4 mRNA expression in the gastric tissue of model rats declined in the high- and low-dose Scutellariae Radix groups, while that in the medium-dose Scutellariae Radix group and each Atractylodis Rhizoma group rose without statistical difference. Conclusion:Scutellariae Radix and Atractylodis Rhizoma display a certain property-efficacy relationship in drying dampness of rats with spleen-stomach dampness-heat syndrome. ABSTRACT

Atractylodis Rhizoma(AR) is a traditional Chinese medicinal material for food and medicine, with the functions of eli-minating dampness, strengthening the spleen, expelling wind evil and dispersing cold. AR contains a variety of compounds, including sesquiterpenoids, alkynes, triterpenoids, aromatic glycosides, polysaccharides and so on. At present, the researches on AR mainly focus on volatile components, with relatively fewer researches on non-volatile components. Polysaccharide from Atractylodis Rhizoma(ARP) is an important material basis among non-volatile components for the efficacy. Due to its many biological activities such as immunomodulatory activity, anti-tumors, anti-virus and anti-oxidation, ARP has certain research value and potential. The diversity of the polysaccharide structure is the basis for biological functions, but it also increases the difficulty of carbohydrate research. The research on the extraction, separation, purification, structure and activity of ARP is in the preliminary exploration stage, still with many shortcomings. Herein, recent advancements in the extraction, purification, structural characteristics and biological activities of ARP were summarized in this article to provide scientific reference for the in-depth systematic research of ARP and the development of AR resources.

ABSTRACT

The fumigation of Chinese herbal medicines, such as Artemisiae Argyi Folium, Atractylodis Rhizoma, and Angelicae Dahuricae Radix, is one of the creations of ancient Chinese medical practitioners during the struggle against the disease for thousands of years. Researches have showed the application of fumigation of Chinese medicinal materials in clinical practice, and there are clear pharmacological studies to prove its related effective ingredients. Therefore, we review the historical evolution and modern application progress of Chinese materia medica fumigation. In the light of the infectious features of COVID-19 including air droplet transmission and even possible aerosol transmission, the corresponding Chinese herbal medicine fumigation should be recommended and used in hospitals, communities, and families, which is expected to play an important role in fighting disease infections.

ABSTRACT

Objective To study the spectrum-effect relationship between GC-MS fingerprint of the volatile oil of Atractylodis Rhizoma and the dryness effect of Atractylodis Rhizoma, and clarify the main dryness components of Atractylodis Rhizoma. Methods The volatile oil of different batches of Atractylodis Rhizoma (S1-S9) were analyzed by GC-MS. Using the daily drinking amount of water, the viscosity of whole blood, and the content of aquaporin 2 (AQP2) in the kidney of rats as the indexes of dryness effect of Atractylodes Rhizome, the spectrum-effect relationship was analyzed by the combination of grey relational grade and orthogonal partial least square method. Results The dryness effect of Atractylodis Rhizoma was the combined action of multiple components, and the peaks of the major contribution to dryness effect were 31, 28, and 33, respectively. The corresponding components of the three peaks were β-eudesmol, (-)-aristolene, and bulnesol, which were identified by the spectral library retrieval analysis. Conclusion The main dryness effect components of Atractylodis Rhizoma are β-eudesmol, (-)-aristolene, and bulnesol, which provide a reference for investigating the material basis of dryness effect of Atractylodis Rhizoma.

ABSTRACT

The research and development of Chinese materia medica classical prescription is an important way to promote the inheritance and innovation of traditional Chinese medicine and improve the level of clinical services. The determination of the classic Chinese herbal medicine origin is particularly important in its research and development work, and it is the source of ensuring its quality, efficacy and safety. “Zhu” has a long history of medicine, and it is listed as the top grade in Shennong Herbal Classic, which is a commonly used Chinese materia medica in the classic prescription. However, before the Northern and Southern Dynasty, “Atractylodis Macrocephalae Rhizoma” and “Atractylodis Rhizoma” could be used as “zhu” medicines, and no clear distinction was made. As a result, the research on herbal medicine is a difficult problem in the development of the classics containing “Atractylodis Macrocephalae Rhizoma” and “Atractylodis Rhizoma”. This study combines the past medicinal herbs, combing the historical changes of “zhu” from the aspects of plant morphology, origin, taste, and efficacy, in order to provide reference for the research and development of the “zhu” and the research and development of the classics.

ABSTRACT

In order to establish a more perfect evaluation system for dryness effect of Atractylodis Rhizoma, determine the main dry parts of Atractylodis Rhizoma,and further define the mechanism of stir-baked Atractylodis Rhizoma in reducing the dryness. The healthy rats were given with different doses of water extract and volatile oil of raw Atractylodis Rhizoma and stir-baked Atractylodis Rhizoma for 21 days. Based on the theory of the dry-dry and dryness-induced Yin deficiency, the amount of drinking water, tissue morphology of submandibular glands, urine volume and the expression of aquaporin 2 (AQP2) in the kidneys, as well as blood rheology, ratio of cAMP/cGMP in serum and the content of Na⁺-K⁺-ATP enzyme were selected as the evaluation indexes. The results indicated that the rats with high dose volatile oil from raw Atractylodis Rhizoma had a significant increase in the amount of drinking water, urine volume, blood viscosity, ratio of cAMP/cGMP and content of Na⁺-K⁺-ATP enzyme in the serum(<0.05)as compared with the soybean oil group; meanwhile, atrophy of submandibular acinar gland was obvious,and the expression of aquaporin 2 was reduced significantly(<0.05). There were significant differences between volatile oil high dose group of raw Atractylodis Rhizoma and volatile oil high dose group of stir-baked Atractylodis Rhizoma. There was no significant difference between the water extract groups of raw and stir-baked Atractylodis Rhizoma and the saline group. A comprehensive evaluation system for the dryness effect of Atractylodis Rhizoma was established. It was confirmed that the volatile oil part was the main dry part of Atractylodis Rhizoma. It revealed that the mechanism of dryness effect of Atractylodis Rhizoma was not only related to the decrease of the total content of the volatile oil, but also may be related to the transformation of dryness components in the volatile oil. It provides references for the study of material basis of Atractylodis Rhizoma dryness, provides an experimental basis for the clinical application of Atractylodis Rhizoma, further clarifies the mechanism of stir-baked Atractylodis Rhizoma in reducing the dryness, and provides thoughts for the evaluation of other dry traditional Chinese medicines.

The pharmacologic action of Atractylodis Rhizoma and its effective constituents in digestive system includes mainly anti-ulcer activity,acceleration of gastric emptying,regulation of gastrointestinal propellant movement,antidiarrheat effect,choleretic effect,hepatoprotection,and promotion of digestive and absorptive function.Atractylodis Rhizoma decreases gastric acid secretion via blockade of H2-receptor,and inhibits overexpression of inflammatory cytokines in gastric tissues to produce anti-ulcer effect.Atractylodis Rhizoma increases gastric mucosal blood flow,and improves growth and reparation of gastric mucosa to produce anti-ulcer effect by blockade of 5-TH receptor,and elevating levels of gastrin and trefoil factor in serum and gastric tissues.Atractylodis Rhizoma accelerating gastric emptying and gastrointestinal propellant movement is relative to decreasing release of central corticotropin-releasing factor,and stimulating vagal nerve,and promoting secretion of gastrin and motilin,and inhibiting secretion of vasoactive intestinal peptide,and blocking 5-HT-3receptor,and increasing amount of interstitial cell of Cajal.

ABSTRACT

Objective: To study the content determination of atractylodin, atractylon, and β-eudesmol in Atractylodis Rhizoma by GC and characteristic spectrum in order to provide a scientific basis for the quality control. Methods: Using GC and Agilent HP-5 capillary column, taking nitrogen as carrier gas, FID as detector, temperature programming, split ratio, injection port temperature: 250℃, detector temperature: 250℃, column temperature: 130℃; The contents of atractylodin, atractylon and β-eudesmol of 25 samples between Chengde and purchased from other markets were determined by external standard method. The characteristic spectrum was set up and the similarity was analyzed by Estimating System of Similarity on the Chinese Medicine Fingerprint Chromatogram. Results: The determination method and characteristic spectrum by GC for atractylodin, atractylon, and β-eudesmol in Atractylodis Rhizoma were established. Nine characteristic peaks were identified; The linear range of β-eudesmol was 20.00 - 406.10 μg/mL (r = 0.999 9), and the average recovery was 100.75%, RSD = 1.17% (n = 6), and the limit of detection was 0.12 ng. The linear range of atractylon was 35.00 - 348.70 μg/mL (r = 0.999 5), and the average recovery was 99.84%, RSD = 1.29% (n = 6), and the limit of detection was 0.04 ng; The linear range of atractylodin was 16.46 - 329.30 μg/mL (r = 0.999 6), and the average recovery was 100.12%, RSD = 0.88% (n = 6), and the limit of detection was 0.06 ng. Conclusion: The concent determination and characteristic spectrum method of atractylodin, atractylon, and β-eudesmol established by this study are sensitive, simple, stability, which could make the determination result accurate and reliable.

ABSTRACT

Objective: To establish a method for the content identification of chemical constituents in Qiling Wenshen Xiaonang Formula by using UPLC-ESI-IT-TOF/MS, which can infer the potential active ingredients in the formula, combining with literature. Methods: The chromatography separation was performed on a Waters ACQUITY UPLC BEH C18 column (100 mm × 2.1 mm, 1.7 μm) with acetonitrile-0.1% formic acid in water by gradient elution. Electrospray ionization (ESI) source was applied and operated in both positive and negative ion mode. Results: A total of 36 chemical constituents were identified by ESI/MS through comparison on chemical standards and literature data, including 19 flavonoids, 11 phenolic acids, four triterpenoid saponins, one sesquiterpene glycoside, and one alkaloid. Conclusion: This study introduces a comprehensive analysis method for chemical ingredients in Qiling Wenshen Xiaonang Formula, and lays a foundation for the further analysis on the substance basis research. The results provide a significant guidance for the substance basis research and quality control of compound Chinese materia medica.

ABSTRACT

This research is to estimate the toxicity of Atractylodis Rhizoma Alba (ARA) in F344 rats and to find a dose level for the 13 weeks toxicity study. A hot water extract of ARA (ARWE) was administered orally to F344 rats at dose levels of 0 (vehicle control), 500, 1000, 2000, 3500, and 5000 mg/kg/day for 2 weeks. Each group was composed to five male and five female F344 rats. According to the result, there were no ARWE-related adverse changes in mortality, body weights, food consumption, urinalysis, hematology, clinical chemistry, gross finding at necropsy, and organ weight examination. Salivation was observed in 3500 and 5000 mg/kg/day in male and female rats but it could not have found any relationship with ARWE administration. Based on our findings, ARWE may not cause toxicity in rats under the experimental conditions. Therefore, dose level of 5000 mg/kg/day as a highest treatment group in 13-week exposure study is recommended for further toxicity assessment.

Objective: To optimize the processing technology for deep-fried Atractylodis Rhizoma. Methods: Taking the contents of tannin and diarrheal index of mice as the indexes, the affecting factors (stir-frying temperature, stir-frying time, and turning frequency) on the processing technology of deep-fried Atractylodis Rhizoma was investigated by L9(34) orthogonal test and multi-index comprehensive weighted mark method. Results: The best deep-fried technology was as following: the processing temperature was 220-230 ℃, turning frequency was 50 times/min, and stir-frying time was 6 min. Conclusion: The processing technology is stable and feasible, and could lay the foundation for the research on the quality of deep-fried Atractylodis Rhizoma.

ABSTRACT

Rikkunshito is comprised of 8 crude drugs and is used for the treatment of gastrointestinal dysfunctions such as anorexia and heavy stomach feeling. These symptoms are often caused by delay in gastric emptying. Cisplatin is a representative cancer chemotherapeutic drug with severe adverse effects such as anorexia and nausea, that gives rise to a delay in gastric emptying. However, it is still unknown whether rikkunshito has effects on improving the delayed gastric emptying induced by cisplatin. In the present study, we examined the effects of rikkunshito (an Atractylodis rhizoma-containing formula) on cisplatin-induced delay in gastric emptying in the rat. Rikkunshito improved this. Among the crude drugs that comprise rikkunshito, Atractylodis rhizoma, Ginseng radix, Poria and Aurantii nobilis pericarpium individually improved the delay in gastric emptying, suggesting that they all contribute to the action of rikkunshito. Moreover, the effects of these 4 crude drugs in combinations were also examined, and as a result, tended to be stronger when Atractylodis rhizoma was included. On the other hand, when Atractylodis rhizoma was excluded from rikkunshito, the effects were weaker. Meanwhile, atractylenolide III, a specific chemical constituent of Atractylodis rhizoma, improved delay in gastric emptying in a manner similar to that of rikkunshito with Atractylodis rhizoma. These results, taken together, suggest that Atractylodis rhizoma likely contributes greatly to the improving effect of rikkunshito on cisplatin-induced delay in gastric emptying.
