ABSTRACT

Roots of plants have immense reducing potential. Ions of noble metals namely Au3+ and Ag+ get reduced easily to form Au and Ag nanoparticles (NPs), respectively. Therefore, we hypothesize that plant roots could have potential to form Au-NPs and Ag-NPs. For present investigations, plants of Pennisetum glaucum L. were used to evaluate if their roots possess capacity to generate metal NPs. The generation of Au-NPs and Ag-NPs was initially presumed based on colour change, and confirmed by UV-vis spectra, TEM and EDX investigations. Pale yellow Au3+ and colourless Ag+ solutions turned purple and brown, respectively, by roots of Pennisetum sp. within 8 h. Absorption spectra of respective solutions showed plasmon resonance band at 560 nm and 420 nm confirming the presence of Au-NPs and Ag-NPs. TEM coupled with SAED revealed the presence of crystalline spherical NPs in the size range of 5-50 nm in these solutions. EDX further confirmed the presence of Au and Ag as NPs of respective solutions. These results confirmed that the roots of P. glaucum possess ideal reducing strength to generate Au-NPs and Ag-NPs exogenously in the aqueous phase.

ABSTRACT

Erythrocytes (red blood cells, RBCs) are the most abundant circulating cells in the blood and have been widely used in drug delivery systems (DDS) because of their features of biocompatibility, biodegradability, and long circulating half-life. Accordingly, a "camouflage" comprised of erythrocyte membranes renders nanoparticles as a platform that combines the advantages of native erythrocyte membranes with those of nanomaterials. Following injection into the blood of animal models, the coated nanoparticles imitate RBCs and interact with the surroundings to achieve long-term circulation. In this review, the biomimetic platform of erythrocyte membrane-coated nano-cores is described with regard to various aspects, with particular focus placed on the coating mechanism, preparation methods, verification methods, and the latest anti-tumor applications. Finally, further functional modifications of the erythrocyte membranes and attempts to fuse the surface properties of multiple cell membranes are discussed, providing a foundation to stimulate extensive research into multifunctional nano-biomimetic systems.

ABSTRACT

Objective: To synthesize bio-inspired gold nanoparticles (AuNPs) using the leaf extract of Justicia adhatoda and evaluate the anti-cancer activity on human lung cancer cell line (A549). Methods: Synthesis of AuNPs was done using an aqueous leaf extract of Justicia adhatoda as a green route. The bio-synthesized AuNPs were confirmed and characterized by using various spectral studies such as UV-Vis spectrum, Scanning Electron Microscope with EDAX, Transmission Electron Microscope, Fourier Transmission Infrared Spectroscope analysis and Surface Enhanced Raman Spectroscopy. The cell viability was determined by MTT reduction assay. In addition, cytomorphology and the nuclear morphological study of A549 cell line was observed under fluorescence microscope. Results: UV-Vis spectrum showed surface plasmon resonance peak at 547 nm, scanning electron microscope and transmission electron microscope studies showed the monodispersed spherical shape and its average size in the range of 40.1 nm was noticed. Fourier Transmission Infrared Spectroscope analysis confirmed that the C=O group of amino acids of proteins had strong ability to bind with the surface of nanoparticle. ABSTRACT

@#A sandwiched electrochemical immunoassay based on the AuNPs@GSH-CdTe as a signal label, which formed by GSH-CdTe QDs and AuNPs, with dual signal amplified by reduced graphene oxide and AuNPs was proposed for the sensitive detection of prostate specific antigen(PSA). Through a sandwich immunoreaction, the target PSA and AuNPs@GSH-CdTe labeled Ab1 were captured to rGO/AuNPs-Ab2 surface. After the HNO3-dissolution step, square wave stripping voltammetry(SWSV)analysis of the captured CdTe QDs was used to quantify the concentration of PSA. In this system, AuNPs possessedlarge specific surface and good biocompatibility, which could effectively expand the amount of antigen and GSH-CdTe QDs loading and signals amplifying, while rGO played a synergistic amplification role due to its large specific surface. The proposed method showed good linearity ranging from 0. 5 to 200 ng/mL with the detection limits of 5. 0 pg/mL. It also showed excellent selectivity, good reproducibility, satisfactory stability. In addition, the method was successfully applied to the determination of real samples. The result was satisfactory and the recovery could fall in 98. 20%- 106. 2%, which represented a novel approach for versatile detection of tumor markers.

ABSTRACT

In this work a new method is presented for simultaneous colorimetric determination of morphine(MOR)and ibuprofen(IBU)based on the aggregation of citrate-capped gold nanoparticles(AuNPs).Citrate-capped AuNPs were aggregated in the presence of MOR and IBU. The difference in kinetics of AuNPs aggregation in the presence of MOR/IBU was used for simultaneous analysis of MOR and IBU. The formation and size of synthesized AuNPs and the aggregated forms were monitored by infra-red(IR)spectroscopy and transmission electron microscopy(TEM),respectively.By adding MOR or IBU the absorbance was decreased at 520 nm and increased at 620 nm.The difference in kinetic profiles of aggregation was applied for simultaneous analysis of MOR and IBU using partial least square(PLS)regression as an efficient multivariate calibration method.The number of PLS latent variables was optimized by leave-one-out cross-validation method using predicted residual error sum of square. The proposed model exhibited a high capability in simultaneous prediction of MOR and IBU concentrations in real samples. The results showed linear ranges of 1.33–33.29μg/mL (R2=0.9904) and 0.28–6.9μg/mL (R2=0.9902) for MOR and IBU respectively with low detection limits of 0.15 and 0.03μg/mL(S/N=5).

ABSTRACT

Gold nanoparticles (AuNPs) have several biomedical applications in diagnosis and treating of disease such as targeted chemotherapy and in pharmaceutical drug delivery due to their multifunctionality and unique characteristics. AuNPs can be conjugated with ligands, imaging labels, therapeutic drugs and other functional moieties for site specific drug delivery application. In this present review we are discussing the synthesis, properties, and forthcoming applications of gold nanoparticle (AuNPs) which is the most studied among all other metallic-nanoparticles. Here our main focus is to explain the AuNPs application in cancer treatment. AuNPs provides non-toxic carrier system for pharmaceutical drug and gene delivery applications. Currently various anticancer drugs are available but these are cause the necrosis of cancerous cell as well as normal cells. AuNPs cause the necrosis of only cancer cells therefore we can utilize it as a delivery vehicle as well as anticancer agent.

ABSTRACT

Mycobacterium tuberculosis, the causing agent of tuberculosis, comes second only after HIV on the list of infectious agents slaughtering many worldwide. Due to the limitations behind the conventional detection methods, it is therefore critical to develop new sensitive sensing systems capable of quick detection of the infectious agent. In the present study, the surface modified cadmium-telluride quantum dots and gold nanoparticles conjunct with two specific oligonucleotides against early secretory antigenic target 6 were used to develop a sandwich-form fluorescence resonance energy transfer-based biosensor to detect M. tuberculosis complex and differentiate M. tuberculosis and M. bovis Bacille Calmette–Guerin simultaneously. The sensitivity and specificity of the newly developed biosensor were 94.2% and 86.6%, respectively, while the sensitivity and specificity of polymerase chain reaction and nested polymerase chain reaction were considerably lower, 74.2%, 73.3% and 82.8%, 80%, respectively. The detection limits of the sandwich-form fluorescence resonance energy transfer-based biosensor were far lower (10 fg) than those of the polymerase chain reaction and nested polymerase chain reaction (100 fg). Although the cost of the developed nanobiosensor was slightly higher than those of the polymerase chain reaction-based techniques, its unique advantages in terms of turnaround time, higher sensitivity and specificity, as well as a 10-fold lower detection limit would clearly recommend this test as a more appropriate and cost-effective tool for large scale operations.
